RESUMEN

A hard X-ray engineering applications beamline (BL-02) was commissioned recently and started operation in March 2019 at the Indian synchrotron source, Indus-2. This bending-magnet-based beamline is capable of operating in various beam modes, viz. white, pink and monochromatic beam. The beamline utilizes the X-ray diffraction technique in energy-dispersive and angle-dispersive modes to carry out experiments mainly focused on engineering problems, viz. stress measurement, texture measurement and determination of elastic constants in a variety of bulk as well as thin-film samples. An open-cradle six-circle diffractometer with ∼12 kg load capacity allows accommodation of a wide variety of engineering samples and qualifies the beamline as a unique facility at Indus-2. The high-resolution mode of this beamline is suitably designed so as to carry out line profile analysis for characterization of micro- and nano-structures. In the present article the beamline is described starting from the beamline design, layout, optics involved, various operational modes and experimental stations. Experiments executed to validate the beamline design parameters and to demonstrate the capabilities of the beamline are also described. The future facilities to be incorporated to enhance the capabilities of the beamline are also discussed.

RESUMEN

A microfocus X-ray fluorescence spectroscopy beamline (BL-16) at the Indian synchrotron radiation facility Indus-2 has been constructed with an experimental emphasis on environmental, archaeological, biomedical and material science applications involving heavy metal speciation and their localization. The beamline offers a combination of different analytical probes, e.g. X-ray fluorescence mapping, X-ray microspectroscopy and total-external-reflection fluorescence characterization. The beamline is installed on a bending-magnet source with a working X-ray energy range of 4-20 keV, enabling it to excite K-edges of all elements from S to Nb and L-edges from Ag to U. The optics of the beamline comprises of a double-crystal monochromator with Si(111) symmetric and asymmetric crystals and a pair of Kirkpatrick-Baez focusing mirrors. This paper describes the performance of the beamline and its capabilities with examples of measured results.

RESUMEN

One hundred thirty soil samples from agricultural fields and animal-inhabited areas were examined for the presence of Listeria. The microorganism was identified in 23 (17.7%) samples. L. monocytogenes was detected in 7 samples (5.4%), L. ivanovii in 2 (1.5%), L. innocua in 10 (7.7%) and L. welshimeri in 4 samples (3.1%). Prevalence of Listeria in soil from agricultural fields (17.5%) did not differ considerably from that in the soil and animal-inhabited area (18.0%), but L. ivanovii was isolated only from the latter source. Frequency of occurrence of different species of Listeria differed from place to place.

RESUMEN

Examination of 90 isolates of Enterococcus sp. revealed production of enterocin by two isolates of E. faecalis which was inhibitory to Listeria monocytogenes. Two isolates of E. gallinarum produced enterocin active against Staphylococcus aureus. None of the isolates antagonized Salmonella enteritidis. The enterocins of E. faecalis isolates were inactivated by alpha-chymotrypsin but not by trypsin and papain, while those of E. gallinarum were resistant to all the three enzymes. Enterocins produced by all 4 strains were resistant to heating at 60 degrees C for 30 min and 80 degrees C for 10 min, but sensitive to 121 degrees C for 15 min. At 100 degrees C for 10 min, two enterocins, one each of E. faecalis and E. gallinarum were inactivated, while the remaining two retained the bactericidal activity.

Cellular and humoral immune responses to Salmonella dublin infection were studied in guinea-pigs given zinc-deficient (ZnD) or zinc-adequate (ZnA) diet, containing 1.03 or 52.4 mg Zn/kg, respectively. On day 30, each animal of each dietary group was inoculated intraperitoneally with 10(6)viable cells of S. dublin strain 493. On the 15th day after infection, the immune responses were assessed by skin hypersensitivity (SH) tests and enzyme-linked immunosorbent assay (ELISA) for serum antibody with Salmonella -extract antigen. ZnD animals had significantly decreased ability to produce Salmonella -specific immediate and delayed type SH responses. These results were further substantiated by histological examination of skin sections. The ELISA indicated significantly lower Salmonella -specific serum antibody titres in ZnD animals than in ZnA animals. Mean viable counts of S. dublin in the liver and spleen of ZnD animals were significantly higher than those in ZnA animals.
