RESUMEN

Objective: To observe the expression, correlation and significance of chemokine (C-X-C motif) ligand 12 (CXCL12) and chemokine (C-X-C motif) receptor 4 (CXCR4) in endometrium and myometrium of adenomyosis. Methods: Totally 38 patients were selected in this study, who underwent hysterectomy for adenomyosis at Beijing Obstetrics and Gynecology Hospital from October 2017 to December 2018 as the adenomyosis group, and, in the same period, selected 31 patients with cervical intraepithelial neoplasia Ⅲ or cervical cancer undergoing hysterectomy served as control group. The expression levels of mRNA and protein for CXCL12, CXCR4 in the endometrium and myometrium of the two groups were detected by immunohistochemistry and real-time PCR. Results: (1) The protein levels of CXCL12 and CXCR4 in endometrium in uterus with adenomyosis (0.229±0.025 and 0.226±0.016) were significantly higher than those in endometrium in uterus without adenomyosis (0.153±0.018 and 0.178±0.026); compared with each other, the differences were statistically significant (all P<0.05). And the expressions of CXCL12 and CXCR4 proteins in uterine myometrium of adenomyosis were 0.222±0.045 and 0.126±0.058, respectively, which were higher than those in the control group (0.091±0.029 and 0.099±0.020); compared with each other, the differences were statistically significant (all P<0.05). (2) The expression levels of CXCL12 and CXCR4 mRNA in endometrium of patients with adenomyosis were 6.31±0.12 and 8.49±0.21, respectively, which were higher than those in the control group (1.23±0.10 and 1.36±0.13); compared with each other, the differences were statistically significant (all P<0.05). Moreover, the expression levels of CXCL12 and CXCR4 mRNA in myometrium of patients with adenomyosis were 9.11±0.12 and 8.45±0.16, respectively, which were higher than those in the control group (1.18±0.08 and 1.46±0.13); compared with each other, the differences were statistically significant (all P<0.05). (3) In endometrium and myometrium of uterus with adenomyosis, CXCL12 and CXCR4 mRNA expression levels were positively associated (r=0.478, 0.542, all P<0.05). Conclusions: The levels of CXCL12 and CXCR4 in the endometrium and myometrium of adenomyosis are increased and positively correlated. The two chemokine may be involved in the development of adenomyosis.

RESUMEN

An RNA/DNA chimeric oligonucleotide was found to be effective in the targeted correction of point mutations in Escherichia coli, plant, and mammalian genomes. This strategy, named chimeraplasty, has the potential for gene therapy of many genetic diseases caused by point mutations. beta-Thalassemia is a very common human genetic disease and in most cases it is caused by point mutations. To test whether the chimeraplasty can be used to correct the point mutations responsible for beta-thalassemia, we introduced one mutated beta-globin gene, betaE, into MEL cells and successfully corrected the point mutation of the betaE gene with the highest correction efficiency of 1.9%. Furthermore, a targeted -202 C-->G mutation of the Ggamma-globin gene, which is associated with the elevated Ggamma-globin gene expression in the adult stage, was introduced into HeLa and CMK cells by an RNA/DNA oligonucleotide. These results indicated that the chimeraplasty has potential for human beta-thalassemia gene therapy.

Embryogenic calli were produced from mature zygotic embryos of Picea meyeri Rehd. et Wils. which were cultured on modified LP medium containing 30 g/L sucrose, 2 mg/L 2,4-D and 1 mg/L 6-BA. The growth rate of embryogenic calli could be improved by way of suspension culture in medium MS containing 20 g/L sucrose, 1 mg/L 2,4-D and 1 mg/L KT. The optimum values of initial cell density and shake rate of flask in suspension culture were 3.0% (fresh weight) and 150 r/min respectively. Several parameters which might be correlated with growth were determined over a 10 day period. Maximum growth in terms of fresh weight, dry weight and number of ESM occurred during 6-10 day. pH and conductivity attained the lowest point during 6-8 day. A lot of somatic embryos were obtained following transfer of the suspension cultures to medium MS containing 5 mg/L ABA. Mature somatic embryos grew into plantlets with well developed cotyledons after being cultured on 1/2 MS medium supplemented with 0.5% activated charcoal but without hormone.

Tc1/mariner transposons are widespread in the eukaryotes. In vitro transposition test indicated that the transposase is the only protein that is needed in transpositions. It was shown later that the reconstructed Tc1-like transposon, "sleeping beauty" in fish, and the Tc1 transposon in C. elegans jumps in human cells. This discovery indicates that the Tc1/mariner transposon may be engineered as a somatic gene therapy vector if coupled with an efficient gene delivery system. We introduced the Tc1 transposon from C. elegans into different mammalian cell lines and detected the transposition events, indicating that Tc1 transposon functions in different mammalian cells. Interestingly, a high inversion frequency of the transposon was also detected, suggesting that this type of transposon may add variations to host genome when it is horizontally transferred into a new species.

Derivative variable-offset synchronous fluorescence spectroscopy is developed to improve the spectral resolution and the selectivity of fluorescence measurements. 1-naphthol and 2-naphthol are employed to evaluate the proposed coupled technique and the various spectral comparisons are conducted. Second derivative variable-offset synchronous scanning permits the rapid simultaneous identification and quantitative determination of 1-naphthol and 2-naphthol in a mixture from a single spectrum. 6.7-2000 ng/ml 1-naphthol and 3.6-500 ng/ml 2-naphthol can be quantified with 1-naphthol and 2-naphthol ratios of 40: 1-1: 10. The determination of 1-naphthol and 2-naphthol in various spiked water samples gave a mean recovery of 100.7% with a relative standard deviation of 2.8% for 1-naphthol and mean recovery of 99.7% with a relative standard deviation (RSD of 2.6% for 2-naphthol, respectively.

This paper conducted a matched case-control study to determine the possible risk factors associated with EPHS. 208 cases and 208 matched controls were selected from two affiliated hospitals of Tianjin Medical College between 1 Dec. 1986 and 30 Oct. 1987. The conditional logistic regression analysis showed that basic diastolic blood pressure, the history of hypertension TH Time Hurry) +CH (Competition and Hostility) scores and quetelet index were the major risk factors. The odds ratio (OR) and 95% confidence of OR for these factors were 1.08 (1.04-1.12), 3.58 (1.88-6.83), 1.05 (1.03-1.09), 1.17 (1.05-1.31), respectively. Compared with Type B behavior, OR for Type A behavior is 3.06 (2.24-4.19). OR for women with history of hypertension of her father is 2.5 (1.49-4.20). The lower the average family income, education level, the higher the risk of EPHS. Basic systolic blood Pressure and body weight were positively related to EPHS. Did not find relationship between fetal sex, maternal age, parity, age of menarche, rest period during pregnancy and EPHS.
