Purpose: To demonstrate advantages of spectral dual-layer computed tomography (CT) in diagnosing pulmonary embolism (PE). To compare D-dimer values in patients with PE and concomitant COVID-19 pneumonia to those in patients without PE and COVID-19 pneumonia. To compare D-dimer values in cases of minor versus extensive PE. Methods: A monocentric retrospective study of 1500 CT pulmonary angiographies (CTPAs). Three groups of 500 consecutive examinations: 1) using conventional multidetector CT (CTC), 2) using spectral dual-layer CT (CTS), and 3) of COVID-19 pneumonia patients using spectral dual-layer CT (COV). Only patients with known D-dimer levels were enrolled in the study. Results: Prevalence of inconclusive PE findings differed significantly between CTS and CTC (0.8 % vs. 5.4 %, p < 0.001). In all groups, D-dimer levels were significantly higher in PE positive patients than in patients without PE (CTC, 8.04 vs. 3.05 mg/L; CTS, 6.92 vs. 2.57 mg/L; COV, 10.26 vs. 2.72 mg/L, p < 0.001). There were also statistically significant differences in D-dimer values between minor and extensive PE in the groups negative for COVID-19 (CTC, 5.16 vs. 8.98 mg/L; CTS 3.52 vs. 9.27 mg/L, p < 0.001). The lowest recorded D-dimer value for proven PE in patients with COVID-19 pneumonia was 1.19 mg/L. Conclusion: CTPAs using spectral dual-layer CT reduce the number of inconclusive PE findings. Plasma D-dimer concentration increases with extent of PE. Cut-off value of D-dimer with 100 % sensitivity for patients with COVID-19 pneumonia could be doubled to 1.0 mg/L. This threshold would have saved 110 (22 %) examinations in our cohort.

Day and nighttime autumn fish abundance and biomass were studied in the pelagic and littoral zones of Lake Sauka in Latvia. Both pelagic methods (hydroacoustics and trawling) revealed significantly higher fish abundance and biomass during the day than at night, especially in deeper zones (below 3 m). Roach (Rutilus rutilus) and Eurasian perch (Perca fluviatilis) dominated the trawl catches during the day, while roach and ruffe dominated at night. Fish smaller than 14 cm strongly dominated in both the trawl catches and hydroacoustic observations. Our hydroacoustic sampling found inhomogeneous pelagic distributions of fish aggregated in big shoals during the day. In the littoral zone, which was sampled by beach seining, both the abundance and biomass were significantly higher at night than during the day. Roach, bleak, and European perch usually dominated in beach seine catches during the day and at night. The daytime pelagic biomass found by hydroacoustics was 62 kg/ha and it decreased to approximately 11 kg/ha at night. The littoral biomass found by beach seining was diurnally opposite, 4 kg/ha during the day and 37 kg/ha at night. It is obvious that diurnal horizontal migrations between pelagic and littoral zones, and shoaling behavior during the daytime are common patterns in the shallow Lake Sauka during the autumn. The study of the spatial distribution of fish is extremely important for the establishment of an appropriate monitoring plan for the purposes of the Water Framework Directive with regard to the morphometry of the lake, the geographical location, and the sampling period of the year. This study also shows that the combination of completely non-invasive hydroacoustic and other methods that are invasive (trawls, beach seines) but not as destructive as gillnets, which are normally used for scientific fish monitoring in Europe, could be a future way forward for fish monitoring.

Stable isotope analysis (SIA) is widely used to study trophic ecology and food webs in aquatic ecosystems. In the case of fish, muscle tissue is generally preferred for SIA, and the method is lethal in most cases. We tested whether blood and fin clips can be used as non-lethal alternatives to muscle tissue for examining the isotopic composition of two freshwater predatory fish, European catfish (Silurus glanis) and Northern pike (Esox lucius), species of high value for many freshwater systems as well as invasive species in many others. Blood samples from the caudal vein, anal fin clips, and dorsal muscle obtained by biopsy punch were collected from four catfish and pike populations (14-18 individuals per population). Subsequently, these samples were analyzed for δ13C and δ15N. The effects of alternative tissues, study site, and fish body mass on the isotopic offset were investigated. Both species showed a correlation between the isotopic offset and the tissue type, as well as the study site, but no significant relationship with the body mass. The isotopic offsets between tissues were used to calculate the conversion equations. The results demonstrated that both blood and fin clips are suitable and less invasive alternative to muscle in SIA studies focused on European catfish and Northern pike. Blood provided better correspondence to muscle isotope values. However, our results clearly demonstrated that isotopic offsets between tissues vary significantly among populations of the same species. Therefore, obtaining a muscle biopsy from several individuals in any population is advisable to gain initial insights and establish a possible population-specific inter-tissue conversion.

RESUMEN

In contrast to conventional diffusion magnetic resonance imaging (MRI), multi-b-value diffusion MRI methods are able to separate the signal from free water, pseudo-diffusion, and non-Gaussian components of water molecule diffusion. These approaches can then be utilised in so-called intravoxel incoherent motion imaging and diffusion kurtosis imaging. Various parameters provided by these methods can describe additional characteristics of the tissue microstructure and potentially help in the diagnosis and classification of various pathological processes. In this review, we present the basic principles and methods of analysing multi-b-value diffusion imaging data and specifically focus on the known possibilities for its use in the diagnosis of brain lesions. We also suggest possible directions for further research.

Intra-species variability in isotopic niches, specifically isotopic total niche width (ITNW), isotopic individual niche width (IINW), and isotopic individual specialization (IIS), was studied using an innovative approach without sacrificing the vertebrates. Stable isotopes (δ13C, δ15N) in four body tissues differing in isotopic half-life were analyzed from four freshwater fish species representing different trophic positions. ITNW was widest for the apex predator (European catfish) and narrowest for the obligate predator (Northern pike). IINW exhibited a polynomial trend for the European catfish, Northern pike, and Eurasian perch (mesopredator), decreasing with body mass and increasing again after exceeding a certain species-dependent body mass threshold. Thus, for ectotherms, apex predator status is linked rather to its size than to the species. In herbivores (rudd), IINW increased with body mass. The IIS of predators negatively correlated with site trophic state. Therefore, eutrophication can significantly change the foraging behavior of certain species. We assume that the observed trends will occur in other species at similar trophic positions in either aquatic or terrestrial systems. For confirmation, we recommend conducting a similar study on other species in different habitats.

To examine the suitability of fish scales as potential tracers of nutrient pollution, we analysed the nitrogen and carbon stable isotope values (δ15N and δ13C) in scales of a generalist fish species, roach Rutilus rutilus, collected from 22 Czech reservoirs covering wide gradients of catchment land use and nutrient enrichment. Using generalised additive mixed models in the first step and generalised linear mixed models in the second step, we evaluated the response of roach scale stable isotope values to catchment land use variables (percentage of agricultural land and human population density) and in-reservoir water quality variables. Roach scale δ15N values varied by 15 ‰ among the reservoirs and were strongly, linearly, and positively associated with the percentage of agricultural land in the reservoir catchments, pointing to agriculture as the dominant source of nitrogen pollution in the investigated systems. Roach scale δ13C values differed by 8 ‰ among the studied reservoirs and were not related to catchment land use variables or in-reservoir primary production (chlorophyll-a levels). Possible variation in roach foraging strategies (littoral versus pelagic) between reservoirs or the contrasting effects of eutrophication-related autotrophic and heterotrophic processes on baseline δ13C values may explain the lack of relationships between roach scale δ13C values and the explanatory variables. In summary, our findings show that fish scale δ15N values are sensitive bioindicators of catchment-derived anthropogenic nitrogen inputs to freshwater ecosystems. Because scales can be sampled in a nonlethal way and δ15N analysis is relatively inexpensive, we suggest that measuring the δ15N values of fish scales could be an effective method for monitoring nitrogen pollution in aquatic environments.

Ligula intestinalis (Linnaeus, 1758) is a tapeworm parasite with a worldwide distribution that uses a wide variety of fish species as its second intermediate host. In the present study, we investigated the prevalence and population genetic structure of plerocercoids of L. intestinalis in five common cyprinoid species, roach Rutilus rutilus (Linnaeus), freshwater bream Abramis brama (Linnaeus), white bream Blicca bjoerkna (Linnaeus), bleak Alburnus alburnus (Linnaeus), and rudd Scardinius erythrophthalmus (Linnaeus), collected in six water bodies of the Czech Republic (Milada, Most, Medard, Jordán, Rímov and Lipno). Of the six study sites, the highest frequency of parasitism was recorded in Lake Medard (15%). The overall prevalence rate among the species was as follows: roach > rudd ≥ freshwater bream > bleak > white bream. Two mitochondrial genes (cytb and COI) were used to compare the population genetic structure of parasite populations using selected samples from the five fish species. The results of the phylogenetic analysis indicated that all populations of L. intestinalis were placed in Clade A, previously identified as the most common in Europe. At a finer scale, haplotype network and PCoA analyses indicated the possible emergence of host specificity of several mtDNA haplotypes to the freshwater bream. Moreover, pairwise Fixation indices (FST) revealed a significant genetic structure between the parasite population in freshwater bream and other host species. Parasite populations in roach not only showed the highest rate of prevalence but also depicted a maximum number of shared haplotypes with populations from bleak and rudd. Our results suggest that recent ecological differentiation might have influenced tapeworm populations at a fine evolutionary scale. Thus, the differences in prevalence between fish host species in different lakes might be influenced not only by the parasite's ecology, but also by its genetic diversity.

Structural complexity is known to influence prey behaviour, mortality and population structure, but the effects on predators have received less attention. We tested whether contrasting structural complexity in two newly colonised lakes (low structural complexity lake-LSC; high structural complexity-HSC) was associated with contrasting behaviour in an aquatic apex predator, Northern pike (Esox lucius; hereafter pike) present in the lakes. Behaviour of pike was studied with whole-lake acoustic telemetry tracking, supplemented by stable isotope analysis of pike prey utilization and survey fishing data on the prey fish community. Pike displayed increased activity, space use, individual growth as well as behavioural differentiation and spent more time in open waters in the LSC lake. Despite observed differences between lakes, stable isotopes analyses indicated a high dependency on littoral food sources in both lakes. We concluded that pike in the HSC lake displayed a behaviour consistent with a prevalent ambush predation behaviour, whereas the higher activity and larger space use in the LSC lake indicated a transition to more active search behaviour. It could lead to increased prey encounter and cause better growth in the LSC lake. Our study demonstrated how differences in structural complexity mediated prominent changes in the foraging behaviour of an apex predator, which in turn may have effects on the prey community.

Temporal and spatial heterogeneity in the distribution of cladocerans in lakes could be caused by abiotic (wind, water currents) and biotic factors (reproduction, food resources, predation). Diel horizontal and vertical distribution of cladoceran assemblages was studied in two deep lakes (Milada and Most Lakes, Czech Republic) in early (June) and late (September) summer. The objective was to study diel vertical migration (DVM) and diel horizontal migration (DHM) of cladocerans under conditions of different macrophyte cover in littoral areas (rich in Milada Lake, poor in Most Lake) and fish assemblages (non-specialised planktivorous species in Milada Lake, and abundant planktivorous fish - maraena whitefish, Coregonus maraena - in open water habitats in Most Lake). Temporal variations in cladoceran assemblages were reported in both lakes in the two sampling periods. DVM was observed in the two lakes, performed by the most vulnerable species to fish predation (the larger Daphnia spp.), but with different patterns (direct and reverse) probably linked with the local fish community and other biotic and abiotic factors in each lake. Horizontal movements were only observed in Most Lake: D. longispina increase its abundance in open waters at night compared to the littoral points; while Ceriodaphnia spp. showed the inverse pattern. In both lakes, higher densities were often found at night in surface layers, producing a great "diurnal deficit": cladocerans remain undetected in some zones during the day (especially in the littoral areas) moving to surface layers at night.

Catfish have spread across Europe and several countries out of this region within the last decades. Basic knowledge of this apex predator has revealed concerns of invasive behaviour and questions regarding its utilization as a biomanipulation species. However, a method enabling its regulation to a required level has not yet been developed. We simulated the impact of angling on the catfish population by method of hook-lines in two post-mining lakes with a monitored population consisting of tagged individuals and in two reservoirs as reference sites. Further, the efficiency of hook-lines as a reducing device was examined and the economic aspects were determined. Catfish population in localities where the species is unwanted or invasive may be efficiently reduced to a harmless level by hook-lines and angling (depending on the approach of anglers). The most efficient time of the year seems to be spring to early summer with catch efficiency of 5.4 individuals per 10 baits in one day. The catch efficiency markedly decreased during the second part of the year and did not exceed 2.8 individuals per 10 baits in one day. Mean size of catfish had negative impact whereas catfish biomass had positive impact on the catch efficiency. Trophic status and number of catfish in the locality had no impact on the catch efficiency. According to model, 11-18 bait-days per 1 ha per season is efficient to decrease catfish population to 10% of the original size. Both angling and hook-lines are very simple, they are financially and time bearable mechanisms of catfish regulation in any condition. However, catfish play an important role as a biomanipulative species in many localities. In this case where catfish is beneficial, angling presents a real threat of population collapse and loss of the biomanipulative effect.

PURPOSE: To compare the accumulation and effect of liposomal doxorubicin in liver tissue treated by radiofrequency ablation (RFA) and irreversible electroporation (IRE) in in vivo porcine models. MATERIALS AND METHODS: Sixteen RFA and 16 IRE procedures were performed in healthy liver of two groups of three pigs. Multi-tined RFA parameters included: 100 W, target temperature 105°C for 7 min. 100 IRE pulses were delivered using two monopolar electrodes at 2250 V, 1 Hz, for 100 µsec. For each group, two pigs received 50 mg liposomal doxorubicin (0.5 mg/kg) as a drip infusion during ablation procedure, with one pig serving as control. Samples were harvested from the central and peripheral zones of the ablation at 24 and 72 h. Immunohistochemical analysis to evaluate the degree of cellular stress, DNA damage, and degree of apoptosis was performed. These and the ablation sizes were compared. Doxorubicin concentrations were also analyzed using fluorescence photometry of homogenized tissue. RESULTS: RFA treatment zones created with concomitant administration of doxorubicin at 24 h were significantly larger than controls (2.5 ± 0.3 cm vs. 2.2 ± 0.2 cm; p = 0.04). By contrast, IRE treatment zones were negatively influenced by chemotherapy (2.2 ± 0.4 cm vs. 2.6 ± 0.4 cm; p = 0.05). At 24 h, doxorubicin concentrations in peripheral and central zones of RFA were significantly increased in comparison with untreated parenchyma (0.431 ± 0.078 µg/g and 0.314 ± 0.055 µg/g vs. 0.18 ± 0.012 µg/g; p < 0.05). Doxorubicin concentrations in IRE zones were not significantly different from untreated liver (0.191 ± 0.049 µg/g and 0.210 ± 0.049 µg/g vs. 0.18 ± 0.012 µg/g). CONCLUSIONS: Whereas there is an increased accumulation of periprocedural doxorubicin and an associated increase in ablation zone following RFA, a contrary effect is noted with IRE. These discrepant findings suggest that different mechanisms and synergies will need to be considered in order to select optimal adjuvants for different classes of ablation devices.

The higher proportion of males of the invasive round goby Neogobius melanostomus in samples from two activity selective passive fishing gears compared with one activity non-selective fishing gear in three Dutch lakes is related to higher male locomotory activity and is a sex-dependent trait. This difference in activity reflects the different ecology of male and female N. melanostomus.

For ethical reasons and animal welfare, it is becoming increasingly more important to carry out ecological surveys with a non-invasive approach. Information about fish distribution and abundance in the upper water column is often fundamental. However, this information is extremely hard to obtain using classical hydroacoustic methods. We developed a rigid frame system for pushing upward looking transducers of the scientific echo sounder (38 and 120 kHz) in front of the research vessel. The efficiency of the new approach for monitoring juvenile fish at night was investigated by comparing the results with a quantitative fry trawl in the Rímov Reservoir in the Czech Republic. The experimental setup enabled comparisons for the 0-3 m and 3-6 m depth layers, which are utilized by almost all juvenile fish in summer. No statistically significant differences in the estimated abundance of juveniles were found between the two sampling methods. The comparison of abundance estimates gathered by the two frequencies were also not significantly different. The predicted mean lengths from acoustic sampling and the trawl catches differed by less than 10 mm in all comparisons. Results suggest that mobile hydroacoustic upward-looking systems can fill the methodological gap in non-invasive surveying of surface fishes.

Echinococcosis (hydatidosis) is a rare severe tissue parasitosis. In the Czech Republic it is caused by two species of tapeworm: Echinococcus granulosus or Echinococcus multilocularis. The species differ in both their usual hosts during their life cycle as well as in the typical form of lesions in infested tissues. In both cases the liver is the most common primary infested organ.We describe an autopsy case of a massive hepatic parasitic lesion in an 81 year-old man. There were metastatic parasitic cysts in both lungs and the hepatic mass spread per continuitatem in the right adrenal gland. The lesion was microscopically represented by small cystic forms with PAS positive laminar membranes and frequent occurrence of protoscoleces in the peripheral parts of the liver and the adrenal gland.The diagnosis of echinococcosis was settled after microscopic exploration of necroptic material. The overall appearance and pattern of spreading corresponded to alveolar echinococcosis, however the massive presence of protoscoleces in the liver is very rare for E. multilocularis. Protoscoleces are usually found in infections caused by E. granulosus. Cystic echinococcosis typically presents with one larger cyst or a small number of bigger cysts with prominent fibrous rim. Because of the unavailability of molecular diagnostic methods (PCR, immunohistochemistry), that are able to distinguish individual species of parasite, the case was closed according to the typical histological findings as disseminated alveolar echinococcosis with the rare appearance of protoscoleces with possible association with immunosuppressive therapy.We have found about ten other cases of alveolar echinococcosis published in last ten years in the Czech Republic. All these cases were diagnosed in living patients. It is assumed that most of these patients were infected in the vicinity of their homes.

Piscivory in cyprinids (Cyprinidae) is extremely rare. Specifically, common bream (Abramis brama) and common carp (Cyprinus carpio) are zooplanktivorous fish in deep lentic waters. Nevertheless, we observed predation by these two cyprinids under natural conditions in the Vír Reservoir, Czech Republic. We conducted diet analysis for cyprinids caught by trawling and gillnets and the large amount of young-of-the-year (YOY) perch (Perca fluviatilis), with sizes of 37-52 mm standard length, were found in their digestive tracts. In 2010, a large amount of YOY perch caused a significant decrease in Daphnia spp. size and abundance in the reservoir. Hence, a food deficit was induced for the cyprinids, apparent also from the poor nutritional condition of common bream which was much worse than the condition of those in similar reservoirs. Common carp and common bream shifted to forced piscivory, and they utilized the YOY perch as an alternative food source. In contrast, smaller species, such as roach (Rutilus rutilus) and bleak (Alburnus alburnus), widely utilized planktonic cyanobacteria. In the following year, YOY perch occurred in significantly lower numbers and conversely, Daphnia spp. size and abundance were significantly higher. The forced piscivory was not observed. Our results indicate a switch to forced piscivory by cyprinids, which was caused by a shortage of their natural food source. Moreover, this phenomenon presents an effective mechanism for reduction in the numbers of YOY perch, ensuring the stability of the ecosystem.

RESUMEN

The European Standard EN 14757 recommends gillnet mesh sizes that range from 5 to 55mm (knot-to-knot) for the standard monitoring of fish assemblages and suggests adding gillnets with larger mesh sizes if necessary. Our research showed that the recommended range of mesh sizes did not provide a representative picture of fish sizes for larger species that commonly occur in continental Europe. We developed a novel, large mesh gillnet which consists of mesh sizes 70, 90, 110 and 135mm (knot to knot, 10m panels) and assessed its added value for monitoring purposes. From selectivity curves obtained by sampling with single mesh size gillnets (11 mesh sizes 6 - 55mm) and large mesh gillnets, we identified the threshold length of bream (Abramis brama) above which this widespread large species was underestimated by European standard gillnet catches. We tested the European Standard gillnet by comparing its size composition with that obtained during concurrent pelagic trawling and purse seining in a cyprinid-dominated reservoir and found that the European Standard underestimated fish larger than 292mm by 26 times. The inclusion of large mesh gillnets in the sampling design removed this underestimation. We analysed the length-age relationship of bream in the Rímov Reservoir, and concluded that catches of bream larger than 292mm and older than five years were seriously underrepresented in European Standard gillnet catches. The Rímov Reservoir is a typical cyprinid-dominated water body where the biomass of bream > 292mm formed 70% of the pelagic trawl and purse seine catch. The species-specific relationships between the large mesh gillnet catch and European Standard catch suggested that the presence of carp (Cyprinus carpio), European catfish (Silurus glanis), tench (Tinca tinca) or bream warrants the use of both gillnet types. We suggest extending the gillnet series in the European Standard to avoid misinterpretation of fish community biomass estimates.
