RESUMEN

The study aimed to estimate genetic and phenotypic parameters for growth traits and Kleiber ratio in Dorper sheep breed. Data used in this study were collected over 12 years (2012-2023) at Debre Birhan Agricultural Research Center sheep research station in Ethiopia. Studied traits were body weights at birth (WT0), weaning (WT3), six month (WT6), and yearling (WT12) age; average daily gains from birth to weaning (ADG0-3), from weaning to six months (ADG3-6), from six months to yearling (ADG6-12); and Kleiber ratios from birth to weaning (KR1) and from weaning to six months (KR2). The (co)variance components were estimated with different animal models using Average Information Restricted Maximum Likelihood (AI-REML) procedure. The best-fitted model for each trait was determined using likelihood ratio tests. Phenotypic performance for WT3, WT6, WT12, ADG0-3 and ADG3-6 showed a decline trend at a rate of 0.216 kg, 0.794 kg, 0.671 kg, 2.601 g and 4.865 g over years respectively. However, WT3, WT6, WT12, ADG0-3 showed a positive genetic improvement trend at a rate of 0.029 kg, 0.043 kg, 0.049 kg and 0.257 g over years respectively. Year of birth had a significant effect (P < 0.001) on all studied traits. Model including direct genetic as well as permanent environmental effect (Model 2) was chosen as the most appropriate model for WT0. Model which included only direct genetic effect (Model 1) was the best-fit model for all other studied traits. Direct heritability estimates based on suitable models were 0.07 ± 0.06, 0.11 ± 0.06, 0.09 ± 0.07, 0.11 ± 0.09 0.11 ± 0.06, 0.00 ± 0.04, 0.15 ± 0.07 and 0.00 ± 0.04 for WT0, WT3, WT6, WT12, ADG0-3, ADG3-6, KR1 and KR2 respectively. The variance ratio for the permanent environmental effect was 0.13 ± 0.04 for WT0. Genetic correlations among the traits ranged from negative (-0.39) for WT0-KR1 to high (0.99) for WT3-ADG0-3 and phenotypic correlations ranged from negative (-0.31) for WT0-KR1 to high (0.98) for WT3-ADG0-3. The low direct heritability estimates for the studied traits indicated that genetic improvement by direct selection might be difficult. Further investigation for the unexpected declined trend of phenotypic performance over years need to be required.

Pre-eclampsia (PE) is a pregnancy-related disorder characterized by hypertension and proteinuria occurring after 20 weeks of gestation. Several studies have been performed to determine the serum magnesium (Mg) level in PE, but most report inconclusive results. Consequently, this study was designed to resolve this controversy among African women. PubMed, Hinari, Google Scholar and African Journals Online electronic databases were searched for studies published in English. The qualities of included articles were appraised using the Newcastle-Ottawa quality assessment tool. Stata 14 software was utilized for analysis and serum Mg levels in cases and normotensive controls were compared through mean and standardized mean difference (SMD) at the 95% confidence interval (CI). In this review, we found that the mean serum Mg level was significantly reduced in cases (0.910±0.762 mmol/L) vs controls (1.167±1.060 mmol/L). The pooled SMD of serum Mg was significantly lower in cases (-1.20 [95% CI -1.64 to -0.75]). Therefore, since serum Mg is reduced in cases vs controls, we propose that Mg is involved in the pathophysiology of PE. Nevertheless, to know the exact mechanisms of Mg in PE development will require large-scale prospective studies.

INTRODUCTION: Three common endothelial nitric oxide synthase (eNOS) gene variants are existed such as; G-894T, T-786C, and variable number tandem repeats in intron-4 (VNTR intron-4) which has been proposed to be linked with PE. However, there is still debate regarding the findings. To address this, a review was conducted to assess the potential association of eNOS gene variants at these positions with the risk of PE. METHODS: PubMed, Scopus, Science Direct, Hinari, and African Journal Online databases and Google Scholar search engines were utilized to search studies published in English-language until 30 January 2023. The Joanna Briggs Institute Meta-Analysis instrument was used for data extraction process and the Newcastle-Ottawa Scale was used to appraise the quality of the included studies. Meta-regression analysis was conducted using Stata 14 statistical software. The pooled odds ratios (ORs) of fixed and random effect models were utilized to evaluate the association of eNOS gene polymorphism with the risk of PE at 95% CI. Publication bias was assessed using Egger's test and a funnel plot. RESULTS: The study included 47 observational studies involving 13,795 pregnant women (6216 cases and 7579 controls). Pregnant women carrying TT and CC genotypes of eNOS gene at 894 and 786 positions were found to have a greater probability of developing PE as compared to GG and TT genotypes (OR = 1.54 vs. 1.43 and CI: 1.12 - 2.14 vs.1.02 - 2.00 at 95% CI), respectively. However, a significant association was not observed between aa genotype of eNOS gene in VNTR intron-4 region and risk of PE as compared to bb genotype (OR =1.26, 95% CI: 0.83 - 1.89). The allelic model of eNOS gene at all positions showed nonsignificant association with the risk of PE. CONCLUSIONS: The women having eNOS gene variants at 894 and 786 positions showed a significant association with the risk of PE. Yet, the women having eNOS gene variant at intron-4 region showed nonsignificant association with the risk of PE. Thus, this study suggests that eNOS gene variants may play a role in the development of PE, but large-scale studies are required to inaugurate concrete evidence on the roles of eNOS gene variants in PE pathogenesis.

Growth curve analysis can help to optimize the management, determine nutritional requirements, predict the weight of animals at a specific age, and to select highly productive animals. Therefore, this study aimed to find the best-fitted nonlinear functions to provide a specific shape of the growth curve from birth to yearling age in different generations of Boer x Central Highland goats. Gompertz, Logistic, Brody, Von Bertalanffy, Monomolecular, Negative exponential, and Richards models were evaluated to quantify their ability to describe the biological growth curve. Root mean square error (RMSE), Bayesian information criterion (BIC), adjusted coefficient of determination (AdjR2), and Akaike's information criterion (AIC) were used to evaluate the goodness of fit and flexibility of the models. Data were analyzed using the nonlinear regression procedure of SAS. High AdjR2 and lower AIC, BIC, and RMSE values are indicators of best-fitted model. The best-fitting model for the first filial generation (F1), second filial generation (F2), and male goats' growth data was Brody function, whereas the Richards model, followed by Brody, best described the growth of third filial generation (F3) and female goats. The values of parameter A (asymptotic weight) for F1, F2, F3, female, and male goats based on the Brody model were 30.5±1.32, 28.2±1.38, 24.4±1.04, 27.8±0.94, and 29.8±1.32 kg for F1, F2, F3, female, and male goats, respectively. As per the best-fitted growth function, the asymptotic weight tended to reduce when the filial generation increased. The asymptotic weight for male goats was higher than for female goats. F1 had a slightly small value of parameter K, followed by F2 and F3. Both males and females had similar maturity rates. Based on the Brody function, the correlation between maturation rate and mature weight was high (-0.98, P<0.001). The correlation estimates for A-B and B-K were 0.27 and -0.15, respectively. Brody was best fitted for most goat categories, although Richards, followed by Brody, was best fitted for female and F3 goats. Besides, Brody could be better than Richards due to the ease of interpretation, convergence, and applicability for a small sample size. Therefore, the Brody function can predict the mature body weight, maturation rate, and growth rate of Boer x Central Highland goats and be used to formulate breeding and management strategies for profitable goat farming.

This study evaluated the reproductive and productivity of local and Dorper crossbred ewes in a community-based management system. We analyzed data collected from 2013 to 2021, taking into account different factors such as dam-breed, location, type of birth, season, and year of lambing. Lambing was observed all year-round, but the majority (35%) occurred in September, October, and December. This suggests that pasture availability, which is influenced by climatic-factors, may play a role in the seasonality of lambing. Litter-size at birth and weaning did not show any significant difference. Age at first lambing varied between breeds (P < 0.001), while lambing-interval and annual reproductive rates were unaffected by dam-breed (P > 0.05). The annual number of lambing per year significantly varied based on location and lambing seasons (P < 0.001), with a higher frequency during the major rainy-season compared to the dry-season (1.58vs1.42), highlighting the influence of feed availability. Productivity indices of ewes were calculated. Location and season of lambing had a significant impact on annual ewe productivity, while the ewe genotype showed no significant influence on productivity indices, except for the weight of lambs produced per kilogram of metabolic weight (0.84vs0.72 lambs per kg ewe and year; P < 0.01: 2.02vs1.77 kg lamb per kg0.75 ewe and year), where local ewes outperformed Dorper crossbred ewes. The difference in annual-productivity indices between local and Dorper crossbred ewes was more evident when considering the postpartum weight, as the ewes exhibited higher postpartum weights. However, both ewe genotypes produced comparable lamb weights per year (20.91vs20.16 kg lamb weaned per ewe and year for local and Dorper crossbred ewes, respectively). In summary, under low-input conditions, Dorper crossbred ewes demonstrated comparable reproductive performances and productivity traits to local ewes. Nevertheless, breed and environmental factors identified in this study should be taken into account to enhance sheep productivity in both local and Dorper crossbred ewes.

Additive genetic and non-additive parameters for reproductive traits of Boer x Central Highland goats were estimated. Pedigree and performance records comprised of Central Highland and their crosses with Boer goats were collected from 2009 to 2018 in the Sirinka Agricultural Research Center sheep and goat breeding station. Least-squares means for genotypes were obtained using a general linear model procedure in SAS. To estimate crossbreeding parameters, breed additive, heterotic, and recombination effects were fitted as fixed covariates instead of genotypes. Variance, heritability, and repeatability estimates were estimated through the AI-REML algorithm using WOMBAT software. Genotype did not significantly (P>0.05) influence most of the reproductive traits studied except for gestation length. The additive effect for litter size at birth (LSB), total litter birth weight (LBW), total litter weaning weight (LWW), litter size at weaning (LSW), and gestation length (GL) was estimated to be -0.004 kid, 0.08 kg, -3.18 kg, -0.54 kid, and 3.69 days, respectively. The contribution of heterosis to LSB, LWW, and GL of crossbred goats was negative, while the estimates for LBW and LSW were positive. However, Boer goats' heterosis effect and direct additive contribution to reproductive traits were insignificant (P>0.05) except for LSW. The recombination effect was negligible and not significant (P>0.05) for all traits examined. The direct heritability estimate for LSB, LWB, LWW, LSW, and GL were 0.050, 0.098, 0.086, 0.018, and 0.00, respectively. The repeatability estimates for LSB, LWB, LWW, LSW, and GL were 0.149, 0.116, 0.099, 0.086, and 0.061, respectively. The result indicates that improvement in reproductive traits would not be expected by crossing Boer with Central Highland goats. In addition, heritability estimates indicate that the improvement of reproductive traits through selection will be small, and the repeatability estimates indicate that multiple records have to be used to make a decision of culling or selection.

Background: Leishmaniasis is a vector-borne protozoan infection that has a wide clinical spectrum in the tropics and subtropics. Kidney damage is frequently associated with increased morbidity and mortality in visceral leishmaniasis (VL) patients. However, up to date, there is a very limited report on the effect of visceral leishmaniasis on kidney function profiling in Ethiopia. Objective: To evaluate the renal function profile in human visceral leishmaniasis (kala-azar) patients. Materials and Methods: Human blood was taken from VL patients (n = 100) and healthy controls (n = 100) attending Kahsay Abera and Mearg Hospitals, Western Tigray of Ethiopia. Serum was separated according to the conventional protocol and kidney function profiling (creatinine, urea, and uric acid) was analyzed by Mindray 200E automated chemistry analyzer. The estimated glomerular filtration rate (eGFR) was also assessed in this study. The obtained data were processed using SPSS Version 23.0. Descriptive statistics, independent-test, and bivariate correlations were used for data analysis. P values <0.05 were considered statistically significant at a 95% confidence level. Results: The mean serum creatinine level was found significantly higher, while respective serum urea and eGFR were significantly lower in VL patients compared to healthy controls. Specifically, from 100 VL cases, an increased level of serum creatinine, urea, and uric acid was found in 10%, 9% and 15% VL cases, respectively; meanwhile, a decreased serum urea and eGFR have been reported from 33% to 44% VL cases, respectively. Conclusion: The finding of this study asserted that visceral leishmaniasis causes derangement in kidney activities characterized by alteration of renal function profile. This may indicate that VL is the determinant factor for developing kidney dysfunction. This study encourages researchers to engage in visceral leishmaniasis and its effect on other organ function profiles in humans and identify potential markers for both prevention and intervention.

Background and Aims: Pre-eclampsia (PE) is a pregnancy-related syndrome that occurs after 20 weeks of pregnancy. The current study was designed to evaluate the determinants of PE in pregnant women following antenatal and delivery services. Methods: An institution-based case-control study was performed in governmental hospitals of Bahir Dar city. The data were collected using an interview-administered questionnaire and analyzed by SPSS version 20 software. Bivariate and multivariable logistic regression models were utilized for the analysis of variables. A p < 0.05 was used to declare the level of significance. Results: In this study, the mean ages (in years) of cases and controls were 28.20 ± 5.66 and 27.52 ± 4.70, respectively. Factors such as: being primiparous (adjusted odds ratio [AOR]: 3.19 at 95% confidence interval [CI]: 1.71, 5.97), family history of hypertension (HTN) (AOR: 4.14 at 95% CI: 1.71, 10.05), previous history of PE (AOR: 7.97 at 95% CI: 2.42, 26.63), number of antenatal care (ANC) visits (AOR: 5.43 at 95% CI: 2.86, 10.33), not taking iron and folic acid supplement (AOR: 4.46 at 95% CI: 1.59, 12.48), body mass index ≥25 kg/m2 (AOR: 3.47 at 95% CI: 1.78, 6.77), not consuming vegetables (AOR: 1.99 at 95% CI: 1.07, 3.69) and not consuming egg, milk and milk products (AOR: 3.00 at 95% CI: 1.47, 6.11) were the determinants of PE. Conclusion: In this study, different determinants of PE were identified. Hence, special attention should be given for primiparous women, women having previous history of PE and family history of HTN. Moreover, nutritional counseling should be given for pregnant women during ANC visits. Besides, higher officials should design an appropriate strategy to increase the number of mothers to complete their ANC visits and to take iron and folic acid supplements. Furthermore, to verify the determinants of PE, community-based cohort studies are warranted.

Introduction: In Ethiopia, small ruminants contribute significantly to livelihoods and food security but productivity is low with high disease burden and essential endoparasite control not widely practiced. The current study assessed worm burden and its control in three districts in Ethiopia. Methods: All sheep older than 3 months in nine villages were treated en-masse with albendazole and triclabendazole twice a year from 2018 to 2021. Treatments were administered under field conditions by animal health workers. Pre- and post-treatment data were assessed looking at fecal egg presence/absence and fecal egg per gram (EPG) count. Results: A total of 1,928 and 735 sheep were examined before and after deworming, respectively. Before treatment worms were detected in 54.4% (95% CI: 52.2-56.6) of sheep. Strongylid (30.4%) and Fasciola (18.2%) were the most frequently identified parasites. Animals living in wet mid-highland environments were more than 23 times more likely to have strongylid eggs in their feces and 5 times more likely to have eggs from any gastrointestinal tract (GIT) parasites detected, as compared to animals living in moist highland agro-ecology. Over the course of the 2018-2021 community intervention there was total elimination of animals with a high worm burden (EPG > 1,500), and elimination of a third of those with moderate infections. Mild infections remained, largely accounted for by strongylid, which remains at low levels in healthy sheep. However, there were signs of emerging drug resistance. Conclusion: Generally, sheep in smallholder systems in Ethiopia experience a needlessly large economic burden from GIT worms. Routine therapy reduces this burden but smart strategies are needed to limit the onset of drug resistance.

Objective: This study aimed to determine the neutrophil-lymphocyte ratio (NLR) as an inflammatory biomarker among type 2 diabetes mellitus (T2DM) patients with diabetic nephropathy (DN). Methods: A comparative cross-sectional study design was conducted on 199 T2DM patients attending Bole 17 Health Center, Addis Ababa, Ethiopia. The urine albumin test was done by the MICRAL-II test strip. Fasting blood sugar was measured by a glucometer. Complete blood count was analyzed using an automated hematology analyzer (HUMAN GmbH, Wiesbaden, Hesse, Germany). The student's t-test, a chi-square test, and Pearson correlation were applied to analyze the data. Results: Out of the 199 diabetes mellitus patients, 45 (22.6%) and 154 (77.4%) were found with DN and without DN, respectively. Interestingly, the mean NLR value (2.66 ± 0.49) was found significantly higher in diabetic patients with DN compared to the mean NLR (1.65 ± 0.20) in diabetes patients without DN (p < 0.0001). The NLR showed positive significant correlation with variables such as age (r = 0.162, p = 0.023), duration of disease (r = 0.52, p < 0.0001), absolute neutrophil count (r = 0.712, p < 0.0001), total white blood cell count (r = 0.162, p = 0.022), systolic blood pressure (r = 0.338, p < 0.0001), and diastolic blood pressure (r = 0.731, p < 0.0001). On the other hand, negatively significant correlation was found between NLR and absolute lymphocyte count (r = -0.770, p < 0.000). Conclusion: The NLR was significantly increased in T2DM patients with DN, suggesting that inflammation and endothelial dysfunction could be an integral part of the pathogenesis of DN, and therefore, this ratio may be considered as a predictor and a prognostic biomarker of DN.

BACKGROUND: Over 90% of Ethiopians still rely on solid fuels for cooking food. The pollution from the burning process causes adverse respiratory outcomes including respiratory infections. This study aimed to assess the association of the pollution with nasopharyngeal occurrence of potential pathogens. METHODS: We conducted a comparative cross-sectional study in urban and rural settings in Ethiopia in 2016. Questionnaire-based data were collected from 168 mothers and 175 children aged below two years. Multiplex real-time PCR assays were performed on nasopharyngeal secretions for detection of bacteria and viruses and for the identification of pneumococcal serotypes/groups. RESULTS: High rates of bacteria and viruses in the nasopharynx were detected by PCR among both the children and the mothers. Among the detected viruses, enterovirus was more commonly detected among rural children than among children from urban areas. Streptococcus pneumoniae and Haemophilus influenzae were both more prevalent among children and mothers from rural areas compared with urban groups and among those using solid fuels compared with cleaner fuel users. Children from rural households using solid fuels and children whose mothers had educational status below high school had four times higher odds for detection of S. pneumoniae compared with those households using cleaner energy or those children having mothers with a higher educational status, respectively. One or more serotype/serogroup was identified in about 40% of the samples that were positive for pneumococci. Out of all identified serotypes/serogroups, 43% in the children and 45% in the mothers belonged to PCV13, indicating the larger majority of detected pneumococci being non-PCV13 serotypes. CONCLUSION: This study presented a high carriage rate of S. pneumoniae and H. influenzae among both children and their mothers, especially in rural areas and among solid fuel users. Thus, interventions should target cleaner energy sources to the public and promote maternal education.

Background: Pre-eclampsia (PE) is a metabolic disorder that adversely affects the lives of mother and their infants. Even though, several studies have been conducted on PE, no effective diagnostic and therapeutic agents were developed so far. Hence, this study was designed to evaluate serum uric acid, blood urea and creatinine levels in the prediction of PE. Methods: A hospital-based case-control study was conducted among pregnant women. A simple random sampling technique was applied to select study participants. The socio-demographic and clinical data were collected using an interview-administered questionnaire. Serum samples were used to determine the maternal uric acid, urea and creatinine levels via an automated chemistry analyzer. Independent sample t-test, Pearson correlation test and receiver operating characteristic (ROC) curve analysis were performed to check the association and diagnostic accuracy of variables to PE. Results: The mean ages (in years) of the case and control groups were 27.98 ± 5.64 and 27.33 ± 4.45, respectively. The mean serum uric acid and blood urea levels were significantly higher in pre-eclamptic women than in normotensive pregnant women (6.27 ± 0.20 vs 4.43 ± 0.15, and 8.50 ± 3.99 vs 5.67 ± 2.19), respectively but the serum creatinine level is non-significantly increased in cases as compared to controls (0.70 ± 0.05 vs 0.50 ± 0.01). The areas under the ROC curve of serum uric acid, creatinine and blood urea levels were 0.785, 0.735 and 0.764 (sensitivity: 69%, 60.7%, 67.9%; specificity: 73.8%, 75%, 71.4%) with the cutoff points of ≥5.25 mg/dL, ≥0.565 mg/dL and ≥6.5 mg/dL, respectively. Conclusion: In this study, we observed a significantly higher concentration of serum uric acid and blood urea values in pre-eclampsia as compared with normotensive pregnant women. Therefore, this suggested that serum uric acid; blood urea and creatinine values can be associated with PE. Moreover, serum uric acid, blood urea and creatinine levels could be carefully utilized as a diagnostic marker for PE, but their inclusion in routine diagnostic test to PE requires large-scale multi-center prospective studies that corroborate our findings.

Background and Aims: Pregnancy-induced hypertension is one of the top three ranked diseases during pregnancy that cause maternal, fetal, and neonatal morbidity and mortality worldwide. To provide adequate information to clinicians and researchers who are striving for potential interventions, biochemical profiling of such patients is required. Methods: A hospital-based case-control study design was conducted from August 2020 to May 2021 to evaluate serum lipid profile, uric acid, and high sensitivity C-reactive protein (hs-CRP) among women with pregnancy-induced hypertension compared to normotensive pregnant women. Data were entered and analyzed using SPSS version 25. Independent t-test and χ 2 were used to compare the relationship of variables between the two groups. A p-value less than 0.05 was used to test statistical significance. Results: The result of this study showed that while the levels (mean ± SD) of serum total cholesterol (TC), triglyceride (TG), low-density lipoprotein-cholesterol (LDL-C), TC/high-density lipoprotein-cholesterol (HDL-C), TG/HDL-C, LDL-C/HDL-C were significantly elevated, HDL-C was decreased among women with pregnancy-induced hypertension than normotensive pregnant women (p < 0.0001). The levels (mean ± SD) of uric acid and hs-CRP were significantly higher among women with pregnancy-induced hypertension compared to normotensive pregnant women (p < 0.0001). Conclusion: This study indicated that pregnancy-induced hypertension women have lipid abnormalities, increased systemic inflammatory markers, and hyperuricemia compared to normotensive pregnant women. Thus, women with PIH showing high dyslipidemia, hyperuricemia, and inflammation are likely to develop hypertension. Therefore, evaluation of these potential biomarkers during early antenatal care services may help seek interventions in PIH.

Nanopore sequencing has brought the technology to the next generation in the science of sequencing. This is achieved through research advancing on: pore efficiency, creating mechanisms to control DNA translocation, enhancing signal-to-noise ratio, and expanding to long-read ranges. Heterogeneity regarding epigenetics would be broad as mutations in the epigenome are sensitive to cause new challenges in cancer research. Epigenetic enzymes which catalyze DNA methylation and histone modification are dysregulated in cancer cells and cause numerous heterogeneous clones to evolve. Detection of this heterogeneity in these clones plays an indispensable role in the treatment of various cancer types. With single-cell profiling, the nanopore sequencing technology could provide a simple sequence at long reads and is expected to be used soon at the bedside or doctor's office. Here, we review the advancements of nanopore sequencing and its use in the detection of epigenetic heterogeneity in cancer.

Background: Blood banks have been suffering a shortage of blood worldwide due to limited donations. By and large, it is widely believed that blood donation has multiple health benefits. However, there are limited studies that support it. As a result, assessing the biochemical profiles of the regular blood donors is indispensable to evaluating an individual's risk for chronic inflammation. Objective: We strived to compare lipid and haematological profiles of the regular and first-time blood donors in the National Blood Bank Service of Ethiopia. Materials and Methods: A comparative cross-sectional study, involving 104 blood samples (52 each of regular and first-time donors), was designed to analyze lipid and haematological profiles and anthropometric parameters were measured. Data were analyzed using SPSS version 25, Chi-square (χ 2) was used to compare the relationship between categorical variables and an independent Student's t-test was used to compare the mean of the two groups. A p-value <0.05 was considered statistically significant. Results: Regular blood donors had lower mean TC (144.3 ± 28 mg/dL), TG (159.3 ± 88.2 mg/dL), LDL-c (75.9 ± 25.9 mg/dL) than the first-time blood donors with values of TC (158.1 ± 38.94 mg/dL), TG (163.9 ± 82.7 mg/dL), LDL-c (93.1 ± 31.5 mg/dL), respectively. The ratio of LDL-c/HDL-c and TC/HDL-c was found to be lower in regular blood donors when compared to the first-time donors (P < 0.05). Even though the level of HDL-c was higher (39.8 ± 8.8 mg/dL) in regular blood donors compared to first-time blood donors (36.8 ± 7.7 mg/dL), it was not statistically significant. The mean of some haematologic parameters like a platelet, RDW, lymphocyte, and MCH was significantly lower in regular blood donors than in first-time blood donors. BMI and WHR in regular donors were less than the first-time donors, albeit statistically insignificant. Conclusion: Interestingly, blood donation has a significant health benefit by lowering TC, TG and LDL-c, which have the potential risk of developing chronic inflammation.

INTRODUCTION: Selenium (Se) is an essential mineral for livestock health and productivity. In cattle, Se deficiency is associated with delayed conception, growth retardation, and increased morbidity and mortality. METHODS: We conducted a survey of cattle serum (n = 224) and feed (n = 81) samples from two areas with contrasting human and cereal grain Se concentration in Ethiopia. The fodder samples include stover, straw, hay and pasture grass. Se concentration of the samples were measured using inductively coupled plasma-mass spectrometry. RESULTS: Serum Se concentration ranged from 14.9 to 167.8 µg L-1 (median, 41.4 µg L-1). Cattle from East Amhara had significantly greater serum Se concentration compared to cattle from West Amhara (median: 68.4 µg L-1 vs 25.7 µg L-1; p < 0.001). Overall, 79.8% of cattle had Se deficiency (<81 µg L-1). All of the cattle from West Amhara were Se deficient compared with 62.5% of those from East Amhara. State of lactation of cows or age of cattle was not associated with serum Se concentration. The Se concentrations of feed samples ranged from 0.05 to 269.3 µg kg-1. Feed samples from East Amhara had greater Se concentration than samples from West Amhara. Cow serum and cattle feed Se concentrations showed strong spatially correlated variation, with a strong trend from East to West Amhara. CONCLUSIONS: This study shows that cattle Se deficiency is likely to be highly prevalent in Ethiopia, which will negatively affect the health and productivity of livestock. The deficiency appears to be geographical dependent. More extensive surveys to map Se concentration in soil-feed-livestock-human cycle are required in Ethiopia and elsewhere.

Proteostasis, also known as protein homeostasis, is critical for cell survival. Autophagy is a cellular process that degrades and recycles damaged or long-lived proteins, misfolded proteins, and damaged or abnormal organelles in order to preserve homeostasis. Among the three forms of autophagy, chaperone-mediated autophagy (CMA) is distinct from macroautophagy and microautophagy; it does not require the formation of vacuoles and only degrades selected individual proteins. CMA helps to maintain cellular homeostasis by regulating protein quality, bioenergetics, and substrate-associated cellular processes at the right moment. This pathway's dysfunction has been linked to several diseases and disorders. Neurodegenerative diseases and cancer have received the most attention. In various neurodegenerative disorders, especially in their later stages, CMA activity declines. CMA has been shown to act as a tumor suppressor in cancer by destroying specific tumor promoters. Once a tumor has grown, it also helps tumor survival and the metastatic cascade. The presence of changes in CMA in these diseases disorders raises the idea of targeting CMA to restore cellular homeostasis as a potential therapeutic method. Manipulation of CMA activity may be effective therapeutic strategies for treating these diseases. Therefore, in this paper; we introduce the basic processes, regulatory mechanisms, and physiological functions of CMA; evidences supporting the role of impaired CMA function in neurodegeneration and cancer; and the potential of how targeting CMA could be a promising therapeutic method for the two diseases.

Mitochondria-associated endoplasmic reticulum membranes (MAM) are specialized subcellular compartments that are shaped by endoplasmic reticulum (ER) subdomains placed side by side to the outer membrane of mitochondria (OMM) being connected by tethering proteins in mammalian cells. Studies showed that MAM has multiple physiological functions. These include regulation of lipid synthesis and transport, Ca2+ transport and signaling, mitochondrial dynamics, apoptosis, autophagy, and formation and activation of an inflammasome. However, alterations of MAM integrity lead to deleterious effects due to an increased generation of mitochondrial reactive oxygen species (ROS) via increased Ca2+ transfer from the ER to mitochondria. This, in turn, causes mitochondrial damage and release of mitochondrial components into the cytosol as damage-associated molecular patterns which rapidly activate MAM-resident Nod-like receptor protein-3 (NLRP3) inflammasome components. This complex induces the release of pro-inflammatory cytokines that initiate low-grade chronic inflammation that subsequently causes the development of metabolic diseases. But, the mechanisms of how MAM is involved in the pathogenesis of these diseases are not exhaustively reviewed. Therefore, this review was aimed to highlight the contribution of MAM to a variety of cellular functions and consider its significance pertaining to the pathogenesis of inflammation-mediated metabolic diseases.

Cancer immunotherapy is an effective treatment option against cancer. One of the approaches of cancer immunotherapy is the modification of T cell-based anti-tumor immune responses. T-cells, a type of adaptive immune response cells responsible for cell-mediated immunity, have long been recognized as key regulators of immune-mediated anti-tumor immunity. T-cell activities have been reported to be suppressed or enhanced by changes in cell metabolism. Moreover, metabolic reprogramming during activation of T cells is required for the development of distinct differentiation profiles of these cells, which may allow the development of long-term cell-mediated anti-tumor immunity. However, T cells have been shown to undergo metabolic exhaustion in tumor microenvironment (TME) as it poses several obstacles to their function. Applications of several mechanistic solutions to improve the efficacy of T cell-based therapies including chimeric antigen receptor (CAR) T cell therapy are yet to be determined. Modifying the metabolic properties of these cells and employing them in cancer immunotherapy is a potential strategy for improving their anti-tumor activity and therapeutic efficacy. To give an insight, in this review paper, we endeavoured to cover metabolic reprogramming in cancer and T cells, signalling mechanisms involved in immuno-metabolic regulation, the effects of the TME on T cell metabolic fitness, and targeting metabolic reprogramming of T cells for an enhanced anti-tumor response.

INTRODUCTION: H. pylori are gram-negative, microaerophilic helical-shaped bacteria with multiple flagella and commonly exist in the stomach. This infection may cause significant mucosal inflammation and damage, leading to ulcers in the stomach. It can also affect organ systems external to the gastrointestinal tract. To assess cardiovascular risk factors and to predict cardiovascular disorders, we are evaluating and comparing lipid profile and inflammatory marker between H. pylori-positive and negative patients. OBJECTIVE: To evaluate and compare lipid profile (TC; TG; LDL; HDL) and inflammatory marker (hs-CRP) in dyspeptic patients with and without H. pylori infection. METHODS: Comparative cross-sectional study was conducted from September 2020 to January 2021 at Debre Markos Referral Hospital, Debre Markos Health Center, and Hidassie Health Center, Ethiopia. Each of 50 H. pylori-positive and negative dyspeptic patients were studied. The data were checked for completeness and analyzed by SPSS version 25.0 Software. A p-value < 0.05 was considered statistically significant. RESULTS: Serum mean high-density lipoprotein (HDL) values were 37.54 ± 7.98 mg/dL and 43.12 ± 7.86 mg/dL (p < 0.05) for H. pylori-positive and negative dyspeptic patients, respectively, and median serum high sensitive C reactive protein (hs-CRP) levels were 6.29 mg/L (1.66-41.34) and 3.35 mg/L (0.39-10.01) (p < 0.05) for H. pylori-positive and negative dyspeptic patients, respectively. CONCLUSION: H. pylori infection significantly alters serum high-density lipoprotein (HDL) and high sensitive C reactive protein (hs-CRP) levels in dyspeptic patients, as a result, increase the potential risk of cardiovascular diseases.
