RESUMEN

Equisetum (Equisetaceae) has long been a focus of attention for botanists and palaeontologists because, given its extensive and well-documented fossil record, it is considered the oldest extant vascular plant and a key element in understanding vascular plant evolution. However, to date, no authentic fossil evidence of Equisetum has been found from the Indian Cenozoic. Here, we describe a new fossil species, namely, E. siwalikum sp. nov., recovered from the middle Siwalik (Late Miocene) sediments of Himachal Pradesh, western Himalaya. We identified fossil specimens based on morphological and epidermal characters. In addition, X-Ray diffraction (XRD) analysis was used to determine the mineral composition of compressed stems of Equisetum. The close affinity of our recovered Siwalik fossils to Equisetum is supported by the presence of both macromorphological and epidermal characters. Because Equisetum generally grows in wet conditions around water reservoirs, our findings indicate that the fossil locality was humid and surrounded by swamp and lowland regions during deposition. Ample fossil evidence indicates that this sphenopsid once existed in the western Himalaya during the Siwalik period. However, at present Equisetum is confined to a particular area of our fossil locality, probably a consequence of severe environmental changes coupled with competition from opportunistic angiosperms. Our discovery of Equisetum fossils in appreciable numbers from the Siwalik sediments of the Himachal Himalayas is unique and constitutes the first reliable recognition of Equisetum from the Indian Cenozoic.

This paper presents the groundwater quality assessment of the upper Brahmaputra floodplains of Assam on a seasonal basis. A total of 88 samples were analyzed for the presence of potentially toxic elements in two seasons. In addition, an attempt is made to identify any possible associated health risks to the residents via the drinking water pathway. The study reveals the presence of various potentially toxic elements, in particular, manganese, iron, nickel, and fluoride concentration exceeding the drinking water specifications set by BIS and WHO drinking water standards. The degree of groundwater contamination was assessed using the Water Quality Index, Heavy metal Pollution Index, Heavy metal Evaluation Index, and Degree of Contamination. The spatial distribution maps of groundwater quality were prepared using geographical information system. The non-carcinogenic health risk was evaluated using hazard quotients and hazard index as per the United States Environmental Protection Agency methodology. The hazard quotient of fluoride and manganese have values > 1, which exceeds USEPA recommended benchmark. The health risk assessment identified that the risk was highest during the pre-monsoon season, and the child population is more vulnerable to non-carcinogenic risk than the adults. Findings of cancer risk identified that pre-monsoon groundwater samples from the Golaghat District pose the highest health risks in the upper Brahmaputra floodplains. The risk is highest in the southwest of the study area, followed by the south and then by the north.

A rich and diverse palynoassemblage recovered from the Churanthi River section (26°53' 59.3" N, 88°34' 17.2" E), Darjeeling foothills Eastern Himalaya, has yielded 87 species assigned to 69 genera. The palynoassemblage is rich in angiosperm taxa (45.63%) followed by gymnosperms (0.45%), pteridophytes (18.49%) and fungal remains (23.88%). Based on their nearest living relatives, a wet evergreen to semi-evergreen forest under a humid tropical to sub-tropical environment during the Mio-Pliocene age has been suggested. A lot of angiosperms such as Palaeosantalaceaepites, Araliaceoipollenites, Malvacearampollis, Zonocostites, Neocouperipollis, Dicolpopollis, Palmidites, Palmaepollenites, isolated salt glands of mangrove plant leaves (Heliospermopsis) and Mediaverrunites type of fungal spores, along with ichnofossils like Planolites, Palaeophycus, Skolithos, Rosselia, Ophiomorpha and Teichichnus associated with rippled mudstone-siltstone suggest an environment strongly influenced by brackish water. Primary sedimentary structures in the associated strata indicate strong wave agitation common in shallow marine setting. Some high elevation components (5.14%) such as Alnipollenites, cf. Corylus (Betulaceae), Juglanspollenites, Engelhardtioipollenites (Juglandaceae), Quercoides, Cupuliferoidaepollenites, Lithocarpus, Castanopsis (Fagaceae), Abietineaepollenites (Pinaceae) represent hinterland vegetation possibly transported to the prograding deltaic coastline by the rivers. Reworked palynotaxa (Striatopodocarpites sp., Striatites sp., Faunipollenites sp., Circumstriatites sp., Crescentipollenites sp., Cuneatisporites sp., Parasaccites sp., Scheuringipollenites sp., Rhizomaspora sp., Marsupipollenites sp., Lophotriletes sp.) of Permian age have also been recorded in the palynoassemblage (11.55%) indicating the abundance of Permian Gondwana strata in the source area.

Thymol, a naturally occurring phenolic compound, has been known for its antioxidant, anti microbial, and anti inflammatory activity. Thymol has also been reported as anti-cancer agent, but its anti-cancer mechanism has not yet been fully elucidated. Thus, we aimed to investigate anticancer activity of thymol on HL-60 (acute promyelotic leukemia) cells. In our study, thymol demonstrated dose dependent cytotoxic effects on HL-60 cells after 24h of exposure. However, thymol did not show any cytotoxic effect in normal human PBMC. The cytotoxic effect of thymol on HL-60 cells appears to be associated with induction of cell cycle arrest at sub G0/G1 phase, and apoptotic cell death based on genomic DNA fragmentation pattern. Thymol also showed significant increase in production of reactive oxygen species (ROS) activity, increase in mitochondrial H(2)O(2) production and depolarization of mitochondrial membrane potential. On performing Western Blot analysis, thymol showed increase in Bax protein level with a concomitant decrease in Bcl2 protein expression in a dose dependent manner. Our study also showed activation of caspase -9, -8 and -3 and concomitant PARP cleavage, which is the hallmark of caspase-dependent apoptosis. Moreover, to rule out the involvement of other mechanisms in apoptosis induction by thymol, we also studied its effect on apoptosis inducing factor (AIF). Thymol induced AIF translocation from mitochondria to cytosol and to nucleus, thus indicating its ability to induce caspase independent apoptosis. We conclude that, thymol-induced apoptosis in HL-60 cells involves both caspase dependent and caspase independent pathways.

The antisecretory and antiulcer effects of aqueous extract of Neem (Azadirachta indica) bark have been studied along with its mechanism of action, standardisation and safety evaluation. The extract can dose dependently inhibit pylorus-ligation and drug (mercaptomethylimidazole)-induced acid secretion with ED(50) value of 2.7 and 2 mg Kg(-1) b.w. respectively. It is highly potent in dose-dependently blocking gastric ulcer induced by restraint-cold stress and indomethacin with ED(50) value of 1.5 and 1.25 mg Kg(-1) b.w. respectively. When compared, bark extract is equipotent to ranitidine but more potent than omeprazole in inhibiting pylorus-ligation induced acid secretion. In a stress ulcer model, it is more effective than ranitidine but almost equipotent to omeprazole. Bark extract inhibits H(+)-K(+)-ATPase activity in vitro in a concentration dependent manner similar to omeprazole. It offers gastroprotection against stress ulcer by significantly preventing adhered mucus and endogenous glutathione depletion. It prevents oxidative damage of the gastric mucosa by significantly blocking lipid peroxidation and by scavenging the endogenous hydroxyl radical ((z.rad;)OH)-the major causative factor for ulcer. The (z.rad;)OH-mediated oxidative damage of human gastric mucosal DNA is also protected by the extract in vitro. Bark extract is more effective than melatonin, vitamin E, desferrioxamine and alpha-phenyl N-tert butylnitrone, the known antioxidants having antiulcer effect. Standardisation of the bioactive extract by high pressure liquid chromatography indicates that peak 1 of the chromatogram coincides with the major bioactive compound, a phenolic glycoside, isolated from the extract. The pharmacological effects of the bark extract are attributed to a phenolic glycoside which is apparently homogeneous by HPLC and which represents 10% of the raw bark extract. A single dose of 1g of raw extract per kg b.w. (mice) given in one day and application of 0.6g raw extract per kg b.w. per day by oral route over 15 days to a cumulative dose of 9g per kg was well tolerated and was below the LD(50). It is also well tolerated by rats with no significant adverse effect. It is concluded that Neem bark extract has therapeutic potential for the control of gastric hyperacidity and ulcer.
