RESUMEN

Coordination cages sustained by metal-ligand interactions feature polyhedral architectures and well-defined hollow structures, which have attracted significant attention in recent years due to a variety of structure-guided promising applications. Sulfonylcalix[4]arenes-based coordination cages, termed metal-organic supercontainers (MOSCs), that possess unique multi-pore architectures containing an endo cavity and multiple exo cavities, are emerging as a new family of coordination cages. The well-defined built-in multiple binding domains of MOSCs allow the efficient encapsulation of guest molecules, especially for drug delivery. Here, we critically discuss the design strategy, and, most importantly, the recent advances in research surrounding cavity-specified host-guest chemistry and biomedical applications of MOSCs.

Combination therapies to induce mixed-type cell death and synthetic lethality have the potential to overcome drug resistance in cancer. In this study, we demonstrated that the curcumin-enhanced cytotoxicity of cisplatin/carboplatin in combination with gemcitabine was associated with Aurora A suppression-mediated G2/M arrest, and thus apoptosis, as well as MEK/ERK-mediated autophagy in human bladder cancer cells. Animal study data confirmed that curcumin combined with cisplatin/gemcitabine reduced tumorigenesis of xenograft in mice and this phenomenon was associated with elevated expressions of p-ERK and reduced p-Aurora A in tumors. Gene analyses using data repositories further revealed that reduced Aurora A expression alone did not significantly elevate the sensitivity of human bladder carcinoma cells to these anticancer drugs. Unlike other major cancer types, human bladder urothelial carcinoma tissue coexpressed higher AURKA and lower MAP1LC3B than normal tissue, and reduced Aurora A and induction of autophagy have been clinically associated with a better prognosis in patients with early but not advanced stage bladder cancer. Therefore, our results suggest that treatment strategies can utilize the synthetic lethal pair to concurrently suppress oncogenic Aurora A and induce autophagy by coadministrating curcumin with anticancer drugs for early-stage bladder cancer with high expression of Aurora A.

With the improvement of public requirements for the health status of aquatic ecosystems, there have been innovative assessment methods developed for aquatic ecosystems. In this study, benthic algae assemblages and water quality variables were analyzed to develop a benthic diatom-based index of biotic integrity (D-IBI) for the assessment of the aquatic environment in the Lalin River. In addition, using redundancy analysis (RDA) based on dominant species and physicochemical indexes, the ecological distribution characteristics of the benthic diatom community were revealed, and the key influencing factors were identified. The results showed that the benthic diatom community structure in the Lalin River basin had obvious spatial differences. The application of the index revealed that the water quality could be described as excellent condition in the upper reaches of the Lalin River, good to common condition in the middle of the sites, and moderate to poor condition in the downstream. The assessment further revealed that the main reason for the degradation of the Lalin River ecosystem was nutrient enrichment through agricultural land use.

Clear vertical variations in phytoplankton community structure are usually observed in deep-water lakes and reservoirs, which is one of the key components of water quality and ecosystem functioning. However, the vertical patterns and ecological drivers of phytoplankton communities in deep-water lakes and reservoirs are still understudied. In this study, we took Qiandao Lake, a deep-water reservoir, as an example to reveal the vertical distribution characteristics of phytoplankton communities and its influencing factors by investigating phytoplankton community structure and the associated water quality index at 12 sites across the whole lake in two seasons (spring and autumn). The results showed that the phytoplankton abundance and chlorophyll a were highest in the surface layer in autumn and then decreased toward deep water, whereas in spring, the maximum value occurred in the subsurface layer (2-5 m), and the dominant phytoplankton species showed obvious vertical stratification characteristics. Specifically, in spring, Cryptomonas and Pseudanabaena dominated the surface and subsurface layers, Cryptomonas dominated in the middle layer, and the abundance of Cyclotella at the bottom layer was significantly higher than that of the other algae genera. The dominant genera in autumn were Pseudanabaena and Aphanizomenon. In the subsurface and middle layers, Leptolyngbya and Pseudanabaena occupied the dominant position, and Leptolyngbya became the only dominant genus. In the bottom layer, Leptolyngbya was the only dominant genus. The key environmental indicators of the water also had obvious vertical changes. The contents of N and P nutrients had a negative correlation with the water depth in spring, whereas the reverse trend was observed in autumn. The correlation analysis showed that the vertical variation in phytoplankton abundance in spring was significantly positively correlated with phosphate concentration, whereas the vertical distribution of phytoplankton abundance in autumn was significantly positively correlated with intensity of light, and the water temperature, NH4+-N, and total nitrogen were the main factors driving the vertical changes in the dominant genera of phytoplankton community in the two seasons. To summarize, environmental conditions such as water temperature, light, and nutrients had strong effects on the vertical distribution of phytoplankton. In the ecological investigation and quality assessment of deep-water lakes and reservoirs, the vertical distribution characteristics of the phytoplankton community structure and the influence of environmental conditions should be fully considered.
