RESUMEN

Background: The limbus is located at a 2-mm-wide area between the bulbar conjunctiva and the cornea and has been suggested to be the niche of corneal epithelial stem cells and immune cells. Like the skin and intestines, the cornea is also an important mucosal surface, and immune cells on the cornea play critical roles in immune surveillance to ensure barrier surface homeostasis and protection from various environmental damage and infections. Single-cell RNA sequencing (scRNA-seq) analysis of protein tyrosine phosphatase receptor type C positive (PTPRC+) hematopoietic cells from the corneal limbus could provide a single cell atlas of all the immune cell subsets. Methods: We performed single-cell RNA sequencing to generate transcriptomic profile for 804 sort-purified hematopoietic cells from the corneal limbus of three healthy donors. Results: Our analysis identified a primary transcriptomic pattern for multiple immune cell subtypes, including naive T cells, antiviral effector CD8+ T cells, and innate immune cells such as IDO1+ mature regulatory dendritic cells (mregDCs), macrophages, monocytes, and basophils in the human corneal limbus. Conclusion: Overall, single-cell transcriptomic analysis of limbal immune cells suggested the possible contribution of these cells on the adaptive and innate immune response of the human cornea.

Most story plots contain multiple characters who are independent, interact, and often have conflicting goals. One would expect that narrative understanding would require monitoring of the goals, concerns, and situations of multiple agents. There is considerable evidence that understanders monitor the primary protagonist's goal plans (e.g., Suh & Trabasso, 1993). However, there is relatively little research on the extent to which understanders monitor the goals of multiple agents. We investigated the impact of characters' roles and prominence on the extent to which understanders monitor the goal plans of multiple characters in a feature length film. In Experiment 1, participants made situation change judgments, and in Experiment 2, they verbally described scenes. Both types of judgments indicated that viewers monitor the goals and plans of multiple agents but do so to a greater extent for characters more prominent to the plotline.

We explored methods of using latent semantic analysis (LSA) to identify reading strategies in students' self-explanations that are collected as part of a Web-based reading trainer. In this study, college students self-explained scientific texts, one sentence at a time. ISA was used to measure the similarity between the self-explanations and semantic benchmarks (groups of words and sentences that together represent reading strategies). Three types of semantic benchmarks were compared: content words, exemplars, and strategies. Discriminant analyses were used to classify global and specific reading strategies using the LSA cosines. All benchmarks contributed to the classification of general reading strategies, but the exemplars did the best in distinguishing subtle semantic differences between reading strategies. Pragmatic and theoretical concerns of using LSA are discussed.
