RESUMEN

When searching over associations between congenital ear abnormalities, especially microtia and affiliated deformities like cleft lip or palate and congenital heart diseases, some clinical analysis and genetic theories are found. A 10-year-old boy sent to the plastic surgery hospital was puzzled by a congenital anterior auricular fistula with fluid trace for more than 9 years. The preoperative diagnoses were branchial cleft fistula and congenital left ear deformity with postoperation of TOF. By browsing over studies on genetic concerns and clinical performance, it may be attributed to a possible association between microtia, branchial cleft fistula, and tetralogy of Fallot, though whose fundamental mechanisms remain concerned.

The gut bacterial microbiota of insects plays a crucial role in physiological, metabolic, and innate immune processes. In the current study, the gut bacterial communities of an insecticide-susceptible (IS), and a resistant (IR) population of a major legume pest, Megalurothrips usitatus (Bagnall), were evaluated. The 16S rDNA V3 + V4 regions of M. usitatus infected with Beauveria brongniartii along with the intestinal flora of both populations were sequenced based on a High-throughput sequencing platform. Toxicological bioassays revealed that the IR population exhibited resistance to acetamiprid and B. brongniartii isolate SB010 at levels of 138.0-fold and 55.6-fold higher, respectively, compared to the IS population. Through 16S High-throughput sequencing, the results indicate that both resistant populations, as well as B. brongniartii infestation, reduce the number of species of M. usitatus gut microbes. Using KEGG function prediction, it was found that most intestinal bacteria were involved in various metabolic activities, and the abundance of resistant populations was higher than that of sensitive populations. The bacteria in the gut of M. usitatus are mainly involved in various metabolic activities to achieve the degradation of B. brongniartii. This study provides valuable insights into the interaction between gut bacteria, insecticide resistance, and Beauveria. brongniartii infection in Megalurothrips usitatus, which can help inform future pest control strategies.

Fluralaner, a systemic pesticide, was originally registered with the US Food and Drug Administration in 2014 under the trade name Bravecto for flea treatment for pets. As a GABA antagonist, the footprint of fluralaner has expended beyond medical and veterinary pests in recent years. In this study, we examined the acute toxicity of fluralaner against three pests of Henosepilachna vigintioctopunctata, Megalurothrips usitatus, and Phyllotreta striolata in the Solanaceae, Fabaceae, and Cruciferae families, respectively, and the sublethal impact of fluralaner on Propylaea japonica, a widely distributed predatory ladybeetle. Based on LC50, fluralaner was effective against H. vigintioctopunctata (0.098 mg a.i. L-1 for the second instar larvae), M. usitatus (0.134 mg a.i. L-1 for adult females), and P. striolata (0.595 mg a.i. L-1 for adults). For P. japonica, however, fluralaner was substantially less effective (1.177 mg a.i. L-1 for the third instar larvae). Furthermore, the LC10 and LC30 of P. japonica were also consistently higher than the LC50 of the three pests. In addition, we did not observe any significant impacts of fluralaner at LC10 and LC30 on the life history traits, including body weight, developmental time, pre-oviposition period, and fecundity of P. japonica. Based on our results from acute toxicities and sublethal impacts, fluralaner is effective against vegetable pests, while potentially friendly to P. japonica when employed as a biological control agent.

Megalurothrips usitatus (Bagrall) is an important pest of legumes worldwide, causing great economic loss every year. Beauveria brongniartii and Akanthomyces attenuatus have shown considerable pathogenicity against M. usitatus in our previous studies. The medial lethal concentration (LC50) and the sublethal lethal concentration (LC25) of B. brongniartii isolate SB010 against M. usitatus were 8.38 × 105 and 1.73 × 105 conidia mL-1, respectively, whereas those of A. attenuatus isolate SCAUDCL-53 against M. usitatus were 4.37 × 105 and 2.97 × 104 conidia mL-1, respectively. This study reports the transcriptome-based explanation of the stress responses of M. usitatus following the application of B. brongniartii and A. attenuatus. The analysis of the transcriptomic data revealed the expression of 254, 207, 195, and 234 immunity-related unigenes by M. usitatus in response to B. brongniartii LC50 (SB1), B. brongniartii LC25 (SB2), A. attenuatus LC50 (V1), and A. attenuatus LC25 (V2), respectively. The biological function and metabolic pathway analyses showed that these unigenes were mainly related to pattern recognition receptors, information transduction factors, and reaction factors, such as scavenger receptor, cytochrome b5, cuticle protein, lysozyme, and serine protease.

A chitinase and a chitosanase were induced from a squid pen powder (SPP)-containing medium of Bacillus cereus TKU030 and purified by precipitation with ammonium sulphate and combined column chromatography. The purified chitinase and chitosanase exhibited optimum activity at 60 °C, pH 5-6 and 40 °C, pH 4, respectively. The chitinase and chitosanase were stable at 25-60 °C, pH 4-7 and 25-50 °C, pH 3-7, respectively. The chitinase and chitosanase showed the highest activity toward ß-chitin and 60% DD chitosan, respectively. The chitinase was significantly inhibited by Mn(2+) and EDTA but activated by Cu(2+), Fe(2+) and Ca(2+). The chitosanase was significantly inhibited by Cu(2+), Fe(2+), Zn(2+), Mn(2+) and EDTA. The chitinase showed high stability in the presence of various surfactants, such as SDS, Tween 20, Tween 40 and Triton X-100. In contrast, these surfactants were inhibitors of the chitosanase. The chitinase and chitosanase were also inhibited by TKUPSP017, a small synthetic boron-containing molecule with a BF3K side-chain. However, TKUPSP017 enhanced the growth of B. cereus TKU030 in SPP-containing medium.
