RESUMEN

Hyperhomocysteinemia is an independent risk factor for atherosclerosis and vascular occlusive diseases. Measurements of total homocysteine (Hcy) require an accurate reproducible method. A high-performance liquid chromatographic (HPCL) method for evaluating plasma Hcy after reduction with 2-mercaptoethanol and precolumn derivatization with iodoacetic acid and o-phthaldialdehyde is described. The resultant derivatives were separated on reverse-phase column C18 in the isocratic HPCL mode with fluorimetric detection. The detection limit for Hcy is 0.8 mumol/liter. The concentration of Hcy after overnight fasting was increased significantly (p < 0.02) in the patients in comparison with the control (11.7 +/- 1.1 vs. 8.3 +/- 0.5 mumol/liter). The method can be used for measuring the concentrations of asparaginic acid, glutamic acid, cysteine, asparagine, serine, and glutamine.

RESUMEN

For studying diagnostic possibilities of a new laboratory test for cerebral ischemia (CIS-test), thirty patients have been examined. The level of autoantibodies to NMDA type glutamate receptors is shown to increase significantly (3-5 times) in patients with stage 2 dyscirculatory encephalopathy. Parallel measurements of free glutamate and homocysteine levels in the patients blood plasma did not reveal any substantial changes of the given parameters, comparing to indices of autoantibodies to NR2a glutamate receptor. The titer of autoantibodies to this receptor type did not change significantly in patients with epilepsy; the titer dependence on dyscirculatory encephalopathy severity level and its decrease during treatment have been found as well. The possibilities of CIS using for diagnosis and therapy control in patients with chronic cerebral circulation disorders are discussed.

RESUMEN

A method for chromatographic analysis of human serum amino acids is proposed. Orthophthalic aldehyde in combination with 2-mercaptoethanol or sodium sulfite as a reagent for amino acid transfer into derivatives permits the identification of 15 amino acids within the framework of a single chromatographic system with an isocratic elution regimen. Glutamic acid, asparagine, serine, glutamine, histidine, taurine, alanine, arginine, methionine, isoleucin, ornithine, leucin, phenylalanine, lysin, and triptophane were measured in the sera of healthy donors and patients with ischemic stroke.

RESUMEN

Concentration of autoantibodies to human brain NMDA-receptor in blood serum of patients with acute brain ischemic stroke was significantly higher than in healthy persons and patients with other neurological diseases. Average content of cholesterol, lipid peroxides and glutamate in the serum were close in all patient groups investigated. It was suggested that the content of autoantibodies to human brain NMDA-receptor could be used as biochemical criteria for acute brain stroke patient's condition control and their treatment efficiency.
