RESUMO

Increasing evidence indicate that the expression of defense genes at the right place and the right time are regulated by host-defense transcription factors. However, the precise mechanisms of this regulation are not well understood. Homeodomain transcription factors, encoded by homeobox genes, play crucial role for the development of multicellular eukaryotes. In this study, we demonstrated that homeodomain transcription factor CEH-37 (known as OTX2 in mammals) was a key transcription factor for host defense in Caenorhabditis elegans. Meanwhile, CEH-37 acted in the intestine to protect C. elegans against pathogen infection. We further showed that the homeodomain transcription factor CEH-37 positively regulated PMK-1/ p38 MAPK activity to promote the intestinal immunity via suppression phosphatase VHP-1. Furthermore, we demonstrated that this function was conserved, because the human homeodomain transcription factor OTX2 also exhibited protective function in lung epithelial cells during Pseudomonas aeruginosa infection. Thus, our work reveal that CEH-37/OTX2 is a evolutionarily conserved transcription factor for defense against pathogen infection. The finding provides a model in which CEH-37 decreases VHP-1 phosphatase activity, allowing increased stimulation of PMK-1/p38 MAPK phosphorylation cascade in the intestine for pathogen resistance.

A field survey and pot experiment were carried out to screen tolerant plants growing in cadmium (Cd)-polluted mining areas which were co-polluted with acid in soil, and the related physiological and biochemical mechanisms were also analyzed. Thirty-seven species of wild plants and their corresponding soil were collected from a farmland around the mining areas. Ageratum (Ageratum conyzoides L.) with strong Cd-accumulative ability was selected, and its tolerance experiment for acid and Cd with different levels were carried out separately or orthogonally, respectively. Furthermore, the activities of superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT), and the contents of malondialdehyde (MDA), photosynthetic pigments, soluble sugar and proline in its leaves were determined. The results showed that the Cd accumulation in ageratum and sticktight (Bidens pilosa L.) was relatively high, but the latter has been well documented, so we focused on ageratum in the present work. In pot experiment, ageratum grew normally at 100 mg kg-1 Cd in soil, and the Cd concentrations in its roots, stems and leaves were 75.37 ± 7.37, 31.01 ± 3.76 and 53.92 ± 10.05 mg kg-1, respectively. In the case of acid tolerance experiment, all plant individuals of ageratum grew normally when soil pH was over 3.5. In the orthogonal experiment, the Cd accumulation in this plant increased with the decrease of soil pH under the same Cd treatment. Under strong acid conditions, the activity of SOD in leaves of ageratum was increased significantly. When the Cd concentration was 10 mg kg-1 and the soil pH was 5.5 or 3.5, the activities of POD and CAT were significantly increased. In addition, based on stepwise regression analysis, the leaf Cd concentration was significantly positive correlated with the activities of SOD and POD in leaves of ageratum. Therefore, ageratum not only had a strong tolerance for Cd and acid pollution in soil, but also had a strong ability to accumulate Cd. As a common plant in the mining area, it has a great potential for the phytoremediation of Cd and acid co-contaminated soil.

Metastasis-associated protein 1 (MTA1) is a critical component of the nucleosome remodeling and histone deacetylase (NuRD) complex. MTA1 has several biological functions, and it is closely associated with the malignant properties of human cancers; however, the mechanisms and subcellular localization of MTA1 in cells remain unclear. Some initial studies indicated that MTA1 was absent from the nucleolus; however, several NuRD components were recently found to be present in the nucleolus, where they regulate preribosomal RNA (pre-rRNA) transcription. In this study, we demonstrated that MTA1 is definitely localized to the nucleolus and regulates pre-rRNA transcription, which is consistent with the recent reports on NuRD. To determine if MTA1 was present in the nucleolus, we utilized the following complementary molecular approaches: immunofluorescence, GFP-tag tracking, immunoelectron microscopy, and immunoprecipitation (IP). To examine the role of MTA1 in rRNA synthesis, we performed quantitative polymerase chain reaction analysis. We revealed that both endogenous and exogenous MTA1 showed apparent granule-like nucleolar subcellular localization. MTA1 interacts with two major resident nucleolar proteins, nucleolin and nucleophosmin. Immunofluorescent colocalization analyses showed that MTA1 localizes to the fibrillarin-deficient regions of the nucleolus, and Co-IP experiments indicated that there was no interaction between MTA1 and fibrillarin; further, fibrillarin was not identified in the MTA1 interactome. Loss- and gain-of-function studies indicated that MTA1 promotes pre-rRNA transcription in cancer cells. Collectively, our data identify MTA1 as a novel nucleolar protein, and activation of pre-rRNA transcription in cancer cells may be an alternative mechanism by which MTA1 promotes malignancies.

In order to understand the mechanisms of arsenic (As) accumulation and detoxification in aquatic plants exposed to different As species, a hydroponic experiment was conducted and the three aquatic plants (Hydrilla verticillata, Pistia stratiotes and Eichhornia crassipes) were exposed to different concentrations of As(III), As(V) and dimethylarsinate (DMA) for 10 days. The biomass, the surface As adsorption and total As adsorption of three plants were determined. Furthermore, As speciation in the culture solution and plant body, as well as the arsenate reductase (AR) activities of roots and shoots, were also analyzed. The results showed that the surface As adsorption of plants was far less than total As absorption. Compared to As(V), the plants showed a lower DMA accumulation. P. stratiotes showed the highest accumulation of inorganic arsenic but E. crassipes showed the lowest at the same As treatment. E. crassipes showed a strong ability to accumulate DMA. Results from As speciation analysis in culture solution showed that As(III) was transformed to As(V) in all As(III) treatments, and the oxidation rates followed as the sequence of H. verticillata>P. stratiotes>E. crassipes>no plant. As(III) was the predominant species in both roots (39.4-88.3%) and shoots (39-86%) of As(III)-exposed plants. As(V) and As(III) were the predominant species in roots (37-94%) and shoots (31.1-85.6%) in As(V)-exposed plants, respectively. DMA was the predominant species in both roots (23.46-100%) and shoots (72.6-100%) in DMA-exposed plants. The As(III) contents and AR activities in the roots of P. stratiotes and in the shoots of H. verticillata were significantly increased when exposed to 1 mg·L-1 or 3 mg·L-1 As(V). Therefore, As accumulation mainly occurred via biological uptake rather than physicochemical adsorption, and AR played an important role in As detoxification in aquatic plants. In the case of As(V)-exposed plants, their As tolerance was attributed to the increase of AR activities.

Two new diterpenoids, ajudecunoid A (1) and ajudecunoid B (14), along with thirteen known diterpenoids, were isolated from the whole plants of Ajuga nipponensis Makino. Their structures were elucidated by the extensive spectroscopic analysis (UV, IR, MS, and NMR). The absolute configurations of ajudecunoid A (1) and ajudecunoid B (14) were defined through analysis of X-ray crystallography. Fifteen compounds were evaluated for inhibition of the formation of osteoclasts in bone marrow-derived macrophages (BMM) cells. Two neo-clerodane diterpenoids ajuganipponin B (5) and (12S)-6α,19-diacetoxy-18-chloro-4α-hydroxy-12-tigloyloxy-neo-clerod-13-en-15,16-olide (12) showed significant inhibition of osteoclastogenesis with IC50 values of 0.88 and 0.79 µM, respectively. Here we firstly reported diterpenoids with anti-osteoclastogenesis activity from A. nipponensis.

Metastasis-associated protein 1 (MTA1) is upregulated in multiple malignancies and promotes cancer proliferation and metastasis, but whether and how MTA1 promotes esophageal squamous cell carcinoma (ESCC) tumorigenesis remain unanswered. Here, we established an ESCC model in MTA1 transgenic mice induced by the chemical carcinogen 4-nitroquinoline 1-oxide (4-NQO) and found that MTA1 promotes ESCC tumorigenesis in mice. MTA1 overexpression was observed in ESCC cells and clinical ESCC samples. Overexpressed MTA1 increased colony formation and the invasiveness and migration of ESCC cells, whereas knock down of MTA1 in ESCC cells significantly decreased colony formation, invasion and migration in vitro and inhibited the growth of xenograft tumors in vivo. RNA sequencing (RNA-seq) analysis combined with western blot assays revealed that MTA1 promotes carcinogenesis by enhancing MEK/ERK/p90RSK signaling. The phosphorylation of MEK, ERK and their downstream target p90RSK was significantly decreased after MTA1 knockdown in ESCC cells and was increased in MTA1-overexpressing cells. Moreover, colony formation, invasion and migration potential were dramatically suppressed when cells overexpressing MTA1 were treated with MEK (PD0325901) or ERK (SCH772948) inhibitors. In conclusion, MTA1 plays a pivotal oncogenic role in ESCC tumorigenesis and development through activating the MEK/ERK/p90RSK pathway.

Heavy metal contamination in soil due to human activities is a global environmental problem. To find a washing solution that can significantly decontaminate heavy metals and minimize damage to soil quality, six washing solutions (H3PO4, K2CO3, CH3COOK, KH2PO4, HNO3 and KNO3) were used at different concentrations to treat contaminated soil collected from the field. Furthermore, changes in soil physicochemical properties and heavy metal speciation among prewashed, postwashed and neutralized samples were tested. Additionally, soil enzyme activities and soil microbial diversities in contaminated soil among the prewashed, postwashed and neutralized samples were also measured. Finally, a pot experiment was conducted with Mentha haplocalyx to test the efficiency of soil washing. The results revealed that the optimum washing solution was 1% HNO3 and that the removal rates of Cd and Pb were 75.7% and 60.6%, respectively, under treatment conditions of 35 °C, 90 min and a solid-liquid ratio of 1:10. The pH, total phosphorous, available potassium, soil enzyme activities and soil microbial diversity decreased significantly after washing. However, after the neutralization of washed soil with Ca(OH)2, the available phosphorous, total nitrogen and some microorganisms increased significantly compared with those of the soil before washing. After treatment with 1% HNO3, the chemical forms of Cd and Pb in soil mainly existed as F1 (exchangeable) fractions, but the main forms of the two metals changed to F5 (residual) and F3 (bound to Fe-Mn oxides) fractions after neutralization with Ca(OH)2. In addition, the plant height, root length, and fresh and dry weight of M. haplocalyx were not significantly affected by soil neutralization, while the Pb, Cu and As concentrations in the aboveground parts significantly decreased. Therefore, although soil washing could effectively remove Pb and Cd in soil, it also resulted in a significant decline in soil quality, but soil neutralization could effectively alleviate the negative effects during soil washing.

To study the effects of indole-3-acetic acid (IAA) on cadmium (Cd) accumulation and the physiological responses of the Cd hyperaccumulator Solanum nigrum and non-hyperaccumulator Solanum melongena, a pot experiment was conducted in soil containing 2 mg kg-1 Cd in which different concentrations of IAA (0, 10, 20, or 40 mg L-1) were sprayed on plant leaves. The results showed that Cd accumulation in shoots of S. nigrum was significantly increased by 30% after the addition of 10 mg L-1 IAA under 2 mg kg-1 Cd stress compared to that in the control, but shoot Cd accumulation showed no significant change in S. melongena after this IAA treatment. Additionally, the growth and the proline content in the two species were significantly increased by 20 mg L-1 IAA. The activities of peroxidase and catalase in leaves of S. nigrum and the activity of superoxide dismutase (SOD) in S. melongena were significantly increased and their malondialdehyde content was significantly decreased compared to those in the control. The root activity of S. nigrum was significantly improved after 10 and 20 mg L-1 IAA treatments, but no significant difference was observed in S. melongena. The correlation analysis results showed that the Cd concentration in leaves of S. nigrum was significantly and positively correlated with the carotenoid and proline contents, and there was also a significant positive correlation between the Cd concentration and SOD activity in leaves of S. melongena. Therefore, S. nigrum is an ideal plant for the phytoextraction of Cd-contaminated soil assisted by IAA. IAA promotes Cd accumulation in plant shoots by enhancing the accumulation of carotenoids and proline in S. nigrum and maintaining a high leaf SOD activity in S. melongena.

Cytokinins (CTKs) are effective in alleviating abiotic stresses on plants, but little information is available regarding the effects of CTKs on arsenic (As) accumulation and changes of chloroplast ultrastructure in plants with different As-accumulating ability. Here a hydroponic experiment was designed to evaluate the effects of different concentration of kinetin (KT, 0-40 mg/L) on growth and chloroplast ultrastructure of As hyperaccumulator Pteris cretica var. nervosa and non-hyperaccumulator Pteris ensiformis treated by 5 mg/L arsenate for 14 days. The growth parameters, As accumulation, contents of photosynthetic pigments and chloroplast ultrastructure were examined. The results showed that KT promoted the growth of two plants, and significantly increased As accumulation and translocation in P. cretica var. nervosa and P. ensiformis at 5 and 20 mg/L, respectively. Additionally, the contents of chlorophyll a and carotenoid in two plants showed no significant difference at 20 mg/L KT compared to the control. Chloroplast ultrastructure of P. cretica var. nervosa was integral with KT application. Comparatively, the swollen chloroplasts were increased, plasmolysis appeared, and chloroplast grana slice layers and stroma lamellas were clearly separated or distorted at 5 mg/L KT in P. ensiformis. The length and width of chloroplasts in P. cretica var. nervosa were significantly increased with KT addition compared to the control. However, the length of chloroplasts in P. ensiformis was significantly decreased but their width showed no significant change. Furthermore, the deterioration of chloroplast ultrastructure in P. ensiformis was ameliorated by 40 mg/L KT. These results suggested that KT increased As accumulation and was beneficial to maintain the photosynthetic pigments for a good growth of plants. Therefore, KT could maintain and reorganize the ultrastructure integrality of As-stressed chloroplasts to some extent for the two plants, especially at high concentration.

A hydroponic experiment was conducted to investigate the effects of oxalic acid (OA) on arsenic (As) uptake and the physiological responses of Hydrilla verticillata exposed to 3 mg L-1 of As in different forms. Plant As(III) uptake was significantly increased by 200-2000 µg L-1 OA. However, an increase of As(V) uptake was only shown with 1000 µg L-1 OA, and no significant difference was observed with dimethylarsinate treatment. Peroxidase and catalase activities, and the contents of photosynthetic pigments, soluble sugar and proline, were significantly increased by 1000 µg L-1 OA during As(III) treatment. Superoxide dismutase and proline were also increased significantly by 1000 µg L-1 OA when plants were exposed to As(V). In DMA treatment, proline was significantly increased by 500 µg L-1 OA. Therefore, As-induced oxidative stress is relieved by OA, but it depends on OA concentration and the form of As. Our results may be useful for the phytoremediation of waste water containing As and OA.

A pot experiment was conducted to compare the content of endogenous trans-zeatin (Z), plant arsenic (As) uptake and physiological indices in the fronds of As-hyperaccumulator (Pteris cretica var. nervosa) and non-hyperaccumulator (Pteris ensiformis). Furthermore, a stepwise regression method was used to study the relationship among determined indices, and the time-course effect of main indices was also investigated under 100mg/kg As stress with time extension. In the 100-200mg/kg As treatments, plant height showed no significant difference and endogenous Z content significantly increased in P. cretica var. nervosa compared to the control, but a significant decrease of height and endogenous Z was observed in P. ensiformis. The concentrations of As (III) and As (V) increased significantly in the fronds of two plants, but this increase was much higher in P. cretica var. nervosa. Compared to the control, the contents of chlorophyll and soluble protein were significantly increased in P. cretica var. nervosa but decreased in P. ensiformis in the 200mg/kg As treatment, respectively. A significant positive correlation was found between the contents of endogenous Z and total As in P. cretica var. nervosa, but such a correlation was not found in P. ensiformis. Additionally, in the time-course effect experiment, a peak value of each index was appeared in the 43rd day in two plants, except for chlorophyll in P. ensiformis, but this value was significantly higher in P. cretica var. nervosa than that in P. ensiformis. In conclusion, a higher endogenous Z content contributed to As accumulation of P. cretica var. nervosa under As stress.

Plants were exposed to 2 mg L-1 As(III), As(V) or dimethylarsinic acid (DMA) and IAA concentrations for 14 d. The biomass and total As in the plants significantly increased at 30 mg L-1 IAA. Superoxide dismutase (SOD) activities significantly increased with IAA addition. Catalase (CAT) activities showed a significant increase in P. ensiformis exposed to three As species at 30 or 50 mg L-1 IAA but varied in P. cretica var. nervosa. Peroxidase (POD) activities were unchanged in P. ensiformis except for a significant decrease at 50 mg L-1 IAA under As(III) treatment. However, a significant increase was observed in P. cretica var. nervosa at 10 mg L-1 IAA under As(III) or DMA treatment and at 50 mg L-1 IAA under As(V) treatment. Under DMA stress, malondialdehyde contents in fronds of P. cretica var. nervosa showed a significant decrease at 10 mg L-1 IAA but remained unchanged in P. ensiformis. Therefore, IAA enhanced As uptake and frond POD activity in P. cretica var. nervosa under As stress.

The aim of this study was to compare the efficacies between adoptive immunotherapy combined chemoradiotherapy and chemoradiotherapy alone in patients with non-small-cell lung cancer (NSCLC). The databases PubMed, EMBASE, and Cochrane database were searched to identify eligible clinical trials. Data analyses were carried out using a comprehensive meta-analysis program, version 2 software. A total of seven articles were finally included in the analysis. Meta-analyses showed that compared with chemoradiotherapy alone, adoptive immunotherapy combined with chemoradiotherapy could improve the 2-year overall survival [odds ratio (OR)=2.45, 95% confidence interval (CI): 1.60-3.75, P<0.001], but not 2-year progression-free survival (OR=1.81, 95% CI: 0.61-5.36, P=0.284). Specifically, early (OR=3.32, 95% CI: 1.38-7.95, P<0.01) but not advanced (OR=3.75, 95% CI: 0.96-14.68, P=0.057) NSCLC patients were likely to gain a large benefit from the adoptive immunotherapy. Most of the adoptive immunotherapy-induced adverse effects were self-limited, mainly including fever, shiver, nausea, fatigue, etc. and severe toxicities were not observed. Adoptive immunotherapy combined with chemoradiotherapy can delay the recurrence of NSCLC and improve survival in patients, where the benefits are even more significant in patients with early-stage NSCLC.

The subcellular localization of a protein is closely linked to and indicates its function. The metastatic tumor antigen (MTA) family has been under continuous investigation since its identification two decades ago. MTA1, MTA2, and MTA3 are the main members of the MTA family. MTA1, as the representative member of this family, has been shown to be widely expressed in both embryonic and adult tissues, as well as in normal and cancerous conditions, indicating that MTA1 has functions both in physiological and pathological contexts. MTA1 is expressed at a higher level in most cancers than in their normal tissue counterparts. Even in normal cells, MTA1 levels vary a great deal from tissue to tissue. Importantly, MTA1 shows a multiple localization pattern in the cell, as do MTA2 and MTA3. Different MTA components in different subcellular compartments may exert different molecular functions in the cell. Previous studies revealed that MTA1 and MTA2 are predominately localized to the nucleus, while MTA3 is observed in both the nucleus and cytoplasm. Recent studies have reported that MTA1 is located in the nucleus, cytoplasm, and the nuclear envelope. In the nucleus, MTA1 dynamically interacts with chromatin in a MTA1-K532 methylation-dependent manner, whereas cytoplasmic MTA1 binds to the microtubule skeleton. MTA1 also shows a dynamic distribution during the cell cycle. Further investigations are needed to identify the exact subcellular localizations of MTA proteins. We review the sub-cellular localization patterns of the MTA family members and give a comprehensive overview of their respective molecular activities in multiple contexts.

As the most common NK/T-cell lymphoma in Asian countries, extranodal NK/T-cell lymphoma, nasal type (ENKTL), has unique clinical features and a strong association with Epstein-Barr virus (EBV). In order to gain a preliminary understanding of the relationship between ENKTL and EBV, we performed genotypic analysis of EBV and investigated LMP1 expression in extranodal NK/T-cell lymphoma. Our study shows that ENKTL is an EBV-associated malignancy and that A, C and F are the predominant EBV genotypes in northern China. LMP1 expression is stronger in extranasal sites than nasal sites, and the expression level is strongly correlated to ENKTL and may play an important role in the development of ENKTL.

BACKGROUND: Crizotinib was granted accelerated approval by the Food and Drug Administration in 2011 for the treatment of anaplastic lymphoma kinase (ALK)-positive non-small cell lung cancer (NSCLC). To evaluate the efficacy and safety of crizotinib, we performed a meta-analysis of published clinical trials using the random effect model. METHODS: The efficacy and safety of crizotinib was evaluated based on 1-year overall survival (OS), progression-free survival (PFS), overall response rate (ORR), partial response, complete response, stable disease, and dose reduction or cessation because of crizotinib toxicity. RESULTS: Six clinical trials were included in the meta-analysis. Crizotinib treatment demonstrated a 1-year OS of 66.8% (95% CI, 52.2-78.8%) and a PFS of 8.6 months (95% CI, 7.3-9.9 months). The aggregate ORR, partial response and complete response rates were 61.2%, 59.8% and 1.5%, respectively. The proportion of patients achieving stable disease was 42.6% (95% CI, 17.3-72.5%). The most frequently reported adverse effects of crizotinib were mild visual disturbances, nausea, vomiting, diarrhea, constipation, edema, reduction in glomerular filtration rate, and generally reversible but sometimes severe elevations in aspartate aminotransferase and alanine aminotransferase. The proportion of patients who required dose reduction or cessation because of crizotinib toxicity was 6.5% (95% CI, 4.1-10.1%). CONCLUSIONS: This meta-analysis revealed extended survival and improved response rates in patients treated with crizotinib. As a novel, targeted anticancer agent, crizotinib appears to be a favorable treatment option for patients with locally advanced or metastatic ALK-positive NSCLC.

Trans-acting small interfering RNAs (tasiRNAs) are 21-nt phased (phased siRNAs) resulting from successive DCL-catalyzed processing from the end of a double-stranded RNA substrate originating from the RDR of an AGO-catalyzed cleaved RNA at a micro RNA target site. Plant tasiRNAs have been synthesized to produce synthetic tasiRNAs (syn-tasiRNAs) targeting viral RNAs that confer viral resistance. In this study, we engineered syn-tasiRNAs to target potato virus Y (PVY) infection by replacing five native siRNAs of TAS1c with 210-bp fragments from the coat protein (CP) region of the PVY genome. The results showed that the transient expression of syn-tasiR-CPpvy2 in Nicotiana benthamiana (N. benthamiana) plants conferred antiviral resistance, supported by the absence of PVY infection symptoms and viral accumulation. This indicated that syn-tasiR-CPpvy2 successfully targeted and silenced the PVY CP gene, effectively inhibiting viral infection. syn-tasiR-CPpvy1 displayed attenuated symptoms and decreased viral accumulation in these plants However, severe symptoms of PVY infection and a similar amount of viral accumulation as the control were observed in plants expressing syn-tasiR-CPpvy3. syn-tasiR-CPpvy/pvx, which targets both PVY and potato virus X (PVX), was engineered using a single precursor. After the transient expression of syn-tasiR-CPpvy/pvx3 and syn-tasiR-CPpvy/pvx5 in N. benthamiana, the plants were resistant to both PVY and PVX. These results suggested that engineered syn-tasiRNAs could not only specifically induce antiviral resistance against one target virus but could also be designed for multi-targeted silencing of different viruses, thereby preventing complex virus infection in plants.

Contamination of leafy vegetables grown in heavy metal(loid)-polluted mining areas pose serious health risks. This study aimed to explore the heavy metal(loid) contamination of leafy vegetables near two mining areas, by collecting samples from 14 different leafy vegetable species in Yunnan Province, China. The lead (Pb), cadmium (Cd), arsenic (As), and copper (Cu) contents of the samples were determined, and risks to human health were calculated using the hazard quotient and hazard index (HI). Moreover, Malabar spinach was identified as a leafy vegetable that exhibits low accumulation of heavy metal(loid)s. The accumulation capacity of different Malabar spinach varieties was verified, and a Cd soil safety threshold was determined using a pot experiment. Overall, Pb and Cd were the main soil and vegetable contaminants found in both study sites. The HI values for all leafy vegetables, apart from Malabar spinach, were greater than 1, indicating the presence of risks to human health; moreover, the health risks were greater for children than adults. The Malabar spinach pot experiment results showed that only some Cd forms exceeded China's maximum permissible standards. Furthermore, Malabar spinach varieties A (instant Malabar spinach), C (extra-large leaf green vine Malabar spinach), and F (large leaf Malabar spinach) displayed the lowest Cd accumulation. We calculated Cd total and bioavailable soil safety thresholds of 4.75 and 0.77 mg kg-1, respectively. However, further research is required to validate soil heavy metal safety thresholds for different vegetables. Ultimately, the heavy metal(loid) contamination of leafy vegetables described here was more serious than anticipated. Finally, the results of this study can inform residents living near these mining areas of a low-risk leafy vegetable, which will reduce the harm caused by heavy metal(loid) contamination in the area.

The identity of the mechanism by which the Baixiangdan capsule (BXD) and the Shuyu capsule (SY) control anger-out (AO) and anger-in (AI) in rodents is unclear. The current study clarified the intervention role of BXD and SY on AO and AI male rats. We further explored the differences between BXD and SY in the treatment of AO and AI rats. Social isolation combined with the resident-intruder paradigm was used to establish the anger-out and AI rats models. On this basis, GABA content in the dorsal raphe nucleus (DRN) and serotonin (5-HT) contents in these brain regions were detected using ELISA after various time courses (0, 1, 3, 5, and 7 days) treated with BXD and SY. Co-expression of 5-HT and GB1 in the DRN was detected. GB1-specific agonist baclofen and GB1-specific inhibitor CGP35348 were injected into the DRN. Changes in 5-HT levels in these brain regions were then detected. After treatment, rats in the BXD group exhibited lower aggressive behavior scores, longer latencies of aggression, lower total distances in the open field test, and a higher sucrose preference coefficient. Meanwhile, rats in the SY group exhibited higher aggressive behavior scores, shorter latencies of aggression, higher total distances in the open field test, and higher sucrose preference coefficients. With increasing medication duration, 5-HT levels in these brain regions were increased gradually, whereas GABA levels in the DRN were decreased gradually, and all recovered to normal levels by the 7th day. A large number of 5-HT-positive cells could be found in the immunofluorescence section in the DRN containing GABABR1 (GB1)-positive cells, indicating that 5-HT neurons in the DRN co-expressed with GB1. Furthermore, after the drug intervention, the 5-HT level in the DRN was elevated to a normal level, and the GB1 level in the DRN was decreased to a normal level. After the microinjection of baclofen into the DRN, the 5-HT contents in these brain regions were decreased. By contrast, the 5-HT contents were increased after injection with CGP35348. BXD and SY could effectively improve the abnormal behavior changes of AO and AI rats, and the optimal duration of action was 7 days. The improvement way is as follows: Decreased abnormal increase of GABA and GB1 in the DRN further mediated synaptic inhibition and increased 5-HT level in the DRN, leading to increased 5-HT levels in the PFC, hypothalamus, and hippocampus. Therefore, GB1-mediated GABA in the DRN could regulate 5-HT levels in these brain regions, which may be one of the ways by which BXD and SY treat AO and AI, respectively.
