RESUMEN

The present study investigates the effects of orange peels derived pectin (OPDP) on skin mucus and serum immune parameters, disease resistance and growth performance of O. niloticus cultured under indoor biofloc system. Six hundred Nile tilapia (average weight 9.09 ±â€¯0.05 g) were distributed into 15 fiber tanks (300 L per tank) assigned to five treatments repeated in triplicate. Fish were fed experimental diets contain different levels OPDP as follows: 0 (control in clear water), 0 (control in biofloc system), 5, 10, and 20 g kg-1 OPDP for 8 weeks. At weeks 4 and 8 post feeding, skin mucus lysozyme (SMLA), peroxidase activities (SMPA), serum lysozyme (SL), serum peroxidase (SP), alternative complement (ACH50), phagocytosis (PI), and respiratory burst activities (RB) as well specific growth rate (SGR), weight gain (WG), final weight (FW), and feed conversion ratio (FCR) were measured. Also, resistance against Streptococcus agalactiae was assessed after 8 weeks post-feeding. Nile tilapia fed OPDP supplemented diets had significantly higher SMLA and SMPA compared to the controls (P < 0.05). The maximum values were observed in tilapia fed 10 g kg-1 OPDP followed by 5 and 20 g kg-1 OPDP. Nevertheless, no significant differences were observed between these two supplemented diets and between the control groups (P > 0.05). Regarding the serum immunological parameters, dietary inclusion of 10 g kg-1 OPDP showed significant higher SL and PI than other supplemented groups and control groups (P < 0.05). However, no significant differences were observed in SL and PI of fish fed 5 and 20 g kg-1 OPDP (P > 0.05). Dietary administration of OPDP significantly increased SP and ACH50 compared to the controls (P < 0.05), regardless of inclusion level. Additionally, non-significant change was found in RB of OPDP fed fish when compared with the controls (P > 0.05). The challenge test revealed that relative percent of survival (RPS) in OPDP treatments were 45.45%, 81.82%, 50%, respectively. The highest RPS was noticed in fish fed 10 g kg-1 OPDP. Furthermore, dietary administration of OPDP significantly improved SGR, WG, FW, and FCR (P < 0.05). Overall, the present findings suggested that OPDP can be taken into account as functional feed additives for O. niloticus.

Infectious diseases have been found to be a major cause of mortality in fish hatcheries. Probiotics have been introduced to replace antibiotics commonly used for treatment of bacterial infection in aquaculture. This study was conducted to isolate, screen, and evaluate the probiotic Bacillus spp. for potential use as a feed supplement to enhance fish growth, disease resistance and innate immunity of Pla-mong Pangasius bocourti. Bacillus aerius strain B81e was selectively isolated from the intestine of healthy catfish and chosen based on its probiotic properties both in vitro and in vivo. This bacterium produced a bacteriocin-like substance and exhibited a broad-spectrum antibacterial activity inhibiting both Gram-positive and Gram-negative bacteria especially the fish pathogens Aeromonas hydrophila and Streptococcus agalactiae. The susceptibility to all 8 antibiotics tested implies that it is unlikely to be an antibiotic-resistant bacterium. B. aerius strain B81e possessed interesting adhesion properties as shown by its high percentages of hydrophobicity, auto-aggregation, co-aggregation with fish pathogens A. hydrophila FW52 and S. agalactiae F3S and mucin binding. The strain B81e survived simulated gastrointestinal conditions, producing protease and lipase but not ß-haemolysin. The study also evaluated the effects of dietary supplementation with strain B81e on growth performance, innate immunity, and the disease resistance of P. bocourti against A. hydrophila infection. Fish with a mean body weight of 69 g were fed strain B81e at 0 (control) and 107 CFU g-1 feed (test) for 60 days. Various growth and immune parameters were examined at 30 and 60 days post-feeding. Fish were challenged with A. hydrophila 60 days post-feeding and mortalities were recorded over 14 days post-infection. Results showed that the administration of strain B81e for 60 days had significant effects (p < 0.05) on weight gain, specific growth rate and feed utilization efficiency of P. bocourti. Dietary administration of strain B81e increased the serum lysozyme and bactericidal activities of P. bocourti significantly throughout the experimental period whereas the alternative complement, phagocytic and respiratory burst activities were significantly (p < 0.05) higher in the test fish compared to the control fish after 60 days of feeding. In addition, the fish fed a strain B81e supplemented diet had a significantly higher (p < 0.05) post-challenge survival rate than the control fish. The results in this study indicate that B. aerius B81e has beneficial effects on growth performance, innate immunity and disease resistance of P. bocourti. This is the first report on the probiotic roles of B. aerius in aquaculture.

The study was conducted to evaluate the effect of dietary phospholipids (PLs) on growth, lipid metabolism, and antioxidative status of hybrid snakehead (Channa argus × Channa maculata). Five isonitrogenous and isolipidic diets with graded levels of PLs (8.5, 19.3, 30.7, 41.5, and 50.8 g kg-1) were fed to triplicate groups of juveniles (initial body weight 12.6 ± 0.23 g) for 8 weeks. Results showed that dietary PL supplementation significantly improved growth of juveniles. The final body weight (FBW) and specific growth rate (SGR) significantly increased with dietary PLs increasing from 8.5 to 41.5 g kg-1 (P < 0.05). Fish fed with the diet containing 8.5 g kg-1 PLs showed higher feed conversion ratio (FCR) compared to the other treatments (P < 0.05). Survival rate (SR) was not affected by dietary PL levels (P > 0.05). Liver lipid contents, serum triglyceride (TG), and low-density lipoprotein cholesterol (LDL-C) contents significantly decreased with the increasing levels of dietary PLs (P < 0.05). However, serum total cholesterol (TC) and high-density lipoprotein cholesterol (HDL-C) contents and HDL-C/TC and HDL-C/LDL-C value significantly increased with increasing dietary PL levels (P < 0.05). The catalase (CAT), superoxide dismutase (SOD), and carnitine palmitoyl transferase I (CPT-1) activities in the liver significantly increased with incremental dietary PL level (P < 0.05), while the liver malondialdehyde (MDA) contents and fatty acid synthase (FAS) activity significantly reduced (P < 0.05). No significant difference was observed in the glutathione peroxidase (GPx) activity among dietary treatments (P > 0.05).These results confirmed that dietary PL supplementation has beneficial effects on growth performance and antioxidant capacity of juvenile hybrid snakehead. Dietary PLs might reduce lipid deposition in the liver of juvenile hybrid snakehead.

The effects of oral administration of Astragalus polysaccharides (APS) and chitooligosaccharides (COS), single or combined, on the growth performance, immunity and disease resistance of M. salmoides were investigated. Largemouth bass juvenile were divided into 4 groups and each group was fed with diets supplemented with or without immunostimulant for 8 weeks. After 8 weeks of feeding trial, five fish per tank were sampled for immunity determination, ten fish per tank were challenged by A. hydrophila. The results showed that the largemouth bass fed with two immunostimulants alone or in combination significantly enhanced the final weight and specific growth rate (SGR), decreased feed conversion ratio (FCR) (P < 0.05). However, there were no significant differences (P < 0.05) in specific growth rate (SGR) between dietary COS and dietary APS. In addition, both COS and APS upregulated respiratory burst activity (RBA), phagocytic activity (PA), lysozyme activity and superoxide dismutase (SOD) activity. Meanwhile, COS also exhibited a increase in total leukocyte count, nitric oxide (NO) content and inducible nitric oxide synthase (iNOS) activity compared to the control. When challenged with A. hydrophila, the mortality of groups fed with COS and/or APS was lower than the control (P < 0.05). Under the experimental conditions, dietary APS and COS had a synergistic effect on lysozme activity, iNOS activity, NO content and disease resistance of fish (P < 0.05).

The use of probiotic bacteria to control bacterial infection in farmed fish is of clear practical interest. The aims of this study were to isolate and select a probiotic Bacillus sp. and to evaluate the effects of its supplementation on the growth and disease resistance of hybrid catfish. Bacillus siamensis strain B44v, selectively isolated from Thai pickled vegetables (Phak-dong), displayed a high potential as a probiotic in catfish culture. This bacterium produced a bacteriocin-like substance and exhibited a broad-spectrum antibacterial activity inhibiting both Gram-positive and Gram-negative bacteria, especially the fish pathogens Aeromonas hydrophila and Streptococcus agalactiae. The susceptibility to all 14 antibiotics tested implies its less possibility to be the antibiotic-resistant bacterium. Bacillus siamensis strain B44v possessed interesting adhesion properties, as shown by its high percentages of hydrophobicity (64.8%), auto-agglutination (73.8%), co-aggregation (67.2% with A. hydrophila FW52 and 63.5% with S. agalactiae F3S), and mucin binding (88.7%). The strain B44v survived simulated gastrointestinal conditions and produced protease and cellulase enzymes. Hybrid catfish (C. macrocephalus × C. gariepinus) were employed in the feed-trial experiments. Fish fed diet containing strain B44v (107 CFU/g feed) displayed not only no mortality but also growth improvement. At the end of the feed trial, fish were challenged by an intraperitoneal injection of Aeromonas hydrophila FW52. The Bacillus siamensis strain B44v fed fish survived (75.0%; p < 0.05) better than the controls (36.7%; p < 0.05) after a two week challenge. These collective results present for the first time the potential of Bacillus siamensis strain B44v for use as a bacterial probiotic in aquaculture.

This study was conducted to evaluate the effects of Eryngii mushroom, Pleurotus eryngii (PE), and Lactobacillus plantarum single or combined on growth, innate immune response and disease resistance of the Pangasius catfish, Pangasius bocourti. Two hundred forty fish were divided into four treatments, i.e., 0 g kg(-1) PE (Control, Diet 1), 3 g kg(-1) PE (Diet 2), 10(8) cfu g(-1) L. plantarum (Diet 3) and 3 g kg(-1) PE + 10(8) cfu g(-1) L. plantarum (Diet 4). Fish were culture in glass tanks with water volume approximately of 150 l, and each treatment had four replications with 15 fish per replication. Following 30, 60 and 90 days of the feeding trial, specific growth rate (SGR), feed conversion ratio (FCR), serum lysozyme, phagocytosis and respiratory burst activities were measured. SGR and FCR were significantly improved in fish fed supplemented diets after 90 days of the feeding trial. Serum lysozyme, phagocytosis and respiratory burst activity of fish were significantly stimulated by both PE and L. plantarum diets; however, the highest innate immune response was observed in fish fed synbiotic diet. At the end of the experiment, five fish were randomly selected for a challenge test against Aeromonas hydrophila. The post-challenge survival rate of the fish fed supplemented diets was significantly greater than the control treatment, and the highest post-challenge survival rate was observed in synbiotic diet. The results revealed that dietary supplementation of PE and L. plantarum stimulated growth, immunity and disease resistance of the P. bocourti.

Two experiments were carried out at the Khon Kaen University, Nong Khai Campus, Thailand during November, 2011 to May, 2012 to search for the most appropriate rates of feeding rations for fingerling and juvenile Asian redtail catfish (Hemibagrus wyckioides). A Completely Randomized Design (CRD) with three replications was used for both experiments. Experiment 1 had four treatments, i.e., T1 (2%), T2 (3%), T3 (4%, control), and T4 (5%). The same experimental design was used for the experiment 2, where four treatments were used, i.e., T1 (1%), T2 (2%), T3 (3%, control) and T4 (4%). The percentages of the amounts of rations used for Experiments 1 and 2 were based on body-live weight per day. Fingerling and juvenile fish were used for the experiments 1 and 2, respectively. Each experiment was carried out for 10 weeks. The results showed that a rate of 5% body live weight per day for fingerling fish of T4 of experiment 1 is the most appropriate rate for the fingerling fish. An increase in ration rate highly increased final body live weight. For juvenile fish of the experiment 2, it was found that there were no significant differences in live weights amongst the treated fish yet weight gained and specific growth rates between T1 and the rest were highly significant, whereas the results of the juvenile fish of T2 up to T4 were similar hence, 2% body live weight per day of T2 is the most appropriate rate for juvenile fish.

Two experiments were carried out at the Department of Fisheries, Khon Kaen University, Khon Kaen, Thailand during September 2013 to February 2014 aiming to pursue both the better growth performance and survival rate of the freshwater prawns (Macrobrachium lanchesteri) where a Completely Randomized Design (CRD) with three replications was used for each experiment. For experiment 1, different percentages of grass shelter were used, i.e. 0% (Control, T1), 25% (T2), 50% (T3) and 75% (T4). For experiment 2, different percentages of commercial chitosan were used, i.e. 0% (Control, T1), 25% (T2), 50% (T3) and 75% (T4). Both experiments were carried out at the same time and each of them was lasted for 75 days. The results showed that the added grass shelter gave significantly higher Total Weight (TW) and Survival Rate (SR) than the control treatment. However, there were no significant differences found on TW and SR among the added grass shelter treatments (T2 up to T4). Chitosan gave significantly higher TW, AW and Number of Molting (NM) than the control treatment. There were no significant differences on TW, NM and AW found amongst the chitosan treatments. The results of the experiment 1 showed that the best rate of grass shelter for the culturing of the Macrobrachium lanchesteri was at a rate of 25% (T 2) and the best rate of the chitosan was at a rate of 4% (Diet 3) for the Experiment 2.

This study evaluated effects of low molecular weight agar (LMWA) and Lactobacillus plantarum singly or combined on growth performance, immunity and disease resistance of basa fish (Pangasius bocourti). Two hundred forty fish were divided into four treatments, i.e. 0 g kg(-1) LMWA (Control), 2 g kg(-1) LMWA, 10(8) cfu g(-1)L. plantarum, and 2 g kg(-1) LMWA + 10(8) cfu g(-1)L. plantarum. Following 7, 14 and 28 days of the treatment, specific growth rate (SGR), feed conversion ratio (FCR), serum lysozyme, phagocytosis, respiratory burst and alternative complement activity (ACP) were measured. A Completely Randomized Design with four replications was applied. At the end of the feeding trial, five fish were randomly selected for a challenge test against Aeromonas hydrophila. The results showed that fish fed diet of 2 g kg(-1) LMWA and 10(8) cfu g(-1) of L. plantarum singly or combined significantly enhanced SGR, FCR, serum lysozyme, phagocytosis, respiratory burst, alternative complement activities and post-challenge survival rate of P. bocourti. The results inferred that dietary of LMWA and L. plantarum stimulated growth, immunity and disease resistance of the P. bocourti.

Random amplified polymorphic DNA (RAPD) and inter-simple sequence repeat (ISSR) markers were used to investigate the genetic structure of four subpopulations of Mystus nemurus in Thailand. The 7 RAPD and 7 ISSR primers were selected. Of 83 total RAPD fragments, 80 (96.39%) were polymorphic loci, and of 81 total ISSR fragments, 75 (92.59%) were polymorphic loci. Genetic variation and genetic differentiation obtained from RAPD fragments or ISSR fragments showed similar results. Percentage of polymorphic loci (%P), observed number of alleles, effective number of alleles, Nei's gene diversity (H) and Shannon's information index revealed moderate to high level of genetic variations within each M. nemurus subpopulation and overall population. High levels of genetic differentiations were received from pairwise unbiased genetic distance (D) and coefficient of differentiation. Mantel test between D or gene flow and geographical distance showed a low to moderate correlation. Analysis of molecular variance indicated that variations among subpopulations were higher than those within subpopulations. The UPGMA dendrograms, based on RAPD and ISSR, showing the genetic relationship among subpopulations are grouped into three clusters; Songkhla (SK) subpopulation was separated from the other subpopulations. The candidate species-specific and subpopulation-specific RAPD fragments were sequenced and used to design sequence-characterized amplified region primers which distinguished M. nemurus from other species and divided SK subpopulation from the other subpopulations. The markers used in this study should be useful for breeding programs and future aquacultural development of this species in Thailand.
