Urolithins are gut microbiota metabolites of ellagic acid. Here, we have identified and chemically characterized a novel urolithin produced from urolithin D (3,4,8,9-tetrahydroxy urolithin) by in vitro incubation with different human gut Enterocloster species under anaerobic conditions. Urolithin G (3,4,8-trihydroxy urolithin) was identified by 1H NMR, 13C NMR, UV, HRMS, and 2D NMR. For the identification, NMR spectra of other known urolithins were also recorded and compared. Urolithin G was present in the feces of 12% of volunteers in an overweight-obese group after consuming an ellagitannin-rich pomegranate extract. The production of urolithin G required a bacterial 9-dehydroxylase activity and was not specific to the known human urolithin metabotypes A and B. The ability to produce urolithin G could be considered an additional metabolic feature for volunteer stratification and bioactivity studies. This is the first urolithin with a catechol group in ring A while having only one hydroxyl in ring B, a unique feature not found in human and animal samples so far.

Introduction: The present study investigated how new words with acquired connotations of disgust and sadness, both negatively valenced but distinctive emotions, modulate the brain dynamics in the context of emotional sentences. Methods: Participants completed a learning session in which pseudowords were repeatedly paired with faces expressing disgust and sadness. An event-related potential (ERP) session followed the next day, in which participants received the learned pseudowords (herein, new words) combined with sentences and were asked to make emotional congruency judgment. Results: Sad new words elicited larger negative waveform than disgusting new words in the 146-228 ms time window, and emotionally congruent trials showed larger positive waveform than emotionally incongruent trials in the 304-462 ms time window. Moreover, the source localization in the latter suggested that congruent trials elicited larger current densities than incongruent trials in a number of emotion-related brain structures (e.g., the orbitofrontal cortex and cingulate gyrus) and language-related brain structures (e.g., the temporal lobe and the lingual gyrus). Discussion: These results suggested that faces are an effective source for the acquisition of words' emotional connotations, and such acquired connotations can generate semantic and emotional congruency effects in sentential contexts.

Introduction: Hyperuricemia and gout are receiving an increasing scientific and medical attention because of their relatively high prevalence and their association with relevant co-morbidities. Recently, it has been suggested that gout patients have an altered gut microbiota. The first objective of this study was to investigate the potential of some Ligilactobacillus salivarius strains to metabolize purine-related metabolites. The second objective was to evaluate the effect of administering a selected potential probiotic strain in individuals with a history of hyperuricemia. Methods: Inosine, guanosine, hypoxanthine, guanine, xanthine, and uric acid were identified and quantified by high-performance liquid chromatography analysis. The uptake and biotransformation of these compounds by a selection of L. salivarius strains were assessed using bacterial whole cells and cell-free extracts, respectively. The efficacy of L. salivarius CECT 30632 to prevent gout was assessed in a pilot randomized controlled clinical trial involving 30 patients with hyperuricemia and a history of recurrent gout episodes. Half of the patients consumed L. salivarius CECT 30632 (9 log10 CFU/day; probiotic group; n = 15) for 6 months while the remaining patients consumed allopurinol (100-300 mg/daily; control group; n = 15) for the same period. The clinical evolution and medical treatment received by the participants were followed, as well as the changes in several blood biochemical parameters. Results: L. salivarius CECT 30632 was the most efficient strain for inosine (100%), guanosine (100%) and uric acid (50%) conversion and, therefore, it was selected for the pilot clinical trial. In comparison with the control group, administration of L. salivarius CECT 30632 resulted in a significant reduction in the number of gout episodes and in the use of gout-related drugs as well as an improvement in some blood parameters related to oxidative stress, liver damage or metabolic syndrome. Conclusion: Regular administration of L. salivarius CECT 30632 reduced serum urate levels, the number of gout episodes and the pharmacological therapy required to control both hyperuricemia and gout episodes in individuals with a history of hyperuricemia and suffering from repeated episodes of gout.

Anaplasma marginale is a blood-borne rickettsia-like organism that infects cattle erythrocytes and causes anaplasmosis. This study reviews diagnostic data of all A. marginale diagnostics performed from 2003 to August 2021 in the Iowa State Veterinary Diagnostic Laboratory. Typically, the referring veterinarian's initial tentative diagnosis was based on presenting clinical signs or necropsy findings. Confirmatory testing at the ISU-VDL consisted of light microscopy evaluation of stained blood smears or molecular diagnostic procedures. A total of 94 cases were submitted with tissue samples from deceased animals, of which 79 were from Iowa and 15 were from other states. The most typical gross lesions were widespread yellow adipose tissue and splenomegaly. Typical histopathological lesions included marked bile stasis and hemosiderin-laden macrophages in the liver and spleen, respectively. Starting in 2013, when PCR was implemented to confirm cases of anaplasmosis, 315/1125 (28%) were positive to A. marginale, and 810 were negative, using a cut-off of 35.0 Ct. The average (±SD) of the positive PCR Ct was 19.5 (±6.0), and the first and third quartiles were 14.9 and 23.4. Most cases occurred between August and November, peaking in September, whether from necropsies or positive blood samples by PCR. The most common tick observed in Iowa, Dermacentor variabilis, is likely the main vector for transmission. Further surveys should be conducted to estimate seroprevalence by geographical location, the density of cattle populations, distribution of known vectors according to season, and strains of A. marginale.

We aimed to elucidate the gut bacteria that characterize the human urolithin metabotypes A and B (UM-A and UM-B). We report here a new bacterium isolated from the feces of a healthy woman, capable of producing the final metabolites urolithins A and B and different intermediates. Besides, we describe two gut bacterial co-cultures that reproduced the urolithin formation pathways upon in vitro fermentation of both UM-A and UM-B. This is the first time that the capacity of pure strains to metabolize ellagic acid cooperatively to yield urolithin profiles associated with UM-A and UM-B has been demonstrated. The urolithin-producing bacteria described herein could have potential as novel probiotics and in the industrial manufacture of bioactive urolithins to develop new ingredients, beverages, nutraceuticals, pharmaceuticals, and (or) functional foods. This is especially relevant in UM-0 individuals since they cannot produce bioactive urolithins.

Recent research has provided evidence that negation processing recruits the neural network of response inhibition (de Vega et al., 2016). Furthermore, inhibition mechanisms also play a role in human memory. In two experiments, we aimed to assess how producing a negation in a verification task may impact long-term memory. Experiment 1 used the same memory paradigm as Mayo et al. (2014), consisting of several phases: first, reading a story describing the activity of a protagonist, immediately followed by a "yes-no" verification task, then a distractive task, and finally an incidental free recall test. Consistent with the previous results, negated sentences were recalled worse than affirmed sentences. Yet, there is a possible confounding between the effect of negation itself and the associative interference of two conflicting predicates - the original and the modified one - in negative trials. To avoid this, Experiment 2 modified the paradigm by including a story describing the activities of two protagonists in such a way that the affirmed and denied verification sentences had the same content, and only differed in the attribution of a specific event to the correct or wrong protagonist. The negation-induced forgetting effect was still powerful, while controlling for potential contaminating variables. Our finding would support that the impaired long-term memory could be ascribed to reusing the inhibitory mechanism of negation.

In this study, the probiotic potential of Ligilactobacillus salivarius CECT 30632 was assessed, including properties specifically related with gynecological targets. This strain displayed co-aggregative and antimicrobial activity against a wide spectrum of vaginal pathogens while being respectful with the growth of vaginal lactobacilli. The strain produced a high concentration of lactic acid and displayed α-amylase activity when assayed in vitro. It showed a noticeable survival rate after exposition to conditions similar to those present in the human digestive tract and was adhesive to both vaginal and intestinal cells. Subsequently, their capacity to increase pregnancy rates among women with habitual abortion or infertility of unknown origin was studied. Administration of L. salivarius CECT 30632 (~9 log10 CFU) daily for a maximum of six months to these women was safe and led to a successful pregnancy rate of 67.5% (80% and 55% for women with repetitive abortion and infertile women, respectively). Significant differences in Nugent score, vaginal pH, and vaginal concentrations of lactobacilli, TGF-ß, and VEFG were observed when the samples collected before the intervention were compared with those collected after the treatment among those women who got pregnant. Therefore, this strain can modulate the vaginal ecosystem and lead to better fertility outcomes.

Motivational congruency has been examined using tasks where participants perform approach or avoidance movements towards socially positive or negative faces. Language is tightly intertwined with interpersonal cognition. Thus, similar situations could be represented by means of language in interpersonal contexts: adjectives furnish valence to people (e.g. someone is cordial or arrogant), and attitudinal verbs define direction to relationship-actions: approach-avoidance (e.g. accept vs. reject). In an Electroencephalography (EEG) study, 40 participants were presented with sentences where a character was valenced (e.g. "Arthur is cordial/arrogant") before being the target of a relationship-actions ("Grisela welcomed/ignored Arthur at the party"). We analyzed both Event-related potential (ERP) amplitude and time-frequency power in response to the attitudinal verb. For ERP amplitudes, we found a significant cluster between 280 and 370 ms, covering part of the development of a N400-like ERP component. This cluster reflects an interaction driven by congruency between motivational direction and target valence. Likewise, time-frequency power analysis revealed an enhancement of theta rhythms under incongruent conditions, most likely indexing conflict processing. Results support that relationship-actions are represented as approach and avoidance and thus involve conflict processing and resolution of incongruent situations. Implications for the interweaving of affective language and social cognition within Embodiment Simulation Theory are discussed.

It has been proposed that processing sentential negation recruits the neural network of inhibitory control (de Vega et al., 2016; Beltrán et al., 2021). In addition, inhibition mechanisms also play a role in switching languages for bilinguals (Kroll et al., 2015). Since both processes may share inhibitory resources, the current study explored for the first time whether and how language-switching influences the processing of negation. To this end, two groups of Spanish-English bilinguals participated in an encoding-verification memory task. They read short stories involving the same two protagonists (Montse and Jordi), referring to their activities in four different scenarios in Spanish or English. Following each story, the participants received verification questions requiring "yes" or "no" responses depending on whether a given fact was correctly referred to one of the protagonists. Some of the verification questions were in the story's original language (non-switch condition) and others in the alternate language (switch condition). Results revealed that language-switching facilitated negative responses compared to affirmative responses, exclusively for questions switching from dominant language (L1) to non-dominant language (L2). This effect might reflect that the domain-general mechanisms of inhibitory control are recruited at least partially for both language switch and negation process simultaneously, although this phenomenon is modulated by language dominance.

We describe here for the first time the consistent observation of two metabotypes associated with resveratrol metabolism by the human gut microbiota, that is, lunularin (LUNU)-producers and LUNU non-producers. In healthy volunteers (n = 195), resveratrol was reduced to dihydroresveratrol, which only in the LUNU-producer metabotype was sequentially dehydroxylated at the 5-position to yield LUNU and the 3-position to produce 4-hydroxydibenzyl. These metabolites (also 3,4'-dihydroxy-trans-stilbene in some LUNU-producers) were detected in the urine and (or) feces of 74% of volunteers after consuming resveratrol, while 26% lacked these dehydroxylase activities. The LUNU non-producer metabotype was more prevalent in females (P < 0.05) but independent of individuals' BMI and age. A 4-styrylphenol reductase in both metabotypes converted stilbenes to their corresponding dibenzyls, while no 4-dehydroxylation in stilbenes or dibenzyls was observed. 4-Hydroxy-trans-stilbene, pinosylvin, dihydropinosylvin, 3-hydroxydibenzyl, and 3-hydroxy-trans-stilbene were not detected in vivo or in vitro. Further research on LUNU metabotypes, their associated gut microbiota, and their impact on health is worthwhile.

Gut microbes have been recognized to convert human bile acids by deconjugation, dehydroxylation, dehydrogenation, and epimerization of the cholesterol core, but the ability to re-conjugate them with amino acids as an additional conversion has been recently described. These new bile acids are known as microbially conjugated bile acids (MCBAs). The aim of this study was to evaluate the MCBAs diversity produced by the gut microbiota through a metabolomics approach. In this study, fresh fecal samples from healthy donors were evaluated to explore the re-conjugation of chenodeoxycholic and 3-oxo-chenodeoxycholic acids by the human gut microbiota. No significant differences were found between the conversion trend of both BAs incubations. The in vitro results showed a clear trend to first accumulate the epimer isoursochenodeoxycholic acid and the dehydroxylated lithocholic acid derivatives in samples incubated with chenodeoxycholic and 3-oxo-chenodeoxycholic acid. They also showed a strong trend for the production of microbially conjugated dehydroxylated bile acids instead of chenodeoxycholic backbone conjugates. Different molecules and isomers of MCBAs were identified, and the new ones, valolithocholate ester and leucolithocholate ester, were identified and confirmed by MS/MS. These results document the gut microbiota's capability to produce esters of MCBAs on hydroxyls of the sterol backbone in addition to amides at the C24 acyl site. This study opens a new perspective to study the BAs diversity produced by the human gut microbiota.

This study examines the neural dynamics underlying the prosodic (duration) and the semantic dimensions in Spanish sentence perception. Specifically, we investigated whether adult listeners are aware of changes in the duration of a pretonic syllable of words that were either semantically predictable or unpredictable from the preceding sentential context. Participants listened to the sentences with instructions to make prosodic or semantic judgments, while their EEG was recorded. For both accuracy and RTs, the results revealed an interaction between duration and semantics. ERP analysis exposed an interactive effect between task, duration and semantic, showing that both processes share neural resources. There was an enhanced negativity on semantic process (N400) and an extended positivity associated with anomalous duration. Source estimation for the N400 component revealed activations in the frontal gyrus for the semantic contrast and in the parietal postcentral gyrus for duration contrast in the metric task, while activation in the sub-lobar insula was observed for the semantic task. The source of the late positive components was located on posterior cingulate. Hence, the ERP data support the idea that semantic and prosodic levels are processed by similar neural networks, and the two linguistic dimensions influence each other during the decision-making stage in the metric and semantic judgment tasks.

Resumen La ivermectina es el antiparasitario más usado en veterinaria a nivel mundial desde que entró al mercado en 1981. La U.S. Food and Drug Administration (FDA) americana en su valoración de riesgos determinó que, si bien es muy tóxica para organismos acuáticos, es improbable que, con las aplicaciones registradas para uso animal y por sus propiedades fisicoquímicas, se lleguen a contaminar cursos de agua. Sin embargo, los efectos de la ivermectina (IVM) sobre la fauna de invertebrados no blanco si pueden tener gran repercusión en la ecología de los pastizales por los costes asociados a la presencia de las boñigas no degradadas. De hecho, las pérdidas económicas por disminuir la calidad de los pastizales y reducir la superficie de los pastos disponible y apetecibles al ganado se han calculado en hasta 380 millones de dólares para la economía americana. La ivermectina afecta a un grupo muy beneficioso y diverso taxonómicamente que habita las boñigas, incluyendo a moscas coprófagas, avispas parasíticas, y escarabajos coprófagos y depredadores. Algunos estudios muestran que las boñigas de animales tratados con IVM pueden permanecer en los pastizales sin muestras apreciables de degradación hasta 340 días, mientras que las de animales no tratados son degradadas casi totalmente después de 80 días. Además, estudios de campo y laboratorio han mostrado que la sensibilidad de muchos insectos ocurre a concentraciones muy por debajo de las que se excretan con las heces de bovinos tratados con IVM. Los efectos sobre la reproducción y desarrollo de larvas de coleópteros ocurren incluso a concentraciones que son de hasta 10 veces por debajo de las que producen mortalidad. En Colombia se han identificado al menos 68 especies de la subfamilia Scarabaeinae que integran la comunidad de las boñigas del bovino. La mayor diversidad de escarabajos estercoleros se ha asociado a bosques y sistemas silvopastoriles que incorporan arboles nativos y proveen hábitats favorables para su supervivencia.


Abstract Ivermectin (IVM) has been the world most widely used antiparasitic agent in veterinary medicine since it came to the market in 1981. The U.S. Food and Drug Administration (FDA) in its risk assessment determined that, although it is very toxic to aquatic organisms, it is unlikely to contaminate water courses from current applications registered for animal use. However, the effects of IVM on non-target invertebrate fauna can have great impact on grassland ecology due to the costs associated with the presence of undegraded dung. In fact, the economic losses from lowering the quality of pastures and reducing the area of pasture available and palatable to livestock have been estimated in $380 million for the American economy. Ivermectin affects a highly beneficial and taxonomically diverse group that inhabits dung, including dung flies, parasitic wasps, and coprophilus and predatory dung beetles. Some studies show that dung from IVM-treated animals can remain in pasture without appreciable signs of degradation for up to 340 days, while those from untreated animals are almost completely degraded after 80 days. In addition, field and laboratory studies have shown the susceptibility of many insects to occur at concentrations well below those excreted in the feces of cattle treated with IVM. Effects on reproduction and development of coleopteran larvae occur at concentrations that can be 10 times lower of those causing mortality. In Colombia, at least 68 species of the subfamily Scarabaeinae have been identified in dung communities. The greater diversity of dung beetles has been associated with forests and silvopastoral systems that incorporate native trees and provide favorable habitats for their survival.


Resumo A ivermectina (IVM) tem sido o agente antiparasitário mais utilizado no mundo na medicina veterinária desde que chegou ao mercado em 1981. A US. Food and Drug Administration (FDA) em sua avaliação de risco determinou que, embora seja muito tóxico para organismos aquáticos, é improvável que contamine cursos d'água a partir de aplicações atuais registradas para uso animal. No entanto, os efeitos do IVM na fauna de invertebrados não-alvo podem ter grande impacto na ecologia das pastagens devido aos custos associados à presença de esterco não degradado. De fato, as perdas econômicas decorrentes da redução da qualidade das pastagens e da redução da área de pastagem disponível e palatável para o gado foram estimadas em US$ 380 milhões para a economia americana. A ivermectina afeta um grupo altamente benéfico e taxonomicamente diverso que habita o esterco, incluindo moscas do esterco, vespas parasitas e coprófilos e escaravelhos predadores. Alguns estudos mostram que o esterco de animais tratados com MIV pode permanecer no pasto sem sinais apreciáveis de degradação por até 340 dias, enquanto os de animais não tratados são quase completamente degradados após 80 dias. Além disso, estudos de campo e de laboratório mostraram que a suscetibilidade de muitos insetos ocorre em concentrações bem abaixo daquelas excretadas nas fezes de bovinos tratados com MIV. Os efeitos sobre a reprodução e desenvolvimento de larvas de coleópteros ocorrem em concentrações que podem ser 10 vezes menores daquelas que causam mortalidade. Na Colômbia, pelo menos 68 espécies da subfamília Scarabaeinae foram identificadas em comunidades de esterco. A maior diversidade de escaravelhos tem sido associada a florestas e sistemas silvipastoris que incorporam árvores nativas e proporcionam habitats favoráveis à sua sobrevivência.

Background: Commensal microflora such as Escherichia coli and Enterococcus spp. are representative indicators of antimicrobial resistance (AMR) as they are part of the normal intestinal microflora and can acquire and disseminate AMR to pathogenic or zoonotic bacteria like Salmonella spp. Objective: To investigate the state of AMR among E. coli and Salmonella spp., potential pathogens in humans, isolated from cecal contents of pigs submitted to a veterinary diagnostic laboratory in Colombia from 2016 to 2019. Methods: Susceptibility testing was conducted using the Kirby-Bauer disk diffusion method according to the Clinical and Laboratory Standards Institute guidelines for antimicrobial zone diameter breakpoints. An E. coli strain (ATCC 25922) was used as the quality control organism. Isolates showing resistance to three or more antimicrobial classes were classified as multidrug-resistant (MDR) as defined by a joint group of the European Centre for Disease prevention and Control and the Center for Disease Control and Prevention of the USA. Results: A total of 112 E. coli and 192 Salmonella spp. colonies were isolated from 557 samples received between 2016 and 2019. In order of decreasing frequency, E. coli was resistant to tetracycline (100%), sulfamethoxazol-trimethoprim (97.5%), amoxicillin (86.4%), enrofloxacin (82.6%), tylosin (82.1%), doxycycline (59%), neomycin (50%), ciprofloxacin (45.5%), ceftiofur (35%), gentamicin (30%), tilmicosin (29%), and fosfomycin (12.5%). When compared with E. coli, Salmonella spp. was generally resistant to the same agents with slightly less resistance (between 10-30%) to eight of the antimicrobials tested. Salmonella spp. showed <20% resistance to three antimicrobials, as follows: neomycin (17%), gentamicin (16%), and fosfomycin (14%). Multi-resistance occurred in 68.7% (77/112) of E. coli and 70.3% (135/192) of Salmonella spp. isolates. Resistance of Salmonella spp. was alarming to all the critically important antimicrobials tested: fluoroquinolones (enrofloxacin, ciprofloxacin), ceftiofur (third- generation cephalosporin), and macrolides (tylosin). Conclusions: According to our results, there is a high level of multi- drug resistance (MDR) in E. coli and Salmonella spp. It is necessary to implement a nationwide antimicrobial resistance monitoring program in Colombia, together with proper antimicrobial prescribing guidelines for pigs. The indiscriminate use of antimicrobial growth promoters by the swine industry is generating widespread bacterial resistance and should be discontinued.

Antecedentes: Flora comensal como espécies de Escherichia coli e Enterococcus são tipicamente escolhidas como indicadores representativos de la resistência antimicrobiana (AMR), pois fazem parte da flora intestinal normal e podem adquirir e disseminar AMR a bactérias patogênicas ou zoonóticas como Salmonella spp. Objetivo: Investigar o estado da AMR entre E. coli e Salmonella spp. isolados do conteúdo cecal de porcos colombianos submetidos ao Laboratório de Diagnóstico Veterinário de 2016 a 2019, ambos sendo patógenos potenciais em humanos. Métodos: O teste de suscetibilidade foi conduzido usando o método de difusão em disco Kirby-Bauer de acordo com as diretrizes do Instituto de Padrões Clínicos e Laboratoriais para pontos de quebra de diâmetro da zona antimicrobiana. A cepa de E. coli (ATCC 25922) foi usada como organismo de controle de qualidade. Os isolados que apresentam resistência a três ou mais classes de antimicrobianos foram classificados como multirresistentes (MDR), conforme definido por um grupo conjunto do Centro Europeu para Prevenção e Controle de Doenças e Centro para Controle e Prevenção de Doenças dos EUA. Resultados: Um total de 112 E. coli e 192 Salmonella spp. foram isolados de 557 amostras submetidas entre 2016 e 2019. Em ordem decrescente de frequência, a resistência a E. coli foi: tetraciclina (100%), sulfametoxazol-trimetoprim (97,5%), amoxicilina (86,4%), enrofloxacina (82,6%), tilosina (82,1%), doxiciclina (59%), neomicina (50%), ciprofloxacina (45,5%), ceftiofur (35%), gentamicina (30%), tilmicosina (29%) e fosfomicina (12,5%). Quando comparada com E. coli, Salmonella spp. foi geralmente resistente aos mesmos agentes com resistência ligeiramente menor (entre 10-30%) a oito dos antimicrobianos. Apenas três antimicrobianos apresentaram resistência a Salmonella spp. abaixo de 20% da seguinte forma: neomicina (17%), gentamicina (16%) e fosfomicina (14%). Multi-resistência ocorreu em 68,7% (77/112) de E. coli e 70,3% (135/192) de Salmonella spp. isolados. Resistência de Salmonella spp. foi alarmante para todos os antimicrobianos criticamente importantes testados: fluoroquinolonas (enrofloxacina, ciprofloxacina), ceftiofur (cefalosporina de terceira geração) e macrolídeos (tilosina). Conclusões: Esses resultados indicam um alto nível de resistência a múltiplos medicamentos (MDR) e que um Programa Nacional de Monitoramento da Resistência Antimicrobiana é necessário para a Colômbia, juntamente com a implementação de diretrizes de prescrição de antimicrobianos para suínos. O uso indiscriminado de antimicrobianos para promoção de crescimento na indústria suína está claramente promovendo resistência generalizada e deve ser interrompido.

In this study, participants listened to first-person statements that mentioned a character who was approaching a geographical location close to (Tenerife, Canary Islands) or distant from the participant (Madrid, Spanish peninsula), pronounced with either the participants' local or a distal regional accent. Participants more often judged approaching statements as coherent when they refer to a close place pronounced with local accent or refer to a distant place with distal accent, rather than when they refer to a close place with distal accent or to a distant place with local accent. These results strongly suggest that the local accent induces listeners to keep their own geographical perspective, whereas the distal accent determines shifting to another's perspective. In sum, a subtle paralinguistic cue, the speaker's regional accent, modulates the participants' geographic perspective when they listen to identical first-person sentences with approaching deictic verbs.

Resveratrol is a natural polyphenol with antioxidant activity and numerous health benefits. However, in vivo application of this compound is still a challenge due to its poor aqueous solubility and rapid metabolism, which leads to an extremely low bioavailability in the target tissues. In this work, rXynSOS-E236G glycosynthase, designed from a GH10 endoxylanase of the fungus Talaromyces amestolkiae, was used to glycosylate resveratrol by using xylobiosyl-fluoride as a sugar donor. The major product from this reaction was identified by NMR as 3-O-ꞵ-d-xylobiosyl resveratrol, together with other glycosides produced in a lower amount as 4'-O-ꞵ-d-xylobiosyl resveratrol and 3-O-ꞵ-d-xylotetraosyl resveratrol. The application of response surface methodology made it possible to optimize the reaction, producing 35% of 3-O-ꞵ-d-xylobiosyl resveratrol. Since other minor glycosides are obtained in addition to this compound, the transformation of the phenolic substrate amounted to 70%. Xylobiosylation decreased the antioxidant capacity of resveratrol by 2.21-fold, but, in return, produced a staggering 4,866-fold improvement in solubility, facilitating the delivery of large amounts of the molecule and its transit to the colon. A preliminary study has also shown that the colonic microbiota is capable of releasing resveratrol from 3-O-ꞵ-d-xylobiosyl resveratrol. These results support the potential of mutagenic variants of glycosyl hydrolases to synthesize highly soluble resveratrol glycosides, which could, in turn, improve the bioavailability and bioactive properties of this polyphenol.

Negation is known to have inhibitory consequences for the information under its scope. However, how it produces such effects remains poorly understood. Recently, it has been proposed that negation processing might be implemented at the neural level by the recruitment of inhibitory and cognitive control mechanisms. On this line, this manuscript offers the hypothesis that negation reuses general-domain mechanisms that subserve inhibition in other non-linguistic cognitive functions. The first two sections describe the inhibitory effects of negation on conceptual representations and its embodied effects, as well as the theoretical foundations for the reuse hypothesis. The next section describes the neurophysiological evidence that linguistic negation interacts with response inhibition, along with the suggestion that both functions share inhibitory mechanisms. Finally, the manuscript concludes that the functional relation between negation and inhibition observed at the mechanistic level could be easily integrated with predominant cognitive models of negation processing.

This study surveyed the prescribing behavior of Colombian companion animal veterinarians and compared the responses to the current guidelines of the International Society for Companion Animals on Infectious Diseases (ISCAID). A convenience sample of 100 primary-care veterinary practitioners was selected from the city of Medellin. A questionnaire was designed to present hypothetical clinical scenarios regarding prescription choices for systemic antimicrobials. The numbers of veterinarians empirically prescribing a course of systemic antimicrobials for each scenario were-perioperative elective surgeries (86%), superficial pyoderma (90%), lower urinary tract disease (52%), acute hemorrhagic diarrhea (50%), and kennel cough (46%). For urinary tract disease, cultures and susceptibility testing were only performed by half of the respondents, suggesting lower diagnostic standards. In superficial pyoderma cases, cytology was performed in the following percent of cases-0% (24), 20% (30), 40% (17), 60% (11), 80% (8), and 100% (10). Antimicrobials were over-prescribed relative to emerging standard for elective surgeries (86%), kennel cough (46%), and acute hemorrhagic diarrhea (50%). Critically important antimicrobials, such as fluoroquinolones, were applied commonly for superficial pyoderma (18%), kennel cough (12%), and lower urinary tract disease in dogs (20%) and cats (26%). In conclusion, antimicrobial prescribing behavior was inconsistent with current guidelines, and antimicrobial use could be improved by appropriate diagnostic steps allowing choice of an optimal antimicrobial drug. Overall, we documented the widespread use of antimicrobials for the treatment of these four common disease conditions.

In this study, the cervicovaginal environment of women with reproductive failure (repetitive abortion, infertility of unknown origin) was assessed and compared to that of healthy fertile women. Subsequently, the ability of Ligilactobacillus salivarius CECT5713 to increase pregnancy rates in women with reproductive failure was evaluated. Vaginal pH and Nugent score were higher in women with reproductive failure than in fertile women. The opposite was observed regarding the immune factors TGF-ß 1, TFG-ß 2, and VEFG. Lactobacilli were detected at a higher frequency and concentration in fertile women than in women with repetitive abortion or infertility. The metataxonomic study revealed that vaginal samples from fertile women were characterized by the high abundance of Lactobacillus sequences, while DNA from this genus was practically absent in one third of samples from women with reproductive failure. Daily oral administration of L. salivarius CECT5713 (~9 log10 CFU/day) to women with reproductive failure for a maximum of 6 months resulted in an overall successful pregnancy rate of 56%. The probiotic intervention modified key microbiological, biochemical, and immunological parameters in women who got pregnant. In conclusion, L. salivarius CECT5713 has proved to be a good candidate to improve reproductive success in women with reproductive failure.

There is abundant literature demonstrating that processing emotional stimuli modulates inhibitory control processes. However, the reverse effects, namely, how cognitive inhibition influences the processing of emotional stimuli, have been considerably neglected. This ERP study tries to fill this gap by studying the bidirectional interactions between emotional language and inhibitory processes. To this end, participants read emotional sentences, embedded in a cue-based Go-NoGo task. In Experiment 1, the critical emotional adjective preceded the Go-NoGo visual cue. The ERPs showed a significant reduction in the inhibition-related N2 component in NoGo trials when they were preceded by negative adjectives, compared to positive or neutral adjectives, indicating a priming-like effect on inhibitory control. Consistently, the estimated source of this interaction was the dorsomedial PFC, a region associated with inhibitory and control processes. In Experiment 2, the Go-NoGo cue preceded the emotional adjective, and the ERPs showed a sustained, broadly distributed LPP-like positivity for NoGo negative trials, relative to all the other conditions. In this case, the presetting of an inhibition state modulated the processing of negatively charged words. Together, the two experiments suggest a mutual facilitation between inhibitory control and negative valence, supporting thereby recent integrative theories of cognition-emotion interactions.
