RESUMEN

Fiber-based implantable electronics are one of promising candidates for in vivo biomedical applications thanks to their unique structural advantages. However, development of fiber-based implantable electronic devices with biodegradable capability remains a challenge due to the lack of biodegradable fiber electrodes with high electrical and mechanical properties. Here, a biocompatible and biodegradable fiber electrode which simultaneously exhibits high electrical conductivity and mechanical robustness is presented. The fiber electrode is fabricated through a facile approach that incorporates a large amount of Mo microparticles into outermost volume of a biodegradable polycaprolactone (PCL) fiber scaffold in a concentrated manner. The biodegradable fiber electrode simultaneously exhibits a remarkable electrical performance (≈43.5 Ω cm-1 ), mechanical robustness, bending stability, and durability for more than 4000 bending cycles based on the Mo/PCL conductive layer and intact PCL core in the fiber electrode. The electrical behavior of the biodegradable fiber electrode under the bending deformation is analyzed by an analytical prediction and a numerical simulation. In addition, the biocompatible properties and degradation behavior of the fiber electrode are systematically investigated. The potential of biodegradable fiber electrode is demonstrated in various applications such as an interconnect, a suturable temperature sensor, and an in vivo electrical stimulator.

Tricholoma matsutake is an ectomycorrhizal fungus that has a symbiotic relationship with the root of Pinus densiflora. Soil microbial communities greatly affect the growth of T. matsutake, however, few studies have examined the characteristics of these communities. In the present study, we analyzed soil fungal communities from Gyeongju and Yeongdeok using metagenomic pyrosequencing to investigate differences in fungal species diversity, richness, and taxonomic composition between the soil under T. matsutake fruiting bodies (Sample 2) and soil where the fairy ring of T. matsutake was no longer present (Sample 1). The same spot was investigated three times at intervals of four months to observe changes in the community. In the samples from Yeongdeok, the number of valid reads was lower than that at Gyeongju. The operational taxonomic units of most Sample 2 groups were less than those of Sample 1 groups, indicating that fungal diversity was low in the T. matsutakedominant soil. The soil under the T. matsutake fruiting bodies was dominated by more than 51% T. matsutake. From fall to the following spring, the ratio of T. matsutake decreased. Basidiomycota was the dominant phylum in most samples. G-F1-2, G-F2-2, and Y-F1-2 had the genera Tricholoma, Umbelopsis, Oidiodendron, Sagenomella, Cladophialophora, and Phialocephala in common. G-F1-1, G-F2-1, and Y-F1-1 had 10 genera including Umbelopsis and Sagenomella in common. From fall to the following spring, the amount of phyla Basidiomycota and Mucoromycota gradually decreased but that of phylum Ascomycota increased. We suggest that the genus Umbelopsis is positively related to T. matsutake.

Despite the multitude of available alternatives, the Grignard coupling-based synthesis of polycarbosilanes remains attractive, offering the benefit of easy structural design. Moreover, this method allows one to obtain a polymer precursor with the stoichiometric Si : C ratio required for SiC ceramic production and is also suited for the synthesis of polymers containing curable functional groups (e.g., allyl and vinyl). Herein, vinyl-substituted hydridopolycarbosilane was synthesized from three different starting materials (Cl3SiCH2Cl, (MeO)3SiCH2Cl, and (EtO)3SiCH2Cl) using Grignard coupling, and the effects of starting material on polymer growth behavior were investigated. The alkoxysilane starting materials had the advantage of being safe to handle, but had a limitation in polymer growth. The largest molecular weight was observed for Cl3SiCH2Cl, although side reactions occurred. This behavior was ascribed to the retention of unreacted -SiCH2Cl groups contributing to polymer growth via coupling with neighboring polymer chains.

A systematic review and meta-analysis of randomized controlled trials (RCTs) and observational studies was conducted to assess the association between menopausal hormone therapy and cardiovascular disease. The PubMed and EMBASE databases were searched for articles published from 2000 to 2019, using review methods based on a previous Cochrane review. Quality assessment of RCTs and observational studies was conducted using the Jadad scale and the Newcastle-Ottawa Scale, respectively. A total of 26 RCTs and 47 observational studies were identified. The study populations in the RCTs were older and had more underlying diseases than those in the observational studies. Increased risks of venous thromboembolism [summary estimate (SE), 95% confidence interval (CI): RCTs, 1.70, 1.33-2.16; observational studies, 1.32, 1.13-1.54] were consistently identified in both study types, whereas an increased risk of stroke in RCTs (SE: 1.14, 95% CI: 1.04-1.25) and a decreased risk of myocardial infarction in observational studies (SE: 0.79, 95% CI: 0.75-0.84) were observed. Differential clinical effects depending on timing of initiation, underlying disease, regimen type, and route of administration were identified through subgroup analyses. These findings suggest that underlying disease and timing of initiation should be carefully considered before starting therapy in postmenopausal women.

Fungal endophytes are symbiotic microorganisms that are often found in asymptomatic plants. This study describes the genetic diversity of the fungal endophytes isolated from the roots of plants sampled from the west coast of Korea. Five halophytic plant species, Limonium tetragonum, Suaeda australis, Suaeda maritima, Suaeda glauca Bunge, and Phragmites australis, were collected from a salt marsh in Gochang and used to isolate and identify culturable, root-associated endophytic fungi. The fungal internal transcribed spacer (ITS) region ITS1-5.8S-ITS2 was used as the DNA barcode for the classification of these specimens. In total, 156 isolates of the fungal strains were identified and categorized into 23 genera and two phyla (Ascomycota and Basidiomycota), with Dothideomycetes and Sordariomycetes as the predominant classes. The genus Alternaria accounted for the largest number of strains, followed by Cladosporium and Fusarium. The highest diversity index was obtained from the endophytic fungal group associated with the plant P. australis. Waito-C rice seedlings were treated with the fungal culture filtrates to analyze their plant growth-promoting capacity. A bioassay of the Sm-3-7-5 fungal strain isolated from S. maritima confirmed that it had the highest plant growth-promoting capacity. Molecular identification of the Sm-3-7-5 strain revealed that it belongs to Alternaria alternata and is a producer of gibberellins. These findings provided a fundamental basis for understanding the symbiotic interactions between plants and fungi.

Tricholoma matsutake is an ectomycorrhizal fungus, related with the host of Pinus densiflora. Most of studies on T. matsutake have focused on mycelial growth, genes and genomics, phylogenetics, symbiosis, and immune activity of this strain. T. matsutake is known for its unique fragrance in Eastern Asia. The most major component of its scent is (R)-(-)-1-octen-3-ol and is biosynthesized from the substrate linoleic acid by the sequential reaction of lipoxygenase and peroxide lyase. Here, we report for the first time the biosynthesis of (R)-(-)- 1-octen-3-ol of T. matsutake using the yeast Saccharomyces cerevisiae as a host. In this study, cDNA genes correlated with these reactions were cloned from T. matsutake, and expression studies of theses genes were carried out in the yeast Saccharomyces cerevisiae. The product of these genes expression study was carried out with Western blotting. The biosynthesis of (R)-(-)- 1-octen-3-ol of T. matsutake in recombinant Saccharomyces cerevisiae was subsequently identified with GC-MS chromatography analysis. The biosynthesis of (R)-(-)-1-octen-3-ol with S. cerevisiae represents a significant step forward.

Dysfunction or progressive degeneration of retinal pigment epithelium (RPE) contributes in the initial pathogenesis of age-related macular degeneration (AMD) causing irreversible vision loss, which makes RPE the prime target of the disease. The present study aimed to identify compounds to protect 4-hydroxynonenal (4-HNE)-induced RPE cell death by inhibiting NADPH oxidase 4 (NOX4) activity, not just as free radical scavengers, using ARPE-19, a human adult retinal pigment epithelial cell line, as a RPE representative. Novel thirty-two 6-ureido/thioureido-2,4,5-trimethylpyridin-3-ol derivatives 17 were synthesized and tested. We found that there was a strong correlation between level of protective effect of compounds 17 against 4-HNE-induced APRE-19 cell death and that of inhibitory activity against 4-HNE-induced superoxide production, and that most of the compounds 17 showed minimal DPPH radical scavenging activity. Compound 17-28 showed the best protective activity against 4-HNE-induced superoxide production (79.5% inhibition) and cell death (85.1% recovery) at 10 µM concentration, which was better than that of VAS2870, a NOX2/4 inhibitor. In addition, compound 17-28 blocked 4-HNE-induced apoptosis of ARPE-19 cells in a concentration-dependent manner. The results indicate that compound 17-28 may be a lead compound to develop AMD therapeutics.

Although current chemotherapeutic agents are active at the beginning of therapy, the most common risk is the development of resistance during later stages in almost all cancer types including breast cancer. Hence, investigation of novel drugs is still a priority goal for cancer treatment. The objective of the present study is to investigate the anticancer effect of a derivative of stilbene, deoxyrhapontigenin (DR) isolated from Rheum undulatum L. root extracts against the chemoresistant MCF-7/adr and its parental MCF-7 human breast cancer cells. The morphological images indicate that DR induces an extensive cytoplasmic vacuolation in breast cancer cells. Mechanistic investigations revealed that DR treatment causes endoplasmic reticulum (ER) dilation and upregulated the expression of ER stress markers GRP78, IRE1α, eIF2α, CHOP, JNK, and p38. Subsequently, we also identified that DR increases the levels of apoptotic fragment of PARP (89 kDa) in breast cancer cells. Blocking the expression of one of the components of the ER stress-mediated apoptosis pathway, CHOP using siRNA significantly decreased DR-induced apoptotic cleavage of PARP. In summary, the present study suggests that the induction of ER stress-mediated apoptosis by DR may account for its cytotoxic effects in human breast cancer cells.

Five halophytic plant species, Suaeda maritima, Limonium tetragonum, Suaeda australis, Phragmites australis, and Suaeda glauca Bunge, which are native to the Muan salt marsh of South Korea, were examined for fungal endophytes by sequencing the internal transcribed spacer (ITS) region containing ITS1, 5.8S rRNA, and ITS2. In total, 160 endophytic fungal strains were isolated and identified from the roots of the 5 plant species. Taxonomically, all 160 strains belonged to the phyla Ascomycota, Basidiomycota, and Zygomycota. The most dominant genus was Fusarium, followed by the genera Penicillium and Alternaria. Subsequently, using 5 statistical methods, the diversity indices of the endophytes were determined at genus level. Among these halophytic plants, P. australis was found to host the greatest diversity of endophytic fungi. Culture filtrates of endophytic fungi were treated to Waito-C rice seedlings for plant growth-promoting effects. The fungal strain Su-3-4-3 isolated from S. glauca Bunge provide the maximum plant length (20.1 cm) in comparison with wild-type Gibberella fujikuroi (19.6 cm). Consequently, chromatographic analysis of the culture filtrate of Su-3-4-3 showed the presence of physiologically active gibberellins, GA1 (0.465 ng/mL), GA3 (1.808 ng/mL) along with other physiologically inactive GA9 (0.054 ng/mL) and GA24 (0.044 ng/mL). The fungal isolate Su-3-4-3 was identified as Talaromyces pinophilus.

Hypochlorous acid treatment of a microalga, Chlorella vulgaris, was investigated to improve the quality of microalgal lipid and to obtain high biodiesel-conversion yield. Because chlorophyll deactivates the catalyst for biodiesel conversion, its removal in the lipid-extraction step enhances biodiesel productivity. When microalgae contacted the hypochlorous acid, chlorophyll was removed, and resultant changes in fatty acid composition of microalgal lipid were observed. The lipid-extraction yield after activated clay treatment was 32.7 mg lipid/g cell; after NaClO treatment at 0.8% available chlorine concentration, it was 95.2 mg lipid/g cell; and after NaCl electrolysis treatment at the 1 g/L cell concentration, it was 102.4 mg lipid/g cell. While the contents of all of the unsaturated fatty acids except oleic acid, in the microalgal lipid, decreased as the result of NaClO treatment, the contents of all of the unsaturated fatty acids including oleic acid decreased as the result of NaCl electrolysis treatment.

Acid-catalyzed hot-water treatment for efficient extraction of lipids from a wet microalga, Chlorella vulgaris, was investigated. For an initial fatty acids content of 381.6mg/g cell, the extracted-lipid yield with no heating and no catalyst was 83.2mg/g cell. Under a 1% H2SO4 concentration heated at 120°C for 60min, however, the lipid-extraction yield was 337.4mg/g cell. The fatty acids content, meanwhile, was 935mg fatty acid/g lipid. According to the severity index formula, 337.5mg/g cell of yield under the 1% H2SO4 concentration heated at 150°C for 8min, and 334.2mg/g cell of yield under the 0.5% H2SO4 concentration heated at 150°C for 16min, were obtained. The lipids extracted by acid-catalyzed hot-water treatment were converted to biodiesel. The biodiesel's fatty acid methyl ester (FAME) content after esterification of the microalgal lipids was increased to 79.2% by the addition of excess methanol and sulfuric acid.
