Background: The mechanical properties of muscles, such as changes in muscle tone and stiffness, are related to sports performance and injuries. Rowers are at increased risk of muscle fatigue and injury during high-repetition and heavy-load cyclic muscle actions. In view of this, the aim of the present study was to investigate the acute effect on muscle tone and stiffness, as well as bilateral muscle asymmetry, in high school rowers after a 2000-meter rowing ergometer test. Methods: Twelve young male rowers (age = 17.1 ± 0.9 years, body weight = 73.5 ± 9.7 kg) were included in the study. The data of muscle tone (frequency) and stiffness of the posterior deltoids (PD), latissimus dorsi (LD), and rectus femoris (RF) (dominant and non-dominant side) before and after a 2000-m rowing ergometer test were collected using a handheld MyotonPRO device. Results: After the rowing ergometer test, the muscle tone of dominant side PD, LD, and RF were significantly increased (p < 0.05). On the other hand, the muscle stiffness of the non-dominant side LD and RF, as well as the dominant side PD, LD, and RF were significantly increased after the rowing ergometer test (p < 0.05). The muscle tone and stiffness results showed that the dominant side PD, LD, and RF were all significantly higher than the non-dominant side after the rowing ergometer test (p < 0.05), where bilateral PD and RF exhibits moderate asymmetry (5% < symmetry index < 10%). Conclusions: After a high-intensity and high-load 2000-m rowing ergometer test, PD, LD, and RF showed increases in muscle tone and stiffness, as well as changes in the symmetry of bilateral muscle mechanical properties.

This research was to analyze the effect of flavored Tongxie Yaofang on diarrheal irritable bowel syndrome (IBS) by the situation of intestinal microecology. The treatment mechanism was analyzed, so as to provide a more effective treatment method for patients clinically. 60 IBS patients were selected as the research objects and were divided according to the different treatment methods. For the control group (n = 20 cases), oral pinaverium bromide tablets were given. For the treatment group (n = 40 cases), the flavored Tongxie Yaofang decoction was given in addition to conventional treatment. The curative effects on the two groups of patients were evaluated in combination with the changes in intestinal microecology. With the syndrome score, the total effective rate of the treatment group (92.5%) was obviously superior to the control group (80%) (P < 0.05). The clinical symptoms such as abdominal pain, abdominal distension, and diarrhea in the treatment group were significantly relieved after treatment in contrast to the control group (P < 0.05). Intestinal Bifidobacterium, Escherichia coli, and Bifidobacterium/Escherichia coli (B/E) ratio were all greatly higher than those in the control group (P < 0.05). In summary, flavored Tongxie Yaofang had a good effect in improving the symptoms of patients with diarrheal IBS and improved the microflora of Bifidobacterium and Escherichia coli in the intestinal tract of patients.

BACKGROUND: Gastric cancer is a common malignant tumor. The aim of the present study was to analyze the application value of serum pepsinogen (PG), carbohydrate antigen 72-4 (CA72-4), and gastrin-17 (G-17) detection in the screening, diagnosis, and evaluation of early gastric cancer. METHODS: In total, 122 patients with gastric cancer treated in our hospital from January 2018 to January 2021 were selected as the gastric cancer group and subdivided into the early gastric cancer (group A) and advanced gastric cancer (group B) groups. Sixty-five patients with benign gastric disease treated in the same hospital during the same period were selected as the control group, and 122 healthy people who underwent physical examination during the same period were allocated to the control group. The differences in the levels of G-17, PGI, PGII, PGI/PGII, and CA72-4 were compared; receiver-operating characteristic curves were drawn; and the efficacy of different factors in the diagnosis of early gastric cancer was calculated. RESULTS: G-17, PGI, and PGI/PGII levels in the gastric cancer group were significantly lower than those in the healthy group, and CA72-4 was significantly higher than that in the healthy group (P<0.05), but there was no significant difference in PGII between the 2 groups (P>0.05). G-17, PGI, and PGI/PGII levels in groups A and B were significantly lower than those in the control group. CA72-4 in groups A and B was significantly higher than that of the control group, and was highest in group B (P<0.05). The areas under the curve (AUC) of G-17, PGI, PGI/PGII, and CA72-4 were 0.671, 0.726, 0.769, and 0.602, respectively, and the AUC of combined detection was 0.883, which was significantly higher than that of single detection. CONCLUSIONS: Serum PG, CA72-4 combined with G-17 detection has high sensitivity and specificity in the screening and diagnosis of early gastric cancer, and has high clinical application value.

RESUMEN

Electro-optical properties of polymer-stabilized sphere phase liquid crystal (PS-SPLC) switching devices are analyzed and validated experimentally. The experimental results show the voltage-dependent transmittance curves of PS-SPLC devices. A diffraction approach, called extended anomalous diffraction approach, is proposed to fit the experimental data. Good agreement between experiment and model is obtained. The scattering model provides practical guidance for the improvement of PS-SPLC displays performance and optimization.

RESUMEN

The safety of home birth is widely debated. The European Court of Human Rights (ECtHR) recently made two conflicting rulings on home birth, Ternovszky v Hungary and Dubská and Krejzová v the Czech Republic. In Ternovszky v Hungary, the ECtHR essentially forced Hungary to change existing legislation to allow home birth. In contrast, in Dubská and Krejzová v the Czech Republic, the Court found Czech Republic's stringent regulations against home birth acceptable. We describe these cases, the different evidence used and interpreted by the court and examine the ethical conflict between maternal autonomy and the rights of the newborn.

The use of fluorescence is ubiquitously found in the detection of immunoreaction; though with good sensitivity, this technique requires labeling as well as other time-consuming steps to perform the measurement. An alternative approach involving liquid crystals (LCs) was proposed, based on the fact that an immunocomplex can disturb the orientation of LCs, leading to an optical texture different from the case when only antigen or antibody exists. This method is label-free, easy to manipulate and low-cost. However, its sensitivity was low for practical usage. In this study, we adopted a high-birefringence liquid crystal (LC) to enhance the sensitivity for the immunodetection. Experiments were performed, targeting at the cancer biomarker CA125. We showed that the larger birefringence (Δn = 0.33 at 20 °C) amplifies the detected signal and, in turn, dramatically improves the detection limit. To avoid signal loss from conventional rinsing steps in immunodetection, CA125 antigen and antibody were reacted before immobilized on substrates. We studied the specific binding events and obtained a detection limit as low as 1 ng/ml. The valid temperature ranges were compared by using the typical single-compound LC 5CB and the high-birefringence LC mixture. We further investigated time dependency of the optical textures and affirmed the capability of LC-based immunodetection in distinguishing between specific and nonspecific antibodies.

OBJECTIVE: Antibodies to CA 125 have been used to predict relapse of ovarian cancer, but have performed poorly as therapeutic agents. One rationale for this is antibody binding to circulating shed antigen. Our aim in this study was to develop antibodies to human CA 125 that have enhanced selectivity for the cell-associated form of the antigen. METHODS: Monoclonal antibodies were raised to a recombinant fragment of CA 125 that included sequence proximal to the putative membrane attachment site. Antibodies were characterized in terms of their binding site, affinity and selectivity for cell-associated CA 125. RESULTS: In assays using patient-derived CA 125, a subset of high-affinity (KD <5 nM) monoclonal antibodies demonstrated a 10- to greater than 200-fold increase in selectivity for cell-associated CA 125 when compared with controls. Based on mapping of the various monoclonal antibodies obtained, it was determined that shedding of CA 125 most likely occurs in the most C-terminal repeat domain. CONCLUSION: Results from competition analysis using patient-derived shed antigen predict that the antibodies described in this study may have significantly enhanced tumor-targeting properties when compared with existing antibodies to CA 125 in a tumor environment having high concentrations (>10,000 CA 125 units) of shed CA 125.

Fibroblast growth factor receptor 4 (FGFR4) is a member of the FGFR family of receptor tyrosine kinases, and plays important roles in a variety of biological functions such as cell proliferation, differentiation, migration, angiogenesis, tissue repair, and tumorigenesis. The human FGFRs share a high degree of sequence homology between themselves, as well as with their murine homologs. Consequently, it has been suggested that it may be difficult to prepare monoclonal antibodies (MAbs) that are specific for the individual receptor types. In this communication, we report on the development and characterization of a panel of anti-human FGFR4 MAbs that were generated in mice using a rapid immunization protocol. Using a modified rapid immunization at multiple sites (RIMMS) protocol with the soluble extracellular domain of human FGFR4 (FGFR4-ECD), the immunized mice developed high levels of polyclonal IgG to the immunogen within 13 days of the first immunization. The lymph node cells isolated from the immunized animals were then fused with mouse myeloma cells for hybridoma generation. Use of an efficient hybridoma cloning protocol in combination with an ELISA screening procedure allowed for early identification of stable hybridomas secreting antihuman FGFR4 IgG. Several identified MAbs specifically reacted with the FGFR4 protein without binding to the other human isoforms (FGFR1, FGFR2, and FGFR3). As evaluated by BIAcore analysis, most anti-FGFR4 MAbs displayed high affinities (8.6 x 10(8) approximately 3.9 x 10(10) M) to FGFR4. Furthermore, these MAbs were able to bind to FGFR4 expressed on human breast tumor cell lines MDA-MB-361 and MDA-MB-453. Taken together, the results demonstrate that the RIMMS strategy is an effective approach for generating class-switched, high-affinity MAbs in mice to evolutionarily conserved proteins such as human FGFR4. These MAbs may be useful tools for further investigation of the biological functions and pathological roles of human FGFR4.

Tissue factor (TF) plays an important, physiological role in hemostasis. Recent studies have demonstrated the over-expression of TF in a number of solid tumor types and its pathological roles in angiogenesis and tumor metastasis. In this study, we report the development and characterization of a panel of murine MAbs that are specific for human TF, but do not inhibit TF-mediated blood coagulation. By using a modified repetitive immunizations at multiple sites (RIMMS) protocol in conjunction with an efficient hybridoma cloning procedure, anti-TF MAbs were generated within a relatively short time frame of 5-6 weeks. Following primary screening by ELISA, the binding of the MAbs to the native form of human TF was demonstrated in flow cytometry using a stable cell line expressing human TF. Several of these TF-specific MAbs did not inhibit blood coagulation in a blood coagulation assay and bound with high affinity (0.5-2 nM) to human TF in BIAcore analyses. Importantly, this study represents an independent evaluation of the RIMMS strategy for MAb generation and demonstrates that class-switched, high-affinity MAbs can be generated rapidly and reliably using RIMMS.

There is considerable interest in developing immunotherapeutic approaches to elicit tumor-specific CTL responses in cancer patients. Epitope-based approaches aim to deliver the antigenic peptides or epitopes recognized by CTLs rather than the intact tumor antigen. Many tumor-associated proteins are nonmutated self proteins for which the dominant peptide epitopes are usually poorly immunogenic. The subdominant epitopes, however, can elicit robust T cell responses if optimized for their ability to bind to class I MHC molecules. Only the epitopes for a few tumor antigens expressed in human cancers have been defined to this level, mainly for technical reasons. The means to rapidly screen and characterize the binding of epitopes derived from complex tumor-associated antigens is an important enabling technology. Here, we have used the high-throughput technology iTopia to identify those peptides derived from the tumor-associated antigen survivin that bind 8 class I alleles. A library of overlapping nonamers spanning the length of the survivin protein was initially screened for peptides capable of binding each allele. Nineteen HLA-A*0201, zero HLA-A*0101, seven HLA-A*0301, twelve HLA-A*1101, twenty-four HLA-A*2402, six HLA-B*0702, six HLA-B*0801, and eight HLA-B*1501 binding peptides were identified based on an arbitrary cutoff. Peptides capable of binding a given allele were further characterized by their affinity for MHC class I molecules and by the rate of dissociation of the complex. This information should help guide functional studies and future epitope-based immunotherapies.

Tunable phase shift up to 360 degrees at 1 THz is achieved with a liquid crystal (LC) device. The key to this design is (1) the use of a nematic LC, E7, which exhibits a birefringence of ~ 0.17 (0.2 - 1.2 THz); (2) a LC cell (3-mm in thickness) with sandwiched structure to increase the interaction length while minimizing interface Fresnel losses; and (3) the use of magnetic field to align the thick LC cell and achieve continuous tuning of phase from 0 to 360 degrees . This device can be operated over a broad range near room temperature.

The optical constants of a nematic liquid crystal, 4'-n-pentyl-4-cyanobiphenyl (5CB), in the frequency range 0.3-1.4 THz were determined by terahertz (THz) time-domain spectroscopy. The real parts of the extraordinary refractive index n(e) and the ordinary refractive index n(o) of 5CB varied from 1.74 to 2.04 and from 1.59 to 1.83, respectively. Liquid-crystal 5CB exhibits a relatively small absorption loss in this frequency range. The birefringence of 5CB was found to be as large as 0.21. The experimental results indicate that liquid crystal 5CB is potentially useful for device applications in the THz frequency range.
