BACKGROUND: Neuromyelitis optica spectrum disorder (NMOSD) is a recurring inflammatory demyelinating disease that is commonly observed in Asian countries like China. Prior investigations have shown that mycophenolate mofetil (MMF) with better biocompatibility compared to azathioprine (AZA), and can prevent relapses of NMOSD, but the efficacy was controversially reported in different NMOSD cases. We aimed to explore the factors that weaken efficacy of MMF in NMOSD. METHODS: A total of 34 NMOSD patients treated with MMF were prospectively enrolled and grouped according to the therapeutic efficacy as effective group (EG, n = 23) versus less-effective group (LEG, n = 11). The purine metabolites were profiled in serum samples and gut microbiota was analyzed using 16S rRNA sequencing with stool samples from the same patients. RESULTS: Purine salvage pathway (PSP) metabolites (inosine, hypoxanthine, xanthine, guanine and uric acid) in the serum of NMOSD patients were elevated in the LEG compared to EG (p < 0.05). Additionally, the richness and microbial diversity of gut microbiota was found to be similar between EG and LEG patients. However, LEG patients had increased presence of Clostridium and Synergistes but decreased abundance of the Coprococcus genus. CONCLUSIONS: The PSP metabolites and composition of the gut microbiota were changed between patients with or without optimal clinical response after MMF treatment. This may help us to understand the pharmacodynamics of MMF in NMOSD.

CONTEXT: This study explores, for the first time, using first principles, the impact of substitutional doping with boron (B), carbon (C), and nitrogen (N) on the adsorption of chromium (Cr) on monolayer MoS2. The effects of doping on the Cr adsorption behavior of MoS2 were investigated using four MoS2 systems, namely, pure, boron (B)-doped, carbon (C)-doped, and nitrogen (N)-doped, in order to gain an in-depth understanding of the mechanism of the effects of doping on the electronic structure and optical properties of Cr adsorbed by MoS2, to optimize the properties of MoS2, to explore new areas of application, and to promote the development of materials science. Four MoS2 adsorption systems of Cr adsorption on pure, B-doped, C-doped, and N-doped MoS2 were optimized, and the optimized results showed that the stable adsorption location of Cr on both pure and doped MoS2 was the hollow location at the top of the folded hexagon. The findings reveal that pure MoS2 has an adsorption effect on Cr, and doped elements B, C, and N can promote the adsorption of Cr on MoS2, and the strong and weak order of this promotion is B > C > N. METHODS: In this paper, we use the CASTEP module in the simulation software Materials Studio to perform simulation calculations and analyses to optimize the simulation of Cr adsorption by MoS2 doped with B, C, and N atoms using the generalized gradient approximation (GGA) plane-wave pseudo-potential method (Perdew et al. Phys Rev Lett 77(18):3865-3968, 1996), as well as Perdew-Burke-Ernzerhof (PBE) generalized functionals (Segall et al. J Phys: Condens Matter 14(11):2717-2744, 2022). The convergence test reveals that it is more reasonable to set the K-point network to 3 × 3 × 1 and the truncation energy to 400 eV. In this paper, a 3 × 3 × 1 supercell structure with 18 S atoms and 9 Mo atoms is selected. The convergence value of the iteration accuracy is 1.0e - 5eV/atom, and all the atomic forces are less than 0.02eV/Å. Additionally, to prevent MoS2 interlayer interaction, a vacuum layer with a thickness of 18 Å is set in the C direction. The geometrical optimization of the model is performed first, and then the corresponding adsorption energies of the model and the nature of the electronic structure are analyzed.

OBJECTIVE: To investigate the agreement of subjective evaluation of orthodontic treatment outcome and to analyze possible factors that may be related to it. METHODS: As a randomized clinical trial, with Angle's classification as a stratification factor, our study contained 48 cases with integrity data, which were randomly extracted from 806 orthodontic treatment cases in Peking University School and Hospital of Stomatology during July 2004 and August 2008, and gathered post-treatment study casts, cephalometrics and photographs of the 48 cases as the research subjects. Similarly with Angle's classification as a stratification factor, the 48 cases were randomly divided into 4 groups. According to the monomial and combined subjects, 12 clinicians were asked to act as the raters to rank the 12 cases in each group. RESULTS: Overall, there were 1 584 pairings between the raters in the examination of evaluation. The mean Spearman r was 0.565 3 ± 0.239 9. Grouping factor was related to the agreement of subjective evaluation (P<0.05). In the third trial item-Post-M+P, the correlations were the greatest among the judge-pairs (P<0.001). The other five items were at the same agreement level. The level of orthodontic treatment outcome was a factor that influenced the agreement level of subjective evaluation (P<0.05). The score stability of the patients, whose treatment duration was longer than 2.5 years, was significantly higher than that of the patients whose treatment duration was between 1.5 years and 2.5 years (P<0.05). The following factors, such as Angle's classification, age of patients and whether the teeth was extracted or not, were the insignificant factors (P>0.05). CONCLUSION: The average correlations present a moderate agreement level. Grouping, experimental item, the length of treatment duration and the level of orthodontic treatment outcome are the factors that affect the agreement of subjective evaluation. Several factors including Angle's classification, age of patients and whether the teeth is extracted or not, do not affect the agreement of subjective evaluation.

OBJECTIVE: To investigate the effects of Xiaoyusan plaster and Daoyin in the treatment of cervical vertebrae disease, to search new methods in the treatment of cervical vertebrae disease. METHODS: From May 2007 to April 2008, 63 patients with disease of cervical vertebrae were treated. By odd or even numbers in random digits table, the patients were randomly divided into two groups included treatment group and control group. Thirty-three patients in the treatment group were treated with Xiaoyusan plaster and Daoyin included 21 males and 12 females with an average age of (30.60+/-7.89) years ranging from 20 to 49. Thirty patients in the control group were treated with Votalin Sustained Release Tablets included 16 males and 14 females with an average age of (32.43+/-8.00) years ranging from 20 to 49. The pain, pressing pain, activity of cervical vertebrae were observed before and after treatment in two groups. RESULTS: Compared the scores before and after treatment in treatment group: pain (t=8.953, P<0.001); pressing pain (t=7.867, P<0.001); activity (t=6.918, P<0.001). Compared the scores before and after treatment in control group: pain (t=8.733, P<0.001); pressing pain (t=7.663, P<0.001); activity (=5.066, P<0.001). The symptoms such as pains, pressing pain, movement restriction improved significantly in two groups. CONCLUSION: Xiaoyusan plaster and Daoyin are one of the effective methods in the treatment of disease of cervical vertebrae, especially can improve symptoms and correct dynamic balance of cervical vertebrae.
