RESUMEN

OBJECTIVE: To compare the efficacy and safety of different anti-vascular endothelial growth factor (VEGF) agents combined with different delivery methods for neovascular glaucoma (NVG). DESIGN: Systematic review and Bayesian network meta-analysis (NMA). DATA SOURCES: PubMed, Embase, Cochrane Library, Web of Science, ClinicalTrials.gov, ISRCTN and Chinese databases including the China National Knowledge Infrastructure, China Science Periodical Database (Wanfang Database), VIP Journal Integration Platform and China Biology Medicine Database were searched from inception to 5 September 2022. ELIGIBILITY CRITERIA: We included randomised controlled trials (RCTs) that investigated the treatment of NVG using different anti-VEGF agents combined with various methods of drug administration, without any language limitations. All patients included underwent panretinal laser photocoagulation and there were no restrictions on prior glaucoma surgery. DATA EXTRACTION AND SYNTHESIS: Two independent reviewers extracted data and assessed the risk of bias. Random-effect Bayesian NMA was conducted to compare the efficacy and safety and rank priority of anti-VEGF regimens. The source of heterogeneity and the related factors affecting the stability of the results were also explored. CINeMA (Confidence in Network Meta-Analysis) was used to assess the certainty of evidence. RESULTS: Our analysis included 17 RCTs involving a total of 1311 eyes from 1228 patients. We examined five different treatment regimens, which used three different anti-VEGF drugs. The following treatments showed a significant decrease in intraocular pressure (IOP) compared with the control group at 1 month after glaucoma surgery: simultaneous intravitreal and intracameral injection of conbercept (ICCIVC) (mean difference (MD)=-11.56, 95% credible interval (CrI) -20.8 to -2.24), intravitreal injection of conbercept (MD=-8.88, 95% CrI -13.93 to -3.78), intravitreal injection of ranibizumab (MD=-7.62, 95% CrI -10.91 to -4.33) and intravitreal injection of bevacizumab IVB) (MD=-5.51, 95% CrI -10.79 to -0.35). The surface under the cumulative ranking curve (SUCRA) analysis indicated that ICCIVC (82.0%) may be the most effective regimen in reducing IOP. In terms of safety, there were no statistically significant differences among the interventions. According to the SUCRA analysis, ICCIVC (68.0%) was considered the safest choice with the fewest complications. Subgroup and meta-regression analyses showed that mean age was the main source of heterogeneity. Sensitivity analysis demonstrated the robustness of the study results. CONCLUSION: ICCIVC was more effective and safer than other anti-VEGF regimens for NVG. Simultaneous intravitreal and intracameral injection was found to be the best route of administration, and conbercept was found to be the superior drug selection when compared with ranibizumab and bevacizumab. Premature ovarian insufficiency (POI) patients experience a decline in ovarian function and a reduction in serum reproductive hormones, leading to a significant impact on the outcomes of assisted reproductive technology. Despite the absence of an effective clinical treatment to restore fertility in POI patients, recent research has indicated that cord blood plasma (CBP) derived from human umbilical cord blood (hUCB) may offer therapeutic benefits for various degenerative diseases. The primary aim of this study is to explore approaches for enhancing ovarian function and serum reproductive hormones through the administration of CBP in a murine model. Initially, hUCB was utilized to obtain CBP (CBP), which was subsequently analyzed for cytokine and growth factor profiles in comparison to adult blood plasma (ABP) by use of flow cytometry. Subsequently, POI mouse models were established through the induction of 4-vinylcyclohexene diepoxide, followed by the injection of CBP into the tail. At 7, 14, and 21 days posttreatment, mouse ovaries and blood were collected, and their estrus cycle, body weight, and ovarian weights were evaluated using precise electronic balance. Finally, ovarian morphology and follicle number were assessed through HE staining, while serum levels of anti-Müllerian hormone (AMH), estradiol (E2) and follicle-stimulating hormone (FSH) were determined by ELISA. Our study revealed that individuals with CBP exhibited significantly lower concentrations of proinflammatory cytokines, including IL-ß (p < 0.01) and IL-2 (p < 0.05), while displaying elevated levels of anti-inflammatory cytokines and chemokines, such as IL-2, IL-4, IL-6, IL-8, IL-12P70, IL-17A, IP-10, interferon-γ, and tumor necrosis factor-α (p < 0.01). Furthermore, CBP demonstrated remarkably higher levels of growth factors, including transforming growth factor-ß1, vascular endothelial growth factor, and insulin-like growth factor-1 (p < 0.01) than ABP. Notably, our investigation also revealed that CBP restored the content of serum reproductive hormones, such as AMH, E2, and FSH (p < 0.05), and increased the number of primordial and primary follicles (p < 0.01) and decreased the number of luteal and atretic follicles (p < 0.01) in vivo. Our findings suggested that CBP-secreted cytokines and growth factors could be restored POI ovarian function, enhanced serum reproductive hormones and rescued follicular development in vivo. These findings further support the potential of CBP as a promising strategy in clinical applications for POI related infertility.

Müller cells play an integral role in the development, maintenance, and photopic signal transmission of the retina. While lower vertebrate Müller cells can differentiate into various types of retinal neurons to support retinal repair following damage, there is limited neurogenic potential of mammalian Müller cells. Therefore, it is of great interest to harness the neurogenic potential of mammalian Müller cells to achieve self-repair of the retina. While multiple studies have endeavored to induce neuronal differentiation and proliferation of mammalian Müller cells under defined conditions, the efficiency and feasibility of these methods often fall short, rendering them inadequate for the requisites of retinal repair. As the mechanisms and methodologies of Müller cell reprogramming have been extensively explored, a summary of the reprogramming process of unlocking the neurogenic potential of Müller cells can provide insight into Müller cell fate development and facilitate their therapeutic use in retinal repair. In this review, we comprehensively summarize the progress in reprogramming mammalian Müller cells and discuss strategies for optimizing methods and enhancing efficiency based on the mechanisms of fate regulation.

AIM: To assess the visual correction of patients with different degrees of astigmatism with toric soft contact lenses (TSC). METHODS: It was a real-world study with prospective and single-arm design. A total of 384 patients with astigmatism who came for TSC fitting and alignment from November 2022 to January 2023 were included. According to the difference in astigmatism, patients were divided into groups A (cylinder degree: -0.75 to -0.50 D), B (cylinder degree: -1.75 to -1.00 D) and C (cylinder degree ≤ -2.00 D), and followed up on the day of wear, 1wk, 1 and 3mo, mainly to observe visual acuity, refraction, lens fit, visual quality and comfort at 1wk after wear. The visual acuity success rate and the overall success rate of the fitting were evaluation indicators (taking into account the four dimensions of visual acuity, fitting, quality of vision and comfort). The visual acuity success rate was calculated by taking "corrected visual acuity with contact lenses is no less than 1 line or better than best spectacle-corrected visual acuity" (i.e. corrected visual acuity with contact lenses is 1 line below, equal to, one line above or more than best spectacle-corrected visual acuity) as the criterion for visual success, and the the overall success rate of the fitting was calculated by using the comprehensive indicators (visual acuity, fit, visual quality, comfort) to meet certain conditions as the judgment criteria for successful fitting. RESULTS: After 1wk of wearing TSC, the visual acuity success rates of patients were 100% (207/207), 98.58% (139/141) and 97.22% (35/36) in the three groups, respectively, with residual cylinder closed to 0. The acceptability of the lens fitting was over 95%; the incidence of adverse visual symptoms was within 10% and the comfort acceptability was over 97%. The overall success rate of fitting for patients with high, medium and low astigmatism was 93.72% (194/207), 90.78% (128/141) and 88.89% (32/36), respectively. CONCLUSION: TSC (model: G&G POP·CT) are effective in correcting astigmatism in patients with different degrees of astigmatism.

Objective: Premature ovarian insufficiency (POI) is a female reproductive disorder of unknown etiology with no definite pathogenesis. Melatonin (MT) is an endogenous hormone synthesized mainly by pineal cells and has strong endogenous effects in regulating ovarian function. To systematically explore the pharmacological mechanism of MT on POI therapy, a literature review approach was conducted at the signaling pathways level. Methods: Relevant literatures were searched and downloaded from databases, including PubMed and China National Knowledge Infrastructure, using the keywords "premature ovarian insufficiency," "Hippo signaling pathways," and "melatonin." The search criteria were from 2010 to 2022. Text mining was also performed. Results: MT is involved in the regulation of Hippo signaling pathway in a variety of modes and has been correlated with ovarian function. Conclusions: The purpose of this review is to summarize the research progress of Hippo signaling pathways and significance of MT in POI, the potential crosstalk between MT and Hippo signaling pathways, and the prospective therapy.

PURPOSES: In order to investigate the association between serum periostin levels and the variation of its encoding gene POSTN and the prevalence of vertebral fractures and bone mineral density (BMD) in Chinese postmenopausal women, an association study was performed. MATERIALS AND METHODS: 385 postmenopausal women were recruited. For participants without a history of vertebral fracture, lateral X-rays of the spine covering the fourth thoracic spine to the fifth lumbar spine were performed to detect any asymptomatic vertebral fractures. Ten tag-single nucleotide polymorphisms (SNP) of POSTN were genotyped. Serum periostin levels, biochemical parameters, and BMD were measured individually. RESULTS: rs9603226 was significantly associated with vertebral fractures. Compared to allele G, the minor allele A carriers of rs9603226 had a 1.722-fold higher prevalence of vertebral fracture (p = 0.037). rs3923854 was significantly associated with the serum periostin level. G/G genotype of rs3923854 had a higher serum periostin level than C/C and C/G (67.26 ± 19.90 ng/mL vs. 54.57 ± 21.44 ng/mL and 54.34 ± 18.23 ng/mL). Furthermore, there was a negative correlation between the serum level of periostin and BMD at trochanter and total hip. CONCLUSION: Our study suggested that genetic variation of POSTN could be a predicting factor for the risk of vertebral fractures. The serum level of periostin could be a potential biochemical parameter for osteoporosis in Chinese postmenopausal women.

PURPOSE: Macular degeneration is the leading cause of blindness worldwide. In this study, we aimed to define a new subtype of macular-retinal dystrophy and its genetic predisposition in 5 families. METHODS: Exome sequencing was performed to determine the putative disease-causing genes in patients with inherited macular disorders confirmed through comprehensive ophthalmic examinations. To validate its functional consequence, adeno-associated virus-mediated mutant gene was delivered into the murine retina, and both structural and functional tests were performed to investigate its pathological effects in vivo. RESULTS: In total, 5 multigenerational families diagnosed with autosomal dominant maculoretinopathy were found to carry a pathogenic variant in a new gene, CLEC3B, which encodes tetranectin, a plasminogen kringle-4 binding protein. Consistent with the disease phenotypes of patients, mice that received subretinal injections with the CLEC3B variant displayed multiple subretinal hyperreflective deposits, reduced retinal thickness, and decreased electroretinographic responses. Moreover, the optokinetic tracking response indicated that spatial frequency was significantly lower (P < .05), implying impaired visual function in these mice. CONCLUSION: We have presented a new subtype of macular-retinal dystrophy in 5 families as well as a new pathogenic gene, CLEC3B, providing new insights into maculoretinopathy etiology.

RESUMEN

Melatonin (MT) is an endogenous hormone mainly synthesized by pineal cells, which has strong endogenous effects of eliminating free radicals and resisting oxidative damages. Melatonin (MT) can not only regulate the body's seasonal and circadian rhythms; but also delay ovarian senescence, regulate ovarian biological rhythm, promote follicles formation, and improve oocyte quality and fertilization rate. This review aimd to provide evidence concerning the synthesis and distribution, ovarian function, and role of MT in development of follicles and oocytes. Moreover, the role of MT as antioxidative, participating in biological rhythm regulation, was also reviewed. Furthermore, the effects of MT on various ovarian related diseases were analyzed, particularly for the ovarian aging and polycystic ovary syndrome (PCOS).

Using an asymmetric tetradentate ligand N-((6-(1H-pyrazol-1-yl)pyridin-2-yl)methylene)benzohydrazide (pypzbeyz), two high-coordinate 3d transition metal compounds [CoII(pypzbeyz)(NO3)2] (1) and [FeII(pypzbeyz)2](BF4)2(CH3CN) (2) have been synthesized and characterized by structural and magnetic measurements. X-ray crystallographic analyses revealed that compound 1 is seven-coordinate with a distorted pentagonal bipyramidal geometry (pseudo-D5h) and compound 2 is eight-coordinate with a triangular dodecahedral geometry (pseudo-D2d). Direct current (dc) magnetic susceptibilities revealed that compound 1 shows easy-plane magnetic anisotropy (D = +29.9 cm-1, E = 0.31 cm-1) and 2 shows easy-axis magnetic anisotropy (D = -6.6 cm-1, E = 0.02 cm-1). Alternating current (ac) magnetic measurements indicate that both compounds exhibit field-induced slow magnetic relaxation behavior. Furthermore, ab initio calculations also demonstrate that compound 1 presents a strong easy-plane magnetic anisotropy and 2 presents an easy-axis magnetic anisotropy, which are further identified by the calculated orientations of the local magnetic axes. These results demonstrate an effective way to achieve the targeted synthesis of high-coordinate 3d SIMs.

Production of oilseed rape/canola (Brassica napus) is increasingly threatened by dry conditions while the demand for vegetable oil is increasing. Brassica rapa is a genetically diverse ancestor of B. napus, and is readily crossed with B. napus. Recently, we reported promising levels of drought tolerance in a wild type of B. rapa which could be a source of drought tolerance for B. napus. We analysed global gene expression by messenger RNA sequencing in seedlings of the drought-tolerant and a drought-sensitive genotype of B. rapa under simulated drought stress and control conditions. A subset of stress-response genes were validated by reverse transcription quantitative PCR. Gene ontology enrichment analysis and pathway enrichment analysis revealed major differences between the two genotypes in the mode and onset of stress responses in the first 12 h of treatment. Drought-tolerant plants reacted uniquely and rapidly by upregulating genes associated with jasmonic acid and salicylic acid metabolism, as well as genes known to cause endoplasmic reticulum stress and induction of programmed cell death. Conversely, active responses in drought-sensitive plants were delayed until 8 or 12 h after stress application. The results may help to identify biomarkers for selection of breeding materials with potentially improved drought tolerance.

To elucidate the influence mechanisms of background electrolytes and ionic strength on Cd(II) removal, the adsorption of Cd(II) onto magnetic graphene oxide-supported sulfanilic acid (MGO-SA) in aqueous solutions containing different types and concentrations of background electrolytes was studied. The results indicate that Cd(II) adsorption was strongly dependent on pH and could be strongly affected by background electrolytes and ionic strength. The Cd(II) removal was decreased with the presence of background electrolyte cations (Na(+), K(+), Ca(2+), Mg(2+), Mn(2+), Zn(2+), and Ni(2+)), and the divalent cations exerted more obvious influences on the Cd(II) uptake than the monovalent cations at pH 6. Both Cl(-) and NO3(-) had negative effects on Cd(II) adsorption because they can form water-soluble metal-anion complexes with Cd(II) ions. The presence of 0.01molL(-1) Na3PO4 reduced the removal percentage of Cd(II) at pH<5 but extremely enhanced the Cd(II) removal when the pH>5. The Cd(II) adsorption was sensitive to changes in the concentration of NaCl, NaNO3, NaClO4, and Na3PO4. Besides, the adsorption isotherm of Cd(II) onto MGO-SA could be well described by the Freundlich model and was also influenced by the type of background electrolyte ions and the ionic strength.

Leaf canopy temperature has been proposed as a rapid selection tool for drought tolerance among crop genotypes. However, floral bud temperature may be a better indicator of drought tolerance than leaf temperature in grain crops. In this study, we examined whether the floral bud and leaves of Brassica rapa L. had similar stomatal characteristics and showed similar water loss during a drying cycle. We also compared the leaf and bud temperatures when the plants were exposed to a 10-day transient water stress during reproductive development that affected flower development, increased flower abortion, increased pod abortion and reduced yield by an average of 85%. The water loss of detached leaves and floral buds showed that the stomata on the leaves closed before those of the floral buds as the leaf water potential decreased. Consistent with the water loss studies, the temperature of the intact bud showed a delayed increase during the drying process compared with the leaves. This suggested that floral bud temperature could be a useful indicator of the water status of the reproductive organs of B. rapa.

OBJECTIVE: To study processing method and mechanism of Calamine. METHOD: Thermogravimetry analysis method and nano-technology were adopted to analyze and synthesize the components in Calamine, Tetracycline was took as the comparison drug to determine the antibacterial activity of Calamine and its components. RESULT: A part of zinc carbonate in Calamine was decomposed into zinc oxide when processing, and the particle size was smaller than before. The antibacterial activity of Calamine is decided by the content and particle size of zinc oxide, and has nothing with zinc carbonate. The more content and the smaller particle size of zinc oxide, the more powerful antibacterial activity of Calamine. CONCLUSION: The content and the particle size of zinc oxide can be the important targets in the processing of Calamine.
