RESUMEN

The aim of this study was to scrutinize the possible mitigating role of leaves' Allium triquetrum L. against the toxicity of lead acetate on liver and kidney markers of Wistar rat. Lead acetate (Pb) and leaves' aqueous extracts (L) were orally administrated for 3 weeks. Rats were divided into the control, Pb group (500 mg/kg body weight/day), positive controls L (2g, 3g, 4g/kg BW/day), along with three combined groups of the same doses (Pb-L1, Pb-L2, Pb-L3). The levels of plasma aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), total proteins (TP), albumin (ALB), urea, creatinine (Cr), and uric acid (UA), as well as the hepatic and the renal malondialdehyde (MDA), glutathione (GSH), and glutathione peroxidase (GPx), were estimated. Results exhibited a significant increase in plasma AST, ALT, ALP, urea, creatinine, uric acid, and MDA levels of the Pb group compared to the control, with the exception of TP, ALB, GSH levels, and GPx activities that were significantly diminished, though the co-administration of garlic extracts (Pb-L) revealed a significant decrease in all mentioned markers, excluding the TP, ALB, GSH, and GPx levels. Likewise, Pb caused histological injuries in the hepatic and renal tissues of rats, while the co-administration of leaves' wild garlic has reduced such effect. Thought, the Pb-L has attenuated the Pb-induced toxicity in a dose-dependent manner. In conclusion, the aqueous extracts of A. triquetrum have the potential to alleviate Pb hepatotoxicity and nephrotoxicity through the modulation of most biomarkers in Wistar rat.

The current study investigates the potential alleviating activity of bulbs (B) and leaves (L) of Allium triquetrum aqueous extract (ATE) on repro-toxicity induced by lead acetate (Pb) in male Wistar rats administrated orally for 3 consecutive weeks. Eighteen groups of rats were divided into the control, Pb (500 mg/kg body weight/day), positive controls of B and L (2 g, 3 g, 4 g, 6 g/kg body weight/day), in addition to four mixtures of each of Pb-B (Pb-B1, Pb-B2, Pb-B3, Pb-B4) and Pb-L (Pb-L1, Pb-L2, Pb-L3, Pb-L4). The two extracts were subjected to phytochemical screening and HPLC analysis. Sperm characteristics were evaluated by CASA system, as well as the serum testosterone, testicular and epididymal levels of glutathione (GSH), glutathione peroxidase (GPx), and malondialdehyde (MDA). The phytochemical screening proved that bulbs' and leaves' extracts were rich in various compounds and the HPLC showed that leaves contain more tannins. Results revealed a significant decrease in the testicular and in the epididymal weights, sperm concentration, motility, testosterone, velocity, vitality, round cells, GSH, and GPx levels in the Pb-intoxicated rats compared to the control, with the exception of MDA concentration that was significantly increased. However, the co-administration of garlic extracts (Pb-B and Pb-L) exhibited a significant increase in all mentioned markers, except for the MDA level which was reduced. Likewise, Pb caused histological injuries in the testicular seminiferous of rats, while the co-administration of wild garlic has reduced such effect, especially in the higher doses. Both extracts of Pb-B and Pb-L have attenuated Pb toxicity in a dose-dependent manner. In conclusion, aqueous extracts of A. triquetrum have the potential to reduce Pb testicular injuries by boosting sperm characteristics and ameliorating oxidative stress markers.

Abstract This study investigates the toxic effects of ethanol (Eth) on the reproductive system of male rats and the possible protective role of Silybum marianum seeds-infused solution (SMI) over six consecutive weeks of administration. Animals were divided into the following groups: control, SMI positive control (200 mg/kg/day), Eth1 (1 g/kg/day), Eth2 (2 g/kg/day), Eth1+SMI, and Eth2+SMI. Plasma testosterone concentration, epididymal spermatozoa biology, and testicular and epididymal MDA, GSH and GPx levels were evaluated. The results indicated a significant decrease in testis and epididymis weight, testosterone level, sperm concentration, sperm vitality and sperm motility (total motility, progressive motility, curvilinear velocity, straight-line velocity, velocity average path, beat cross frequency, and lateral head displacement) in both Eth1 and Eth2 compared to the control groups and the combined-treatment groups (Eth1+SMI and Eth2+SMI). Furthermore, results showed a significant elevation in MDA concentration with a significant decrease of testicular and epididymal GSH concentration and GPx activity in theEth1 and Eth2 groups compared to the combined-treatment groups. The administration of SMI succeeded in improving the parameters cited above in the combined-treatment groups compared to the Eth1 and Eth2 groups, and bring them to the levels seen in the control groups. To conclude, SMI has clearly protected reproductive indices against ethanol-induced reprotoxicity in male rats

BACKGROUND: Nitrate (NO3) is the most common chemical contaminant in the world's ground water aquifer. Oxidative stress has been proposed as a possible mechanism involved in NO3 toxicity on non-target organism. OBJECTIVES: The current study aimed to elucidate the potential protective effect of Telfairia occidentalis (pumpkin seed oil, PSO) against hepatotoxicity induced by sodium nitrate. METHODS: Wistar rats were exposed either to NaNO3 (200 mg/kg bw) in drinking water in drinking water, or to 4ml PSO/kg bw by gavage or to their combination. Oxidative stress parameters, biochemical biomarkers and liver histopathological examination were determined. RESULTS: Our data showed that the exposure of rats to NaNO3 caused significant changes of some haematological parameters compared to the control. In addition, there was a significant elevation of the levels of biochemical markers as that of aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase and lactate dehydrogenase when compared with the control. Furthermore, exposure of rats to NaNO3 induced liver oxidative stress as indicated by the increase of malondialdehyde, progressive oxidation of protein products and protein carbonyl levels. In addition, a reduction in anti-oxidant status (catalase, glutathione peroxidase, glutathione-S-transferase and superoxide dismutase, reduced glutathione and vitamin C) was observed. CONCLUSION: Co-administration of PSO to the NaNO3 restored most parameters cited above to near-normal values. Therefore, the present investigation revealed the ability of PSO to attenuate NaNO3-induced oxidative damage.

AIM: This study aimed to compare the physicochemical, the biochemical, and the antioxidant characteristics of unifloral bitter honey and polyfloral sweet honey. MATERIALS AND METHODS: Unifloral bitter and polyfloral sweet honey samples were, respectively, harvested in January and July, and then, they were extracted by the traditional method. The markers of refractive index, pH, free acidity, Brix index, density, reducing sugars, total polyphenols, flavonoids, tannins, 2,2-diphenyl-1-picrylhydrazyl (DPPH), and ferric reducing/antioxidant power (FRAP) were evaluated. RESULTS: The obtained results showed that the physicochemical parameters are within the normal ranges, in which they meet the international standards (Codex Alimentarius). For biochemical constituents, matching concentrations of reducing sugars (glucose+fructose) were observed in both samples, while that of sucrose were very low in unifloral than polyfloral honey. The levels of the active ingredients showed a difference in total polyphenols and tannins of the two types of honey studied, whereas that of flavonoids were almost similar. The antioxidant activity of various samples evaluated by DPPH and FRAP indicated that unifloral has a superior activity. CONCLUSION: Compared to polyfloral honey, unifloral bitter honey has lower sucrose, high total polyphenols, and tannins levels, in addition to higher antioxidant potential.

The aim of this work is to assess biomarker responses to stresses caused by pollution of the marine environment in the Gulf of Annaba, using the bivalve mollusc Donax trunculus and Mytilus galloprovincialis. Males and females were collected during the summer from site 1 considered to be far from pollution sources (SM1 and SD1), site 2 (SM2 and SD2) exposed to a mixture of contaminants, and site 3 (SM3 and SD3) receiving the untreated sewage. The whole body was used for the determination of malonaldehyde (MDA), reduced glutathione (GSH) and DNA damage. The MDA concentration from site 2 of both species has indicated significant augmentation. The two bivalves from S2 and S3 have recorded significant decrease of GSH level, with the highest number of Comet scores. A significant site and sex effect was measured for the level of GSH and MDA, accompanied with a significant site effect for DNA damage. The genotoxicity seems correlated with the stress biomarkers and dependent on the effluent types.

Bisphenol A, an everywhere chemical, is applied as a plasticizer in polycarbonate plastics, which often used in our everyday products and in epoxy resins as protective coatings and linings for food and beverage cans for decades. Human exposure to BPA may lead to adverse effects by interfering with oestrogen receptors. Our present study was conducted to investigate the protective effects of selenium (Se) and vitamin E (Vit E) on BPA-induced damage in the liver of male rats. Animals were randomly divided into four groups: the first group received olive oil and served as control. The second group received both (Se + Vit E) (0.5 mg/kg diet; 100 mg/kg of diet). The third one treated orally by (10 mg/kg b.w.) of BPA. The last group received (Se + Vit E) (0.5 mg/kg diet; 100 mg/kg of diet) concomitantly with (10 mg/kg b.w.) BPA. Exposure to BPA for three weeks engendered a hepatic disorder. An increased AST and ALT enzymatic activity was noticed in BPA-treated group as compared to other groups. Furthermore, a change in glucose, cholesterol, LDL-C, HDL-C, albumin, and bilirubin level was remarkable. Moreover, exposure to BPA increased malondialdehyde levels while reduced gluthatione content was decreased in the liver homogenate. A decrease in glutathione peroxidase, glutathione s-transferase and catalase activities was observed in the same group. Administration of selenium and vitamin E through the diet in BPA treated rats ameliorated the biochemical parameters cited above. In addition, an improvement in activities of liver enzymes was recorded. The histological findings confirmed the biochemical results. The model of this study that we employed characterized the relationships between BPA-induced hepatotoxicity and its alleviation by natural antioxidants like selenium and vitamin E.

The objective of this study was to investigate the protective effects of pomegranate (Punica granatum) juice (PGJ) on oxidative damages in liver tissue and erythrocytes of rats intoxicated by sodium fluoride (NaF). Rats were randomly divided into two groups: group I received standard diet and group II received orally 1 mL of PGJ. After 5 weeks of pretreatment, each group was divided again into two subgroups and treated for another 3 weeks as follows: group I was subdivided into a control group and a group that was treated with 100 ppm of NaF (in drinking water); group II was subdivided into one group that was treated daily with both 100 ppm NaF and PGJ (1 mL orally) and one that received daily 1 mL of pomegranate juice. Exposure to NaF decreased hematological parameters, changed the total protein, albumin, bilirubin levels, and increased the activities of hepatic marker enzymes. We also noted an increase in lipid peroxidation contents, accompanied by a decrease of reduced glutathione levels. Antioxidant enzyme activities in both tissues were modified in the NaF group compared with the control group. However, the administration of PGJ juice caused an amelioration of the previous parameters. Our results indicated the potential effects of NaF to induce oxidative damage in tissues and the ability of PGJ to attenuate NaF-induced oxidative injury.

CONTEXT: Pistacia lentiscus L. (Anacardiaceae) is an evergreen shrub widely distributed throughout the Mediterranean region. Pistacia lentiscus oil (PLo) was particularly known in North African traditional medicine. Thus, people of these regions have used it externally to treat sore throats, burns and wounds, as well as they employed it internally for respiratory allergies. PLo is rich in essential fatty acids, vitamin E and polyphenols. As a very active site of metabolism, liver is reported to be susceptible to arsenic (As) intoxication. OBJECTIVE: The present study evaluates the protective effect of PLo against sodium arsenite-induced hepatic dysfunction and oxidative stress in experimental Wistar rats. MATERIALS AND METHODS: Twenty-eight rats were equally divided into four groups; the first served as a control, the remaining groups were respectively treated with PLo (3.3 mL/kg body weight), sodium arsenite (5.55 mg/kg body weight) and a combination of sodium arsenite and PLo. After 21 consecutive days, cellular functions were evaluated by hematological, biochemical and oxidative stress markers. RESULTS: A significant decrease in the levels of red blood cells, haemoglobin (p ≤ 0.001), hematocrit (p ≤ 0.001), reduced glutathione and metallothionein (p ≤ 0.05) associated with a significant increase of malondialdehyde (p ≤ 0.001) were noticed in the arsenic-exposed group when compared to the control. The As-treated group also exhibited an increase in hepatic antioxidant enzymes namely superoxide dismutase, glutathione peroxidase (p ≤ 0.01) and catalase (p ≤ 0.05). However, the co-administration of PLo has relatively reduced arsenic effect. CONCLUSION: The results showed that arsenic intoxication disturbed the liver pro-oxidant/antioxidant status. PLo co-administration mitigates arsenic-induced oxidative damage in rat.

AIM: The objective of this study was to investigate the possible protective role of nettle Urtica dioica (UD) against Hg-induced toxicity. MATERIALS AND METHODS: A total of 28 rats were equally divided into four groups: the control, the Hg (0.8 g HgCl2/kg in the diet), the UD (1.5 ml UD/rat by gavage), and the Hg+UD group. HgCl2 was daily dissolved in distilled water and immediately mixed with the standard diet. A solution of daily infused fresh nettle leaves in boiling water (16 g in 25 ml) was obtained and then it was administrated by gavage. Biochemical and reproductive markers, in addition to glutathione (GSH) level (liver, kidney and testis) and the histological profiles (testis and epididymis) were evaluated after 1 month exposure. RESULTS: Compared to the control, the levels of glucose, triglycerides, urea, creatinine, alanine aminotransferase (ALT), aspartate aminotransferase (AST), and alkaline phosphatase (ALP) were significantly raised in the Hg group. In the latter group, the concentrations of Mg, Fe, and Ca were significantly decreased. Besides, Hg+UD group has only showed raised AST activity and reduced Mg level. Concerning the fertility markers, Hg has provoked a significant decrease in the spermatozoa's concentration and motility and in plasma testosterone level as well. Furthermore, hepatic, renal and testicular GSH concentrations have declined significantly in the Hg treated rat compared to the control. A remarkable enhancement of the GSH level was observed in all organs of the UD group. The histological examinations of the Hg group have revealed marked testicular degeneration of the most seminiferous tubules, and showed few sperms in the lumen of epididymis ducts. However, the Hg+UD rats have demonstrated an improved histological structure with the presence of important numbers of sperms in the lumen. In addition, a clear stabilization of organized seminiferous tubules and an increased sperms' numbers were noted in the UD supplemented rats. CONCLUSION: Nettle leaves have not only played a clear protective role during Hg intoxication, but it also enhanced hepatic, renal and testicular GSH level of Wistar rats.

The present study was planned to investigate the protective effects of Nigella sativa oil (NSO) supplementation against aluminium chloride (AlCl3)-induced oxidative damage in liver and erythrocytes of rats. Simultaneously, a preliminary phytochemical study was affected in order to characterize the bioactive components containing in the NSO using chemical assays. The antioxidant capacities of NSO were evaluated by DPPH assay. The results showed that NSO was found to contain large amounts of total phenolics, flavonoids and tannins. Twenty-four rats were equally divided into two groups, in which group A received standard diet, whereas group B treated daily with an oral gavage dose of 2 ml NSO/kg body weight. After 5 weeks pretreatment, both groups were divided again into two subgroups (A and B) of six animals each and treated for other 3 weeks. Therefore, subgroup A1 was served as a control which received standard diet, but subgroup A2 received AlCl3 (34 mg/kg bw mixed with food). Subgroup B1 received both AlCl3 and NSO; however, subgroup B2 received NSO only. Results showed that AlCl3 exhibited an increase in white blood cell counts and a marked decrease in erythrocyte counts and haemoglobin content. Plasma aspartate transaminase, alanine transaminase, alkaline phosphatase and lactate dehydrogenase activities and total bilirubin concentration were higher in AlCl3 group than those of the control, while albumin and total protein concentration were significantly lower. Compared to the control, a significant raise of hepatic and erythrocyte malondialdehyde level associated with a decrease in reduced glutathione content, glutathione peroxidase, superoxide dismutase and catalase, activities of AlCl3 treated rats. However, the administration of NSO alone or combined with AlCl3 has improved the status of all parameters studied. It can be concluded that AlCl3 has induced the oxidative stress, altered the biochemical parameters and the hepatic histological profile, but the supplementation of NSO has alleviated such toxicity.

The aim of this study was to evaluate the protective effects of vitamin E and/or curcumin against diazinon (DZN) (an organophosphorus insecticide)-induced toxicity of blood, liver and erythrocyte markers of male Wistar rats. The exposure of rats to DZN for 21 days provoked significant changes in red blood cell counts and hemoglobin. Results showed that lipid peroxidation increased significantly in DZN-treated rats, as evidenced by high liver and erythrocyte thiobarbituric acid reactive substance levels. Alteration of the antioxidant system in DZN-treated rats was confirmed by the significant decrease in the activity of catalase, glutathione peroxidase and glutathione-S-transferase, accompanied by a decline in reduced glutathione content in both tissues. On the other hand, a significant increase in the activities of plasma aspartate transaminase, alanine transaminase, lactate dehydrogenase and alkaline phosphatase was observed in the rats treated with DZN. However, the administration of vitamin E and curcumin has ameliorated the previous markers. In conclusion, our results indicate that the natural antioxidants like vitamin E and curcumin can effectively lower the erythrocytes and hepatic injuries induced by DZN as monitored by lipid peroxides, antioxidant enzyme activities and sensitive serum enzyme levels.

Males were randomly divided into four groups where the first was served as a control, whereas the remaining groups were respectively treated with sodium selenite (3 mg/kg b.w.), sodium arsenite (5.55 mg/kg b.w.) and a combination of sodium arsenite and sodium selenite. Changes in liver enzyme activities, thiobarbituric acid reactive substances (TBARS) level, antioxidants and reduced glutathione (GSH) contents were determined after 3 weeks experimental period. Exposure of rats to As caused a significant increase in liver TBARS compared to control, but the co-administration of Se was effective in reducing its level. The activities of glutathione peroxidase (GPx) and glutathione-S-transferase (GST) of As-treated group were found lower compared to the control and the Se-treated group. The co-administration of Se had an additive protective effect on liver enzyme activities compared to As-treated animals. On the other hand, a significant increase in plasmatic activities of AST, ALT and ALP was observed in As-treated group. The latter was also exhibited a decrease in body weight and an increase in liver weight compared to the control. The co-administration of Se has decreased the activities of AST, AST and ALP and improved the antioxidant status as well. Liver histological studies have confirmed the changes observed in biochemical parameters and proved the beneficial role of Se. To conclude, results suggest that As exposure enhanced an oxidative stress by disturbing the tissue antioxidant defense system, but the Se co-administration protected liver tissues against As intoxication probably owing to its antioxidant properties.

The purpose of this study was to evaluate the effects of dysthyroidism on lipid peroxidation, antioxidants status, liver, and serum dysfunction parameters in the hypo-/hyperthyroidism-induced rats. Hypothyroidism and hyperthyroidism conditions were induced for 5 weeks by administration of 0.05% benzythiouracile (BTU) and l-thyroxine sodium salt (0.0012%), in drinking water, respectively. The enzymatic activities of glutathione peroxidase (GPx), superoxide dismutase (SOD), catalase (CAT) and the lipid peroxidation product; thiobarbituric acid reacting substances (TBARS) were measured in liver as indicators of oxidative damage. However, liver dysfunction parameters represented by the activities of aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), and gamma glutamyl transferase (GGT), were measured in serum. In hyperthyroidism rats, the TBARS contents of liver have significantly increased compared to those in hypothyroid rats and the controls (p<0.001), associated with a fall of the total antioxidant status (TAS) in the serum of the hyperthyroid rats. The SOD, CAT, and GPx activities in liver of hyperthyroid rats have significantly increased compared to hypothyroid rats and the controls (p<0.001). The AST, ALT, LDH, GGT, and ALP activities increased in the hyperthyroidism rats (p<0.05). We conclude that thyroid dysfunction induces oxidative stress and modifies some biochemical parameters of liver. Our results show the occurrence of a state of oxidizing stress in relation to hyperthyroidism.

The effect of the thyroid activity on the formation of lipid peroxidation and on liver and heart antioxidant enzyme activities was investigated in Wistar rats. Hypothyroidism and hyperthyroidism conditions were induced for five weeks by the administration of 0.05% benzythiouracile (BTU) and L-thyroxine sodium salt (0.0012%), in drinking water, respectively. No significant effect was observed on the rates of both lipid peroxidation and the vitamin E in hepatic and cardiac tissues of hypothyroidism rats compared to the controls, contrary to the hyperthyroidism rats, which expressed a pronounced increase. The increased glutathione peroxidase activity in rats suffering from hyperthyroidism was associated with a fall of the reduced glutathione in the homogenate and a marked increase in the glutathione reductase activity. An increase in superoxide dismutase and catalase activities was also recorded in hyperthyroidism. Our results explain the thyroid activity variation in relation to the lipid peroxidation and the tissular contents of the enzymatic and the non-enzymatic antioxidants. To conclude, our results show the occurrence of a state of oxidizing stress in relation to hyperthyroidism.
