RESUMEN

Antibody-drug conjugates (ADCs), integrating high specificity of antigen-targeting antibodies and high potency of cell-killing chemical drugs, have become one of the most rapidly expanding therapeutic biologics in oncology. Although ADCs were widely studied from multiple aspects, overall structural elucidation with comprehensive understanding of variants is scarcely reported. Here, for the first time, we present a holistic and in-depth characterization of an interchain cysteine-conjugated ADC, focusing on conjugation and charge heterogeneity, and in vitro biological activities. Conjugation mapping utilized a bottom-up approach, unraveled positional isomer composition, provided insights into the conjugation process, and elucidated how conjugation affects the physicochemical and biological properties of an ADC. Charge profiling combined bottom-up and top-down approaches to interrogate the origin of charge heterogeneity, its impact on function, and best practice for characterization. Specifically, we pioneered the utilization of capillary isoelectric focusing-mass spectrometry to decode not only critical post-translational modifications but also drug load and positional isomer distribution. The study design provides general guidance for in-depth characterization of ADCs, and the analytical findings in turn benefit the discovery and development of future ADCs.

Even the lowest concentration level of lead (Pb) in the human body is dangerous to health due to its bioaccumulation and high toxicity. Therefore, it is very important to develop selective and fast adsorption methods for the removal of Pb(ii) from various samples. In this paper, a new Pb(ii) ion-imprinted polymer (Pb(ii)-IIP) was prepared with surface imprinting technology by using lead nitrate as a template, for the solid-phase extraction of trace Pb(ii) ions in environmental water samples. The imprinted polymer was characterized by X-ray diffraction, Fourier transform infrared spectroscopy, Raman spectroscopy, scanning electron microscopy and N2 adsorption-desorption isotherms. The separation/pre-concentration conditions for Pb(ii) were investigated, including the effects of pH, shaking time, sample flow rate, elution conditions and interfering ions. Compared with non-imprinted particles, the ion-imprinted polymer had a higher selectivity and adsorption capacity for Pb(ii). The pseudo-second-order kinetics model and Langmuir isotherm model fitted well with the adsorption data. The relative selectivity factor values (α r) of Pb(ii)/Zn(ii), Pb(ii)/Ni(ii), Pb(ii)/Co(ii) and Pb(ii)/Cu(ii) were 168.20, 192.71, 126.13 and 229.39, respectively, which were all much greater than 1. The prepared Pb(ii)-imprinted polymer was shown to be promising for the separation/pre-concentration of trace Pb(ii) from natural water samples. The adsorption and desorption mechanisms were also proposed.

Lectin A (LecA) from Pseudomonas aeruginosa is an established virulence factor. Glycoclusters that target LecA and are able to compete with human glycoconjugates present on epithelial cells are promising candidates to treat P. aeruginosa infection. A family of 32 glycodendrimers of generation 0 and 1 based on a bifurcated bis-galactoside motif have been designed to interact with LecA. The influences both of the central multivalent core and of the aglycon of these glycodendrimers on their affinity toward LecA have been evaluated by use of a microarray technique, both qualitatively for rapid screening of the binding properties and also quantitatively (Kd ). This has led to high-affinity LecA ligands with Kd values in the low nanomolar range (Kd =22 nm for the best one).

RESUMEN

In this paper, we investigated the wettability and adhesive behavior of the natural honeycomb wall for water and honey droplets. The cell walls have hydrophobic and highly adhesive properties for both water and honey in air. This highly adhesive cell wall was used as a "mechanical hand" to transfer micro-droplets. These findings will help us to comprehensively understand the surface properties of honeycomb walls, and will provide a novel strategy for achieving functional biomimetics based on honeycombs.

RESUMEN

The patterned honeycomb structure film with the aggregation-induced emission property was prepared successfully by the breath figure method and photopolymerization method. Characterization of the HeLa and HepG2 cell culture on this surface indicates the porous honeycomb structures show anticancer cells growth function. So this kind of honeycomb structure will be promising for the control of cancer cell growth behaviors and achieving the application of anticancer.

RESUMEN

Creatures in nature possess almost perfect structures and properties, and exhibit harmonization and unification between structure and function. Biomimetics, mimicking nature for engineering solutions, provides a model for the development of functional surfaces with special properties. Recently, honeycomb structure materials have attracted wide attention for both fundamental research and practical applications and have become an increasingly hot research topic. Though progress in the field of breath-figure formation has been reviewed, the advance in the fabrication materials of bio-inspired honeycomb structure films has not been discussed. Here we review the recent progress of honeycomb structure fabrication materials which were prepared by the breath-figure method. The application of breath figures for the generation of all kinds of honeycomb is discussed.

Since 1996, 16 states and the District of Columbia in the United States have enacted legislation to decriminalize marijuana for medical use. Although marijuana is the most commonly detected nonalcohol drug in drivers, its role in crash causation remains unsettled. To assess the association between marijuana use and crash risk, the authors performed a meta-analysis of 9 epidemiologic studies published in English in the past 2 decades identified through a systematic search of bibliographic databases. Estimated odds ratios relating marijuana use to crash risk reported in these studies ranged from 0.85 to 7.16. Pooled analysis based on the random-effects model yielded a summary odds ratio of 2.66 (95% confidence interval: 2.07, 3.41). Analysis of individual studies indicated that the heightened risk of crash involvement associated with marijuana use persisted after adjustment for confounding variables and that the risk of crash involvement increased in a dose-response fashion with the concentration of 11-nor-9-carboxy-delta-9-tetrahydrocannabinol detected in the urine and the frequency of self-reported marijuana use. The results of this meta-analysis suggest that marijuana use by drivers is associated with a significantly increased risk of being involved in motor vehicle crashes.
