RESUMEN

We evaluated the effect of cyclosporine A (CsA) administration on the level of miR-26-5p in rat gingival tissues and human gingival fibroblasts (HGFs) by qRT-PCR assay. Further, we conducted Western blotting and immunohistochemical analysis to assess the expressions of PTEN, PI3K, and p-AKT, and evaluated cell proliferation of HGFs by MTT assay. CsA treatment significantly downregulated the expressions of miR-26-5p and PTEN and upregulated the expressions of PI3K and p-AKT in both rat gingival tissues and HGFs. Overexpression of miR-26-5p inhibited CsA-induced overgrowth of HGFs, whereas knockdown of miR-26-5p promoted the overgrowth. PTEN knockdown not only promoted CsA-induced overgrowth of human HGFs but also reversed the repressive effects of miR-26-5p on CsA-induced overgrowth of HGFs. Our results revealed that miRNA-26-5p could repress CsA-induced overgrowth of human HGFs by regulating PTEN/PI3K/AKT pathway.

PURPOSE: To investigate the status of dental caries in children of primary school in Hangzhou City from 2009 to 2011. METHODS: The status of dental caries was examined annually from 2009 to 2011, and the caries prevalence, filling rates, mean DMFT/dmft were recorded. SPSS 13.0 software package was used for statistical analysis. RESULTS: From 2009 to 2011, the caries prevalence of deciduous teeth were 49.27%, 48.09% and 48.33%, mean dmft were 2.78, 2.81 and 2.84, filling rates of deciduous teeth were 3.92%, 4.31% and 4.28%, respectively. No significant difference was found in the caries prevalence of temporary teeth, filling rates of temporary teeth, mean dmft. For the permanent teeth, the caries prevalence were 20.24%, 18.48% and 15.85%, mean DMFT were 0.46, 0.41 and 0.33, filling rates were 10.17%, 15.67% and 23.00%, respectively. From 2009 to 2011, the caries prevalence and mean DMFT of permanent teeth was decreased, while the filling rate was increased. CONCLUSIONS: In the past three years, the status of dental caries of permanent teeth shows a remarkable improving tendency in children of primary school in Hangzhou City. However, the status of dental caries of deciduous teeth presents no significant improvement.

OBJECTIVE: To investigate the effects of Drynaria fortunei J. Smith naringin (DFSN) on proliferation and differentiation of human periodontal ligament cells (hPDLC). METHODS: Cultured human periodontal ligament cells were treated with DFSN at different concentrations. Cell proliferation was determined by MTT method; cell differentiation was evaluated through the examination of alkaline phosphate (ALP) activities. RESULTS: DFSN in a concentration ranged from 0.01 mu mol/L to 10 mu mol/L showed a promoting effect on proliferation of hPDLC (P< 0.05), and it also promoted ALP activities of hPDLC (P<0.05). CONCLUSION: Drynaria fortunei J. Smith naringin can promotes the proliferation and differentiation of human periodontal ligament cells.

During mouse eye development, the correct formation of the lens occurs as a result of reciprocal interactions between the neuroectoderm that forms the retina and surface ectoderm that forms the lens. Although many transcription factors required for early lens development have been identified, the mechanism and genetic interactions mediated by them remain poorly understood. Foxe3 encodes a winged helix-forkhead transcription factor that is initially expressed in the developing brain and in the lens placode and later restricted exclusively to the anterior lens epithelium. Here, we show that targeted disruption of Foxe3 results in abnormal development of the eye. Cells of the anterior lens epithelium show a decreased rate of proliferation, resulting in a smaller than normal lens. The anterior lens epithelium does not properly separate from the cornea and frequently forms an unusual, multilayered tissue. Because of the abnormal differentiation, lens fiber cells do not form properly, and the morphogenesis of the lens is greatly affected. The abnormally differentiated lens cells remain irregular in shape, and the lens becomes vacuolated. The defects in lens development correlate with changes in the expression of growth and differentiation factor genes, including DNase II-like acid DNase, Prox1, p57, and PDGFalpha receptor. As a result of abnormal lens development, the cornea and the retina are also affected. While Foxe3 is also expressed in a distinct region of the embryonic brain, we have not observed abnormal development of the brain in Foxe3(-/-) animals.

Bone morphogenetic protein (BMP) signals pattern the dorsal neural tube, defining distinct neuronal progenitor cell domains along the dorsoventral axis of the developing spinal cord. These dorsally expressed BMPs appear to have a limited range of action. The mechanisms that regulate this range of action are unclear. We created a GAL4/UAS bigenic mouse system to overexpress BMP4 or a mutant form of BMP4 (mutBMP4), which lacks a subset of amino-terminal basic amino acids that limits its range of action, in the dorsal neural tube. UAS-Bmp4 and UAS-mutBmp4 responder genes were activated in the dorsal neural tube by a Wnt1-GAL4 transgene. Analysis of the spinal cords of bigenic embryos that expressed comparable levels of transgenic transcripts revealed that mutBMP4 acted more ventrally in the neural tube than BMP4. This suggests that the amino-terminal basic amino-acid motif of mature BMP4 controls long-range activity for dorsoventral patterning of the vertebrate neural tube.
