RESUMEN

Due to the exclusive maternal transmission, oocyte mitochondrial dysfunction reduces fertility rates, affects embryonic development, and programs offspring to metabolic diseases. However, mitochondrial DNA (mtDNA) are vulnerable to mutations during oocyte maturation, leading to mitochondrial nucleotide variations (mtSNVs) within a single oocyte, referring to mtDNA heteroplasmy. Obesity (OB) accounts for more than 40% of women at the reproductive age in the USA, but little is known about impacts of OB on mtSNVs in mature oocytes. It is found that OB reduces mtDNA content and increases mtSNVs in mature oocytes, which impairs mitochondrial energetic functions and oocyte quality. In mature oocytes, OB suppresses AMPK activity, aligned with an increased binding affinity of the ATF5-POLG protein complex to mutated mtDNA D-loop and protein-coding regions. Similarly, AMPK knockout increases the binding affinity of ATF5-POLG proteins to mutated mtDNA, leading to the replication of heteroplasmic mtDNA and impairing oocyte quality. Consistently, AMPK activation blocks the detrimental impacts of OB by preventing ATF5-POLG protein recruitment, improving oocyte maturation and mitochondrial energetics. Overall, the data uncover key features of AMPK activation in suppressing mtSNVs, and improving mitochondrial biogenesis and oocyte maturation in obese females.

Diazinon (DZN) is a refractory organophosphorus pesticide (OP) in the surrounding environment due to its overuse in agriculture. The antioxidant activity of Epigallocatechin gallate (EGCG) from green tea is at least 100 times greater than that of vitamin C. This study aimed to study the effects of DZN on the meiotic maturation of porcine oocytes, as well as the protective roles of EGCG. Firstly, the effects of DZN and EGCG on meiotic nuclear maturation of porcine oocytes were detected, and then embryonic development was investigated by chemical parthenogenetic activation. Next, the spindle assembly, mitochondrial membrane potential (MMP), reactive oxygen species (ROS), DNA damage, and finally the early apoptosis of oocytes were examined by immunofluorescence staining. The results revealed that DZN exposure significantly reduced the quality of porcine oocytes, such as failure of nuclear and cytoplasmic maturation, evidenced by abnormal spindle assembly, disordered chromosome alignment, low MMP, observably increased ROS, severe DNA damage, and early apoptosis. Appropriate EGCG could significantly reduce all these defects caused by DZN. In conclusion, EGCG can help prevent the harm that DZN exposure can do. These findings offer convincing support for enhancing the oocyte quality from EGCG through daily ordinary beverages.

AIMS: Organophosphorus pesticide diazinon (DZN) has adverse effects on animals and humans by direct contact or the spread of food chain. The antioxidant melatonin has protective effects on female reproduction. This study aimed to explore the effects of DZN on meiosis maturation in mouse cumulus oocyte complexes (COCs) and the effects of melatonin. MAIN METHODS: Different concentrations of DZN and melatonin were added during the in vitro maturation of COCs. Then we detected the extrusion rate of the first polar body, the number of sperms binding to oocyte, mitochondrial membrane potential, reactive oxygen species (ROS), early apoptosis. Subsequently, the expression of Juno, CX37, CX43 and ERK1/2 were detected by immunofluorescence staining and Western blotting. KEY FINDINGS: DZN exposure results in the failure of nuclear and cytoplasmic maturation of oocyte meiosis. Destruction of repositioning and function of mitochondria increases the levels of ROS and early apoptosis. The DZN-exposed oocytes express less Juno resulting to bind less sperms than normal. The loss of gap junctions and failure to activate ERK1/2 also contribute to the failure of cytoplasmic maturation. All these ultimately lead to the poor oocyte quality and low fertility. Appropriate melatonin can effectively restore all these defects. SIGNIFICANCE: Under DZN exposure, melatonin can significantly improve the quality of oocytes, and melatonin promotes oocyte maturation by protecting gap junction and restoring ERK1/2 pathway, which is a new breakthrough for improving female fertility.

The aim of this study was to examine the interactive effects of situational variables (competition stage, match location, and match outcome) on teams' technical performance in the UEFA Champion League. Match data of 19 technical actions and events were collected and classified into three groups (variables related to goal scoring, offense, and defense) during eight seasons (2009/2010-2016/2017). Repeated-measures analysis of variance (RMANOVA) was used to identify the differences in the technical performances among teams. Results showed that the significant differences in the technical performance between the group stage and the knockout stage were only found in dribble and yellow card. However, differences in the variables related to goal scoring and offense were more significant than in the variables related to defense under the comparisons across competition situations, and the differences in the variables related to defense were mainly detected in the yellow card. The number of variables that showed significant differences among match location and match outcome in the group stage were less than in the knockout stage. Therefore, the identified key performance indicators by considering the interactive effects of situational variables may provide detailed and practical insights for coaches to develop useful training interventions and match strategies for upcoming match playing in specific competition situations.

The characterization of monolignols H-, G- and S- units composition Vanholme et al. (2010) of lignin is important in agriculture, forestry, herb medicine, livestock, and health care research Vanholme et al. (2008) and Sticklen (2008). The conventional methods often require a great deal of samples and reagents and are time-consuming. Here, we present a newly developed method with fewer operations. The optimized method is suitable for detecting and characterizing lignin composition of cell wall in different plant species and has the advantages of: •Avoiding the influence of plasticizer by plasticware and enhancing the accuracy of monolignols analysis.•Lowering the required samples from grams to milligrams, and organic reagents from milliliters to microliters.•Reducing the time required from a few days to 6 h.

The aim of the present study was to investigate the validity of match variables and the reliability of Champdas Master System used by trained operators in live association football match. Twenty professional football coaches voluntarily participated in the validation of match variables used in the System. Four well-trained operators divided into two groups that independently analyzed a match of Spanish La Liga. The Aiken's V averaged at 0.84 ± 0.03 and 0.85 ± 0.03 for the validation of indicators. The high Kappa values (Operator 1: 0.92, 0.90; Operator 2: 0.91, 0.88), high intra-class correlation coefficients (varied from 0.93 to 1.00), and low typical errors (varied from 0.01 to 0.34) between the first and second data collection represented a high level of intra-operator reliability. The Kappa values for the inter-operator reliability of were 0.97 and 0.89. The intra-class correlation coefficients and typical errors ranged from 0.90 to 1.00 and ranged from 0.01 to 0.24 for two independent operators within two data collections. The results suggest that the Champdas Master system can be used validly and reliably to gather live football match statistics by well-trained operators. Therefore, the data obtained by the company can be used by coaches, managers, researchers and performance analysts as valid match statistics from players and teams during their professional tasks and investigations.

Physical demands and technical skills of different playing positions within soccer match-play have been rarely studied in competitions from Asia that have unique restrictions that limit the number of foreign players per team. Therefore, the aims of this study were to identify the technical and physical differences between domestic and foreign soccer players according to playing-positions in the China Super League (CSL); and to classify domestic and foreign players (best/worst) based on their match performance characteristics. Data were provided by Amisco Sports Analysis Services (n = 3468 observations). Discriminant and ANOVA analyses showed important differences between domestic and foreign players in the CSL in terms of physical and technical performance indicators for various playing positions. The unique match performance profiles of domestic and foreign players within the CSL highlighted important features for coaches and managers to improve the recruitment process within a league that implements a restrictive foreign player policy.
