The diversity observed in canine breed phenotypes, together with their risk for heritabily disorders of relevance to dogs and humans, makes the species an ideal subject for studies aimed at understanding the genetic basis of complex traits and human biomedical models. Dog10K is an ongoing international collaboration that aims to uncover the genetic basis of phenotypic diversity, disease, behavior, and domestication history of dogs. To best present and make the extensive data accessible and user friendly, we have established the Dog10K (http://dog10k.kiz.ac.cn/) database, a comprehensive-omics resource summarizing multiple types of data. This database integrates single nucleotide variants (SNVs) from 1987 canine genomes, de-novo mutations (DNMs) from 43 dog breeds with >40× sequence, RNA-seq data of 105057 single nuclei from hippocampus, 74067 single cells from leukocytes and 30 blood samples from published canid studies. We provide clear visualization, statistics, browse, searching, and downloading functions for all data. We have integrated three analysis tools, Selscan, LiftOver and AgeConversion, to aid researchers in custom exploration of the comprehensive-omics data. The Dog10K database will serve as a foundational platform for analyzing, presenting and utilizing canine multi-omics data.

AIM: To evaluate the incidence of facial pressure injuries in health-care professionals during the COVID-19 pandemic in a meta-analysis. METHODS: Related studies were obtained through electronic databases, including PubMed, Embase, Cochrane Library, Web of Science, China National Knowledge Infrastructure (CNKI) Chinese Scientific Journal (VIP) China Biomedical Literature service systems (CBM) and Wanfang Data (from inception to 27 November 2021). The pooled incidence and the 95% confidence interval of facial pressure injuries were calculated with Review Manager v5.4 software. RESULTS: Overall, 16 studies with 14 430 health-care professionals were included. Pooled results showed that the pooled incidence of facial pressure injury in health-care professionals was 58.8% (95% CI: 49.0%-68.7%; p < 0.01). The results of the subgroup analysis showed that the incidence of facial pressure injury in these staff was high, and predominantly stage I pressure injury, in the following cases: in health-care professionals who wore personal protective equipment for longer than 4 h, in those without any training experience, and on the nose. CONCLUSION: Administrators and researchers should pay attention to preventing facial pressure injury related to the wearing of personal protective equipment (PPE) by ensuring all health-care professionals receive training and by limiting prolonged periods of use.

BACKGROUND: Better protection can be provided during neurosurgery due to the establishment of somatosensory-evoked potential (SEP) and motor-evoked potential (MEP) monitoring technologies. However, some studies have showed that inhaled halogenated anesthetics have a significant impact on neurophysiological monitoring. METHODS: A total of 40 consecutive patients undergoing neurosurgery were randomly assigned to two groups receiving inhaled anesthetics, either desflurane or sevoflurane. Multiples levels (concentrations of 0.3, 0.6 and 0.9) of anesthetics were administered at minimum alveolar concentration (MAC), and then the latencies and amplitudes of SEPs and MEPs were recorded. RESULTS: SEP and MEP signals were well preserved in patients who underwent neurosurgery under general anesthesia supplemented with desflurane or sevoflurane at concentrations of 0.3, 0.6 and 0.9 MAC. In each desflurane or sevoflurane group, the amplitudes of SEPs and MEPs decreased and the latencies of SEPs were prolonged significantly as the MAC increased (P < 0.05). The SEP latencies of both the upper and lower limbs in the desflurane group were significantly longer, and the SEP amplitudes were significantly lower than those in the sevoflurane group (P < 0.05). The MEP amplitudes in the desflurane group were significantly lower than those in the sevoflurane group (P < 0.05), only the amplitudes of the upper limbs at 0.3 MAC did not vary significantly. CONCLUSIONS: SEPs and MEPs were inhibited in a dose-dependent manner by both desflurane and sevoflurane. At the same MAC concentration, desflurane appeared to have a stronger inhibitory effect than sevoflurane. All patients studied had normal neurological examination findings, hence, these results may not be applicable to patients with preexisting deficits. Research suggests that physical cleansing leads individuals to perceive themselves as morally purer, thereby influencing their moral activities, but findings are inconsistent. Some researchers have found that individuals act more morally after physical cleansing, whereas others have observed the reverse effect. Building on embodied cognition theory, which holds that cognition is developed from physical experience and that this experience in turn guides their responses, we hypothesized that physical cleansing would have a general impact on individuals' moral activities by highlighting themselves. In contrast to previous research, we did not expect that physical cleansing would directly make people more (im)moral. Two experiments were conducted to test our hypotheses. In experiment 1, participants who had bathed showed reduced susceptibility to external moral priming (morality-related descriptions of themselves) than did control participants. Experiment 2 showed that physical cleansing (but not the control condition) amplified the interplay among participants' moral activities, depending on the order of immoral behavior and intention measurement. Taken together, these results suggest that physical cleansing strengthens the connection between individuals' morality and bodily experiences, weakening the effect of external priming (experiment 1) and enhancing the influence of moral activities performed by themselves (experiment 2). Although further research is required to provide direct evidence for these findings, our study contributes to the integration of inconsistencies in previous findings by suggesting a new perspective for the understanding of the embodied effect of physical cleansing.

This study established a model of mineral element homeostatic imbalance and examined the respiratory burst function of peripheral blood neutrophils and inflammatory and antioxidant indicators before and after the imbalance in sheep. The results showed that after an EDTA injection, the number of activated neutrophils in the peripheral blood was significantly higher than that in the control group (p < 0.01). In addition, the serum IL-6 level was significantly increased (p < 0.05) and matrix metalloproteinase 7 (MMP7) was inhibited (p < 0.05), but returned to a normal level one week after the injection. Tissue inhibitor of metalloproteinase 1 (TIMP1) levels were consistently higher after the injection and significantly higher than in the control group (p < 0.05). CuZn-SOD, TNOS activity, serum creatinine and urea nitrogen levels were significantly higher than before the injection (p < 0.05). Combining the results of previous studies, the EDTA injection altered the metabolism and transcription of peripheral blood neutrophils. These changes enhance the respiratory burst function of neutrophils and alter the status of inflammatory and antioxidant indicators such as IL-6 and CuZn-SOD.
RESUMO
AIM: This systematic review evaluated the quality of evidence for the prevention and management of facial pressure injuries in medical staff. DESIGN: This review was presented in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses guidelines. METHODS: We retrieved the relevant studies from 19 databases. Using the literature evaluation standards and evidence grading system of the Australian Joanna Briggs Institute Evidence-Based Health Care Center, we evaluated the quality of the literature encompassing different types of research and assessed their levels of evidence. RESULTS: A total of 13 studies were included, including seven expert consensuses, two recommended practices, one clinical decision, one best practice information booklet, one systematic review and one randomized controlled trial. In the end, 31 best evidence were summarized, including skin cleaning and care, PPE placement and movement, reasonable use of dressings, treatment measures and education and training.

BACKGROUND: Thyroid cancer (TC) is the most common endocrine malignancy that has rapidly increased in global incidence. Prunella vulgaris (PV) has manifested therapeutic effects in patients with TC. We aimed to investigate its molecular mechanisms against TC and provide potential drug targets by using network pharmacology and molecular docking. METHODS: The ingredients of PV were retrieved from Traditional Chinese Medicine Systematic Pharmacology Database. TC-related gene sets were established using the GeneCard and OMIM databases. The establishment of the TC-PV target gene interaction network was accomplished using the STRING database. Cytoscape constructed networks for visualization. Protein-protein interaction, gene ontology and the biological pathway Kyoto encyclopedia of genes and genomes enrichment analyses were performed to discover the potential mechanism. Molecular docking technology was used to analyze the effective compounds from PV for treating TC. RESULTS: 11 active compounds and 192 target genes were screened from PV. 177 potential targets were obtained by intersecting PV and TC gene sets. Network pharmacological analysis showed that the PV active ingredients including Vulgaxanthin-I, quercetin, Morin, Stigmasterol, poriferasterol monoglucoside, Spinasterol, kaempferol, delphinidin, stigmast-7-enol, beta-sitosterol and luteolin showed better correlation with TC target genes such as JUN, AKT1, mitogen-activated protein kinase 1, IL-6 and RELA. The gene ontology and Kyoto encyclopedia of genes and genomes indicated that PV can act by regulating the host defense and response to oxidative stress immune response and several signaling pathways are closely associated with TC, such as the TNF and IL-17. Protein-protein interaction network identified 8 hub genes. The molecular docking was conducted on the most significant gene MYC. Eleven active compounds of PV can enter the active pocket of MYC, namely poriferasterol monoglucoside, stigmasterol, beta-sitosterol, vulgaxanthin-I, spinasterol, stigmast-7-enol, luteolin, delphinidin, morin, quercetin and kaempferol. Further analysis showed that oriferasterol monoglucoside, followed by tigmasterol, were the potential therapeutic compound identified in PV for the treatment of TC. CONCLUSION: The network pharmacological strategy integrates molecular docking to unravel the molecular mechanism of PV. MYC is a promising drug target to reduce oxidative stress damage and potential anti-tumor effect. Oriferasterol monoglucoside and kaempferol were 2 bioactive compounds of PV to treat TC. This provides a basis to understand the mechanism of the anti-TC activity of PV.

The mismatch between the supply and demand of religious places has resulted from a lack of management, population mobility, and urban socio-spatial differentiation. Furthermore, the identity of religious groups and the localized characteristics of their behavioral practices continue to dominate the evolution of religious places. Therefore, this study analyzed the spatio-temporal patterns of Christian activity places, and further studied the characteristics and mechanisms of various influencing factors on the spatial distribution of places using a geographically weighted regression model from a socio-spatial perspective. The following results were obtained: (1) Christian activity places form a distribution pattern of circling aggregation within the core area and polycentric dispersion outside the core area. There is a certain coupling relationship between spatio-temporal patterns of places and marketization in Yiwu City. (2) The spatial differentiation pattern of influencing factors has an obvious circle-layer characteristic. The regression coefficients of population mobility, age structure, public services, and jobs-housing space are larger, and social structure and spatial resource allocation are becoming the main driving forces of spatial differentiation. (3) Due to the differences in spatial resource allocation and believers' behavioral choices, the above factors have distinct differences in urban core areas, urban-rural transition areas and rural areas.

The imbalance of mineral element homeostasis in animals is common, causing animal immune dysfunction. Ten female sheep were randomly selected and injected with 4% (w/v) Na2EDTA through a central venous catheter to establish the mineral element imbalance model, then divided into control group (before injection) and Ethylene Diamine Tetraacetic Acid (EDTA) group (after injection). Isolation of peripheral blood neutrophils for mineral elements content determination was done using Inductively coupled plasma-mass spectrometry (ICP-MS) and nontargeted metabolomics analysis using liquid chromatography-tandem mass spectrometry (LC-MS/MS). The ICP-MS results showed that Hg and Cs levels in neutrophils were significantly lower after injection than before (P < 0.05), but had no significant effects on other elements. Our previous study showed that serum Zn, P, K, and other 11 elements were significantly lower after the injection of Na2EDTA than before. LC-MS/MS results showed that differential metabolites are mainly involved in amino acid metabolism, lipid metabolism, and nucleotide metabolism; monoamine metabolism was weakened; and polyamine metabolism was enhanced. Under positive and negative ion modes, the mineral elements P, K, Ca, Mn, Cu, and Zn had the highest correlation with the differential metabolites of neutrophils, followed by Se, and the correlation between each mineral element and different differential metabolites was also different. The results indicated that the imbalance in mineral elements affected the metabolism of sheep neutrophils, these may affect neutrophil function, and Na2EDTA could help to reduce the level of heavy metals in the body of sheep. Our data may provide a theoretical basis for the precise regulation of animal immune metabolism by modern animal husbandry nutrition.

This experiment was designed to investigate the effects of mineral element imbalance on gene transcription in peripheral circulating neutrophils of sheep. Ten female sheep weighing 30 kg of similar age and physiological condition were randomly selected and injected via central venous catheterization with EDTA at a concentration of 4% (W/V) to construct a model of mineral element imbalance. As self-control, the sheep were divided into control [Con, before EDTA injection, time point 0 (T0)], EDTA [12 h after EDTA injection, time point 12 h (T12h)] and recovery [HF, 7 days after injection, time point 7 days (T7d)] groups. Whole blood was collected, and serum and neutrophils were separated for ionomic and transcriptomic analysis. The results showed that levels of P (P < 0.05), Zn (P < 0.01), K (P < 0.01) and some other elements were significantly lower in sheep in the EDTA group than in those in the control group, but levels of P (P > 0.05), Zn (P > 0.05) and K (P > 0.05) were similar in the recovery and control groups. Levels of Ca (P > 0.05) and Cu (P < 0.05) were higher after injection, and levels of Cu (P < 0.01) continued to increase during the recovery period. There were 1561 genes, including S100 family genes, significantly differentially expressed in neutrophils between the control and trial groups, and these genes were mainly enriched in the categories defense response, immune response, regulation of interleukin production and other functions. The top enriched signaling pathways were phagosome, NF-κB and Toll-like receptor. Mineral element homeostasis imbalance affected the gene transcriptome of circulating neutrophils, demonstrating the importance of carefully controlling the addition of mineral elements.

The process of domestication has led to dramatic differences in behavioral traits between domestic dogs and gray wolves. Whole-genome research found that a class of putative positively selected genes were related to various aspects of learning and memory, such as long-term potentiation and long-term depression. In this study, we constructed a single-nucleus transcriptomic atlas of the dog hippocampus to illustrate its cell types, cell lineage and molecular features. Using the transcriptomes of 105 057 nuclei from the hippocampus of a Beagle dog, we identified 26 cell clusters and a putative trajectory of oligodendrocyte development. Comparative analysis revealed a significant convergence between dog differentially expressed genes (DEGs) and putative positively selected genes (PSGs). Forty putative PSGs were DEGs in glutamatergic neurons, especially in Cluster 14, which is related to the regulation of nervous system development. In summary, this study provides a blueprint to understand the cellular mechanism of dog domestication.

Vertebrate evolution was accompanied by two rounds of whole-genome duplication followed by functional divergence in terms of regulatory circuits and gene expression patterns. As a basal and slow-evolving chordate species, amphioxus is an ideal paradigm for exploring the origin and evolution of vertebrates. Single-cell sequencing has been widely used to construct the developmental cell atlas of several representative species of vertebrates (human, mouse, zebrafish, and frog) and tunicates (sea squirts). Here, we perform single-nucleus RNA sequencing (snRNA-seq) and single-cell assay for transposase accessible chromatin sequencing (scATAC-seq) for different stages of amphioxus (covering embryogenesis and adult tissues). With the datasets generated, we constructed a developmental tree for amphioxus cell fate commitment and lineage specification and characterize the underlying key regulators and genetic regulatory networks. The data are publicly available on the online platform AmphioxusAtlas.

The grey wolf (Canis lupus) is one of the most widely distributed terrestrial mammals, and its distribution and ecology in Europe and North America are largely well described. However, the distribution of grey wolf in southern China is still highly controversial. Several well-known western literatures stated that there are no grey wolves in southern China, while the presence of grey wolf across China has been indicated in A Guide to the Mammals of China, published by Princeton University Press. It is essential to solve this discrepancy since dogs may have originated from grey wolfs in southern China. Therefore, we systematically investigated Chinese literatures about wild animal surveys and identified more than 100 articles and books that included information of the distribution of grey wolves in China. We also surveyed the collections of three Chinese natural museums and found 26 grey wolf skins specimens collected across China. Moreover, we investigated the fossil records of wolf in China and identified 25 archaeological sites with wolf remains including south China. In conclusion, with the comprehensive summary of Chinese literatures, museum specimens and fossil records, we demonstrate that grey wolves does distribute across all parts of the Chinese mainland, including the most southern parts of China.
