Concentrations of eleven representative metals (Al, Ca, Cd, Cr, Cu, K, Mg, Mn, Na, Pb and Zn) in forty-two propolis samples were measured by electrothermal atomization and flame atomic absorption spectrometry after calcination in a muffle furnace. Samples were collected from different regions from Paraná State - Brazil where apiculture is an important economic activity. Results showed that the average content of Al, Ca, K, Mg, Mn, Na and Zn in propolis was 0.68, 1.66, 7.59, 1.27, 0.08, 0.58 and 0.02 mg g(-1), respectively. Levels of Al, Ca, and Mg were statistically different in some regions of Paraná and could be used to assign the geographical origin of the propolis. The average concentration of the Cd, Cr, and Pb in raw propolis was 0.13, 5.53 and 9.85 µg g(-1), respectively, and allowed for identification of specific areas with environmental contamination.

The increased incidence of thyroid diseases raises a series of questions about what the main predisposing factors are nowadays. If dietary restriction of iodine was once a major global health concern, today, the processes of industrialization of food and high exposure to a wide variety of environmental chemicals may be affecting, directly or indirectly, thyroid function. The homeostasis of hypothalamus-pituitary-thyroid (HPT) axis is finely regulated through the negative feedback mechanism exerted by thyroid hormones. Allostatic mechanisms are triggered to adjust the physiology of HPT axis in chronic conditions. Glyphosate and glyphosate-based herbicides are pesticides with controversial endocrine disrupting activities and only few studies have approached their effects on HPT axis and thyroid function. However, glyphosate has an electrophilic and nucleophilic zwitterion chemical structure that may affect the mechanisms involved in iodide oxidation and organification, as well as the oxidative phosphorylation in the ATP synthesis. Thus, in this review, we aimed to: (1) discuss the critical points in the regulation of HPT axis and thyroid hormones levels balance, which may be susceptible to the toxic action of glyphosate and glyphosate-based herbicides, correlating the molecular mechanisms involved in glyphosate toxicity described in the literature that may, directly or indirectly, be associated to the higher incidence of thyroid diseases; and (2) present the literature regarding glyphosate toxicity in HPT axis.

The volatile composition of honeys produced by eight species of stingless bees collected in three municipalities in the state of Paraná (Brazil) was compared by combining static headspace GC-MS and chemometrics methods. Forty-four compounds were identified using NIST library and linear retention index relative to n-alkanes (C8-C40). Linalool derivatives were the most abundant peaks in most honeys regardless geographical or entomological origin. However, Principal Component Analysis discriminated honeys from different geographical origins considering their distinctive minor volatile components. Honey samples from Guaraqueçaba were characterized by the presence of hotrienol while those from Cambará showed epoxylinalol, benzaldehyde and TDN as minor discriminating compounds. Punctual species such as Borá showed similar fingerprints regardless geographical origin, with ethyl octanoate and ethyl decanoate as characteristic intense chromatographic peaks, which may suggest a specialized behavior for nectar collection. Discriminant Analysis allowed correct geographic discrimination of most honeys produced in the three spots tested. We concluded that volatile profile of stingless bee honeys can be used to attest authenticity related to regional origin of honeys.

Corn syrups, important ingredients used in food and beverage industries, often contain high levels of 5-hydroxymethyl-2-furfural (HMF), a toxic contaminant. In this work, an in house validation of a difference spectrophotometric method for HMF analysis in corn syrups was developed using sophisticated statistical tools by the first time. The methodology showed excellent analytical performance with good selectivity, linearity (R2=99.9%, r>0.99), accuracy and low limits (LOD=0.10mgL-1 and LOQ=0.34mgL-1). An excellent precision was confirmed by repeatability (RSD (%)=0.30) and intermediate precision (RSD (%)=0.36) estimates and by Horrat value (0.07). A detailed study of method precision using a nested design demonstrated that variation sources such as instruments, operators and time did not interfere in the variability of results within laboratory and consequently in its intermediate precision. The developed method is environmentally friendly, fast, cheap and easy to implement resulting in an attractive alternative for corn syrups quality control in industries and official laboratories.

Propolis is a resin produced by bees from raw material collected from plants, salivary secretions, and beeswax. New therapeutic properties for the Central Nervous System have emerged. We explored the neurobehavioral and antioxidant effects of an ethanolic extract of yellow propolis (EEYP) rich in triterpenoids, primarily lupeol and ß-amyrin. Male Wistar rats, 3 months old, were intraperitoneally treated with Tween 5% (control), EEYP (1, 3, 10, and 30 mg/kg), or diazepam, fluoxetine, and caffeine (positive controls) 30 min before the assays. Animals were submitted to open field, elevated plus maze, forced swimming, and inhibitory avoidance tests. After behavioral tasks, blood samples were collected through intracardiac pathway, to evaluate the oxidative balance. The results obtained in the open field and in the elevated plus maze assay showed spontaneous locomotion preserved and anxiolytic-like activity. In the forced swimming test, EEYP demonstrated antidepressant-like activity. In the inhibitory avoidance test, EEYP showed mnemonic activity at 30 mg/kg. In the evaluation of oxidative biochemistry, the extract reduced the production of nitric oxide and malondialdehyde without changing level of total antioxidant, catalase, and superoxide dismutase, induced by behavioral stress. Our results highlight that EEYP emerges as a promising anxiolytic, antidepressant, mnemonic, and antioxidant natural product.

The chemical composition and biological activity of a sample of yellow propolis from Mato Grosso do Sul, Brazil (EEP-Y MS), were investigated for the first time and compared with green, brown, and red types of Brazilian propolis and with a sample of yellow propolis from Cuba. Overall, EEP-Y MS had different qualitative chemical profiles, as well as different cytotoxic and antimicrobial activities when compared to the other types of propolis assessed in this study and it is a different chemotype of Brazilian propolis. Absence of phenolic compounds and the presence of mixtures of aliphatic compounds in yellow propolis were determined by analysing (1)H-NMR spectra and fifteen terpenes were identified by GC-MS. EEP-Y MS showed cytotoxic activity against human tumour strain OVCAR-8 but was not active against Gram-negative or Gram-positive bacteria. Our results confirm the difficulty of establishing a uniform quality standard for propolis from diverse geographical origins. The most appropriate pharmacological applications of yellow types of propolis must be further investigated.

The aim of this study was to determine the trace elements, Pb, Cd, and Cr in honey samples from eight different regions from the state of Paraná (Brazil), using slurry sampling graphite furnace atomic absorption spectrometry. Chemometric analysis (principal component analysis (PCA)) was applied to classify honey samples according to their levels of the trace elements Pb, Cd, and Cr, which is also related to the geographical origin of honey samples. The mean concentration for the elements followed the order Pb > Cr > > Cd. The mean values were 200 ± 76, 88 ± 14, and 4.1 ± 4 ng g(-1) for Pb, Cr, and Cd, respectively. It could be verified that honey samples are geographically separated, especially with regard to Pb and Cd contents. Thus, honey can be considered a bioindicator of environmental contamination, suggesting possible contamination in soil, water, and air. This contamination can be related to natural or anthropogenic sources present in the study regions.

Slurry sampling electrothermal atomic absorption spectrometry was used for direct determination of Cr, Pb and Cd in honey without sample pretreatment. The honey slurries were prepared in aqueous solution containing hydrogen peroxide and nitric acid. The slurries were directly introduced in the pyrolytic graphite tubes. Pd-Mg was used as a chemical modifier only for Cd determination. Analytical curves were performed with aqueous standards for Pb and Cr and with addition of fructose for Cd. The quantification limits for Cd, Pb and Cr were 2.0, 5.4 and 9.4ngg(-1), respectively. Acceptable precision of the methodology was obtained through repeatability and intermediate precision. In the accuracy study, recoveries were satisfactory (94-101%) for the three elements. The methodology was applied in honey from Paraná (Brazil). The concentrations of Pb, Cd and Cr ranged from 141 to 228ngg(-1), <2.0 to 8ngg(-1) and 83 to 94ngg(-1), respectively.

OBJECTIVE: As propolis is a highly valued bee product, we aimed to verify the quality of aged propolis, investigating their phenolic and flavonoid composition, levels of toxic metals, radical scavenging and antimicrobial activities. MATERIAL AND METHODS: Samples of fresh and aged propolis of six different beekeepers, from the same geographical location, were investigated in terms of their phenolic and flavonoid composition and levels of Pb, Cd, and Cr, as well as radical scavenging and antimicrobial activities. RESULTS: The two groups of propolis had similar qualitative composition by HPLC-PDA and ESI(-)-MS. Fresh propolis and aged propolis show no differences when average values of extraction yield, flavonoids, EC50, or MIC were compared and both types of propolis showed good antimicrobial activity at low concentrations. Only levels of phenolic compounds were higher in fresh propolis. CONCLUSION: The propolis samples considered in this study, aged or fresh, had similar qualitative composition, although they were collected in different periods. Samples only differed in their levels of total phenolic content. Moreover, aged propolis conserves significant radical scavenging and antimicrobial properties. We suggest that aged propolis should not be discarded but explored for alternative applications.
