Nonalcoholic fatty liver disease (NAFLD), as the commonest chronic liver disease, is accompanied by liver oxidative stress and inflammatory responses. Herein, the extract obtained from Rubus corchorifolius fruits was purified and characterized for its polyphenol composition. The liver protective effect of the purified R. corchorifolius fruit extract (RCE) on mice with high-fat-diet (HFD)-induced NAFLD were investigated, and the potential mechanisms were explored through the integration of transcriptomics and metabolomics. Results showed that the polyphenolic compounds in RCE mainly included (-)-epigallocatechin, procyanidin B2, keracyanin, vanillin, dihydromyricetin, and ellagic acid. In addition, RCE intervention ameliorated liver and mitochondrial damage, which was evidenced by decreased indices of oxidative stress, liver function markers, and lipid profile levels. The liver metabonomics research revealed that RCE intervention affected the metabolic pathways of metabolites, including linoleic acid metabolism, galactose metabolism, alanine, aspartate and glutamate metabolism, retinol metabolism, glycine, serine and threonine metabolism, tryptophan metabolism, aminoacyl-tRNA biosynthesis, riboflavin metabolism, starch and sucrose metabolism, and arachidonic acid metabolism. Additionally, liver transcriptomics research indicated that pathways like fatty acid degradation, circadian rhythm, valine, leucine and isoleucine degradation, primary bile acid biosynthesis, cytokine-cytokine receptor interaction, adipocytokine signaling pathway, glutathione metabolism, lipid and atherosclerosis were significantly enriched. The transcriptomics and metabolomics analysis demonstrated that RCE intervention had significant modulatory effects on the metabolic pathways associated with glycolipid metabolism. Moreover, RT-PCR results verified that RCE intervention regulated liver mRNA levels associated with the inflammatory response. Therefore, our findings suggest that the intake of RCE might be an effective strategy to alleviate liver damage.

Plant-derived phenolics as natural α-glucosidase (α-GLU) inhibitors have attached great attention in the treatment of type-II diabetes mellitus currently. In this study, trans-polydatin and its aglycone resveratrol were found to show a notable inhibitory activity on α-GLU in a mixed-type manner with IC50 values of 18.07 and 16.73 µg/mL, respectively, which were further stronger than anti-diabetic drug acrabose (IC50 = 179.86 µg/mL). Multi-spectroscopic analysis results indicated that polydatin/resveratrol bound to α-GLU with one affinity binding site which was mainly driven by hydrogen bonds and van der Waals forces, and this binding process resulted in conformational alteration of α-GLU. In silico docking study showed that polydatin/resveratrol can well interact with the surrounding amino acid residues in the active cavity of α-GLU. Molecular dynamics simulation further clarified the structure and characterization of α-GLU-polydatin/resveratrol complexes. This study might supply a theoretical basis for the designing of novel functional foods with polydatin/resveratrol.

Surface treatment technology is an effective method to reinforce the durability of concrete. In this study, cement-based materials containing industrial solid wastes were modified by hybrid nano-silica (HN), then applied as a novel surface protection material (SPM-HN). The effect of SPM-HN on surface hardness of mortar matrix exposed to seawater was investigated. Further, the microstructure was characterized by X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), and mercury intrusion porosimetry (MIP). The results show SPM-HN could significantly enhance the surface hardness of matrix in seawater curing, and the rebound number is increased by 94%.The microstructure analysis demonstrates that the incorporation of HN inhibits the formation of ettringite, thaumasite, and Friedel's salt. In addition, thermodynamic modeling shows the incorporation of hybrid nano-silica could generate more C-S-H, and decrease the maximum volume of Friedel's salt when SPM is exposed to seawater. This research indicates SPM-HN can be applied as a concrete protective layer in the marine environment.

ETHNOPHARMACOLOGICAL RELEVANCE: Dengue is one of the most important pervasive diseases in many regions of the world, including China. There is an urgent need for new repellents, including plant derivatives, due to the resistance, toxicity, and non-degradability of synthetic insecticides. Traditional plant-based remedies may provide potential avenues for developing new strategies. AIMS OF THE STUDY: The aims of this study were to 1) document the traditional mosquitoes repellent plants used by the Dai people of Xishuangbanna, China; 2) screen out new efficient mosquito repellent plants as candidates for further study. MATERIALS AND METHODS: During the period August 2016 to July 2017, five field surveys were conducted in 16 villages of Xishuangbanna. A total of 81 informants (44 males and 37 females) were interviewed using semi-structured questions to collect detailed information on the plants they use to prevent mosquito bites. Ten plants with higher popularity and larger resource were collected and extracts were prepared by hydro-distillation or with petroleum ether. Extracts were tested for adult Aedes albopictus repellency using a human-bait cage. Firstly, repellency was determined as the Minimum Effective Dosage (MED) per minute at which 1% of the mosquito bite through the treated cloth. Secondly, five plant extracts with lower MEDs were tested the repellent longevity of different concentrations. RESULTS: Eighteen plants were documented as being used in traditional remedies against mosquitoes. The methods for controlling mosquitoes were diverse: direct burning was used for most plants (16 species), followed by smearing (5 species), and placing (5 species). Laboratory analyses confirmed that ten plants did exhibit mosquito repellent activity. Of them, Artemisia indica, Nicotiana tabacum, Blumea balsamifera, Vitex trifolia, and Chromolaena odorata showed good mosquito repellency with MEDs of 0.015, 0.061, 0.090, 0.090, and 0.105 mg/cm2, respectively. The protection rate provided by A. indica is also the highest among five plants. Although it provides complete protection time of only 30 min at 0.45 mg/cm2 concentration, its repellency within 2 h is not significantly different from that of DEET. CONCLUSION: Dai villagers in Xishuangbanna have a rich, diverse and scientific knowledge of plant-based mosquito repellents. Laboratory experiments screened out several plants as candidates for mosquito repellents, of which Artemisia indica was the most promising candidate plant.

Hematophagous invertebrates such as mosquitoes, leeches, mites, ticks, lice and bugs cause various problems for humans. Considering reports on insecticide resistance and requirements for improved environmental and toxicological profiles, there is a continuing need to discover and develop new insecticides and repellents. Ethnobotanical surveys of traditional plant-based repellents provide a direct method of identifying plants for potential use. During five field surveys in Bulang, Jinuo and Lahu villages between August 2018 and July 2019, semi-structured interviews were conducted with 237 informants (151 male, 86 female; mean age 63). Frequency of citation, use value, informant consensus factor and Jaccard index were employed to statistically analyze the collected data. A total of 709 use reports relating to 32 plant species and 71 remedies were collected. Similarities and differences between the three groups, as well as the Dai and Hani of Xishuangbanna, who were studied earlier, were shown through network analysis. These five ethnic groups living in the same area have a common understanding of traditional botanical knowledge against hematophagous invertebrates, but each group also possesses unique knowledge. Recording and protecting this traditional knowledge is potentially useful for protecting this cultural diversity and related biodiversity and can also have important practical applications. In this study, traditional knowledge provided us with many new potential plants for follow-up research for the development of new insecticides and repellents, among which Artemisia indica, Nicotiana tabacum and Clausena excavata are the most promising.

BACKGROUND: Indigo-dyed textiles have been central to the cultural identity of Landian Yao (literally "blue clothes Yao") people in Southwest China for centuries, driving a significant local market for naturally dyed indigo cloth. In the past two decades, local indigo production for traditional textiles has declined for several reasons: Firstly, the younger generation of Landian Yao has shifted to using western style jeans and T-shirts. Secondly, due to its labor-intensive nature. In contrast, at a global scale, including in China, there has been a revival of interest in natural indigo use. This is due to a growing awareness in the fashion industry about human and environmental health issues related to synthetic dye production. Ironically, this new awareness comes at a time when traditional knowledge of indigo dyeing is being lost in many places in China, with weaving and use of natural dyes now limited to some remote areas. In this study, we recorded indigo dyeing processes used by Landian Yao people and documented the plant species used for indigo dyeing. METHODS: Field surveys were conducted to the study area from September 2015 to November 2016, supplemented by follow-up visits in July 2018 and November 2018. We interviewed 46 key informants between 36 and 82 years old who still continued traditional indigo dyeing practices. Most were elderly people. Semi-structured interviews were used. During the field study, we kept a detailed account of the methods used by Landian Yao dyers. The data were then analyzed by using utilization frequency to determine the best traditional recipe of indigo dye extraction. All the specimens of documented species were collected and deposited at the herbarium of Kunming Institute of Botany. RESULTS: Our results showed that indigo dyeing was divided into two main steps: (1) indigo pigment extraction and (2) dyeing cloth. The general procedures of indigo dye extraction included building or buying a dye vat, fermentation, removal of the leaves of indigo producing plant species, addition of lime, oxygenation, followed by collection, and the storage of the indigo paste. The procedures of dyeing cloth included preparing the dye solutions, dyeing cloth, washing, and air drying. It is notable that Landian Yao dyers formerly only performed the dyeing process on the goat days in the lunar calendar from June to October. After comparing the range of local indigo extraction methods, our results showed that the following was best of these traditional recipes: a indigo-yielding plant material to tap water ratio of 30 kg: 200 l, lime 3 kg, a fermentation time of 2-3 d, aeration by agitation for up to 60 min, and a precipitation time of 2-3 h. Our results show that 17 plant species in 11 families were recorded in the indigo dyeing process. With the exception of the indigo sources, only Dioscorea cirrhosa Lour. and Artemisia argyi H.Lév. & Vaniot were previously recorded in dyeing processes. Other species given in this paper are recorded for the first time in terms of their use in the indigo dyeing process. In the study area, Landian Yao men were in charge of indigo dye extraction, and the women were responsible for dyeing cloth. CONCLUSIONS: The Landian Yao has completely mastered the traditional indigo dyeing craft and are one of the well-deserved identity blues. Indigo production from plants using traditional methods is a slow process compared to synthetic dyes and is not suitable for modern and rapid industrial production. Therefore, our study records the detailed information of traditional indigo dyeing to protect and inherit it. Strobilanthes cusia (Nees) Kuntze is the main indigo source in Landian Yao that is widely used in the world and can be commercially exploited as an indigo plant. For commercial and environment benefits, we suggest that producing natural indigo for the commercial market is a good choice.

BACKGROUND: Fleagrass, Adenosma buchneroides, is an aromatic perennial herb that occupies an important position in the life of the Akha people. They regard it as a tribal symbol and a gift of love. Fleagrass also has many medicinal uses, and there is considerable potential for its development as an insect repellent. Traditionally, Akha people plant it in swidden fields, but there are few swidden fields in China now. Therefore, the first question this study aims to answer is as follows: how is fleagrass planted and utilized now? At present, fleagrass is only reported to be used by Akha people in Mengla. We also try to understand the following questions: Is fleagrass used in nearby area? If so, how is fleagrass used in nearby area? Furthermore, why is fleagrass used in that way? METHODS: From August 2016 to July 2018, field surveys were conducted six times. The ethnobotanical and ethnopharmacological uses of A. buchneroides in 13 Akha villages were investigated by means of semi-structured interviews. We assessed the responses of a total of 64 interviewees (32 men and 32 women; mean age, 58.6) from the Xishuangbanna Dai Autonomous Prefecture, southwest China, and from Phongsaly Province, Laos. To explain the bases for the ethnobotanical uses of fleagrass, we used Google Scholar, Web of Science, and China National Knowledge Infrastructure to review the bioactivities of the chemical constituents of A. buchneroides. RESULTS: With the vanishing of swidden agriculture and the development of modern products, fleagrass cultivation is disappearing in China. However, most Akha people in Xishuangbanna still remember and yearn for its traditional uses, and Akha people in a nearby area (northern Laos) continue to plant and utilize it. We documented ten uses of A. buchneroides within five discrete categories. The whole plant of fleagrass has a distinct strong aroma, of which Akha villagers are particularly fond. Akha villagers mostly use this aromatic property as a decoration, perfume, and insect repellent. A. buchneroides is also used as a condiment and for medicinal and ritual purposes, including its use as a cure for insect bites, headaches, influenza, and diarrhoea, and as a part of pray ritual for a bumper harvest. From our literature review, we identified many major chemical compounds contained in the essential oil of A. buchneroides, including thymol, carvacrol, 3-carene, and p-cymene, which have insecticidal or insect-repellent, antimicrobial, and anti-inflammatory properties. CONCLUSION: Fleagrass is an aromatic plant that is widely used by Aka people. Its chemical composition also has a variety of biological activities. With the vanishing of swidden agriculture and the development of modern products, fleagrass utilization in China is disappearing and its cultural importance is reduced. However, its economic and medicinal value is assignable.

OBJECTIVE: To evaluate the immediate effect of moxibustion at Shenshu (BL 23) on renal hemodynamics in patients with chronic kidney disease(CKD)ï¼. METHODS: Thirty-two non-dialysis patients with CKD were recruited in this study. Moxibustion was applied at bilateral BL 23 for 30 min. The peak systolic velocity (PSV), end diastolic velocity (EDV) of the interlobar renal artery were measured by Doppler ultrasonography (US) before, and 0 and 15 min after moxibustion. The renal resistance index (RI) was equal to (PSVï¼EDV)/PSV. The body temperature (T), systolic blood pressure (SBP), diastolic blood pressure (DBP), heart rate (HR) were also monitored at the same time points. RESULTS: After moxibustion, the EVD level at 15 min after moxibustion was significantly increased (P<0.05), and the renal RI levels at 0 and 15 min after moxibustion were significantly down-regulated in comparison with their own pre-treatment (P<0.01). No significant changes were found in the levels of PSV, T, SBP, DBP and HR after moxibustion (P>0.05). CONCLUSION: Moxibustion at BL 23 can increase EDV and reduce renal RI of the interlobar renal artery in patients with CKD, which may be useful in relieving kidney disorders by improving renal blood flow.

OBJECTIVES: Regulatory cytokines are associated with viral infection. The objective of this study was to evaluate the relation between serum regulatory cytokines concentrations and respiratory syncytial virus (RSV) disease. METHODS: We enrolled 325 children agedâ¯<â¯24â¯months who were diagnosed with acute respiratory tract infection. Twenty age-matched healthy children were enrolled as controls. Nasopharyngeal swabs were analyzed to identify virus by reverse transcription polymerase chain reaction, and blood samples were taken to quantify the regulatory cytokine concentrations, including interleukin (IL)-35, IL-10 and transforming growth factor (TGF)-ß1 using the Bio-Plex immunoassay or enzyme-linked immunosorbent assay. RESULTS: RSV disease was associated with a great regulatory cytokine response than healthy children, among 89 RSV-infected patients, serum IL-35 (Pâ¯=â¯.0001) and IL-10 (Pâ¯=â¯.006) was significantly elevated in comparison with healthy controls. Young children (0< age ≤6 months) with RSV infection had significantly lower IL-35 and IL-10 expression but needed more oxygen therapy and more severe disease comparing with older children (12< age <24â¯months). Comparing with mild group, the expression levels of IL-10 were significantly lower in children with moderate and severe disease (Pâ¯=â¯.012 and Pâ¯=â¯.005, respectively). And levels of IL-10 was inversely associated with total duration of RSV infection symptoms (râ¯=â¯-â¯0.311, Pâ¯=â¯.019). CONCLUSION: Children with RSV infected had increased serum regulatory cytokine IL-10 and IL-35 concentrations. Elevated expression of IL-10 and IL-35 were contributed to protect hypoxia and reduce the severity of disease.

Respiratory syncytial virus (RSV) infection is the leading cause of acute respiratory tract disease in children less than 5 years old. The aim of this study was to further elucidate the molecular properties and clinical characteristics of RSV infection. The study sample included 238 patients <5 years old who were hospitalized with clinical symptoms of upper or lower respiratory tract infection (URTI or LRTI) in the Pediatric Department at the First People's Hospital of Chenzhou, South China in 2014. We subjected nasopharyngeal aspirate (NPA) or nasal swab (NS) samples from the patients to indirect fluorescence assay screens. RSV G genes were amplified by reverse transcription-PCR (RT-PCR) and sequenced. Of the 238 patients screened, 64 (26.8%) were confirmed to have RSV infections. Of those 64 confirmed RSV infection cases, 39 (60.9%) had subtype BA9, 13 (20.3%) had the recently identified subtype ON1, 11 (17.2%) had subtype NA1, and 1 (1.6%) had subtype GB2. The predominant presentation was LRTI with coughing, sputum production, fever, and wheezing. RSV subtype NA1 and BA9 infections were found mostly in infants, whereas the age distribution of subtype ON1 infections was more uniform across the age bands. Phylogenetic analysis indicated that, compared with the prototype strain A2, all ON1 and most NA1 isolates had lost one potential N-glycosylation site at amino acid 251 and 249 due to T251K and N249Y substitution, respectively. These findings suggest that NA1, BA9, and ON1 are the dominant RSV subtypes causing respiratory tract infections in young children presenting to the hospital in South China. J. Med. BACKGROUND/AIMS: Coronavirus (CoV) infections induce respiratory tract illnesses and central nervous system (CNS) diseases. We aimed to explore the cytokine expression profiles in hospitalized children with CoV-CNS and CoV-respiratory tract infections. METHODS: A total of 183 and 236 hospitalized children with acute encephalitis-like syndrome and respiratory tract infection, respectively, were screened for anti-CoV IgM antibodies. The expression profiles of multiple cytokines were determined in CoV-positive patients. RESULTS: Anti-CoV IgM antibodies were detected in 22/183 (12.02%) and 26/236 (11.02%) patients with acute encephalitis-like syndrome and respiratory tract infection, respectively. Cytokine analysis revealed that the level of serum granulocyte colony-stimulating factor (G-CSF) was significantly higher in both CoV-CNS and CoV-respiratory tract infection compared with healthy controls. Additionally, the serum level of granulocyte macrophage colony-stimulating factor (GM-CSF) was significantly higher in CoV-CNS infection than in CoV-respiratory tract infection. In patients with CoV-CNS infection, the levels of IL-6, IL-8, MCP-1, and GM-CSF were significantly higher in their cerebrospinal fluid samples than in matched serum samples. CONCLUSION: To the best of our knowledge, this is the first report showing a high incidence of CoV infection in hospitalized children, especially with CNS illness. The characteristic cytokine expression profiles in CoV infection indicate the importance of host immune response in disease progression.

ETHNOPHARMACOLOGICAL RELEVANCE: Traditional and local medicinal knowledge would be useful for finding pharmaceutical resources. Ethnopharmacological methods, especially quantitative analysis could help us to pre-screen herbs in field studies. "Using different plants as the same herb" is common in both traditional and local medicinal systems in China. In terms of medicine safety, it is not best choice, for it would lead to difficulty in controlling the quality, safety and efficacy of herbs. However, from the perspective of finding new resources for pharmaceutical industry, it would be beneficial. The usage of Huang-lian is one of the typical examples of "using different plants as the same herb". According to the previous Phytochemical and Pharmacological studies, berberine is the common effective compound of most of the species used as Huang-lian. Recently, berberine and other effective compounds of Huang-lian have gained much more attention and will become more popular in both medicinal researches and pharmaceutical industry. In our preliminary field work, we found that dozens of plant species might be used as Huang-lian by local people in Northwest Yunnan, an area well known by its rich biodiversity and culture diversity. These herbs might have potential value for pharmaceutical industry, for example, it could be used as the new resources to extract berberine and other effective compounds. Due to this, it is very necessary to collect, identify, document, and analyze the herbs used as Huang-lian in NW Yunnan. In the present study, we focused on that how to use traditional and local medicinal knowledge to find resources for pharmaceutical industry. MATERIAL AND METHODS: In the field work, interviews and participative observation were used. In the quantitative analysis of the local knowledge, Informant consensus factor (Fic), Use value (UV) and Relative frequency of citation (RFC) were used. RESULTS AND DISCUSSION: A total of 230 key informants were interviewed and 29 plant species belonging to 8 families and 11 genera used as Huang-lian were collected in the study area. Diarrhoea had the highest value of Fic.xiana had the highest value of UV and RFC. The main effective compounds of most of these species were related to the protoberberine group of isoquinoline alkaloids, e.g. berberine, jatrorrhizin and palmatine, according to the previous phytochemical studies. CONCLUSION: The range of sources of Huang-lian were very wide in NW Yunnan. Treating diarrhoea was the most common use of these species, most of which contained berberine. Based on the results of quantitative analysis, M. duclouxiana may had the greatest potential to future uses, e.g. as a resource for pharmaceutical industry. In the present study, we did not discuss whether the herbs used as Huang-lian could replace the standard Huang-lian in traditional or local medicine or not, and we just wanted to explore how this phenomenon could be used to find new resources for pharmaceutical industry.
