RESUMEN

Multiple myeloma (MM) is still an incurable hematologic malignancy, which is eagerly to the discovery of novel therapeutic targets and methods. N-acetyltransferase 10 (NAT10) is the first reported regulator of mRNA acetylation that is activated in many cancers. However, the function of NAT10 in MM remains unclear. We found significant upregulation of NAT10 in MM patients compared to normal plasma cells, which was also highly correlated with MM poor outcome. Further enforced NAT10 expression promoted MM growth in vitro and in vivo, while knockdown of NAT10 reversed those effects. The correlation analysis of acetylated RNA immunoprecipitation sequencing (acRIP-seq) and ribosome profiling sequencing (Ribo-seq) combined with RIP-PCR tests identified centrosomal protein 170 (CEP170) as an important downstream target of NAT10. Interfering CEP170 expression in NAT10-OE cells attenuated the acceleration of cellular growth caused by elevated NAT10. Moreover, CEP170 overexpression promoted cellular proliferation and chromosomal instability (CIN) in MM. Intriguingly, remodelin, a selective NAT10 inhibitor, suppressed MM cellular growth, induced cellular apoptosis in vitro and prolonged the survival of 5TMM3VT mice in vivo. Collectively, our data indicate that NAT10 acetylates CEP1 70 mRNA to enhance CEP170 translation efficiency, which suggests that NAT10 may serve as a promising therapeutic target in MM.

With three grape varieties, Red Barbara, Summer Black and Hutai No. 8 as test mate-rials, we investigated the effects of foliar spraying of organic selenium fertilizer on greenhouse grape quality and selenium content. The results showed that spraying 12 mg·L-1 amino acid chelated selenium-enriched foliar fertilizer on grape leaves significantly increased selenium content and the quantity and quality of grape yield, including the contents of soluble sugar, organic acid, soluble protein, soluble solids, vitamin C and proanthocyanidins. However, there was no increase in resveratrol. Among the three varieties, selenium content of Summer Black in 2017 and 2018 was increased by 36.7% and 37.1%, respectively, being higher than that of Red Barbara and Hutai No. 8. Red Barbara sprayed with selenium fertilizer had better quality due to high sugar and low acid contents, as well as high health-care components. Moreover, the selenium content of Hutai No. 8 in 2018 was 53.26 µg·kg-1, higher than the others, indicating a stronger ability of selenium enrichment. We concluded that the increase range of Se content was larger in Summer Black, Red Barbara showed the better nutrition value and quality, and Hutai No. 8 was a suitable variety for selenium-rich grape production.

BACKGROUND: Spraying selenium (Se) fertilizer is an effective method for Se-enriched fruit production. Sugar content in fruit is the major factor determining berry quality. However, changes in sugar metabolism in response to Se fertilizer are unclear. Hence, this study was conducted to identify the effects of Se fertilizer on sugar metabolism and related enzyme activities of grape berries. Additionally, production of leaves with and without Se fertilizer was also investigated. RESULTS: Acid invertase (AI) activity, total soluble sugar and Se content in berries, and photosynthetic rate in leaves produced under Se fertilizer treatments were higher than that of control. Glucose and fructose were the primary sugars in berries, with a trace of sucrose. In both berries and leaves, neutral invertase activity was lower than AI, there was no significant difference in neutral invertase, sucrose synthase and sucrose phosphate synthase between Se fertilizer-treated and control. In berries, AI showed a significant positive correlation with glucose and fructose; also Se content was significantly correlated with sugar content. CONCLUSION: AI played an important role in the process of sugar accumulation in berries; high AI activity in berries and photosynthetic rate in leaves could explain the mechanism by which Se fertilizer affected sugar accumulation in berries. © 2017 Society of Chemical Industry.

Through field investigation and indoor analysis, the organic carbon content and organic carbon mineralization characteristics of six kinds of natural forest soil were studied, including the pine forests, evergreen broad-leaved forest, deciduous broad-leaved forest, mixed needle leaf and Korean pine and Chinese pine forest. The results showed that the organic carbon content in the forest soil showed trends of gradual decrease with the increase of soil depth; Double exponential equation fitted well with the organic carbon mineralization process in natural forest soil, accurately reflecting the mineralization reaction characteristics of the natural forest soil. Natural forest soil in each layer had the same mineralization reaction trend, but different intensity. Among them, the reaction intensity in the 0-10 cm soil of the Korean pine forest was the highest, and the intensities of mineralization reaction in its lower layers were also significantly higher than those in the same layers of other natural forest soil; comparison of soil mineralization characteristics of the deciduous broad-leaved forest and coniferous and broad-leaved mixed forest found that the differences of litter species had a relatively strong impact on the active organic carbon content in soil, leading to different characteristics of mineralization reaction.
