RESUMEN

Probiotics and phytobiotics have demonstrated effective improvement of gut health in broiler chickens when individually administered in-ovo. However, their combined use in-ovo, has not been studied to date. We coined the term "prophybiotic" (probiotic + phytobiotic) for such a combination. The current study therefore, aimed to elucidate the effects of combined use of a selected probiotic and a phytobiotic in-ovo, on broiler gut health and production parameters, as opposed to use of probiotics alone. ROSS 308 hatching eggs were injected with either Leuconostoc mesenteroides (probiotic: PB) or L. mesenteroides with garlic aqueous extract (prophyiotic: PPB) on the 12th day of incubation. Relative abundances of bacteria in feces and cecal content (qPCR), immune related gene expression in cecal mucosa (qPCR) and histomorphology of cecal tissue (PAS staining) were analyzed along with production parameters (hatch quality, body weight, feed efficiency and slaughter and meat quality). PPB treatment increased the abundance of faecalibacteria and bifidobacteria in feces (d 7) and Akkermansia sp. in cecal content. Moreover, it decreased Escherichia coli abundance in both feces (d 34) and cecal content. PB treatment only increased the faecalibacteria in feces (d 7) and Akkermansia sp. in the cecal content. Moreover, PPB treatment resulted in up-regulation of immune related genes (Avian beta defensing 1, Free fatty acid receptor 2 and Mucin 6) and increased the crypt depth in ceca whereas PB treatment demonstrated a higher crypt depth and a tendency to increase Mucin 6 gene expression. Both treatments did not impair the production parameters studied. In conclusion, our results suggest that in-ovo PPB treatment may have enhanced potential in boosting the immune system without compromising broiler production and efficiency, as compared to the use of probiotic alone. Our study, highlights the potential of carefully selected PPB combinations for better results in improving gut health of broiler chickens.

The aim of this study was to investigate whether injecting synbiotics to the egg air chamber on d 12 of embryo incubation will affect the processes of angiogenesis, and thus the share of histopathological changes in superficial pectoral muscle, as well as Ca and P in blood of 42-day-old broiler chickens. The eggs containing viable embryos were injected with 0.2 mL suspension of 1/physiological saline, 2/SYN1 composed of galactooligosaccharide (GOS) (trade name: Bi2tos®, Clasado Biosciences Ltd, UK) and L. salivarius or 3/SYN2 composed of RFO and L. plantarum. All birds were fed ad libitum the standard commercial feed mixtures: starter, grower, and finisher, with a constant access to water and feed. Injecting synbiotics in ovo on d 12 of the embryonal development significantly affected the blood supply to superficial breast muscle in broiler chickens. The highest density of capillaries in the muscle area under study and per muscle fiber were identified in the group of birds the egg air chamber of which was provided with synbiotic GOS+L. salivarius. Consequently, for the muscles of the birds injected with the same synbiotic there was found the highest share of normal fibers and least necrosis and splitting, as compared with the control. The conducted research confirms the relationship between the blood supply to the muscle and the occurrence of pathological changes. We have observed a positive effect of synbiotics on the microvascularization and the size of histopathological changes in the chicken muscle, which, from a practical perspective, can affect the health status and the meat quality. Blood biochemical analyses showed that the in ovo injection of synbiotics did not significantly affect the level of parameters, except for Ca and P. A significant increase in the concentration of these minerals in the blood of chickens injected with SYN1 could have a positive effect on the angiogenesis process.

The aim of the study was to compare production results and quality of meat, as well as histological features of the jejunum in broiler chickens administered feed with 1% addition of zeolite or halloysite, with the addition of aluminosilicates to litter (4.50 kg/m2) throughout the rearing. In the experiment, 300 male broilers were used for 6 wk. They were divided into 3 groups, each of 10 repetitions (10 birds each). Group 1 was a control, halloysite was added to feed and litter in group 2, while zeolite was applied instead of halloysite in group 3. After rearing, 10 selected birds from each group were slaughtered. Selected production properties and degree of footpad dermatitis were examined, and histomorphometric examination of the jejunum was performed. The lowest yield and the highest proportion of neck with skin in the carcass were demonstrated in group 2 (P < 0.05). The lowest percentage of skin with subcutaneous fat was found in group 3 (P < 0.05). A decrease in lightness (L∗) and yellowness (b∗) was demonstrated in group 2, while redness (a∗) was the lowest in group 3 (P < 0.05). Group 2 was characterized by the lowest water-holding capacity in breast muscles, and in group 3, in leg muscles (P < 0.05). In group 3, the highest fat content and the lowest water content in the breast muscles (P < 0.05) were found. The leg muscles in groups 2 and 3 were characterized by the highest a∗, and in group 2, by b∗. The control group had the lowest protein and the highest fat content in leg muscles. In the intestine from group 2, a higher height (P < 0.05) and surface area (P < 0.01) of intestinal villi were found, in comparison to group 3. The width of intestinal villi was higher in groups 2 and 1 than in group 3 (P < 0.05), similarly the depth of intestinal crypts. The addition of zeolite could be proposed because of the obtained production results, while the halloysite had a positive effect on the histomorphometric features of the jejunum.

: The aim of the study was to evaluate the effects of a trans-galactooligosaccharide prebiotic (GOS) on the growth performance, biochemical blood parameters, and intestine morphometric parameters of common carp. The 60-day-long experiment was performed on one-year-old fish with a mean body weight of 180 g (±5 g). Three diets were used: control diet 1 (C) with no microbiota affecting feed additives, diet 2 (B1) with 1% of prebiotic, and diet 3 (B2) with 2% of prebiotic, in four replications (tanks) per treatment and 25 fish per tank. At the end of the trial, 16 individuals from each group were used for analyses. The study showed that GOS supplementation did not affect growth performance. In turn, the prebiotic had a positive effect on the development of the intestine, and increased the height, width, and surface of the villi in B1 and B2 groups. The content of phosphorus (P) was significantly higher in B1 group compared with B2 group, which indicated that 1% addition of prebiotic causes better absorption of P from the intestine. The other biochemical indicators-namely lipid, protein and hepatic parameters, insulin, and Ca-were not affected by GOS treatment, which may indicate similar metabolic balance of fish in each experimental group. Serum triiodothyronine (TT3) and glucose (stress markers) concentrations were not significantly different among treatments groups. GOS may be recommended as a feed additive for common carp due to its positive effects on fish physiology and development of the gastrointestinal tract. However, our results suggest that 1% diet supplementation causes satisfactory reactions for the abovementioned aspects in comparison to control or 2% supplementation.

The aim of the study was to determine the effect of dietary supplementation of a probiotic and a synbiotic on the morphometric parameters of the small intestine of broiler chickens. The experiment was conducted on three hundred sixty, one-day-old female Ross 308 chicks, which were randomly selected from 20,000 birds and divided into three treatment groups (n = 120) with ten replicates per treatment. The control group (C) was fed a commercial diet, the probiotic group (PRO) was fed the same diet with an added 1% of the probiotic Lavipan® (Lactococcus lactis, Carnobacterium divergens, Lactobacillus casei, Lactobacillus plantarum and Saccharomyces cerevisiae), and the synbiotic group (SYN) was fed the commercial diet with an added synbiotic: 0.8% of the prebiotic RFO (extracted from lupin seeds) and 1% Lavipan®. According to the manufacturer's data, apart from the typical probiotic action,microorganisms contained in the preparation release anti-bacterial substances (hydrogen peroxide and bacteriocins) and, therefore, are antagonistic towards pathogenic bacteria present in the gut of animals. Supplementation took place in the first seven days of rearing, and all birds had ad libitum access to water and feed during the 42 days of the experiment. On the last day, all birds were slaughtered and samples from three segments of the small intestine were taken. Villi area, height, width and crypt depth ratios were read using Multiscan software. Synbiotic supplementation increased the BWG of broilers from first to tenth day of rearing, compared to the control group. The PRO group had improved villi morphometric parameters of the duodenum. In the jejunum and ileum, both bioactive substances improved villus width and villus surface area. Crypts were deeper in the small intestine of birds supplemented with bioactive substances, which allows greater renewal of the villi. As expected, the intestinal morphometric parameters of broiler chickens benefited from bioactive substance supplementation.

BACKGROUND: Among various feed additives currently used in poultry nutrition, an important role is played by bioactive substances, including prebiotics. The beneficial effect of these bioactive substances on the gastrointestinal tract and immune system give rise to improvements in broiler health and performance nutrition, thus increasing the productivity of these birds. An innovative method for introducing bioactive substances into chickens is the in ovo injection into eggs intended for hatching. The aim of the study was to evaluate the development of histomorphological parameters of the duodenum and productivity in chickens injected in ovo with the prebiotic DiNovo® (extract of Laminaria species of seaweed, BioAtlantis Ltd., Ireland) on d 12 of incubation, under large - scale, high density poultry production conditions. RESULTS: There was no significant impact of the injection of DiNovo® prebiotic on the production parameters of broiler chickens (body weight, FCR, EBI and mortality) obtained on d 42 of rearing. No significant impact of the DiNovo® injection on the duodenum weight and length was observed, as well as on the CSA, diameter and muscular layer thickness of the duodenum. The in ovo injection of DiNovo® significantly increased the width of the duodenal villi (P < 0.05) and crypt depth (P < 0.01) of chickens on d 21 of rearing. Other histomorphological parameters of duodenal villi at d 42 of chickens rearing such as: the height, width, and cross section area of villi were significantly greater in chickens from the control group compared to those from the DiNovo® group (P < 0.05 and P < 0.01). CONCLUSIONS: In conclusion, this study demonstrates that injection of DiNovo® prebiotic into the air chamber of egg significantly influences the histomorphological parameters on d 21 of rearing without negatively affecting productivity in chickens at the end of rearing.

The objective of the study was to determine the effect of prebiotics and synbiotics administered in ovo on the 12th day of incubation, on the development of the intestinal villi and the number of goblet cells in the small intestine of broiler chickens on the Ist and the 4th days of life of chicks. Two prebiotics: inulin (PI) or Bi2tos (PB) and two synbiotics: inulin + L. lactis subsp. lactis (SI) or Bi2tos + L. lactis subsp. cremoris (SB) were injected in ovo on the 12th day of embryonic development. The control group of the embryos was injected with physiological saline (C). On the 1st day of life, an increase in the height of the villi in the jejunum was reported as a result of the injection of pre- and synbiotics, moreover an increase in the surface area of the villi in the jejunum and the duodenum in chicks from the SB group was also observed. A stimulatory effect of synbiotics on the morphology of the duodenum and thejejunum was also observed on the 4th dh day after hatching. Conversely, in the ileum, in the SB group, a reduction in the height of villi was found both on thse 1t and thte 4h days of life. In contrast, injection of inulin and synbiotic with the addition of inulin resulted in an increase in the number of goblet cells in the duodenum and the jejunum on thst 1" day of life, and caused a significant decrease on the 4th day after hatching.
