This study compared the effect of four levels of K Humate S100R (potassium humate) supplementation on the quality, shelf-life, and nutritional properties of beef. Angus steers (n = 40) were individually housed and fed either 0, 35, 70, or 140 g K Humate S100R/animal/day for 100 days, following a 30 day adjustment period. The steers were slaughtered at the completion of the feeding study. The left m. longissimus lumborum (LL) was collected at 24 h post-mortem and aged for either 2 or 6 weeks before analysis. K Humate S100R supplementation did not affect beef drip loss, cooking loss, shear force, sarcomere length, ultimate pH, intramuscular fat content, or total volatile basic nitrogen concentrations (P > 0.05). Steers supplemented with 70 g/day K Humate S100R produced beef with higher a* values on Days 1 and 3 of retail display (P < 0.05). Beef mineral composition was unchanged by K Humate S100R supplementation (P > 0.05), but there were minor changes to the fatty acid profile. Specifically, the ratio of omega-6 to omega-3 (P < 0.05) and C20:2n-6 concentrations (P < 0.05) increased with supplementation level. Together, these results demonstrate no detrimental effects on beef quality and shelf-life as a result of K Humate S100R supplementation.

There is limited information on changes of pasture mineral concentrations over the long-term in response to liming. A long-term field experiment was conducted to assess the influence of lime application on (a) changes in pasture mineral composition over time; and (b) key pasture mineral concentrations and ratios important to animal health. Perennial and annual pastures with or without lime application were sampled annually over 12 years and analysed for macro- and micro-minerals. Mineral ratios and indices were calculated to assess the potential impact on animal health. Liming increased the concentrations of calcium, sodium and silicon, but decreased the concentrations of micro-nutrients including copper, zinc and manganese. The same trend was found in both annual and perennial pastures although there were some fluctuations between years. Liming increased the calcium:phosphorus ratio and the dietary cation-anion difference but reduced the tetany index on both annual and perennial pastures. These findings suggest a potential benefit to improve animal health outcomes for some disorders on the limed pastures. However, the reduced concentrations of some trace elements following liming potentially decreases antioxidant capacity and requires further research.

Australia's primary production sector operates in one of the world's most variable climates with future climate change posing a challenge to its ongoing sustainability. Recognising this, Australia has invested in understanding climate change risks to primary production with a substantial amount of research produced. Recently, focus on this research space has broadened, with interests from the financial sector and expanded scopes of works from government and industry. These expanded needs require sector- and country-wide assessments to assist with the implementation of climate strategies. We considered the applicability of the current research body for these needs by reviewing 188 peer-reviewed studies that considered the quantitative impacts of climate change on Australia's primary industries. Our broad review includes cropping, livestock, horticulture, forestry and fisheries and biosecurity threats. This is the first such review for Australia, and no other similar country-wide review was found. We reviewed the studies through three lenses, industry diversity, geographic coverage and study comparability. Our results show that all three areas are lacking for sector- and country-wide assessments. Industry diversity was skewed towards cropping and biosecurity threats (64% of all studies) with wheat in particular a major focus (25% of all studies). Geographic coverage at a state level appeared to be evenly distributed across the country; however, when considered in conjunction with industry focus, gaps emerged. Study comparability was found to be very limited due to the use of different historical baseline periods and different impact models. We make several recommendations to assist with future research directions, being (1) co-development of a standard set of method guidelines for impact assessments, (2) filling industry and geographic knowledge gaps, and (3) improving transparency in study method descriptions. Uptake of these recommendations will improve study application and transparency enabling and enhancing responses to climate change in Australia's primary industries.

This study investigated the effect of supplementation of ewes with calcium (Ca) and magnesium (Mg) in late gestation and early lactation on the plasma mineral concentration, hormone profile and immunity responses of ewes and their lambs. Twin bearing ewes were allocated between four dietary treatment groups (n = 11 per treatment): control (0.33% DM Ca and 0.28% DM Mg); high Ca (0.72% DM Ca and 0.28% DM Mg); high Mg (0.33% DM Ca, 0.48% DM Mg); and high Ca+Mg (0.66% DM Ca and 0.47% DM Mg), offered as part of a whole ration. Ewes were fed the treatment supplement from around one month prior to lambing to one month after lactation. Blood and urine samples were collected at seven time-points: weekly starting at 5 weeks prior to lambing; within 12 h post lambing (+12 h); and then at fortnightly intervals at 2 week (+2 W) and at 4 week (+4 W) post lambing. Colostrum/milk samples from ewes and blood samples from lambs were collected at +12 h, +2 W and +4 W. Live weight of lambs were measured at +12 h, +2 W and +4 W. The plasma concentration of PTH, 1,25(OH)2D3 and 25(OH)D3 was lowest at +2 W (P ≤ 0.002). Ewes from the Ca+Mg group had the lowest mean concentration of 1,25(OH)2D3 than the other groups (P = 0.005). Magnesium supplementation improved the plasma Mg concentration over time in ewes (P <  0.001) and lambs from the control group had lower plasma Mg concentration compared to the treatment groups at +4 W (P = 0.001). Oxidative burst response in lambs supplemented with Ca tended to be greater at +4 W than the other groups at the same time point (P = 0.051) and Mg supplementation increased total antioxidant capacity (TAC) concentration in lambs (P = 0.040). The average daily weight gain of lambs was 204 g/lamb/d for the Ca group, 207 g/lamb/d for the Mg group, 245 g/lamb/d for the Ca+Mg group which were greater than the control group (148 g/lamb/d) (P < 0.001). Despite the normal concentration of Ca and Mg in the plasma, supplementation of ewes with Ca and Mg from one month prior to lambing to one month post lambing improved TAC concentration and weight gain in lambs.

Extracellular matrices have drawn attention in tissue engineering as potential biomaterials for scaffold fabrication because of their bioactive components. Noninvasive techniques of scaffold fabrication and cross-linking treatments are believed to maintain the integrity of bioactive molecules while providing proper architectural and mechanical properties. Cartilage matrix derived scaffolds are designed to support the maintenance of chondrocytes and provide proper signals for differentiation of chondroinducible cells. Chondroinductive potential of bovine articular cartilage matrix derived porous scaffolds on human dermal fibroblasts and the effect of scaffold shrinkage on chondrogenesis were investigated. An increase in sulfated glycosaminoglycans production along with upregulation of chondrogenic genes confirmed that physically treated cartilage matrix derived scaffolds have chondrogenic potential on human dermal fibroblasts.

Scaffold design from xenogeneic bone has the potential for tissue engineering (TE). However, major difficulties impede this potential, such as the wide range of properties in natural bone. In this study, sintered cortical bones from different parts of a bovine-femur impregnated with biodegradable poly(ethylene glycol) (PEG) binder by liquid phase adsorption were investigated. Flexural mechanical properties of the PEG-treated scaffolds showed that the scaffold is stiffer and stronger at a sintering condition of 1000°C compared with 900°C. In vitro cytotoxicity of the scaffolds evaluated by Alamar Blue assay and microscopic tests on human fibroblast cells is better at 1000°C compared with that at 900°C. Furthermore, in vitro biocompatibility and flexural property of scaffolds derived from different parts of a femur depend on morphology and heat-treatment condition. Therefore, the fabricated scaffolds from the distal and proximal parts at 1000°C are potential candidates for hard and soft TE applications, respectively.

Extracellular environments can regulate cell behavior because cells can actively sense their mechanical environments. This study evaluated the adhesion, proliferation and morphology of endothelial cells on polydimethylsiloxane (PDMS)/alumina (Al2 O3 ) composites and pure PDMS. The substrates were prepared from pure PDMS and its composites with 2.5, 5, 7.5, and 10 wt % Al2 O3 at a curing temperature of 50°C for 4 h. The substrates were then characterized by mechanical, structural, and morphological analyses. The cell adhesion, proliferation, and morphology of cultured bovine aortic endothelial (BAEC) cells on substrate materials were evaluated by using resazurin assay and 1,1'-dioctadecyl-1,3,3,3',3'-tetramethylindocarbocyanine perchlorate-acetylated LDL (Dil-Ac-LDL) cell staining, respectively. The composites (PDMS/2.5, 5, 7.5, and 10 wt % Al2 O3 ) exhibited higher stiffness than the pure PDMS substrate. The results also revealed that stiffer substrates promoted endothelial cell adhesion and proliferation and also induced spread morphology in the endothelial cells compared with lesser stiff substrates. Statistical analysis showed that the effect of time on cell proliferation depended on stiffness. Therefore, this study concludes that the addition of different Al2 O3 percentages to PDMS elevated substrate stiffness which in turn increased endothelial cell adhesion and proliferation significantly and induced spindle shape morphology in endothelial cells.

BACKGROUND AIMS: Numerous protocols for the isolation of bovine aortic endothelial cells have been described in the previous literature. However, these protocols prevent researchers from obtaining the pure population of endothelial cells. Thus, this study aimed to develop a new and economical method for the isolation of pure endothelial cells by introducing a new strategy to the enzymatic digestion method proposed by previous researchers. METHODS: With the use of this method, the lumen of a bovine aorta was filled with wash medium and the outer surface of the sample was washed with alcohol for 30 seconds. Under a laminar flow hood, the inner surface of the sample was covered with filter paper. Collagenase type II was dripped onto the filter paper as a digestion enzyme. The digestion fluid was seeded in T25 flasks and fed with complete medium every 3 days. RESULTS: The isolated cells were characterized by markers such as CD31, von Willebrand factor, 1,1'-dioctadecyl-1,3,3,3',3'-tetramethylindocarbocyanine perchlorate acetylated low-density lipoprotein and angiogenesis behavior. The purity of endothelial cells was detected by flow cytometry to be of nearly 90% purity; these results were confirmed by immunostaining. Moreover, endothelial cells formed blood vessel-like tubes in a three-dimensional environment, which is specific dynamic behavior for endothelial cells. CONCLUSIONS: The new strategy presented in the current report enables isolation of a highly pure population of endothelial cells that can survive long-term culture without inducing an overgrowth of fibroblast cells.

Native cartilage matrix derived (CMD) scaffolds from various animal and human sources have drawn attention in cartilage tissue engineering due to the demonstrable presence of bioactive components. Different chemical and physical treatments have been employed to enhance the micro-architecture of CMD scaffolds. In this study we have assessed the typical effects of physical cross-linking methods, namely ultraviolet (UV) light, dehydrothermal (DHT) treatment, and combinations of them on bovine articular CMD porous scaffolds with three different matrix concentrations (5%, 15% and 30%) to assess the relative strengths of each treatment. Our findings suggest that UV and UV-DHT treatments on 15% CMD scaffolds can yield architecturally optimal scaffolds for cartilage tissue engineering.

The present study aims to evaluate the serum level of copper (Cu) in lambs suffering from subclinical forms of white muscle disease (WMD) and its relationship with cardiac troponin I (cTn-I) as a novel biomarker of cardiovascular disorders. Ten milliliters of jugular blood were taken from 200 lambs less than one year old to measure serum concentrations of Cu, selenium (Se), and cTn-I. The subjects were divided into 2 groups, namely, the deficient group which included 36 lambs, and the control group which included 164 lambs according to the reference serum Se concentration (50 ng/mL). Serum Se levels in the deficient group were lower than 50 ng/mL. By contrast, the control group showed Se levels higher than 50 ng/mL. Differences among the serum Cu and cTn-I levels were determined in both groups. The mean ±SD and median of serum Cu and cTn-I levels in the deficient group were lower and higher than those in the control group, respectively. A significant positive correlation was observed between serum Cu and Se levels, and also serum Cu and Se levels showed a negative correlation with serum cTn-I concentrations. Stepwise linear regression analysis showed that serum Cu levels were correlated positively with serum Se levels (p<0.05). Receiver operating characteristic (ROC) curve analysis indicated that the area under curve (AUC) of Cu was significantly higher than that of cTn-I based on the reference diagonal line. It is important to keep in mind that the value of AUC for the ROC curve is between 0.5 and 1.00, in which the lowest accuracy is related to the reference diagonal line with AUC of 0.5. A cut-off was determined to indicate which Cu level can discriminate between affected and healthy lambs. The cut-off level, sensitivity, and specificity of Cu in this study were 144.5 ng/mL, 74%, and 61%, respectively.
