RESUMEN

An inter-laboratory exercise was carried out by a consortium of five European laboratories to establish a set of compounds, suitable for calibrating gas chromatography/combustion/isotope ratio mass spectrometry (GC-C-IRMS) devices, to be used as isotopic reference materials for hydrogen, carbon, nitrogen and oxygen stable isotope measurements. The set of compounds was chosen with the aim of developing a mixture of reference materials to be used in analytical protocols to check for food and beverage authentication. The exercise was organized in several steps to achieve the certification level: the first step consisted of the a priori selection of chemical compounds on the basis of the scientific literature and successive GC tests to set the analytical conditions for each single compound and the mixture. After elimination of the compounds that turned out to be unsuitable in a multi-compound mixture, some additional oxygen- and nitrogen-containing substances were added to complete the range of calibration isotopes. The results of delta(13)C determinations for the entire set of reference compounds have previously been published, while the deltaD and delta(18)O determinations were unsuccessful and after statistical analysis of the data the results did not reach the level required for certification. In the present paper we present the results of an inter-laboratory exercise to identify and test the set of nitrogen-containing compounds present in the mixture developed for use as reference materials for the validation of GC-C-IRMS analyses in individual laboratories.

RESUMEN

Tetramethylurea (TMU) with a certified D/H ratio is the internal standard for Site-specific Natural Isotope Fractionation measured by Nuclear Magnetic Resonance (SNIF-NMR) analysis of wine ethanol for detection of possible adulterations (Commission Regulation 2676/90). A new batch of a TMU certified reference material (CRM) is currently being prepared. Whereas SNIF-NMR has been employed up to now, Elemental Analysis/Isotope Ratio Mass Spectrometry ((2)H-EA-IRMS) was envisaged as the method of choice for value assignment of the new CRM, as more precise (better repeatable) data might be obtained, resulting in lower uncertainty of the certified value. In order to evaluate the accuracy and intra- and inter-laboratory reproducibility of (2)H-EA-IRMS methods, a laboratory inter-comparison was carried out by analysing TMU and other organic compounds, as well as some waters. The results revealed that experienced laboratories are capable of generating robust and well comparable data, which highlights the emerging potential of IRMS in food authenticity testing. However, a systematic bias between IRMS and SNIF-NMR reference data was observed for TMU; this lack of data consistency rules out the (2)H-IRMS technique for the characterisation measurement of the new TMU CRM.

Three stable isotope ratios, D/H, (13)C/(12)C and (18)O/(16)O, are measurable in ethanol, an important organic compound that is used as a material for food and beverages, fuel and chemical feedstock, and as a substance related to metabolism. We developed a simple and rapid method of measurement of three isotope ratios of ethanol in aqueous solution at millimole levels using gas chromatography-high-temperature conversion or combustion-isotope ratio mass spectrometry (GC-TC/C-IRMS) combined with solid-phase microextraction (SPME). Using this method, the delta value for ethanol was determined in 30 min for deltaD and delta(13)C, and in 75 min for delta(18)O with precisions of +/-9 per thousand, +/-0.3 per thousand and +/-0.7 per thousand, respectively, for deltaD, delta(13)C, and delta(18)O. An advantage of this process is that it requires no distillation for ethanol purification. The method is useful for small quantities of analyte with low ethanol concentrations, which is expected for environmental and metabolic studies.

RESUMEN

A simple and rapid method to measure naturally occurring delta(13)C values of headspace CO(2) of sparkling drinks has been set up, using direct injections on a gas chromatograph coupled to an isotope ratio mass spectrometer, through a combustion interface (GC/C/IRMS). We tested the method on CO(2) gas from several origins. No significant isotopic fractionation was observed nor influences by secondary compounds eventually present in the gas phase. Standard deviation for these measurements was found to be <0.1 per thousand.

RESUMEN

Glycerol of about 170 European wines was analyzed using gas chromatography-combustion-isotope ratio mass spectrometry technique. (13)C/(12)C isotopic ratio measurements were performed to characterize glycerol's delta(13)C values of genuine wine samples from European Union wine-producing countries. Glycerol was also successfully dosed using an internal reference, 1,5-pentanediol.
