RESUMEN

Peripheral nerve sheath tumours are rare in pigs. In the present case, a juvenile female hybrid pig showed a solitary, pigmented, cutaneous mass. Histologically, it consisted of clustered melanin-laden, epithelioid cells as well as spindle cells forming bundles and nodules. The latter were surrounded by perineurial-like cells. Single Wagner-Meissner-like corpuscles were present. Immunohistochemically, the epithelioid cells expressed S100 protein, melan A and the p75 neurotrophin receptor (p75NTR). The spindle cells expressed S100, sex determining region Y-box 2, p75NTR, Krox20, growth associated protein 43 and glial fibrillary acidic protein. Perineurial-like cells were positive for p75NTR, α-smooth muscle actin and cytokeratin. Taken together, the histological und immunohistochemical findings support the diagnosis of a cutaneous pigmented neurofibroma.

RESUMEN

Congenital teratomas arising in the coccygeal region have not been reported in domestic animals. This report describes a congenital coccygeal teratoma in a male Burmese cat. A round to oval expansile mass with solid and cystic areas was found ventral to the 5th to 7th caudal vertebrae. Microscopically, the tumour was composed of derivatives of all three primordial germ layers with neuroectodermal predominance. Immunohistochemical double labelling identified immature tissue components in combination with Sox2, a regulator of stem cell proliferation and differentiation. Few Sox2-positive cells co-expressed the neural crest stem cell markers vimentin and neurotrophin receptor p75(NTR). Not all Sox2- and p75(NTR)-positive cells expressed vimentin. An overlapping expression of vimentin-negative and Sox2-, p75(NTR)-positive cells and GFAP- and p75(NTR)-positive cells may indicate a transition stage from immature to mature non-myelinating Schwann cells. Periaxin-positive myelinating Schwann cells surrounding neurofilament-positive axons were observed. Sox2 was additionally expressed in immature odontogenic epithelial cells and in immature cells of endodermal origin. Sox2 was not observed in mature cells, with the exception of satellite glial cells and mucous glands. Despite the presence of immature tissue components, no recurrence was observed 1 year after surgical removal.

RESUMEN

Schwannomas arising in the parotid gland are rare in man and are not documented in domestic animals. This report describes the clinical, morphological and immunohistochemical findings of a benign schwannoma in the parotid gland of a 12-year-old warmblood gelding. A slow-growing mass was surgically excised from the parotid gland and did not recur within the following 6 months. The tumour was well circumscribed and was composed of densely packed spindle cells partly arranged in an Antoni A pattern and intermixed with hypocellular areas resembling the Antoni B pattern. Tumour cells expressed vimentin, S100 protein, glial fibrillary acidic protein and variably neuron-specific enolase. Less than 5% of the neoplastic cells were immunolabelled with the proliferation marker Ki67. Although malignant melanomas frequently involve the parotid gland in horses, schwannomas should be included in the differential diagnosis of parotid masses.

RESUMEN

A 2-year-old female boxer dog was presented with a vaginal serosanguineous discharge not associated with oestrus. There was a friable mass occupying the upper caudal part of the vagina. Cytological and histological examination revealed a monomorphic population of neoplastic round cells consistent with canine transmissible venereal tumour (TVT). In addition, Leishmania spp. amastigotes were found within the neoplastic tissue. In order to characterize whether the amastigotes were present inside macrophages and/or neoplastic cells, a co-localization study using cell- and pathogen-specific markers was performed. To detect Leishmania spp. a 5.8S ribosomal RNA (rRNA) parasite-specific sequence was used for in-situ hybridization and Mac387 was used as a macrophage marker for immunohistochemistry. Leishmania spp. rRNA was detected inside Mac387(+) macrophages and within the cytoplasm of some neoplastic cells. DNA isolation and polymerase chain reaction using specific primers and sequencing analysis identified the organism as Leishmania infantum (syn. Leishmania chagasi). This is the first report describing infection of tumour cells by L. infantum in a genital TVT from an asymptomatic bitch. Transplantation of Leishmania-laden neoplastic cells could represent an alternative route of venereal transmission of leishmaniasis among dogs.

RESUMEN

Optical tweezers are experimental tools with extraordinary resolution in positioning (± 1 nm) a micron-sized colloid and in the measurement of forces (± 50 fN) acting on it-without any mechanical contact. This enables one to carry out a multitude of novel experiments in nano- and microfluidics, of which the following will be presented in this review: (i) forces within single pairs of colloids in media of varying concentration and valency of the surrounding ionic solution, (ii) measurements of the electrophoretic mobility of single colloids in different solvents (concentration, valency of the ionic solution and pH), (iii) similar experiments as in (i) with DNA-grafted colloids, (iv) the nonlinear response of single DNA-grafted colloids in shear flow and (v) the drag force on single colloids pulled through a polymer solution. The experiments will be described in detail and their analysis discussed.

RESUMEN

Forces between single pairs of negatively charged micrometer-sized colloids in aqueous solutions of monovalent, divalent, or trivalent counter-ions at varying concentrations have been measured by employing optical tweezers. The experimental data have been analyzed by using the Derjaguin-Landau-Verwey-Overbeek (DLVO) theory and a numerical solution of the Poisson-Boltzmann (PB) equation. With monovalent counterions, the data are well described by the DLVO and PB theories, suggesting that the DLVO theory is adequate to describe the colloidal forces at these conditions. At higher counter-ion valence, the approximations within the two theories become evident.

RESUMEN

Optical tweezers are employed to measure the forces of interaction between single DNA-grafted colloids. Parameters to be varied are the length of the DNA, the grafting density, and the ion concentration of the surrounding medium. From the measured force-separation dependence an interaction length at a given force is deduced. It shows in the mushroom regime a scaling with the grafting density which levels off for brushes. For the latter the transition from an osmotic to a salted brush can be traced in detail by varying the ion concentration in accordance with mean field theories.

RESUMEN

Optical tweezers are employed to study the action of the histone-like protein from Thermotoga maritima (TmHU) on DNA at a single molecule level. Binding and disruption of TmHU to and from DNA are found to take place in discrete steps of 4-5 nm length and a net binding enthalpy of about 16kBT. This is in reasonable agreement with a microscopic model that estimates the extension of the binding sites of the protein and evaluates the energetics mainly for bending of the DNA in the course of interaction.

RESUMEN

Optical tweezers are microscopic tools with extraordinary precision in the determination of the position (±2 nm) of a colloid (diameter: ∼2.0 µm) in 3D-space and in the measurement of small forces in the range between 0.1 and 100 pN (pN=10-12 N). Experiments are reported in which single double-stranded (ds)-DNA chains of different length [2,000 base pairs (bp), 3,000, 4,000, and 6,000 bp] are spanned between two colloidal particles by use of appropriate molecular linkers. For the forces applied (≤40 pN) a fully reversible and well reproducible force-extension dependence is found. The data can be well described by both the worm-like chain model or by an approach developed by R. G. Winkler. For the resulting persistence length, a pronounced dependence on the ionic concentration in the surrounding medium is found.
