Southern and in situ hybridization were used to examine the chromosome constitution, genomic relationships, repetitive DNA sequences, and nuclear architecture in durum wheat x tritordeum hybrids (2n = 5x = 35), where tritordeum is the fertile amphiploid (2n = 6x = 42) between Hordeum chilense and durum wheat. Using in situ hybridization, H. chilense total genomic DNA hybridized strongly to the H. chilense chromosomes and weakly to the wheat chromosomes, which showed some strongly labelled bands. pHcKB6, a cloned repetitive sequence isolated from H. chilense, enabled the unequivocal identification of each H. chilense chromosome at metaphase. Analysis of chromosome disposition in prophase nuclei, using the same probes, showed that the chromosomes of H. chilense origin were in individual domains with only limited intermixing with chromosomes of wheat origin. Six major sites of 18S-26S rDNA genes were detected on the chromosomes of the hybrids. Hybridization to Southern transfers of restriction enzyme digests using genomic DNA showed some variants of tandem repeats, perhaps owing to methylation. Both techniques gave complementary information, extending that available from phenotypic, chromosome morphology, or isozyme analysis, and perhaps are useful for following chromosomes or chromosome segments during further crossing of the lines in plant breeding programs.

Retrotransposons make up a major fraction--sometimes more than 40%--of all plant genomes investigated so far. We have isolated the reverse transcriptase domains of the Ty1-copia group elements from several species, ranging in genome size from some 100 Mbp to 23,000 Mbp, and determined the distribution patterns of these retrotransposons on metaphase chromosomes and within interphase nuclei by DNA:DNA in situ hybridization. With some exceptions, the reverse transcriptase domains were distributed over the length of the chromosomes. Exclusion from rDNA sites and some centromeres (e.g., slash pine, 23,000 Mbp, or barley, 5500 Mbp) is frequent, whereas many species exclude retrotransposons from other sites of heterochromatin (e.g., intercalary and centromeric sites in broad bean). In contrast, in the plant Arabidopsis thaliana, widely used for plant molecular genetic studies because of its small genome (c. 100 Mbp), the Ty1-copia group reverse transcriptase gene domains are concentrated in the centromeric regions, colocalizing with the 180 bp satellite sequence pAL1. Unlike the pAL1 sequence, however, the Ty1-copia signal is also detectable as weaker, diffuse hybridization along the lengths of the chromosomes. Possible mechanisms for evolution of the contrasting distributions are discussed. Understanding the physical distribution of retrotransposons and comparisons of the distribution between species is critical to understanding their evolution and the significance for generation of the new patterns of variability and in speciation.

Representatives of two major repetitive DNA sequence families from the diploid Brassica species B. campestris and B. oleracea were isolated, sequenced and localized to chromosomes by in situ hybridization. Both sequences were located near the centromeres of many chromosome pairs in both diploid species, but major sites of the two probes were all on different chromosome pairs. Such chromosome specificity is unusual for plant paracentromeric repetitive DNA. Reduction of stringency of hybridization gave centromeric hybridization sites on more chromosomes, indicating that there are divergent sequences present on other chromosomes. In tetraploid species derived from the diploids, the number of hybridization sites was different from the sum of the diploid ancestors, and some chromosomes had both sequences, indicating relatively rapid homogenization and copy number evolution since the origin of the tetraploid species.

Genomic in situ hybridization was used to identify alien chromatin in chromosome spreads of wheat, Triticum aestivum L., lines incorporating chromosomes from Leymus multicaulis (Kar. and Kir.) Tzvelev and Thinopyrum bessarabicum (Savul. and Rayss) Löve, and chromosome arms from Hordeum chilense Roem. and Schult, H. vulgare L. and Secale cereale L. Total genomic DNA from the introgressed alien species was used as a probe, together with excess amounts of unlabelled blocking DNA from wheat, for DNA:DNA in-situ hybridization. The method labelled the alien chromatin yellow-green, while the wheat chromosomes showed only the orange-red fluorescence of the DNA counterstain. Nuclei were screened from seedling root-tips (including those from half-grains) and anther wall tissue. The genomic probing method identified alien chromosomes and chromosome arms and allowed counting in nuclei at all stages of the cell cycle, so complete metaphases were not needed. At prophase or interphase, two labelled domains were visible in most nuclei from disomic lines, while only one labelled domain was visible in monosomic lines. At metaphase, direct visualization of the morphology of the alien chromosome or chromosome segment was possible and allowed identification of the relationship of the alien chromatin to the wheat chromosomes. The genomic in-situ hybridization method is fast, sensitive, accurate and informative. Hence it is likely to be of great value for both cytogenetic analysis and in plant breeding programmes.

Data are presented on the early metabolism and long-term retention of 133Ba (half-life 10.74 y) injected into six healthy male volunteers at ages 25-81 y. The tracer appeared to be mainly skeletal within several days, much earlier than predicted by the ICRP's model of alkaline earth metabolism. Excretion was mainly fecal, the relative fecal:urinary clearance up to 14 d ranging from 6 to 15 in the six subjects. The whole-body retention at 50 d (mean 8%, range 4.5 to 12%) was similar to that inferred from published data on the retention of injected Ra in man (mean 7%, range 3-13%). For about 1 y thereafter, the retention of Ba could be represented by simple power functions of time, with the rate of loss correlating with the excretory plasma clearance rate inferred over the first 4 d. In the subject aged 81 y, the pattern differed from that established following an earlier injection at age 60, but the differences were not necessarily related to advancing age.

Data are presented on the metabolism of Ca and Sr in a healthy male volunteer who, in a series of investigations conducted between the ages of 53 and 82 y, received controlled intakes of 45Ca, 47Ca, or 85Sr. No age-related trends were established, either in factors affecting the skeletal deposition of the tracers or in their subsequent retention studied for up to 462 d after intake. The data thus lend support to an important working postulate in the ICRP's model of alkaline earth metabolism.

RESUMEN

Although the nominal diameters of fibres produced in the glass and rock wool industries are usually 6-15 micrometers, these products contain a small proportion of respirable fibres (less than 3 micrometers diameter). Particular significance has been attached to the biological risk arising from the long (greater than 10 micrometers), fine (less than 1 micrometers) fibres. Therefore, the medical research sponsored by the Joint European Medical Research Board into the effects of man-made fibres on health has included detailed environmental studies on both the exposure levels to respirable fibres and the fibre size distributions at the European plants selected for epidemiological work. This paper summarizes the results obtained so far. The size distributions (length and diameter) of the airborne man-made mineral fibres are compared with similar data previously reported for airborne asbestos fibres.

1. The daily losses from the skin of Ca and Na have been measured for two healthy men. The losses included Ca and Na in exfoliated skin cells as well as in insensible perspiration. The daily output of Ca in urine was also measured.2. In five measurements, the total daily skin loss of Ca ranged from 8.0 to 21.1 mg with mean values of 16.7 and 10.8 mg for the two subjects. The total daily skin loss of Na under the same conditions ranged from 45 to 146 mg with mean values of 97 and 77 mg respectively.3. The ratio of the daily skin loss to the urinary excretion of Ca was 6.8 and 3.4%.4. There was no correlation between the daily skin loss of Ca and atmospheric humidity.5. In the last experiment, the fraction of the total skin loss of Ca due to fluid secretion was assessed by giving an I.V. dose of (47)Ca to each subject. The values derived for this fraction were 0.37 and 0.38.

RESUMEN

Rats were fed a stock diet supplemented with calcium phosphate and sodium alginate, both separately and simultaneously, over a period of 4-5 months. Observations were made at intervals on the growth rate and on visual and histological abnormalities in some body tissues. No toxic effects were observed. The absorption and skeletal retention of calcium for rats on the combined supplements remained almost unchanged but the absorption and retention of strontium was reduced 7-fold.
