RESUMEN

Enhancing the initial stages of plant growth by using polymeric gels for seed priming presents a significant challenge. This study aimed to investigate a microgel derived from polyetheramine-poly(propylene oxide) (PPO) and a bisepoxide (referred to as micro-PPO) as a promising alternative to optimize the seed germination process. The micro-PPO integrated with an iron micronutrient showed a positive impact on seed germination compared with control (Fe solutions) in which the root length yield improved up to 39%. Therefore, the element map by synchrotron-based X-ray fluorescence shows that the Fe intensities in the seed primers with the micro-PPO-Fe gel are about 3-fold higher than those in the control group, leading to a gradual distribution of Fe species through most internal embryo tissues. The use of micro-PPO for seed priming underscores their potential for industrial applications due to the nontoxicity results in zebrafish assays and environmentally friendly synthesis of the water-dispersible monomers employed.

The present study examines the potential of sequencing a neurocognitive intervention with behavioral parent training (BPT) to improve executive functions (EFs), psychiatric symptoms, and multiple indices of functional impairment in school-age children aged 7 to 11 years who have been diagnosed with attention-deficit/hyperactivity disorder (ADHD). Specifically, in a randomized controlled trial design, 85 children were assigned to either Cogmed Working Memory Training (CWMT) followed by an empirically supported, manualized BPT intervention, or to a placebo version of CWMT followed by the same BPT intervention. Working memory maintenance (i.e., attention control/short-term memory), working memory processing and manipulation, ADHD and oppositional defiant disorder (ODD) symptoms, impairment in parent-child dynamics, familial impairment, and overall functional compromise were evaluated as outcomes. The results suggest specific effects of the combined CWMT and BPT program on verbal and nonverbal working memory storage and nonverbal working memory processing and manipulation but no incremental benefits in regard to ADHD symptoms, ODD symptoms, and functional outcomes. The present findings do not support the hypothesis regarding the complementary and augmentative benefits of sequenced neurocognitive and BPT interventions for the treatment of ADHD. These results, the study's limitations, and future directions for research are further discussed.

Boron neutron capture therapy (BNCT) is a treatment modality that combines different radiation qualities. Since the severity of biological damage following irradiation depends on the radiation type, a quantity different from absorbed dose is required to explain the effects observed in the clinical BNCT in terms of outcome compared with conventional photon radiation therapy. A new approach for calculating photon iso-effective doses in BNCT was introduced previously. The present work extends this model to include information from dose-response assessments in animal models and humans. Parameters of the model were determined for tumour and precancerous tissue using dose-response curves obtained from BNCT and photon studies performed in the hamster cheek pouch in vivo models of oral cancer and/or pre-cancer, and from head and neck cancer radiotherapy data with photons. To this end, suitable expressions of the dose-limiting Normal Tissue Complication and Tumour Control Probabilities for the reference radiation and for the mixed field BNCT radiation were developed. Pearson's correlation coefficients and p-values showed that TCP and NTCP models agreed with experimental data (with r > 0.87 and p-values >0.57). The photon iso-effective dose model was applied retrospectively to evaluate the dosimetry in tumours and mucosa for head and neck cancer patients treated with BNCT in Finland. Photon iso-effective doses in tumour were lower than those obtained with the standard RBE-weighted model (between 10% to 45%). The results also suggested that the probabilities of tumour control derived from photon iso-effective doses are more adequate to explain the clinical responses than those obtained with the RBE-weighted values. The dosimetry in the mucosa revealed that the photon iso-effective doses were about 30% to 50% higher than the corresponding RBE-weighted values. While the RBE-weighted doses are unable to predict mucosa toxicity, predictions based on the proposed model are compatible with the observed clinical outcome. The extension of the photon iso-effective dose model has allowed, for the first time, the determination of the photon iso-effective dose for unacceptable complications in the dose-limiting normal tissue. Finally, the formalism developed in this work to compute photon-equivalent doses can be applied to other therapies that combine mixed radiation fields, such as hadron therapy.

OBJECTIVES: Searching for more effective and selective therapies for head and neck cancer, we demonstrated the therapeutic effect of boron neutron capture therapy (BNCT) to treat oral cancer and inhibit long-term tumor development from field-cancerized tissue in the hamster cheek pouch model. However, BNCT-induced mucositis in field-cancerized tissue was dose limiting. In a clinical scenario, oral mucositis affects patients' treatment and quality of life. Our aim was to evaluate different radioprotectors, seeking to reduce the incidence of BNCT-induced severe mucositis in field-cancerized tissue. MATERIALS AND METHODS: Cancerized pouches treated with BNCT mediated by boronophenylalanine at 5 Gy were treated as follows: control: saline solution; Hishigh : histamine 5 mg kg(-1) ; Hislow : histamine 1 mg kg(-1) ; and JNJ7777120: 10 mg kg(-1). RESULTS: Hislow reduced the incidence of severe mucositis in field-cancerized tissue to 17% vs CONTROL: 55%; Hishigh : 67%; JNJ7777120: 57%. Hislow was non-toxic and did not compromise the long-term therapeutic effect of BNCT or alter gross boron concentration. CONCLUSION: Histamine reduces BNCT-induced mucositis in experimental oral precancer without jeopardizing therapeutic efficacy. The fact that both histamine and boronophenylalanine are approved for use in humans bridges the gap between experimental work and potential clinical application to reduce BNCT-induced radiotoxicity in patients with head and neck cancer.

Newcastle disease virus (NDV) causes a fast-spreading, highly contagious infectious disease in several bird species. Commercial poultry farms in Brazil were considered free of virulent NDV. Data on NDV infection levels in backyard poultry flocks and the epidemiology of the disease are limited. The aim of this study was to perform a NDV survey in backyard poultry from households flocks located around one of the main wintering sites for migratory wild birds in Brazil, and to identify potential risk factors associated with NDV. Backyard poultry may be sentinels and a source of infection for commercial poultry, since they may have as much contact with these birds as with migratory wild birds. Data were collected from 48 randomly selected households using an epidemiological questionnaire. Serum samples from poultry were tested for NDV antibodies using an ELISA, and tracheal and cloacal swabs were collected for NDV molecular detection. The risk factors were assessed using a multivariate Poisson regression with robust variance. The ELISA showed that 33.8% of the serum samples were positive for anti-NDV antibodies and in 42 households (87.5%) at least one NDV-positive bird was found. Tracheal and cloacal swabs were negative for NDV by real time RT-PCR, possible because within this region there might flow a low pathogenicity NDV strain, which can induce seroconversion with innaparent clinical findings. The prevalence ratio (PR) increased when farmers used their own replacement poultry to restock their flock (PR=1.64; 95% CI: 1.11-2.42). Furthermore, the increasing distance of the household flock from the "Laguna do Peixe" estuary was associated with decreasing NDV seropositivity (PR=0.94; 95% CI: 0.90-0.99). This is the first study in Brazil evaluating the presence of NDV and the associated risk factors in households with backyard poultry flocks. The great number of farms with seropositive birds indicates that the virus circulates in backyard flocks, and this breeding system may be a source of NDV. These data can be used to establish appropriate biosecurity and husbandry measures for this type of breeding system to prevent NDV spread in Brazil.

A ovinocultura sempre foi uma atividade de grande importância econômica e de tradição para o Estado do Rio Grande do Sul (RS), mesmo com as crises da lã ocorridas nas décadas de 80 e 90, o rebanho ovino Gaúcho continua sendo o maior a nível nacional. Com a escassez de dados sobre essa atividade, o presente estudo possui como objetivo caracterizar a ovinocultura do RS. Para isso, foi utilizada uma amostragem planejada, caracterizada pela aleatoriedade e estratificação da amostra pelas sete Mesorregiões do Estado. Foram analisadas 705 propriedades rurais através de um questionário epidemiológico, aplicado por 25 veterinários do Departamento de Defesa Animal, da Secretaria da Agricultura, Pecuária e Agronegócio do Estado do Rio Grande do Sul. Conforme os resultados obtidos, a ovinocultura gaúcha é explorada extensivamente e baseada na produção conjunta de carne e lã, cuja principal finalidade é a subsistência. Assim, demonstrando que essa atividade ainda mantém padrões de sua origem, com pouca tecnificação, tanto em aspectos sanitários quanto reprodutivos, revelando, portanto, que a ovinocultura gaúcha ainda é vista como uma produção secundária pelos produtores rurais gaúchos, o que pode ser explicado pelos baixos investimentos neste setor.


The sheep industry has always been an important economic and traditional activity in the State of Rio Grande do Sul. In spite of the wool crises occurred in the 1980ies and 1990ies, the sheep flock remains the largest flock in Brazil. Due to scarcity of recent data on this activity, the present study aims to characterize the sheep industry of Rio Grande do Sul. For this reason, we performed a planned and probabilistic stratified cluster sampling with flocks selected propotionally by seven regions of the State. We analyzed 705 farms through an epidemiological questionnaire, applied by 25 veterinarians from the Department of Animal Health (DDA), of the Secretary of Agriculture, Livestock and Agribusiness of the state of Rio Grande do Sul (SEAPA/RS). According to the results, the sheep industry is essentially extensive and based on production of meat and wool, the main objective of which is subsistence. This indicates that the activity still maintains older standards, with almost no technical improvement regarding sanitary and reproduction aspects. This revels that sheep production in Rio Grande do Sul is still considered as a secondary exploitation of farms, which explains the low investment in this sector.

OBJECTIVES: Field-cancerized tissue can give rise to second primary tumours, causing therapeutic failure. Boron neutron capture therapy (BNCT) is based on biological targeting and would serve to treat undetectable foci of malignant transformation. The aim of this study was to optimize BNCT for the integral treatment for oral cancer, with particular emphasis on the inhibitory effect on tumour development originating in precancerous conditions, and radiotoxicity of different BNCT protocols in a hamster cheek pouch oral precancer model. MATERIALS AND METHODS: Groups of cancerized hamsters were locally exposed to single or double (2 or 4 weeks apart) applications of BNCT at different dose levels, mediated by the boron compounds boronophenylalanine (BPA) or BPA and decahydrodecaborate (GB-10) administered jointly. Cancerized, sham-irradiated hamsters served as controls. Clinical status, tumour development from field-cancerized tissue and mucositis were followed for 8 months. RESULTS: A double application (4 weeks apart) of BNCT mediated by GB-10+ BPA at a total dose of 10 Gy in two 5-Gy doses rendered the best therapeutic advantage (63-100% inhibition of tumour development from field-cancerized tissue), minimizing dose-limiting mucositis. CONCLUSION: BNCT can be optimized for the integral treatment for head and neck cancer, considering the implications for field-cancerized tissue.

O protozoário Nespora caninum é um parasito que causa grandes perdas reprodutivas e econômicas em bovinos no mundo inteiro. Os objetivos deste estudo foram verificar tanto a associação entre o histórico de aborto e a presença de anticorpos contra N. caninum, quanto a transmissão vertical como forma de manutenção da infecção nos rebanhos bovinos em regiões do Rio Grande do Sul, através da sorologia pareada de mães e filhas. Foi realizada amostragem de 60 propriedades distribuídas em duas regiões do Rio Grande do Sul, das quais foi coletado sangue de 40% dos animais presentes para a detecção de anticorpos anti-N. caninum por imunofluorescência indireta (IFI). Para verificar a relação aborto e soropositividade foi utilizado o teste de regressão logística univariada, e para sorologia de mães e filhas o teste de qui-quadrado de McNemar para dados pareados. Foram confrontados os dados de sorologia e aborto, sendo encontrada a frequência de 58,5% (24/41) de soropositivos quando havia histórico de aborto, e 16,4% (199/1215) dentre os sem histórico de aborto. Os animais soropositivos apresentaram um risco 7,21 (IC 95%, 3,65-14,32) vezes maior de possuir histórico de abortamento (estatística de Wald χ2=44,93, P<0,001). A fração atribuível à neosporose como causa de aborto na população em risco nas duas regiões foi estimada em 9,73% (λpop). O resultado sorológico de cada mãe foi pareado com o de sua filha e, pelo teste de qui-quadrado de McNemar (χ2=59,84, P<0,001), houve associação significativa entre as sorologias de mães e filhas, sugerindo transmissão vertical. Ressalta-se ainda a importância do acompanhamento sorológico para N. caninum, evitando assim manutenção de animais portadores que sirvam como reservatório do protozoário nas propriedades.


The protozoa Nesporora caninum is a parasite that causes great economic and reproductive losses in cattle worldwide. The objective of this study was to verify the association between abortion and the presence of antibodies against N. caninum and the vertical transmission as a means of maintaining the infection in cattle herds in Rio Grande do Sul by matching the serology of mothers and daughters. Sampling was performed in 60 dairy farms of two regions of the state, where blood was collected from 40% of the herds for the detection of anti-N. caninum by indirect immunofluorescence (IFI). To verify the association between abortion and seropositivity we used the univariate logistic regression test, and for the serology of mothers and daughters the chi-square McNemar test for paired data. In comparing serology and abortion data, a prevalence of 58.5% (24/41) among cattle with history of abortion, and 16.4% (199/1215) prevalence between those with no history of abortion was found; seropositive animals were 7.21 times more likely (95% CI, 3.65-14.32) to have previously aborted (Wald statistic χ 2=44.93, P<0.001). The fraction affected by neosporosis in the population studied was estimated as 9.73% (λpop). The serologic result of each mother was paired with her daughter and showed by the McNemar chi-square (χ2=59.84, P<0.001) significant association between the serology of mothers and daughters, suggesting vertical transmission. It is worth to note the importance of serological monitoring for N. caninu to avoid maintenance of animals that may serve as carries of the parasite reservoir on the farms.

We previously demonstrated the efficacy of boron neutron capture therapy (BNCT) mediated by boronophenylalanine (BPA), GB-10 (Na(2)(10)B(10)H(10)) and (GB-10+BPA) to control tumors, with no normal tissue radiotoxicity, in the hamster cheek pouch oral cancer model. Herein we developed a novel experimental model of field-cancerization and precancerous lesions (globally termed herein precancerous tissue) in the hamster cheek pouch to explore the long-term potential inhibitory effect of the same BNCT protocols on the development of second primary tumors from precancerous tissue. Clinically, second primary tumor recurrences occur in field-cancerized tissue, causing therapeutic failure. We performed boron biodistribution studies followed by in vivo BNCT studies, with 8 months follow-up. All 3 BNCT protocols induced a statistically significant reduction in tumor development from precancerous tissue, reaching a maximum inhibition of 77-100%. The inhibitory effect of BPA-BNCT and (GB-10+BPA)-BNCT persisted at 51% at the end of follow-up (8 months), whereas for GB-10-BNCT it faded after 2 months. Likewise, beam-only elicited a significant but transient reduction in tumor development. No normal tissue radiotoxicity was observed. At 8 months post-treatment with BPA-BNCT or (GB-10+BPA)-BNCT, the precancerous pouches that did not develop tumors had regained the macroscopic and histological appearance of normal (non-cancerized) pouches. A potential new clinical application of BNCT would lie in its capacity to inhibit local regional recurrences.

The National Atomic Energy Commission of Argentina (CNEA) constructed a novel thermal neutron source for use in boron neutron capture therapy (BNCT) applications at the RA-3 research reactor facility located in Buenos Aires. The aim of the present study was to perform a dosimetric characterization of the facility and undertake radiobiological studies of BNCT in an experimental model of oral cancer in the hamster cheek pouch. The free-field thermal flux was 7.1 x 10(9) n cm(-2)s(-1) and the fast neutron flux was 2.5 x 10(6) n cm(-2)s(-1), indicating a very well-thermalized neutron field with negligible fast neutron dose. For radiobiological studies it was necessary to shield the body of the hamster from the neutron flux while exposing the everted cheek pouch bearing the tumors. To that end we developed a lithium (enriched to 95% in (6)Li) carbonate enclosure. Groups of tumor-bearing hamsters were submitted to BPA-BNCT, GB-10-BNCT, (GB-10+BPA)-BNCT or beam only treatments. Normal (non-cancerized) hamsters were treated similarly to evaluate normal tissue radiotoxicity. The total physical dose delivered to tumor with the BNCT treatments ranged from 6 to 8.5 Gy. Tumor control at 30 days ranged from 73% to 85%, with no normal tissue radiotoxicity. Significant but reversible mucositis in precancerous tissue surrounding tumors was associated to BPA-BNCT. The therapeutic success of different BNCT protocols in treating experimental oral cancer at this novel facility was unequivocally demonstrated.

Ex-situ BNCT for multifocal unresectable liver metastases employing whole or partial autograft techniques requires knowledge of boron concentrations in healthy liver and metastases following perfusion and immersion in Wisconsin solution (W), the procedure employed for organ preservation during ex-situ irradiation. Measurements of boron concentration in blood, liver and metastases following an intravenous infusion of BPA-F in five colorectal liver metastases patients scheduled for surgery were performed. Tissue samples were evaluated for boron content pre and post perfusion and immersion in W. Complementary histological studies were performed. The data showed a dose-dependent BPA uptake in liver, a boron concentration ratio liver/blood close to 1 and a wide spread in the metastases/liver concentration ratios in the range 0.8-3.6, partially attributable to histological variations between samples. Based on the boron concentrations and dose considerations (liver < or =15 Gy-Eq and tumor> or =40 Gy-Eq) at the RA-3 thermal neutron facility (mean flux of about (6+/-1) x 10(9) n cm(-2)s(-1)), ex-situ treatment of liver metastases at RA-3 would be feasible.

Microdistributions of the prospective BNCT-compound CuTCPH, a carborane-containing tetraphenylporphyrin with one Cu atom in its molecular structure, have been obtained in tissue sections of different organs of tumor-bearing and normal Syrian hamsters injected with the boron compound by employing a heavy ion microbeam. High resolution X-ray spectroscopy following micro-PIXE (Particle Induced X-ray Emission with micrometer-sized beams) with a focused (16)O ion beam was used. Focusing was performed with a heavy-ion scanning high-precision magnetic quadrupole triplet microprobe. Squamous Cell Carcinomas were induced on the right Cheek Pouch of Syrian Hamsters (HCP), sampled, cryo-sectioned and freeze-dried. Two-dimensional maps of elemental concentration were obtained by scanning the beam over the samples. Very non-uniform Cu concentrations were found in all sections.

The possibility of detection of incipient cellular alterations is central to early diagnosis and to clinician's capacity to discriminate between samples that appear similar on routine preparations. We examined the value of silver-stained nucleolar organizer regions (AgNOR) in detecting radio-induced alterations in a model of squamous epithelium biologically similar to oral mucosa. Morphometry of AgNOR has been proven to be of value in the detection of incipient cellular alterations. This method allows for the quantitative evaluation of lesions induced by high doses of radiation long before they become apparent in routine preparations. We herein examine the capacity of AgNOR to reveal the response to low doses of radiation, closer to the therapeutic or accidental dose to which the epithelium of oral mucosa may be exposed.

The model of hamster cheek pouch carcinogenesis closely mimics the development of human oral cancer. The study of the interaction between chemical carcinogens and radiation in the process of oral carcinogenesis is of interest given that the oral cavity is frequently exposed to chemical carcinogens such as alcohol and tobacco and is the route of entry of therapeutic radiation. In this context, markers of incipient alterations associated to a process of malignant transformation would contribute to early diagnosis and follow-up. The aim of the present study was to assess the early changes produced by carcinogenic agents applied separately or combined in a two-stage carcinogenesis protocol in hamster cheek pouch. The cheek pouch of the hamsters was treated with a single dose of radiation (20 Gy) or 7,12-dimethylbenz(a)anthracene (DMBA) as initiating agents and 12-O-tetradecanoylphorbol-13-acetate (TPA) as a promoting agent for 1 or 2 weeks. The end-points chosen to identify early alterations were hyperplastic foci and silver-stained nucleolar organizer regions (Ag NOR). The data show that both markers are useful in the detection of early alterations compatible with a process of malignant transformation.

Report about an electronic data processing system for gynaecology. The developed data document design and data flowchart are shown. The accumulated data allowed a detailed interpretation record. For all clinical treated patients the computer printed out a final gynaecological epicrisis. The system is an improvement of the information and the typewriting work of medial staff has been reduced.
