Local indigenous knowledge (IK) informs decision-making about fundamental aspects of life. The inclusion of indigenous knowledge is increasingly becoming a topical subject to enhance livestock veterinary care. The objective of the current study was to explore the extent of utilisation of indigenous knowledge to control ticks in goats. There was an association (P < 0.05) between the use of IK and gender, with males using IK (76.58%) more than females. The association between age distribution and IK use was (P < 0.05); however, farmers above the age of 50 years were using IK more than all group ages. Farmers ranked the purposes of using IK differently (P < 0.05). Ectoparasites were ranked as the most important constraint limiting goat productivity. Ticks were ranked as the most important external parasites. Amblyomma tick species were ranked as the most important amongst the tick species, followed by Rhipicephalus evertsi evertsi ticks. A significant population of farmers (80.7%) are dependent on the use of tick sprays, whereas others used injections (3.3%). Cissus quadrangularis L. (Inhlashwana) was singled out as the most used ethno-veterinary plant to control ticks with a frequency of (64%), followed by Gomphocarpus physocarpus E. Mey (Uphehlacwathi) (55.9%). The probability of keeping goats in wet rangelands (P < 0.05) was 3.04 times more likely to influence the extent of IK use compared to their contemporaries in the dry rangeland. Male farmers keeping goats (P < 0.01) were 2.95 more likely to influence the extent of use of IK than females. The type of rangeland, gender, age, residing on farm and also having the herbalist in the locality were the most common factors that influenced the extent of IK utilisation.

BACKGROUND: Ticks are able to transmit important diseases to humans, including Rocky Mountain spotted fever, Q fever, Crimean-Congo haemorrhagic fever, summer Russian encephalitis, and relapsing fever. AIMS: To determine the repellency effect of 1% flumethrin pour-on formulation against hard ticks. METHODS: The concentration of flumethrin pour-on formulation was 1 mg/10 kg body weight and was administered on the dorsal midline from the head to the base of the tail. The livestock included cows, goats, oxen and sheep in 2 villages in Ardabil Province, Islamic Republic of Iran. RESULTS: We studied 200 livestock comprising 5 age groups (< 2, 3-4, 5-6, 7-8 and >8 years). The main hard ticks identified were Hyalomma species (62.5%) and Rhipicephalus bursa (37.5%). In the treatment village, the maximum number of ticks per animal was 11.6 in oxen, 9.5 in sheep, 8.9 in goats and 8.6 in cattle. The repellency effect of flumethrin remained for 2 months. CONCLUSIONS: Flumethrin provided 2 months protection against hard ticks. Therefore, it could be used in the livestock industry. Control of ticks is important for prevention of disease transmission.

RESUMO

Acaricide resistance is a global problem that has impacts worldwide. Tick populations with broad resistance to all commercially available acaricides have been reported. Since resistance selection in ticks and their role in pathogen transmission to animals and humans result in important economic and public health burden, it is essential to develop new strategies for their control (i.e., novel chemical compounds, vaccines, biological control). The synganglion is the tick central nervous system and it is responsible for synthesizing and releasing signaling molecules with different physiological functions. Synganglion proteins are the targets of the majority of available acaricides. In this review we provide an overview of the mode-of-action and resistance mechanisms against neurotoxic acaricides in ticks, as well as putative target sites in synganglion, as a supporting tool to identify new target proteins and to develop new strategies for tick control.

The exotic southern cattle fever tick, Rhipicephalus (Boophilus) microplus (Canestrini) (Ixodida: Ixodidae), since its eradication from the United States in 1943, made a strong incursion into Texas, beginning 2016. The pest is arguably the most economically detrimental ectoparasite of cattle, Bos taurus L., worldwide. Current R. (B.) microplus control mostly relies on conventional synthetic acaricides to which the ixodid has been developing resistance. Our study demonstrates that commercially available desiccant dust products, with and without bioactive botanical additives, are strongly lethal, when applied dry, against larval R. (B.) microplus in the laboratory, and after being released on dust-treated cattle. Deadzone (renamed Celite 610, a diatomaceous earth product), Drione (silica gel + pyrethrins + piperonyl butoxide synergist), and EcoVia (silica gel + thyme oil), each prophylactically prevented larval R. (B.) microplus from attaching to and feeding on stanchioned calves. Desiccant dust-based products are less likely than conventional synthetic acaricides to decline in terms of efficacy as a result of ixodid resistance, and other desiccant dust advantages, including extended residual, flexibility in terms of application methods, environmental, animal, and human safety, and possible compatibility with organic, or 'green', production systems, are discussed. We anticipate that the desiccant dusts we evaluated, and others not included in this study (e.g., kaolin, perlite, and silica gel) will be effective when used with other control tactics in integrated pest management approaches for controlling R. (B.) microplus (and other ixodid species).

Ticks have medical and economic importance due to their ability to transmit pathogens to humans and animals. In tropical and sub-tropical countries, tick-borne diseases (TBD) are among the most important diseases affecting livestock and humans. The fast spread of ticks and TBD requires a quick development and application of efficient prevention and/or control programs. Therefore, prior investigations on TBD and related vectors epidemiology, for instance, through accurate epidemiological models, are mandatory. This study aims to develop models to forecast suitable habitat for Rhipicephalus microplus distribution in West Africa. Tick occurrences were assembled from 10 different studies carried out in six West African countries in the past decade. Six statistical models (maximum entropy in a single model and generalised linear model, generalised additive model, random forest, boosted regression tree and support vector machine model in an ensemble model) were applied and compared to predict the habitat suitability of R. microplus distribution in West Africa. Each model was evaluated with the area under the receiver operating characteristic curve (AUC), the true skill statistic (TSS) and the Boyce index (BI). The selected models had good performance according to their AUC (above .8), TSS (above .7) and BI (above .8). Temperature played a key role in MaxEnt model, whereas normalised difference vegetation index (NDVI) was the most important variable in the ensemble model. The model predictions showed coastal countries of West Africa as more suitable for R. microplus. However, some Sahelian areas seems also favourable. We stress the importance of vector surveillance and control in countries that have not yet detected R. microplus but are in the areas predicted to host suitable habitat. Indeed, awareness-raising and training of different stakeholders must be reinforced for better prevention and control of this tick in these different countries according to their status.

This study aimed to evaluate, by means of artificial feeding, the interaction between a pathogenic rickettsia and the hard tick R. microplus. We used partially engorged females fed on calves free of Rickettsia spp. Group 1 (G1), containing 20 ticks, was fed bovine blood only. Group 2 (G2), containing 20 ticks, was fed blood containing uninfected VERO cells, and group 3 (G3), containing 40 ticks, was fed blood containing VERO cells infected with Rickettsia parkeri. Biological parameters of the non-parasitic phase and a possible bacterial transmission to the tick eggs and to guinea pigs were evaluated. At the end of oviposition, all G3 females were PCR-positive for genes specific for the genus Rickettsia. Although no guinea pigs were infected, the experimental infection of R. microplus by R. parkeri caused a deleterious effect on the oviposition and provided the first report of transovarian transmission of rickettsia in this tick...

As blood-sucking parasites, ticks inflict great damage to animals and humans in many parts of the world. The continued use of chemical acaricides is not sustainable due to increasing tick resistance, growing public concern over drug residues in food and in the environment, and the high cost of developing new acaricides. Therefore, an alternative control strategy is urgently needed. Vaccines against ticks have been shown to be functionally feasible, as highlighted by the success of Bm86 vaccines against Rhipicephalus (Boophilus) microplus and closely related tick species. However, a limited number of tick antigens with cross-protective epitopes have been characterized so far, limiting widespread deployment of the available vaccines, including those derived from Bm86. Therefore, identifying tick antigens with potential broad-spectrum protection against multiple tick species is subject of vigorous research at present. In this paper, progress towards effective anti-tick vaccines is reviewed in the light of emerging data from studies including heterologous tick challenge. Taken together, these studies indicate that the decades-long search for a universal tick vaccine is making progress, with such a vaccine likely to be based on multiple cross-reactive antigens.

Chimeric yellow fever 17D/DENV-1-4 viruses (CYD-1-4) have been developed as a tetravalent dengue vaccine candidate which is currently being evaluated in efficacy trials in Asia and America. While YF 17D and DENV are mosquito-borne flaviviruses, it has been shown that CYD-1-4 do not replicate after oral infection in mosquitoes and are not transmitted to new hosts. To further document the risk of environmental dissemination of these viruses, we evaluated the replication of CYD-1-4 in ticks, the vector of tick-borne encephalitis virus (TBEV), another member of the flavivirus family. Females of two hard tick species, Ixodes ricinus and Rhipicephalus appendiculatus, were inoculated intracoelomically with CYD-1-4 viruses and parent viruses (DENV-1-4 and YF 17D). Virus persistence and replication was assessed 2, 16, and 44 days post-inoculation by plaque titration and qRT-PCR. CYD-1-4 viruses were detected in I. ricinus ticks at early time points post-inoculation, but with infectious titers at least 100-fold lower than those observed in TBEV-infected ticks. Unlike TBEV, complete viral clearance occurred by day 44 in most ticks except for CYD-2, which had a tendency to decline. In addition, while about 70% of TBEV-infected I. ricinus nymphs acquired infection by co-feeding with infected tick females on non-viremic hosts, no co-feeding transmission of CYD-2 virus was detected. Based on these results, we conclude that the risk of dissemination of the candidate vaccine viruses by tick bite is highly unlikely.

Sand flies are recognized as the major vector of canine visceral leishmaniasis. However, in some areas of Brazil where sand flies do not occur, this disease is found in humans and dogs. There has been speculation that ticks might play a role in transmission of canine visceral leishmaniasis and the DNA of Leishmania spp. has been reported in whole ticks. We investigated the presence of Leishmania spp. promastigotes in the intestines, ovaries, and salivary glands of Rhipicephalus sanguineus ticks collected from tick-infested dogs in two cities of Brazil. We used 66 dogs that tested positive and 33 that tested negative for Leishmania spp. according to direct cytological examination assays. Ten ticks were collected from each dog and dissected to collect the intestines, ovaries, and salivary glands for immunohistochemistry (IHC) and diagnostic real-time polymerase chain reaction (RT-PCR). IHC results showed Leishmania spp. in 98, 14, and 8 % of the intestines, ovaries, and salivary glands, respectively. Real-time PCR showed that 89, 41, and 33 % of the tick intestine, ovary, and salivary glands, respectively, were positive for Leishmania spp. The verification of promastigotes of Leishmania spp. by two independent techniques in ticks collected from these urban region dogs showed that there is need for clarification of the role of ticks in the transmission of canine visceral leishmaniasis in Brazil.
