RESUMEN

There is an increased interest from transit agencies to replace fixed-route transit services with on-demand public transits (ODT). However, it is still unclear when and where such a service is efficient and sustainable. To this end, we provide a comprehensive framework for assessing the sustainability of ODT systems from the perspective of overall efficiency, environmental footprint, and social equity and inclusion. The proposed framework is illustrated by applying it to the Town of Innisfil, Ontario, where an ODT system has been implemented since 2017. It can be concluded that when there is adequate supply and no surge pricing, crowdsourced ODTs are the most cost-effective transit system when the demand is below 3.37 riders/km2/day. With surge pricing applied to crowdsourced ODTs, hybrid systems become the most cost-effective transit solution when demand ranges between 1.18 and 3.37 riders/km2/day. The use of private vehicles is more environmentally sustainable than providing public transit service at all demand levels below 3.37 riders/km2/day. However, the electrification of the public transit fleet along with optimized charging strategies can reduce total yearly GHG emissions by more than 98%. Furthermore, transit systems have similar equity distributions for waiting and in-vehicle travel times.

To curb the spread of the ongoing 2019 novel coronavirus (COVID-19), authorities have adopted several non-pharmaceutical (NPIs) and pharmaceutical interventions, which significantly affected our daily activities and mobility patterns. However, it is still unclear how severity of NPIs, COVID-19-related variables, and vaccination rates have affected demand for ridesourcing services, and whether these effects vary across small towns and large cities. We analyzed over 220 million ride requests in the City of Chicago (population: 2.7 million), Illinois, and 52 thousand in the Town of Innisfil (population: 37 thousand), Ontario, to investigate the impact of the COVID-19 pandemic on the ridesourcing demand in the two locations. Overall, the pandemic resulted in fewer trips in areas with higher proportions of seniors and more trips to parks and green spaces. Ridesourcing demand was adversely affected by the stringency index and COVID-19-related variables, and positively affected by vaccination rates. However, compared to Innisfil, ridesourcing services in Chicago experienced higher reductions in demand, were more affected by the number of hospitalizations and deaths, were less impacted by vaccination rates, and had lower recovery rates.

Applications of neuroimaging methods have substantially contributed to the scientific understanding of human factors during driving by providing a deeper insight into the neuro-cognitive aspects of driver brain. This has been achieved by conducting simulated (and occasionally, field) driving experiments while collecting driver brain signals of various types. Here, this sector of studies is comprehensively reviewed at both macro and micro scales. At the macro scale, bibliometric aspects of these studies are analysed. At the micro scale, different themes of neuroimaging driving behaviour research are identified and the findings within each theme are synthesised. The surveyed literature has reported on applications of four major brain imaging methods. These include Functional Magnetic Resonance Imaging (fMRI), Electroencephalography (EEG), Functional Near-Infrared Spectroscopy (fNIRS) and Magnetoencephalography (MEG), with the first two being the most common methods in this domain. While collecting driver fMRI signal has been particularly instrumental in studying neural correlates of intoxicated driving (e.g. alcohol or cannabis) or distracted driving, the EEG method has been predominantly utilised in relation to the efforts aiming at development of automatic fatigue/drowsiness detection systems, a topic to which the literature on neuro-ergonomics of driving particularly has shown a spike of interest within the last few years. The survey also reveals that topics such as driver brain activity in semi-automated settings or neural activity of drivers with brain injuries or chronic neurological conditions have by contrast been investigated to a very limited extent. Potential topics in driving behaviour research are identified that could benefit from the adoption of neuroimaging methods in future studies. In terms of practicality, while fMRI and MEG experiments have proven rather invasive and technologically challenging for adoption in driving behaviour research, EEG and fNIRS applications have been more diverse. They have even been tested beyond simulated driving settings, in field driving experiments. Advantages and limitations of each of these four neuroimaging methods in the context of driving behaviour experiments are outlined in the paper.

Rheumatoid arthritis is a chronic, systemic joint disease in which an autoimmune response translates into an inflammatory attack resulting in joint damage, disability and decreased quality of life. Despite recent introduction of therapeutic agents such as anti-TNFα, even the best current therapies fail to achieve disease remission in most arthritis patients. Therefore, research into the mechanisms governing the destructive inflammatory process in rheumatoid arthritis is of great importance and may reveal novel strategies for the therapeutic interventions. To gain deeper insight into its pathogensis, we have developed for the first time a three-dimensional synovium-on-a-chip system in order to monitor the onset and progression of inflammatory synovial tissue responses. In our study, patient-derived primary synovial organoids are cultivated on a single chip platform containing embedded organic-photodetector arrays for over a week in the absence and presence of tumor-necrosis-factor. Using a label-free and non-invasive optical light-scatter biosensing strategy inflammation-induced 3D tissue-level architectural changes were already detected after two days. We demonstrate that the integration of complex human synovial organ cultures in a lab-on-a-chip provides reproducible and reliable information on how systemic stress factors affect synovial tissue architectures.
