RESUMEN

Parkinson's disease (PD), a chronic and progressive neurodegenerative disease, can reduce the population of dopaminergic neurons in the substantia nigra. The cause of this neuronal death remains unclear. 1-Methyl-4-phenylpyridinium ion (MPP+) is a potent neurotoxin that can destroy dopaminergic (DA) neurons and promote PD. Garcinol, a polyisoprenylated benzophenone derivative, was extracted from Garcinia indica and is an important active compound it has been used as an anticancer, antioxidant, and anti-inflammatory, agent and it can suppress reactive oxygen species (ROS) mediated cell death in a PD model. Human neuroblastoma (SH-SY5Y) cells (1 × 105 cells) were treated with MPP+ (1 mM) for 24 h to induce cellular ROS production. The formation of ROS was suppressed by pretreatment with different concentrations of garcinol (0.5 and 1.0 µM) for 3 h in SH-SY5Y cells. The present study found that MPP+ treatment increased the formation of reactive oxygen species (ROS), and the increased ROS began to promote cell death in SH-SY5Y cells. However, our natural compound garcinol effectively blocked MPP+-mediated ROS formation by activating the DJ-1/SIRT1 and PGC-1α mediated antioxidant pathway. Further findings indicate that the activated SIRT1 can also regulate p-AMPK-mediated autophagy to protect the neurons from the damage it concludes that garcinol sub-sequential regulates intracellular autophagy in this model, and the productive efficacy of garcinol was confirmed by western blot analysis and MitoSOX DCFDA and MTT assays. The results showed garcinol increased protection due to the prevention of MPP+-induced ROS and the promotion of cell survival.

Vermicomposting uses less energy and requires fewer infrastructures, and it is capable of restoring soil nutrition and carbon. Banana cultivation produces lots of trash in a single crop season, with 30 tonnes of waste generated per acre. The biodegradable fraction of banana leaf waste is thrown out in large quantities from temples, markets place wedding halls, hotels, and residential areas. Vermicomposting can be used for recovering lignin, cellulose, pectin, and hemicellulose from banana leaves. Earthworm digests organic materials with the enzymes produced in gut microflora. Biochar adds bulk to vermicomposting, increases its value as fertilizer. The goal of this study was to amend biochar (0, 2, 4 and 6%) with banana leaf waste (BLW) + cow dung (CD) in three different combinations (1:1, 2:1 and 3:1) using Eisenia fetida to produce enriched vermicompost. In the vermicompost with biochar groups, there were higher levels of physicochemical parameters, as well as macro- and micronutrient contents. The growth and reproduction of earthworms were higher in groups with biochar. A maximum of 1.82, 1.18 and 1.67% of total nitrogen, total phosphorus and total potassium was found in the final vermicompost recovered from BLW + CD (1:1) amended with 4% biochar; while the other treatments showed lower levels of nutrients. A lower C/N ratio of 18.14 was observed in BLW + CD (1:1) + 4% biochar followed by BLW + CD (1:1) + 2% biochar amendment (19.92). The FTIR and humification index studies show that degradation of organic matter has occurred in the final vermicompost and the substrates with 4% biochar in 1:1 combination showed better degradation and this combination can be used for nutrient rich vermicompost production.

Purpose: To compare the clinical efficacy of classical inferior alveolar nerve block (CIANB) and Vazirani-Akinosi (VA) injection technique in patients indicated for bilateral mandibular premolar teeth extraction for orthodontic treatment. Methods: This split-mouth comparative study was conducted on 20 patients randomly selected for bilateral extraction of mandibular premolar indicated for orthodontic treatment to receive CIANB and VA injection technique alternatively. The clinical parameters evaluated were pain during injection, onset of anesthesia, duration of anesthesia, quality of anesthesia, nerve anesthetized with single nerve block, need for re-injection and frequency of positive aspiration. Results: No statistically significant differences were observed regarding the onset of anesthesia, duration of anesthesia, quality of anesthesia, nerves anesthetized with single nerve block and need for re-injection. However, pain experienced during injection was statistically significant and was lesser in VA technique than CIANB. Positive aspiration was not found in both the injection techniques. Conclusion: VA technique showed a statistically significant difference in terms of less pain experienced during injection than CIANB. However, though not significant, VA technique was more clinically superior over the CIANB in terms of duration of anesthesia, quality of anesthesia and the need for re-injection. Also in this study, there were no complications associated with any of the injection techniques and the prevalence of positive aspiration was not found in both the techniques.

Three spotted crab (Portunus sanguinolentus) is a preferred variety of crab among seafood consumers. Biochemical, microbial and sensory characters along with formation of biogenic amines of whole crabs stored under iced and refrigerated condition was evaluated. Cadaverine, putrescine and spermidine were the major amines formed during spoilage and a strong correlation between cadaverine and psychrotrophs was observed in refrigerated (0.92) and iced storage (0.86) respectively. Histamine was at lower level and not likely to cause any toxicological problems to consumers. Although TVB-N and TMA-N increased significantly (p < 0.01), TVBN was observed as a good indicator of spoilage than TMA-N. Mesophilic and psychrotrophic bacteria crossed 7 log CFU/g on 8th day, rejection day of refrigeration, while it was below spoilage limit in iced storage. A shelf life of 6 and 10 days were observed in storage under refrigeration and icing respectively based on microbiological, biochemical and sensory attributes.

Air frying technique was used for the preparation of fish cutlet-a popular fish snack with low fat content, better protein content and color. The process conditions viz: temperature varying from 160 to 200 °C and time varying from 5 to 15 min were optimized using response surface methodology. A factorial design with 9 runs satisfying rotatability conditions under correlated errors was formulated for the experiment. The parameters of first order response surface model with interaction were estimated by generalized least square method. The optimum temperature-time combination for air-frying condition was found to be 180 °C and 12 min, respectively for low fat content, better protein content and colour, comparable texture profile and sensory acceptability when compared to deep fried fish cutlet samples. Air-fried fish cutlets can be a healthier protein rich snack product as an alternative to deep fat fried fish cutlets.

In the treatment of mid-facial fractures circum-zygomatic suspension wiring is one of the treatment modality. Earlier zygomatic awls were used to pass wire, which used to cause conspicuous trauma. In the present case we have used 16 gauge lumbar puncture needle for the suspension wiring for Lefort 1 fracture, which is inconspicuous as compared to an awl. The needle was passed in close proximity to bone to prevent soft tissue impaction between the wire and bone as it might lead to the necrosis of soft tissue, and the wire was twisted around the maxillary arch bar. The fragments were stable and occlusion was maintained. Six weeks post-operatively the bone healing was satisfactory, and the wires and arch bar were removed.

Hypertension (HTN) is one of the most prevalent chronic conditions; it can damage blood vessels and rupture blood vessels can trap in small vessels. This blockage can prevent blood flow and oxygen delivery to brain cells and can result in Alzheimer's disease (AD). HTN- and AD-mediated long-time memory loss and its treatment remain poorly understood. Plant-derived natural compounds are alternative solutions for effectively treating diseases without any side effects. This study revealed that bioactive peptides extracted from potato hydrolysis suppress HTN-mediated long-term memory (LTM) loss and cell apoptosis, thus improving memory formation and neuronal cell survival in the spontaneously hypertensive rat (SHR) rat model. SHR rats were treated with bioactive peptide IF (10 mg/kg orally) and angiotensin-converting enzyme inhibitors (5 mg/kg orally). In this study, we evaluated the molecular expression levels of BDNF-, GluR1-, and CREB-mediated markers protein expression in 24-week-old SHR rats. The study result showed that HTN-induced AD regulated long-term memory (LTM) loss and neuronal degeneration in the SHR animals. The bioactive peptide-treated animals showed an elevated level of survival proteins. Bioactive peptide IF activate CREB-mediated downstream proteins to regulate synaptic plasticity and neuronal survival in the SHR rat model.

BACKGROUND: Fish oils, which are rich in health-promoting polyunsaturated fatty acids (PUFA), have emerged as promising functional foods in the global health and wellness food market. Their source regarding the fish type, season, and location of harvesting might influence the nutritional value of such bioactive oils and determine their market price. The differences in price among such oils often lead to economically motivated mislabeling and adulteration. OBJECTIVE: In this study, our objective was to demonstrate how a qualitative targeted shotgun lipid profile workflow using an electrospray ionization-quadrupole-linear ion trap MS (QTrap) could differentiate fish oils originating from two different species. METHODS: Five samples each of sardine (Sardinella longiceps) oil and shark (Echinorhinus brucus) liver oil were diluted to a concentration of 80 µg/mL in chloroform-methanol (1 + 2, v/v) with 5 mM ammonium acetate. These samples were directly infused into a QTrap MS. The data were acquired for 23 precursor ion and 4 neutral loss scan experiments in the positive ionization mode and compared. RESULTS: We identified the following major lipid classes: cholesteryl ester, diacyl glycerol, triacylglycerol, monoalkyldiacylglycerol, and phophatydyl choline. The relative peak areas of the identified lipid species, when subjected to supervised multivariate analysis, could effectively distinguish the sardine oil and shark liver oil. CONCLUSIONS: The approach will be useful in establishing authenticity of fish oil and to support the regulatory agencies in dispute resolution. It can also be extended to establish authenticity in other agricultural and food commodities. HIGHLIGHTS: This paper reports a proof of concept for authenticating PUFA-rich fish supplements. A shotgun targeted lipidomics profile and chemometrics modeling successfully discriminated sardine oil and shark liver oil.

Metal sulfides are of great interest for future electrode materials in supercapacitor and solar cell applications owing to their superior electrochemical activity and excellent electrical conductivity. With this scope, a binary transition metal sulfide (CoS) is prepared via one-step hydrothermal synthesis. Hexagonal phase of CoS with space group of P63/mmc(194) is confirmed by XRD analysis. Additional cubic Co3S4 phase in the prepared sample originates the mixed valence state of Co (Co2+ and Co3+) is affirmed from XPS analysis. Morphological features are visualized using HRSEM images that shows nanoflower shaped star-anise structure. Employing the prepared CoS as active electrode material, interfacial charge transport kinetics is examined by EIS-Nyquist plot. The supercapacitive performances are tested in two and three-electrode system which exhibited respective specific capacitances of 57 F/g and 348 F/g for 1 A/g. Further, the fabricated asymmetric CoS//AC supercapacitor device delivers an appreciable energy density of 15.58 Wh/kg and power density of 700.12 W/kg with excellent cyclic stability of 97.9% and Coulombic efficiency of 95% over 2000 charge-discharge cycles. In addition, dye-sensitized solar cells are fabricated with CoS counter electrode and the obtained power conversion efficiency of 5.7% is comparable with standard platinum based counter electrode (6.45%). Curie-Weiss plot confirms the transition of paramagnetic nature into ferrimagnetic behavior at 85 K and Pauli-paramagnetic nature at 20 K respectively. Temperature dependent resistivity plot affirms the metallic nature of CoS sample till 20 K and transition to semiconducting nature occurs at <20 K owing to Peierl's transition effect.

Temporomandibular joint (TMJ) ankylosis in children is one of the most complex and challenging problems managed by oral and maxillofacial surgeons. TMJ ankylosis in growing children often leads to facial deformity, difficulty in chewing and swallowing, severe malocclusion, poor oral hygiene, multiple decayed teeth, and impairment of speech. A good functional and esthetic outcome can be achieved after reconstruction with the autogenous grafts. Here, we present a case of a 9-year-old patient treated with condylectomy and ipsilateral coronoidectomy, followed by reconstruction with costochondral graft with 5 years of follow-up. During this period, the reconstructed graft remodeled into a neocondyle and also regrowth of the coronoid process. The mouth opening and facial symmetry were acceptable.

This paper reports a selective and sensitive method for multiresidue determination of 119 chemical residues including pesticides and polyaromatic hydrocarbons (PAH) in high fatty fish matrix. The novel sample preparation method involved extraction of the target analytes from homogenized fish meat (5 g) in acetonitrile (15 mL, 1% acetic acid) after three-phase partitioning with hexane (2 mL) and the remaining aqueous layer. An aliquot (1.5 mL) of the acetonitrile layer was aspirated and subjected to two-stage dispersive solid phase extraction (dSPE) cleanup and the residues were finally estimated by gas chromatography mass spectrometry with selected reaction monitoring (GC-MS/MS). The co-eluted matrix components were identified on the basis of their accurate mass by GC with quadrupole time of flight MS. Addition of hexane during extraction and optimized dSPE cleanup significantly minimized the matrix effects. Recoveries at 10, 25 and 50 µg/kg were within 60-120% with associated precision, RSD<11%.

Anorectal malformations (ARMs) are one of the most common congenital anomalies dealt by surgeons. The reported incidence of ARMs range between 1:3,300 and 1:5,000 live births. These defects are invariably detected and treated in infancy or early childhood. There is a group of patients among these who have fistulous external opening from the rectum. These may not present in child hood and may continue to live with fecal incontinence till adult hood. One of such anomalies is rectovaginal fistulas which comprises of only 4% of all anomalies. Delayed management in such cases increases surgical and functional complications. Traditionally high and intermediate anorectal anomalies are treated by posterior sagittal anorectoplasty (PSARP). This involves cutting of sphincter muscles in the midline and then placement of rectum in the sphincter complex. The continence results of this operation are less than ideal. Laparoscopically assisted anorectal pull-through (LAARP) has potential advantage of precise placement of the rectum inside the sphincter complex without dividing and weakening the muscles, diminished soft tissue scarring around the rectum leading to improved rectal compliance. Three adult female patients with ARMs were managed through LAARP procedure. It involves dissection around rectum, identification and ligation of fistula tract, creation of neoanus and pull through of rectum into neoanus. Results-Continence was good in all our patients which they regained after 3 to 4 days of surgery. On follow up which ranged from 6 months to 2 years all were passing well formed stools 1-2 times a day and have symmetric anal contraction with strong squeeze on digital rectal examination. Conclusion-LAARP offers an excellent option to the patients of ARM over conventional posterior sagittal anorectal approach because if its theoretical advantages of early recovery and better continence. Long term followup is needed to substantiate these results.

Not many cases have been reported from India. We report a case of a young woman who presented with persistent back pain having large tumour of body of pancreas treated successfully by left pancreatectomy.

BACKGROUND: A surgical cut-down may be necessary when an emergency intra-aortic balloon counterpulsation catheter insertion has to be performed. Closure requires reopening the wound to extract the catheter and close the arteriotomy, or a graft has to be presutured to allow simple ligation. These methods are cumbersome. A method permitting rapid surgical access and easy removal in an intensive care unit is highly desirable. Such an insertion technique that, after insertion, includes placement of transcutaneous hemostatic purse-string sutures was evaluated. METHOD: All intra-aortic balloon catheter insertions done between August 1996 and March 2000, where all patients underwent an attempt at percutaneous intra-aortic balloon insertion, were evaluated. Failure to insert the balloon percutaneously was followed by direct surgical exposure and insertion with the placement of 2 transcutaneous purse-string sutures. The balloon pump catheter was removed when deemed appropriate and hemostasis was achieved by compression (percutaneous), or by tying of the preplaced sutures followed by compression. Patients were followed and both groups were compared with respect to mortality and vascular complications and any other complications were noted. RESULTS: There were 157 intra-aortic balloon insertions. Surgical introduction was required in 9 (5.7%) cases. These represent 17.3% of all emergency introductions. There was no statistical difference in original diagnosis, mortality, or vascular complications. The survivors (8/9) have been followed up for a mean of 2.8 years with no late complications. CONCLUSIONS: The removable transcutaneous suture method permits easy intra-aortic balloon catheter insertion and easy removal in an intensive care unit. This method has not been associated with complications in the follow-up period (mean 2.8 years).

Motor evoked potentials were recorded in 25 patients with ischaemic stroke within 72 hours of onset by transcranial magnetic stimulation. Motor power was graded according to MRC grading and the disability by Ranking scale. The patients could be grouped into those with normal MEPs (Group I) - 5 (20 per cent), those with prolonged CMCT and/or abnormal MEP/CMAP ratios or prolonged interside difference in CMCT on the hemiparetic side (Group II) - 12 (48 per cent) and those with no elicitable cortical MEPs on the hemiparetic side (Group III) - 8 (32 per cent). Patients with greater weakness and disability tended to have more abnormal or absent MEPs. No definite correlation could be seen with the site of infarct and MEP abnormalities. Patients with no elicitable cortical MEPs (Group III) showed no significant improvement in motor deficit at six months follow up. Where as patients in whom normal or abnormal cortical MEPs could be elicited (Group I & II) showed significant improvement. Evaluation of MEPs by transcranial magnetic stimulation early in acute ischaemic stroke can be used to predict stroke outcome.
