BACKGROUND: Sharing individual phenotype and genotype data between countries is complex and fraught with potential errors, while sharing summary statistics of genome-wide association studies (GWAS) is relatively straightforward, and thus would be especially useful for traits that are expensive or difficult-to-measure, such as feed efficiency. Here we examined: (1) the sharing of individual cow data from international partners; and (2) the use of sequence variants selected from GWAS of international cow data to evaluate the accuracy of genomic estimated breeding values (GEBV) for residual feed intake (RFI) in Australian cows. RESULTS: GEBV for RFI were estimated using genomic best linear unbiased prediction (GBLUP) with 50k or high-density single nucleotide polymorphisms (SNPs), from a training population of 3797 individuals in univariate to trivariate analyses where the three traits were RFI phenotypes calculated using 584 Australian lactating cows (AUSc), 824 growing heifers (AUSh), and 2526 international lactating cows (OVE). Accuracies of GEBV in AUSc were evaluated by either cohort-by-birth-year or fourfold random cross-validations. GEBV of AUSc were also predicted using only the AUS training population with a weighted genomic relationship matrix constructed with SNPs from the 50k array and sequence variants selected from a meta-GWAS that included only international datasets. The genomic heritabilities estimated using the AUSc, OVE and AUSh datasets were moderate, ranging from 0.20 to 0.36. The genetic correlations (rg) of traits between heifers and cows ranged from 0.30 to 0.95 but were associated with large standard errors. The mean accuracies of GEBV in Australian cows were up to 0.32 and almost doubled when either overseas cows, or both overseas cows and AUS heifers were included in the training population. They also increased when selected sequence variants were combined with 50k SNPs, but with a smaller relative increase. CONCLUSIONS: The accuracy of RFI GEBV increased when international data were used or when selected sequence variants were combined with 50k SNP array data. This suggests that if direct sharing of data is not feasible, a meta-analysis of summary GWAS statistics could provide selected SNPs for custom panels to use in genomic selection programs. However, since this finding is based on a small cross-validation study, confirmation through a larger study is recommended.

Feed efficiency and energy balance are important traits underpinning profitability and environmental sustainability in animal production. They are complex traits, and our understanding of their underlying biology is currently limited. One measure of feed efficiency is residual feed intake (RFI), which is the difference between actual and predicted intake. Variation in RFI among individuals is attributable to the metabolic efficiency of energy utilization. High RFI (H_RFI) animals require more energy per unit of weight gain or milk produced compared with low RFI (L_RFI) animals. Energy balance (EB) is a closely related trait calculated very similarly to RFI. Cellular energy metabolism in mitochondria involves mitochondrial protein (MiP) encoded by both nuclear (NuMiP) and mitochondrial (MtMiP) genomes. We hypothesized that MiP genes are differentially expressed (DE) between H_RFI and L_RFI animal groups and similarly between negative and positive EB groups. Our study aimed to characterize MiP gene expression in white blood cells of H_RFI and L_RFI cows using RNA sequencing to identify genes and biological pathways associated with feed efficiency in dairy cattle. We used the top and bottom 14 cows ranked for RFI and EB out of 109 animals as H_RFI and L_RFI, and positive and negative EB groups, respectively. The gene expression counts across all nuclear and mitochondrial genes for animals in each group were used for differential gene expression analyses, weighted gene correlation network analysis, functional enrichment, and identification of hub genes. Out of 244 DE genes between RFI groups, 38 were MiP genes. The DE genes were enriched for the oxidative phosphorylation (OXPHOS) and ribosome pathways. The DE MiP genes were underexpressed in L_RFI (and negative EB) compared with the H_RFI (and positive EB) groups, suggestive of reduced mitochondrial activity in the L_RFI group. None of the MtMiP genes were among the DE MiP genes between the groups, which suggests a non-rate limiting role of MtMiP genes in feed efficiency and warrants further investigation. The role of MiP, particularly the NuMiP and OXPHOS pathways in RFI, was also supported by our gene correlation network analysis and the hub gene identification. We validated the findings in an independent data set. Overall, our study suggested that differences in feed efficiency in dairy cows may be linked to differences in cellular energy demand. This study broadens our knowledge of the biology of feed efficiency in dairy cattle.

The fortification of processed foods including dairy products is increasingly commonplace with phytosterols among many compounds used to improve the nutritional value of food products. It is also increasingly common practice for some dairy cattle feeds to be fortified for their potential to increase phytosterol levels in milk. In this study, a combined, streamlined protocol using acid hydrolysis, saponification and sample clean-up was developed to enable the rapid and reliable measurement of phytosterols. The method was developed with focus on streamlining the overall technique to make it suitable for commercial laboratories, to reduce labor and consumable costs, while maintaining accuracy. A total of 12 different feed types commonly used in the dairy industry were analyzed with the highest and lowest sterol contents found in cotton seed oil and tannin with average phytosterol contents of 256 and <30 mg per 100 g, respectively. With a limit of reporting of 30 mg/kg for individual sterols and a correlation coefficient > 0.99, the method was validated for milk to enable feed comparison studies with respect to the total phytosterol content in raw milk.

The influence of different pasture-based feeding systems on fatty acids, organic acids and volatile organic flavour compounds in yoghurt was studied. Pasture is the main source of nutrients for dairy cows in many parts of the world, including southeast Australia. Milk and milk products produced in these systems are known to contain a number of compounds with positive effects on human health. In the current study, 260 cows were fed supplementary grain and forage according to one of 3 different systems; Control (a traditional pasture based diet offered to the cows during milking and in paddock), PMR1 (a partial mixed ration which contained the same supplement as Control but was offered to the cows as a partial mixed ration on a feedpad), PMR 2 (a differently formulated partial mixed ration compared to Control and PMR1 which was offered to the cows on a feedpad). Most of the yoghurt fatty acids were influenced by feeding systems; however, those effects were minor on organic acids. The differences in feeding systems did not lead to the formation of different volatile organic flavour compounds in yoghurt. Yet, it did influence the relative abundance of these components.

This study investigated the effects of different strategies for feeding supplements to grazing dairy cows on the proportions of fatty acids in milk. Two hundred and sixteen cows were fed supplementary grain and forage according to one of 3 different strategies; (1) CONTROL: cows grazed perennial ryegrass pasture (14 kg dry matter/d) supplemented with milled barley grain fed in the milking parlour and pasture silage offered in the paddock; (2) Partial mixed ration 1 (PMR1): same pasture allotment and supplement as CONTROL strategy, but the supplements presented as a mixed ration after each milking in feedpad, and; (3) Partial mixed ration 2 (PMR2): same pasture allotment, supplemented with a mixed ration of milled barley grain, alfalfa hay, corn silage and crushed corn grain fed in a feedpad. Within each strategy, cows were assigned to receive either 6, 8, 10 or 12 kg dry matter supplement/cow per d. Milk fatty acid proportions from cows fed CONTROL and PMR1 strategies were similar and different from those fed PMR2, particularly at 10 to 12 kg dry matter supplement/cow per d. The reduction in milk fat yield and concentration in cows fed high amounts of supplement as CONTROL and PMR1 was coincident with 4 × increase in 10t-18:1 proportion. The composition of the partial mixed ration (PMR) and the amount offered affected milk fatty acid proportions and milk fat content, however, the method of supplementation did not.

This experiment was conducted to determine the effects of diets supplemented with cottonseed oil, Acacia mearnsii-condensed tannin extract, and a combination of both on composition of bovine milk. Treatment diets included addition of cottonseed oil (800 g/d; CSO), condensed tannin from Acacia mearnsii (400 g/d; TAN) or a combination of cottonseed oil (800 g/d) and condensed tannin (400 g/d; CPT) with a diet consisting of 6·0 kg dry matter (DM) of concentrates and alfalfa hay ad libitum, which also served as the control diet (CON). Relative to the CON diet, feeding CSO and CPT diets had a minor impact on feed intake and yield of lactose in milk. These diets increased yields of milk and protein in milk. In contrast to the TAN diet, the CSO and CPT diets significantly decreased milk fat concentration and altered milk fatty acid composition by decreasing the proportion of saturated fatty acids but increasing proportions of monounsaturated and polyunsaturated fatty acids. The CPT diet had a similar effect to the CSO diet in modifying fatty acid profile. Overall, reduction in milk fat concentration and changes in milk fatty acid profile were probably due to supplementation of linoleic acid-rich cottonseed oil. The TAN diet had no effect on feed intake, milk yield and milk protein concentration. However, a reduction in the yields of protein and lactose occurred when cows were fed this diet. Supplemented tannin had no significant effect on fat concentration and changes in fatty acid profile in milk. All supplemented diets did not affect protein concentration or composition, nitrogen concentration, or casein to total protein ratio of the resulting milk.

A detailed understanding of the relationships between the distinct metabolic compartments of blood and milk would be of potential benefit to our understanding of the physiology of lactation, and potentially for development of biomarkers for health and commercially relevant traits in dairy cattle. NMR methods were used to measure metabolic profiles from blood and milk samples from Holstein cows. Data were analyzed using PLS regression to identify quantitative relationships between metabolic profiles and important traits. Statistical Heterospectroscopy (SHY), a powerful approach to recovering latent biological information in NMR spectroscopic data sets from multiple complementary samples, was employed to explore the metabolic relationships between blood and milk from these animals. The study confirms milk is a distinct metabolic compartment with a metabolite composition largely not influenced by plasma composition under normal circumstances. However, several significant relationships were identified, including a high correlation for trimethylamine (TMA) and dimethylsulfone (DMSO(2)) across plasma and milk compartments, and evidence plasma valine levels are linked to differences in amino acid catabolism in the mammary gland. The findings provide insights into the physiological mechanisms underlying lactation and identification of links between key metabolites and milk traits such as the protein and fat content of milk. The approach has the potential to enable measurement of health, metabolic status and other important phenotypes with milk sampling.

This experiment investigated the preservation effects of two preweaning milk feeding nutritional treatments (High: 8 L and Low: 4 L milk per day) on 20, 12-month-old Holstein-Friesian dairy heifers (Bos taurus). A vaccination immune challenge was initially implemented on these 20 heifers at 6 weeks of age and the findings indicated superior growth, immune competence and favorable metabolic characteristics from the calves that had been fed 8 L milk per day. Postweaning, all heifers were treated the same under non-experimental conditions, and the immune challenge was repeated at 12 months of age for the current experiment. Consistent with the first immune challenge, heifers from the High preweaning treatment group still had higher white cell count and neutrophil count, indicating superior immune competence. The differences found in metabolic biomarkers, including beta-hydroxybutyrate, glucose and insulin, in the preweaning phase had disappeared, suggesting these biomarkers were influenced directly by the nutritional input at the time. There were no differences in NEFA levels between treatments at either stage of development. Postweaning, the heifers from the Low preweaning treatment group experienced accelerated growth with slightly numerically higher ADG (0.83 kg/day vs. 0.89 kg/day), resulting in the initial differences in bodyweight recorded at weaning being eliminated by 13 months of age. These results are evidence of a form of immunological developmental programming as a result of accelerated preweaning nutrition and therefore, are not supportive of restricted milk feeding of calves.

Feeding increased volumes of milk in the preweaning phase has been shown to improve growth, morbidity and mortality rates in calves (Bos Taurus). This experiment enlisted 20 Holstein-Friesian dairy replacement calves from birth until weaning (at 10 weeks of age) and assessed the effect of feeding either 4 L (Low) or 8 L (High) of milk per calf per day on their growth, immune competence and metabolic characteristics. The responsiveness of these systems was compared through a vaccination immune challenge. Calves in the High treatment group were significantly heavier from two weeks of age and were 19 kg heavier than calves in the Low treatment group at weaning. Calves in the High treatment group also exhibited greater immune responses, with significantly higher white cell counts and neutrophil counts than calves in the Low treatment group post-vaccination. Calves in the High treatment group also had lower beta-hydroxybutyrate both pre- and post-vaccination, and higher glucose and insulin levels post-vaccination, indicating superior metabolic characteristics. Calves had ad libitum access to lucerne hay (Medicago sativa) and a commercial concentrate. Solid feed intakes were mostly the same between treatments, with differences in hay intake only detected at 7 and 8 weeks of age. Results from this experiment are indicative of a positive influence of accelerated preweaning nutrition on growth, immune response and metabolic characteristics.

The frequency, duration, and intensity of heat waves in Australia are increasing. To reduce the impact of heat waves on milk production, novel management strategies are required. Altering the forage type and amount offered affect the heat load on dairy cows and offer potential strategies to ameliorate the effects of hot weather. Thirty-two multiparous, lactating Holstein-Friesian cows were assigned one of four dietary treatments: chicory high amount, chicory low amount, pasture silage high amount, or pasture silage low amount. These cows were exposed to a heat wave in controlled-environment chambers. Cows that were offered fresh chicory had similar feed intake to cows that were offered pasture silage (15.3 kg DM/d). However, cows that were offered chicory produced greater energy-corrected milk (21.9 vs. 17.2 kg/d) and had a lower maximum body temperature (39.4 vs. 39.6 °C) than cows that were offered pasture silage overall. Cows that were offered the high amount of forage had greater feed intake (16.5 vs. 14.1 kg DM/d) and energy corrected milk yield (20.0 vs. 17.9 kg/d) than cows that were offered the low amount, as intended, but with no difference in maximum body temperature (39.5 °C). We conclude that feeding chicory instead of pasture silage to dairy cows shows promise as a dietary strategy to ameliorate the effect of heat exposure, and there was no advantage in restricting feed amount.

Cereal grains that differ in the rate and extent of ruminal fermentation differ in heat increment and may be used to improve thermoregulation during heat stress. This experiment investigated the responses of dairy cows in late lactation to a heat challenge when offered wheat-grain or corn-grain. Eighteen lactating cows, 220 ± 94 (mean ± standard deviation) days in milk, 3.7 ± 0.17 years of age and 558 ± 37 kg bodyweight, were allocated treatments containing 6 kg dry matter (DM)/day of wheat grain or 6 kg DM/day corn grain (9 per treatment) plus 14 kg DM/day of alfalfa hay. Measurements were made during a 7-day pre-challenge period at ambient conditions in individual stalls, during a 4-day heat challenge (temperature humidity index of 74 to 84) in individual controlled-climate chambers, then during a 7-day recovery period at ambient conditions in individual stalls. During the heat challenge, cows offered corn had lower respiration rates (p = 0.017) and greater feed intake (p = 0.021) but energy-corrected milk (p = 0.097) was not different to that of cows offered wheat. Feeding corn grain to dairy cows during a heat challenge reduced some of the negative impacts of heat stress, enabling the cows to consume more forage compared with supplementing with wheat grain.

Human immunodeficiency virus type 1 (HIV-1) vaccination of cows has elicited broadly neutralizing antibodies (bNAbs). In this study, monoclonal antibodies (mAbs) are isolated from a clade A (KNH1144 and BG505) vaccinated cow using a heterologous clade B antigen (AD8). CD4 binding site (CD4bs) bNAb (MEL-1872) is more potent than a majority of CD4bs bNAbs isolated so far. MEL-1872 mAb with CDRH3 of 57 amino acids shows more potency (geometric mean half-maximal inhibitory concentration [IC50]: 0.009 µg/mL; breadth: 66%) than VRC01 against clade B viruses (29-fold) and than CHO1-31 against tested clade A viruses (21-fold). It also shows more breadth and potency than NC-Cow1, the only other reported anti-HIV-1 bovine bNAb, which has 60% breadth with geometric mean IC50 of 0.090 µg/mL in this study. Using successive different stable-structured SOSIP trimers in bovines can elicit bNAbs focusing on epitopes ubiquitous across subtypes. Furthermore, the cross-clade selection strategy also results in ultra-potent bNAbs.

Body condition scoring is a valuable tool used to assess the changes in subcutaneous tissue reserves of dairy cows throughout the lactation resulting from changes to management or nutritional interventions. A subjective visual method is typically used to assign a body condition score (BCS) to a cow following a standardized scale, but this method is subject to operator bias and is labor intensive, limiting the number of animals that can be scored and frequency of measurement. An automated three-dimensional body condition scoring camera system is commercially available (DeLaval Body Condition Scoring, BCS DeLaval International AB, Tumba, Sweden), but the reliability of the BCS data for research applications is still unknown, as the system's sensitivity to change in BCS over time within cows has yet to be investigated. The objective of this study was to evaluate the suitability of an automated body condition scoring system for dairy cows for research applications as an alternative to visual body condition scoring. Thirty-two multiparous Holstein-Friesian cows (9 ± 6.8 days in milk) were body condition scored visually by three trained staff weekly and automatically twice each day by the camera for at least 7 consecutive weeks. Measurements were performed in early lactation, when the greatest differences in BCS of a cow over the lactation are normally present, and changes in BCS occur rapidly compared with later stages, allowing for detectable changes in a short timeframe by each method. Two data sets were obtained from the automatic body condition scoring camera: (1) raw daily BCS camera values and (2) a refined data set obtained from the raw daily BCS camera data by fitting a robust smooth loess function to identify and remove outliers. Agreement, precision, and sensitivity properties of the three data sets (visual, raw, and refined camera BCS) were compared in terms of the weekly average for each cow. Sensitivity was estimated as the ratio of response to precision, providing an objective performance criterion for independent comparison of methods. The camera body condition scoring method, using raw or refined camera data, performed better on this criterion compared with the visual method. Sensitivities of the raw BCS camera method, the refined BCS camera method, and the visual BCS method for changes in weekly mean score were 3.6, 6.2, and 1.7, respectively. To detect a change in BCS of an animal, assuming a decline of about 0.2 BCS (1-8 scale) per month, as was observed on average in this experiment, it would take around 44 days with the visual method, 21 days with the raw camera method, or 12 days with the refined camera method. This represents an increased capacity of both camera methods to detect changes in BCS over time compared with the visual method, which improved further when raw camera data were refined as per our proposed method. We recommend the use of the proposed refinement of the camera's daily BCS data for research applications.

Feed is the largest variable cost for dairy farms in Australia, and dairy farmers are faced with the challenge of profitably feeding their cows in situations where there is significant variation in input costs and milk price. In theory, the addition of 5.2 MJ of metabolisable energy to a lactating cow's diet should be capable of supporting an increase in milk production of one litre of milk of 4.0% fat, 3.2% protein and 4.9% lactose. However, this is almost never seen in practice, due to competition for energy from other processes (e.g., body tissue gain), forage substitution, associative effects and imbalances in rumen fermentation. Pasture species, stage of maturity, pasture mass, allowance and intake, stage of lactation, cow body condition and type of supplement can all affect the milk protein plus fat production response to additional feed consumed by grazing dairy cows. We developed a model to predict marginal milk protein plus fat response/kg DM intake when lactating dairy cows consume concentrates and pasture + forages. Data from peer reviewed published experiments undertaken in Australia were collated into a database. Meta-analysis techniques were applied to the data and a two-variable quadratic polynomial production function was developed. Production economic theory was used to estimate the level of output for given quantities of input, the marginal physical productivity of each input, the isoquants for any specified level of output and the optimal input combination for given costs and prices of inputs and output. The application of the model and economic overlay was demonstrated using four scenarios based on a farm in Gippsland, Victoria. Given that feed accounts for the largest input cost in dairying, allocation of pasture and supplements that are based on better estimates of marginal milk responses to supplements should deliver increased profit from either savings in feed costs, or in some cases, increased output to approach the point where marginal revenue equals marginal costs. Such data are critical if the industry is to take advantage of the opportunities to use supplements to improve both productivity and profitability.

To increase the dry matter and metabolisable energy intake of cows, dairy farmers often supplement pasture with concentrates and conserved fodder. Feeding large amounts of highly fermentable concentrates to cows can result in metabolic issues, such as ruminal acidosis, and thus safer but more efficient introduction strategies are desirable. We assessed the role that forages play in ruminal, behavioural and production responses to a wheat grain challenge in dairy cows with no previous wheat adaptation. Multiparous lactating Holstein dairy cows (n = 16) were fed a forage-only diet of either lucerne (Medicago sativa) hay, perennial ryegrass (Lolium perenne L.) hay or one of two cultivars of zero-grazing fresh perennial ryegrass herbage (Bealey or Base), for 3 weeks. The forage diet was then supplemented with crushed wheat grain at 8 kg dry matter/cow day-1, with no adaptation period. Wheat comprised between 32 and 43% of total dry matter intake. Cows fed hay maintained a higher mean ruminal fluid pH than those fed herbage, on both the forage-only diet (6.43 vs. 6.17) and the forage plus wheat diet (6.03 vs. 5.58). Following supplementation of wheat, cows fed herbage exhibited minimum ruminal fluid pH levels indicative of acute ruminal acidosis, at 5.15 and 5.06 for cultivars Bealey and Base, respectively. Furthermore, for both herbage cultivars, adding wheat resulted in a ruminal fluid pH under 6 for >20 h/day. The ruminal environment of cows fed lucerne hay remained most stable throughout the grain challenge, spending the least amount of time below pH 6.0 (9.0 h/day). Hay created a ruminal environment that was better able to cope with the accumulation of acid as wheat was digested. A combination of increased ruminating time and a slower rate of fermentation, due to higher neutral detergent fiber and lower metabolisable energy concentrations in the hays, is likely responsible for the higher ruminal fluid pH values. Forage plays a critical role in wheat introduction strategies; aggressive adaptation strategies could be implemented when a hay such as lucerne is used as the base forage.

The economics of grazing dairy cows offered a range of herbage allowances and fed supplements as a partial mixed ration (PMR) were examined where profit was defined as the margin between total milk income and the cost of pasture plus PMR supplement. The analysis made use of milk production and feed intake data from two dairy cow nutrition experiments, one in early lactation and the other in late lactation. In early lactation and at a PMR intake of 6 kg DM/cow per day, the profit from the cows with access to a medium herbage allowance (25 kg DM/cow per day) was AUD 1.40/cow per day higher than that for cows on a low allowance (15 kg DM/cow per day). At a higher PMR intake of 14 kg DM/cow per day, the profit from the cows on a medium herbage allowance was AUD 0.45/cow per day higher than the cows on a low allowance; there was no additional profit from increasing the herbage allowance from medium to high (40 kg DM/cow per day). In late lactation, the profit from the cows fed a PMR with a medium herbage allowance (20 kg DM/cow per day) was only higher than the cows on a low allowance (12 kg DM/cow per day) when the PMR intake was between 6 and 12 kg DM/cow per day. There was also a difference of AUD +0.50/cow per day between the PMR with medium and high herbage allowance (32 kg DM/cow per day). It was concluded that farmers who feed a PMR to dairy cows should offer at least a medium herbage allowance to optimize profit. While feeding additional PMR increases milk production and profit, further gains would be available by offering a higher herbage allowance. These findings provide an estimate of the net benefits of different herbage allowances when feeding a PMR and will enable farmers to manage their feeding systems more profitably.

Supplementing the diet of lactating cows with ingredients that increase energy density, or reduce internal heat production, may reduce some of the negative impacts of hot weather on milk yield. Thirty-two dairy cows were assigned either: (1) basal diet only, (2) basal diet plus canola oil, (3) basal diet plus betaine, or (4) basal diet plus canola oil and betaine. The basal diet was lucerne hay, pasture silage, and grain. Cows were exposed to a four-day heat challenge (temperature-humidity index 74 to 84) in controlled-environment chambers. Canola oil supplementation increased milk production (22.0 vs. 18.7 kg/d) across all periods of our experiment and increased body temperature (39.6 vs. 39.0 °C) during the heat challenge. Betaine supplementation reduced maximum body temperature during the pre-challenge period (39.2 vs. 39.6 °C) but not during the heat challenge (40.3 °C). Cows fed canola oil had greater declines in dry matter intake (5.4 vs 2.7 kg DM) and energy corrected milk (1.3 vs. 1.0 kg) from the pre-challenge to the heat challenge than other cows. Contrary to our expectations, the combination of fat and betaine supplements did not result in a clear benefit in terms of milk production or body temperature. Further work is warranted to understand the interactions between diet and hot weather.

Despite the fact that the sulphur hexafluoride (SF6) tracer technique was developed over 25 years ago to measure methane production from grazing and non-housed animals, no studies have specifically investigated whether ambient wind speed, temperature, relative humidity and rainfall influence the accuracy of the method. The aim of this research was to investigate how these weather factors influence the measurement of enteric methane production by the SF6 technique. Six different cohorts of dairy cows (40 per cohort) were kept outdoors and fed a common diet during spring in 3 consecutive years. Methane production from individual cows was measured daily over the last 5 days of each 32-day period. An automated weather station measured air temperature, wind speed, relative humidity and rainfall every 10 min. Regression analyses were used to relate the average daily wind speed, average daily temperature, average daily relative humidity and total daily rainfall measurements to dry matter intake, average daily methane production and methane yield of each cohort of cows. It was concluded that the modified SF6 technique can be used outdoors during a range of wind speeds, ambient temperatures, relative humidities and rainfall conditions without causing a significant effect on the measurement of methane production or methane yield of dairy cows.

PURPOSE: Among healthcare professionals, burnout is one of the key challenges affecting organizational outcomes, employee productivity and quality of care. The knowledge of burnout and its root causes and primary contributors continues to grow yet remains limited. In many environments, an entrepreneurial orientation (EO) has been shown to dramatically improve organizational outcomes and performance. The purpose of this paper is to illustrate critical research areas at the intersection of organizational EO and employee burnout within the healthcare sector. DESIGN/METHODOLOGY/APPROACH: A conceptual model which considers how EO has the potential to provide an operational context that may negate, lessen or delay the negative effects of burnout among healthcare professionals, is advanced as a useful focal point to foster research exploring connections between organizational orientation and employee well-being. FINDINGS: Insights into how an opportunity-embracing EO characteristic of high-tech firms may shape how stress is experienced and address burnout when applied to healthcare organizations. A decrease in burnout stands to improve quality of care as well as the satisfaction of staff and patients alike, including a greater sense of autonomy, engagement, motivation and passion. ORIGINALITY/VALUE: This research agenda proposes new insights and the need for additional research into how the manifestation of organizational EO may contribute to the field of medicine, influence burnout and enhance the well-being among healthcare professionals.

Disorders of energy metabolism, which can result from a failure to adapt to the period of negative energy balance immediately after calving, have significant negative effects on the health, welfare and profitability of dairy cows. The most common biomarkers of energy balance in dairy cows are ß-hydroxybutyrate (BHBA) and non-esterified fatty acids (NEFA). While elevated concentrations of these biomarkers are associated with similar negative health and production outcomes, the phenotypic and genetic correlations between them are weak. In this study, we used an untargeted 1H NMR metabolomics approach to investigate the serum metabolomic fingerprints of BHBA and NEFA. Serum samples were collected from 298 cows in early lactation (calibration dataset N = 248, validation N = 50). Metabolomic fingerprinting was done by regressing 1H NMR spectra against BHBA and NEFA concentrations (determined using colorimetric assays) using orthogonal partial least squares regression. Prediction accuracies were high for BHBA models, and moderately high for NEFA models (R2 of external validation of 0.88 and 0.75, respectively). We identified 16 metabolites that were significantly (variable importance of projection score > 1) correlated with the concentration of one or both biomarkers. These metabolites were primarily intermediates of energy, phospholipid, and/or methyl donor metabolism. Of the significant metabolites identified; (1) two (acetate and creatine) were positively correlated with BHBA but negatively correlated with NEFA, (2) nine had similar associations with both BHBA and NEFA, (3) two were correlated with only BHBA concentration, and (4) three were only correlated with NEFA concentration. Overall, our results suggest that BHBA and NEFA are indicative of similar metabolic states in clinically healthy animals, but that several significant metabolic differences exist that help to explain the weak correlations between them. We also identified several metabolites that may be useful intermediate phenotypes in genomic selection for improved metabolic health.
