RESUMEN

We present a coarse-grained computer model designed to study the growth of fibres in a synthetic self-assembling peptide system. The system consists of two 28 residue α-helical sequences, denoted AB and CD, in which the interactions between the half peptides, A, B, C and D, may be tuned individually to promote different types of growth behaviour. In the model, AB and CD are represented by double ended rods, with interaction sites distributed along their lengths. Monte Carlo simulations are performed to follow fibre growth. It is found that lateral and longitudinal growth of the fibre are governed by different mechanisms--the former is diffusion limited with a very small activation energy for the addition of units, whereas the latter occurs via a process of secondary nucleation at the fibre ends. As a result, longitudinal growth generally proceeds more slowly than lateral growth. Furthermore, it is shown that the aspect ratio of the growing fibre may be controlled by adjusting the temperature and the relative strengths of the interactions. The predictions of the model are discussed in the context of published data from real peptide systems.

Stearic acid and triethanolamine (TEA) in a molar ratio of 2:1 were mixed in aqueous solution at 80 degrees C and subsequently cooled to ambient temperature. The structural evolution of the resultant sample during storage was characterized by using light microscopy, Cryo-SEM, differential scanning calorimetery, pH, infrared spectroscopy, elemental analysis, and simultaneous small and wide-angle X-ray diffraction. It was found that a lamellar liquid crystalline phase was formed when stearic acid and TEA solution were mixed at 80 degrees C and multilamellar spheres of a few microns diameter were formed initially after cooling. A hydrolysis reaction (i.e., the reverse reaction of neutralization between stearic acid and TEA) occurred thereafter that caused the breakdown of the lamellar gel phase and the formation of platelet stearic acid crystals. Three polymorphs of stearic acid (defined following previous work as the A, C, and E forms) were formed as the result of hydrolysis reaction, which gave rise to a strong optically pearlescent appearance.

Arginine is a conditionally essential amino acid with many physiologic roles. Its role in immune function has been one of major focus with conflicting results. Early in vitro immune studies demonstrated increased mitogen-induced lymphocyte proliferation with dietary arginine supplementation; however, not all studies confirmed this effect. Even less is known about the effect of arginine supplementation on in vivo immune responses. To test whether arginine supplementation enhances in vivo indicators of immune function, young female BALB/c mice were fed either the AIN-93G rodent diet (6.4 g arginine/kg diet) or the same diet with 20 g total arginine/kg diet for 15 d before delayed-type hypersensitivity (DTH) testing with 2,4-dinitrofluorobenzene (n = 16-18/diet group). The same mice were challenged with influenza virus A/Port Chalmers/1/73 (H3N2) 15 d later. Mice were killed 3, 6, or 31 d postinfluenza challenge (5-6/diet group on each day). Mitogen-induced splenocyte proliferation, body weight, anti-influenza serum antibody, lung viral titers, and serum arginine were measured. DTH did not differ between diet groups. On d 6 and 31 postchallenge, mitogen-induced proliferation of splenocytes from mice fed the arginine diet was >1.5-fold that of mice fed the control diet (P < 0.05). Body weight and influenza lung viral and serum antibody titers did not differ between diet groups. These data suggest that despite significant enhancement of in vitro mitogen-induced splenocyte proliferation, arginine supplementation does not have a biologically significant effect on antigen-specific in vivo indicators of immune function in this model.

Crystals of partially neutralized stearic acid with triethanolamine (TEA) were prepared by mixing these two materials above 80 degrees C and then cooling. The crystalline composition and the structure and melting behavior of the resultant products were characterized with small-angle and wide-angle X-ray diffraction, thermal analysis, microscopy, and infrared spectroscopy. It was discovered that an acid-soap complex of 2:1 fixed stoichiometric ratio exists between stearic acid and TEA stearate. A binary phase diagram of stearic acid and TEA soap is built based on the experimental results; this is the first published record of a binary phase diagram for amine-based soap. Its behavior is significantly different from that of binary systems of fatty acid and alkali soap.

The kinetics of phase separation were observed in the gelatin/maltodextrin and gelatin/maltodextrin/gum arabic systems, where gum arabic was added as a minority component, using small-angle light scattering, turbidity measurement, and confocal scanning laser microscopy. Phase separation occurred by spinodal decomposition for quenches both above and below the temperature at which gelatin gelled. Coarsening of the phase-separated microstructure was hindered by gelation, and a hydrodynamic mechanism, observed when the gelatin remained in the liquid state, was suppressed. Gum arabic, containing both polysaccharide and polypeptide components, was hypothesized to be potentially interfacially active in the gelatin/maltodextrin system, in analogy with synthetic block copolymer compatibilizers in demixed synthetic polymer systems. The hypothesis was experimentally refuted under the chosen experimental conditions, as no evidence was found to suggest that it altered the phase separation behavior.

RESUMEN

A 15-year-old girl presented with a cystic swelling since birth behind the ramus of the right mandible and diagnosed clinically as a dermoid cyst. Surgical exploration, however, showed that it was closely related to the external auditory canal, with an extension running medially behind the parotid gland and ending in the bony middle ear. The facial nerve was closely related to the deep part of the cyst. Such an anatomical position indicates that this was a first branchial cleft anomaly. Surgical excision of the cyst was performed.

The effect of 16 pure metals on the in vitro growth of a form of dental caries producing Streptococcus mutans was studied under both aerobic and anaerobic conditions. Cobalt and copper were consistently observed to be inhibitory. With less consistency nickel, titanium, iron, and vanadium also exhibited ability to inhibit growth of the organism. Bacteriostasis apparently is contingent upon the presence of a corrosion process. The bacteriostatic agent is of uncertain identity and could be a corrosion product or a process secondary to the occurrence of corrosion. Concentrations of metals after 6 days of electrochemical dissolution in the growth medium were measured via electron microprobe analysis and compared with the amount of inhibition which resulted. Threshold concentrations above which growth did not occur were identified. As measured by threshold concentrations, wide variability between metals exists in the ability to inhibit the growth, with cobalt being particularly effective at small concentrations. This implies a sensitivity on the part of the organism that is different for different metals. The threshold concentration varied between aerobic and anaerobic conditions. Streptococcus mutans appeared more resistant to the effects of the metals under anaerobic conditions even though little difference in the amount of corrosion was detected.
