RESUMEN

Conventional imaging systems can only capture light intensity. Meanwhile, the lost phase information may be critical for a variety of applications such as label-free microscopy and optical metrology. Existing phase retrieval techniques typically require a bulky setup, multiframe measurements, or prior information of the target scene. Here, we proposed an extremely compact system for complex amplitude imaging, leveraging the extreme versatility of a single-layer metalens to generate spatially multiplexed and polarization phase-shifted point spread functions. Combining the metalens with a polarization camera, the system can simultaneously record four polarization shearing interference patterns along both in-plane directions, thus allowing the deterministic reconstruction of the complex amplitude light field in a single shot. Using an incoherent light-emitting diode as the illumination, we experimentally demonstrated speckle-noise-free complex amplitude imaging for both static and moving objects with tailored magnification ratio and field of view. The miniaturized and robust system may open the door for complex amplitude imaging in portable devices for point-of-care applications.

Pixel super-resolution (PSR) has emerged as a promising technique to break the sampling limit for phase imaging systems. However, due to the inherent nonconvexity of phase retrieval problem and super-resolution process, PSR algorithms are sensitive to noise, leading to reconstruction quality inevitably deteriorating. Following the plug-and-play framework, we introduce the nonlocal low-rank (NLR) regularization for accurate and robust PSR, achieving a state-of-the-art performance. Inspired by the NLR prior, we further develop the complex-domain nonlocal low-rank network (CNLNet) regularization to perform nonlocal similarity matching and low-rank approximation in the deep feature domain rather than the spatial domain of conventional NLR. Through visual and quantitative comparisons, CNLNet-based reconstruction shows an average 1.4 dB PSNR improvement over conventional NLR, outperforming existing algorithms under various scenarios.

As the foundation of virtual content generation, cameras are crucial for augmented reality (AR) applications, yet their integration with transparent displays has remained a challenge. Prior efforts to develop see-through cameras have struggled to achieve high resolution and seamless integration with AR displays. In this work, we present LightguideCam, a compact and flexible see-through camera based on an AR lightguide. To address the overlapping artifacts in measurement, we present a compressive sensing algorithm based on an equivalent imaging model that minimizes computational consumption and calibration complexity. We validate our design using a commercial AR lightguide and demonstrate a field of view of 23.1° and an angular resolution of 0.1° in the prototype. Our LightguideCam has great potential as a plug-and-play extensional imaging component in AR head-mounted displays, with promising applications for eye-gaze tracking, eye-position perspective photography, and improved human-computer interaction devices, such as full-screen mobile phones.

Empowered by pixel super-resolution (PSR) and phase retrieval techniques, lensless on-chip microscopy opens up new possibilities for high-throughput biomedical imaging. However, the current PSR phase retrieval approaches are time consuming in terms of both the measurement and reconstruction procedures. In this work, we present a novel computational framework for PSR phase retrieval to address these concerns. Specifically, a sparsity-promoting regularizer is introduced to enhance the well posedness of the nonconvex problem under limited measurements, and Nesterov's momentum is used to accelerate the iterations. The resulting algorithm, termed accelerated Wirtinger flow (AWF), achieves at least an order of magnitude faster rate of convergence and allows a twofold reduction in the measurement number while maintaining competitive reconstruction quality. Furthermore, we provide general guidance for step size selection based on theoretical analyses, facilitating simple implementation without the need for complicated parameter tuning. The proposed AWF algorithm is compatible with most of the existing lensless on-chip microscopes and could help achieve label-free rapid whole slide imaging of dynamic biological activities at subpixel resolution.

In order to increase signal-to-noise ratio in optical imaging, most detectors sacrifice resolution to increase pixel size in a confined area, which impedes further development of high throughput holographic imaging. Although the pixel super-resolution technique (PSR) enables resolution enhancement, it suffers from the trade-off between reconstruction quality and super-resolution ratio. In this work, we report a high-fidelity PSR phase retrieval method with plug-and-play optimization, termed PNP-PSR. It decomposes PSR reconstruction into independent sub-problems based on generalized alternating projection framework. An alternating projection operator and an enhancing neural network are employed to tackle the measurement fidelity and statistical prior regularization, respectively. PNP-PSR incorporates the advantages of individual operators, achieving both high efficiency and noise robustness. Extensive experiments show that PNP-PSR outperforms the existing techniques in both resolution enhancement and noise suppression.

Multi-wavelength phase retrieval provides a competitive solution to lensless holographic imaging that features a low-cost, compact design and high data acquisition speed. However, the existence of phase wraps poses a unique challenge for iterative reconstruction, and the resulting algorithms often suffer from limited generalizability and increased computational complexity. Here, we propose a projected refractive index framework for multi-wavelength phase retrieval that directly recovers the amplitude and unwrapped phase of the object. General assumptions are linearized and integrated into the forward model. Based on an inverse problem formulation, physical constraints and sparsity priors are incorporated, which ensures imaging quality under noisy measurements. We experimentally demonstrate high-quality quantitative phase imaging on a lensless on-chip holographic imaging system using three color LEDs.

The imaging quality of in-line digital holography is challenged by the twin-image and aliasing effects because sensors only respond to intensity and pixels are of finite size. As a result, phase retrieval and pixel super-resolution techniques serve as two essential ingredients for high-fidelity and high-resolution holographic imaging. In this work, we combine the two as a unified optimization problem and propose a generalized algorithmic framework for pixel-super-resolved phase retrieval. In particular, we introduce the iterative projection algorithms and gradient descent algorithms for solving this problem. The basic building blocks, namely the projection operator and the Wirtinger gradient, are derived and analyzed. As an example, the Wirtinger gradient descent algorithm for pixel-super-resolved phase retrieval, termed as Wirtinger-PSR, is proposed and compared with the classical error-reduction algorithm. The Wirtinger-PSR algorithm is verified with both simulated and experimental data. The proposed framework generalizes well to various physical settings and helps bridging the gap between empirical studies and theoretical analyses.

Pixel super-resolution (PSR) techniques have been developed to overcome the sampling limit in lensless digital holographic imaging. However, the inherent non-convexity of the PSR phase retrieval problem can potentially degrade reconstruction quality by causing the iterations to tend toward a false local minimum. Furthermore, the ill posedness of the up-sampling procedure renders PSR algorithms highly susceptible to noise. In this Letter, we propose a heuristic PSR algorithm with adaptive smoothing (AS-PSR) to achieve high-fidelity reconstruction. By automatically adjusting the intensity constraints on the estimated field, the algorithm can effectively locate the optimal solution and converge with high reconstruction quality, pushing the resolution toward the diffraction limit. The proposed method is verified experimentally within a coherent modulation phase retrieval framework, achieving a twofold improvement in resolution. The AS-PSR algorithm can be further applied to other phase retrieval methods based on alternating projection.

Podophyllotoxin (PPT), a toxic polyphenol derived from the roots of genus Podophyllum, had been reported with strong inhibition on both normal human cells and tumor cells, which hindered the development of PPT as the candidate antitumor agent. In the present work, multiple NQO1-activatable PPT prodrugs were synthesized for reducing normal cell toxicity and keeping tumor cell toxicity. The antiproliferative activities in vitro showed prodrug 3 was greatly selectively toxic to tumor cells over-expressing NQO1, taxol-resistant A549, hypoxia A549 and HepG2, and lower damage to normal cells in comparison with podophyllotoxin, prodrug 1 and 2. As elucidated by further mechanistic research, prodrug 3 was activated via NQO1 to efficiently while gently produce cytotoxic PPT units and kill tumor cells. In additions, in vivo study revealed that 3 significantly suppressed cancer growth in HepG2 xenograft models without obvious toxicity. Therefore, this NQO1-activatable prodrug delivery system exhibits good biosafety and provides a novel strategy for the development of drug delivery systems.

Phase-only liquid crystal spatial light modulator has wide ranging applications that require accurate phase retardance. The phase calibration of the spatial light modulator is therefore of vital importance. Available self-referenced calibration methods face the challenges of high time consumption, low efficiency, and low stability against the conditions. A self-referenced multiple-beam interferometric method is proposed to derive the global grayscale-phase response. As is presented theoretically and experimentally, the proposed method reduces the measuring time and improves the calibration efficiency by encoding multiple fringes in a single hologram. Results also show that the method is equally accurate when compared with traditional two-beam interferometric method, whereas providing a greater robustness against measuring errors since the standard deviation is only 56% of that of the traditional method.

LncRNA plays a vital role in many diseases, and abnormal expression of LncRNA has been reported in many types of tumors. In this study, we analyzed the available public TCGA and GEO databases, and found that the expression of SNHG3 was increased in clear cell renal cell carcinoma (ccRCC), which was positively correlated with many clinicopathological parameters, and the higher expression of SNHG3 predicted worse clinical prognosis. Functional experiments indicated that knockdown of SNHG3 could significantly inhibit the proliferation and metastasis of ccRCC in vitro and in vivo. Subsequently, through luciferase reporter assays, qPCR and rescue experiments, it was found that SNHG3 could bind to miR-139-5p, thereby up-regulating the expression of its target gene TOP2A, and play a role in promoting tumor progression in ccRCC. The correlation analysis showed that there was a significant positive correlation between the expression of SNHG3 and TOP2A, and both of them were significantly negative correlated with the expression of miR-139-5p. Our work suggested that the SNHG3/miR-139-5p/TOP2A axis plays an important role in the proliferation and metastasis of ccRCC, and was expected to be a new biomarker for diagnosis, prognosis and a target for treatment of ccRCC.
