RESUMEN

Durio kutejensis, commonly known as Lai durian, has a unique characteristics of a creamy texture and a combination of sweet and bitter tastes. This study aimed to isolate and screen yeast from fruits as a potential probiotic agent. The tests consisted of tolerance to bile salt and gastric acid at pH 2, antibacterial activity against Listeria monocytogenes and Salmonella enteriditis, and hemolytic activity on blood agar medium. The results showed that 40 yeasts isolated from Lai durian fruit and 34 of these isolates grew on YMA medium. The two isolates showed high significance in the probiotic tests. These two isolates were able to grow on bile salt up to a concentration of 2% and gastric acid for up to 6 h, with survival rates of 99.06% and 100%, respectively. Two isolates were identified as Pichia kudriavzevii UNJCC Y-137 and Candida tropicalis UN-JCC Y-140 Therefore, these two yeast isolates can be used as potential probiotic agents.

This study aims to determine the eco-friendliness of microalgae-based renewable energy production in several scenarios based on life cycle assessment (LCA). The LCA provides critical data for sustainable decision-making and energy requirement analysis, including net energy ratio (NER) and cumulative energy demand (CED). The Centrum voor Milieuwetenschappen Leiden (CML) IA-Baseline was used on environmental impact assessment method by SimaPro v9.3.0.3® software and energy analysis of biofuel production using native polyculture microalgae biomass in municipal wastewater treatment plants (WWTP) Bojongsoang, Bandung, Indonesia. The study was analyzed under three scenarios: (1) the current scenario; (2) the algae scenario without waste heat and carbon dioxide (CO2); and (3) the algae scenario with waste heat and carbon dioxide (CO2). Waste heat and CO2 were obtained from an industrial zone near the WWTP. The results disclosed that the microalgae scenario with waste heat and CO2 utilization is the most promising scenario with the lowest environmental impact (- 0.139 kg CO2eq/MJ), positive energy balance of 1.23 MJ/m3 wastewater (NER > 1), and lower CED value across various impact categories. It indicates that utilizing the waste heat and CO2 has a positive impact on energy efficiency. Based on the environmental impact, NER and CED values, this study suggests that the microalgae scenario with waste heat and CO2 is more feasible and sustainable to adopt and could be implemented at the Bojongsoang WWTP.

BACKGROUND: Apples often experience postharvest damage due to being attacked by mold organisms. Several groups of molds such as Aspergillus sp., Penicilium expansum, Botrytis cinerea, and Venturia sp. can cause a serious postharvest disease exhibited as watery regions where areas of blue-green tufts of spores develop. Current methods using fungicides to control pathogenic fungi can cause resistance if applied in the long term. An alternative procedure using yeast as a biological agent has been found. OBJECTIVE: The aim of this study is to screen potential yeast, which has the ability to inhibit the growth of Aspergillus brasielensis (isolate A1) and Aspergillus flavus section flavi (isolate A17) isolated from apple fruits. METHODS: Antagonism test using YMA dual culture medium using in vitro assays and ITS rDNA identification were performed. RESULTS: The result showed that 3 out of 19 yeast isolated from Cerbera manghas L, T1, T3 and T4, demonstrated the potential ability as a biocontrol agent. ITS rDNA identification demonstrated that T1 has a similarity to Rhodotorula mucilaginosa while T3 and T4 were identified as Aureobasidium sp. nov. The 3 isolates exhibited the ability to reduce the growth of A. brasiliensis sensu lato better than dithane 0.3% with a Disease Incidence (DI) of 100% and a Disease Severity (DS) value of 45%. Only isolate T1 and T3 were able to reduce decay symptoms in apples inoculated with A. flavus sensu lato (with DO and DS were 100% and 25%, respectively) compared to dithane pesticides 0.3%. CONCLUSION: This study indicated that competition between nutrients occurs between pathogenic molds and under-yeast in vitro and in vivo conditions. However, further studies in the future might be able to elucidate the 'killer' activity and interaction with the pathogen cells and the bio-product production using Rhodotorula mucilaginosa and Aureoubasidium namibiae strains to control postharvest diseases.
