RESUMEN

Maternal dietary factors have been reported to influence Clostridioides difficile colonisation in the offspring. Twenty suckling piglets from sows fed diets supplemented with high-fermentable sugar beet pulp (SBP) or low-fermentable lignocellulose (LNC) fibres during gestation and lactation were dissected in the first week after birth. Postmortem analysis included clinical mesocolon and faecal scoring, concentration of C. difficile and respective toxins in colon digesta and faeces, immunoglobulins in serum and inflammatory markers in serum and colon tissues. Sow colostrum was assessed for nutrients, immunoglobulins and biogenic amines. Toxin-neutralising IgG antibodies were measured in colostrum and serum of the sows, and in colon digesta and serum of the piglets. Mesocolonic oedema of different severity was present in most of the piglets from both sows' feeding groups. Concentrations of C. difficile, toxins and calprotectin in colon digesta and faecal contents did not differ between the study piglets. Calprotectin correlated positively with mesocolon score (rho = 413, P = 0.07). Piglets from sows fed LNC vs SBP tended to have higher IgA (P = 0.089), IgG (P = 0.053), total Ig (P = 0.053), albumin (P = 0.075) and total protein content (P = 0.007) in serum. Colon tissues of piglets from the SFB vs LNC had upregulated expression of ZO-1 (P = 0.021), PCNA (P = 0.015) and TGF-ß (P = 0.014). Titers of anti-toxin-IgG-antibodies in serum and colostrum and in piglet colon digesta and serum did not differ between sows from both dietary groups, but they all showed strong positive correlations. In conclusion, dietary sugar beet pulp or lignocellulose fed to sows did not influence the concentrations of C. difficile and toxins titers in colon digesta and faeces of neonatal piglets.

The supplementation of feed with phytases enables broilers to utilize more efficiently phosphorus (P) from phytic acid (IP6), the main storage form of P in plants. The current study evaluated the addition of 500, 1000, and 3000 FTU of phytase per kg to a phytate-containing diet with low P level (LP) fed to broilers from 1 to 21 days of age and compared it to a hypoallergenic phytate-free diet (HPF). There was a linear improvement in performance parameters with increasing levels of phytase in the LP diet (p < 0.001). Apparent ileal digestibility of crude protein, P, and some amino acids, increased with phytase. Crude ash, P, and the calcium content of tibia bones linearly increased with increasing levels of phytase (p < 0.001). Crypt depth (related to body weight) in the jejunum epithelium linearly decreased with phytase addition (p < 0.001). Cecal crypt depth decreased with phytase supplementation (p = 0.002). Cecum tissue showed lower counts of CD3-positive intraepithelial lymphocytes in broilers receiving the phytase in comparison to LP (p < 0.001), achieving similar counts to HPF-fed broilers. Although results from the current study seem to point out some mechanisms related to the immune response and mucosal morphology contributing to those overall beneficial effects, no clear differences between different phytase doses could be demonstrated in these specific parameters.

Dietary fiber has a potential to modulate the gut microbiota in sows. We hypothesized that a maternal diet rich in either high- or low-fermentable fiber during gestation and lactation influences Clostridioides difficile gut colonization in suckling piglets. Twenty sows were fed gestation and lactation diets enriched with either high-fermentable sugar beet pulp (SBP) or low-fermentable lignocellulose (LNC) fibers. C. difficile, toxin B (TcdB), fecal score, microbial abundance (16S-rDNA sequencing) and metabolites were measured in the feces from the sows and their piglets. C. difficile concentration was higher in piglets from the sows fed LNC than SBP along the study (P ≤ 0.05). Higher prevalence of C. difficile was noted in three-week-old piglets from sows fed LNC vs. SBP (45% vs. 0%, P = 0.001). TcdB prevalence was higher in six-day-old piglets from the sows fed LNC vs. SBP (60% vs. 17%, P = 0.009). In sows, fecal microbial metabolites were higher in SBP than LNC, while C. difficile concentration showed no difference. Higher microbial diversity Shannon index was noted in sows from SBP vs. LNC one week before parturition and at the parturition (P ≤ 0.05). Piglets from SBP vs. LNC tended to have higher microbial diversity Shannon index at two and three weeks of age. Diets enriched with high-fermentable fiber compared to low-fermentable fiber in sows reduced C. difficile colonization in their piglets. Susceptibility to colonization by C. difficile in neonatal piglets can be modulated by the sows' diet, supporting the hypothesis of the early microbial programming in the offspring and the importance of the sow-piglet couple.

BACKGROUND: Iron (Fe) is traditionally supplemented in poultry and swine diets using inorganic forms (e.g. sulfates, oxides). However, research suggests that organic sources are more beneficial due to greater bioavailability. In this paper, we present results from four studies aimed at assessing ferric citrate (CI-FER™, Akeso Biomedical Inc., Burlington, MA, USA) as a safe and effective source of Fe for broilers and piglets. METHODS: A total of four studies were performed in Germany following standard farming practices for each species. One study in day-old broiler chicks and one study in weaned piglets were designed as target animal safety studies where animals were randomly allocated to one of three treatment groups: a negative control group, the proposed dose group and a multifold dose group. Broilers and pigs were fed the experimental diets for 35 and 42 days, respectively. In each study, average daily feed intake, average daily weight gain and feed conversion ratio were measured, and blood samples were taken at study end for routine biochemistry and haematology. The other two studies were designed to evaluate different sources of dietary Fe for weaned piglets bred and managed under standard farm conditions. All piglets received routine Fe injections (200 mg Fe dextran, intramuscular) on day 3 of age, as well as the experimental diets for 42 days. In both studies, performance parameters were measured. In one study, Fe digestibility and serum Fe, superoxide dismutase and haptoglobin were also measured. In all studies, the general health status of the animals was monitored daily and all culls and mortality recorded. Each study followed a complete randomised block design. RESULTS: In broilers, ferric citrate was well tolerated up to 2,000 mg/kg feed (×10 the recommended inclusion rate) and no adverse effects on growth, blood parameters or mortality were observed. In piglets, ferric citrate was well tolerated up to 5,000 mg/kg feed (×10 the recommended inclusion rate) with no adverse effects on growth, blood parameters or mortality. In addition, piglets fed ferric citrate performed significantly better than animals fed the negative control diet (containing only endogenous Fe) and those fed inorganic forms of Fe. Moreover, piglets fed ferric citrate demonstrated improved Fe digestibility and improved oxidative status. Altogether, these findings show that ferric citrate is a safe and easily digestible source of dietary Fe for broilers and piglets.

This study was carried out to evaluate the effect of essential oils or saponins alone or in combination on productive performance, intestinal morphology, and digestive enzymes' activity in broilers. Four hundred one-day-old male broiler chickens were randomly assigned to four treatments, each with 5 replicates. The birds were fed experimental diets contained either no phytogenic feed additives (control) or were supplemented with 25.0 mg/kg of an essential oil blend from star anise, rosemary, thyme and oregano (essential oils); 46.0 mg/kg of a Quillaja saponin blend (saponins); or a combination of both phytogenic preparations (essential oils plus saponins). The experiment lasted for 42 days. Body weight, feed intake and weight gain were recorded weekly, and the feed conversion ratio was calculated, throughout the experiment. Intestinal morphology and digestive enzymes' activity were determined at the end of the experiment. It was found that the body weight was increased by all dietary supplements at the end of the experiment. There was no change in performance parameters among the treatments during the starter period. At the grower period and the overall experiment, the weight gain of birds was higher in all supplemented groups compared to the control, but the feed conversion was better in the essential oils with saponins treatment. Supplementation of essential oils or saponins alone or in combination increased the villus height and the villus/crypt ratio of the intestine. There was an increase in trypsin, chymotrypsin and elastase activities in the essential oils plus saponins group only. The obtained results indicate that the phytogenic feed additives, especially essential oils with saponins, can be considered as new promising agents in promoting the growth performance of broilers. The effect could be related to its improving impact in intestinal development and increasing the protease enzymes' activity, resulting in increased protein digestibility and absorption of dietary nutrients.

The current study investigated the impacts of supplementation of post-weaning piglet diets with Macleaya cordata extract (MCE) and naringin (NAR) on performance, nutrient digestibility and intestinal histomorphology. Post-weaning crossbred piglets (28 males and 28 females; age at weaning 25 d) were randomly allotted to 28 pens. The experiment consisted of a control and three treatment groups (MCE60, MCE120 and NAR). For diets MCE60 and MCE120, the control diet was supplemented with 60 and 120 mg Sangrovit® Extra (a standardised premixture of MCE) per kg diet, respectively. Group NAR received 50 mg pure NAR per kg diet. The experiment lasted 42 d (d 25 - 66 of age). At d 66, apparent pre-caecal digestibility (APD) of nutrients was determined and histomorphological changes in mid-jejunum were evaluated. Feeding diets MCE120 and NAR improved body weight gain and feed conversion ratio of piglets. After feeding diets MCE120 and NAR, the APD of phosphorus and different single and total amino acids were greater than after feeding the control diet. The present data demonstrated that supplementation of post-weaning piglet diets with 120 mg MCE or 50 mg NAR per kg diet could improve growth performance and nutrient digestibility and had no impact on histomorphological variables in the jejunum. These findings indicate the potential of these products to be used as growth promoters in pig nutrition.

Phytogenic feed additives represent a potential alternative to antibiotics with attributed health and growth-promoting effects. Chickens supplemented with an essential oil blend, a Quillaja saponin blend, or a combination of both phytogenic preparations showed a comprehensively and significantly improved apparent ileal digestibility of crude protein and amino acids compared to control birds. Accordingly, holistic transcriptomic analyses of jejunum and liver samples indicated alterations of macromolecule transporters and processing pathways likely culminating in an increased uptake and metabolizing of carbohydrates and fatty acids. Complementary analyses in Caco-2 showed a significant increase in transporter recruitment to the membrane (SGLT1 and PEPT1) after addition of essential oils and saponins. Although the penetrance of effects differed for the used phytogenic feed additives, the results indicate for an overlapping mode of action including local effects at the intestinal border and systemic alterations of macronutrient metabolism resulting in an improved performance of broilers.

Lutein is an essential dietary carotenoid with health benefits and is inter alia responsible for the colouration of egg yolk. The relationship between lutein accumulation and egg yolk colouration was therefore studied in more detail. After feeding a low-luteine diet for 21 days, 14 birds (Lohmann brown hens aged 20 weeks) were fed a diet containing marigold (80 mg lutein/kg feed) and 14 other birds were fed a diet containing oleoresin (45 mg lutein/kg feed) for 21 days; for both groups of birds, this feeding period was followed by withdrawal for 21 days. The Roche Yolk Colour Fan (RYCF) score (0 to 15, where higher values denote greater colour intensity; R2=0.87; P<0.01) and redness (R2=0.89; P<0.01) increased with increasing lutein content of egg yolk. Total carotenoid content had a poor relationship with lightness (R2=0.13; P>0.05) and yellowness (R2=0.12; P>0.05) of the yolk. It may be concluded that increased lutein is potentially responsible for an increased RYCF score and redness (a*), but decreased yellowness (b*) and lightness (L*), of egg yolk.

Fenugreek seed has been shown to affect the intestinal microbiota and immunological responses in animals. A feeding trial with male castrated piglets was performed over 28 d without or with the addition of 1·5 g fenugreek seeds/kg complete diet in ten and eleven piglets, weaned at 21 d. In the intestinal tract, pH, lactate and SCFA were measured as major bacterial metabolites. Immune cell phenotypes, phagocytic activity and lymphocyte proliferation after stimulation with pokeweed mitogen, concanavalin A and phytohaemagglutinin M were measured by flow cytometry. Health status and performance of the piglets were not affected by fenugreek. The pH in the caecum and colon were reduced compared with the control (P< 0·05). Higher concentrations of l-lactic acid were recorded in the small-intestinal digesta (average concentrations from the duodenum, jejunum and ileum; P< 0·05), while the concentrations of SCFA remained unchanged except an increase in n-butyric acid in colon contents (P< 0·05). The piglets fed the fenugreek diet had higher Lactobacillus and clostridium cluster I concentrations and lower Escherichia, Hafnia and Shigella concentrations in the small intestine. The addition of fenugreek increased the relative concentration of the γδ T-cell population (TCR1+CD8α-) in the blood with a simultaneous reduction of antigen-presenting cells (MHCII+CD5-) (P< 0·05). Proliferation rate and phagocytosis activity of monocytes were not affected by the additive. In conclusion, fenugreek seeds might be interesting as a feed ingredient for young piglets due to their effects on the intestinal microbiota and immunological variables. The impact on performance and animal health has to be further evaluated.

The influence of low dietary levels of free and encapsulated medium-chain fatty acids on their concentrations in the digesta, the gastric microbial ecology and bacterial metabolic products in the gastrointestinal tract (GIT) in weaned piglets was studied. Starting after weaning, 36 piglets were fed a diet without (Control) or with medium-chain fatty acids uncoated (MCFA) or coated with vegetable fat and lecithin (MCFAc). After 4 weeks, the animals were killed, and digesta from the stomach and different sections of the GIT were collected. The concentrations of caprylic (p < 0.001) and capric (p = 0.001) acids were higher in the stomachs of piglets fed diets MCFA and MCFAc compared to the Control group. The concentrations dropped rapidly along the GIT, regardless of encapsulation, and tended to be higher in groups MCFA and MCFAc compared to the Control. Compared to the Control group, ingestion of diet MCFAc led to an increase in the number of eubacteria (p = 0.001), enterobacteriaceae (p < 0.001), clostridial clusters I (p = 0.001) and IV (p = 0.019), Lactobacillus johnsonii (p < 0.001) and Lactobacillus amylovorus (p = 0.001) in gastric contents. A similar trend was seen with diet MCFA. Relative concentrations of short-chain fatty acids were characterised by lower propionic acid levels (p = 0.045), numerically (p < 0.1) higher acetic, lower n-butyric and i-valeric acid concentrations in the small intestine. Lactic acid concentrations were not significantly changed in the GIT, but ammonia concentrations increased (p < 0.001) in the distal small intestine in the MCFA and MCFAc groups. In conclusion, medium-chain fatty acids affected microbial ecology parameters in the gastric contents and bacterial metabolites in the small intestine. At low dietary levels, medium-chain fatty acids may be regarded as modulators of the gastric microbiota in weaned piglets.

We evaluated the impact of the dietary cation-anion difference (DCAD) on the influence of anionic salts (AS) on the metabolism of dairy cows using a study-design that included control of feed intake. Ten mature, non-lactating, non-pregnant, Holstein-Friesian-crossbreed cows received 2000 mEq of either one of the seven anionic salts tested, two combinations of the anionic salts or water as control via a rumen cannula. Salts and controls were assigned in a 10x10 Latin square design. Whole blood, serum and urine samples were taken during treatment (TP) and washout period. Samples of whole blood were tested for pH, base-excess and bicarbonate concentrations. In urine, pH and net acid-base excretion (NABE) were analysed. Calcium was measured in serum and urine. According to the different batches of hay, five groups of DCAD were created regarding cluster analysis. Changes in urine and blood parameters were statistically analysed for each DCAD group separately. The different DCAD had an impact on the amount of change in acid-base balance (ABB) and calcium metabolism and for how long these changes lasted. In the DCAD group receiving the highest amount of AS (239 mEq/kg dry matter with AS), changes of ABB were only noticeable in urine and these changes only differed from day zero in the first week of TP (P<0.05). In the other four groups changes of ABB were also visible in blood parameters, but only on a few days of TP did the deviations differ significantly (P<0.05) from day zero. Changes of ABB parameters in urine samples were more pronounced than those in blood and differed clearly from day zero (P<0.05). Parallel to the changes of ABB, calcium concentrations in these samples were significantly increased (P<0.001) in all DCAD groups. Except for the highest DCAD group, ionized calcium concentrations changed over time (P<0.020). However, the differences were very small and only differed from day zero on a few TP days. We conclude that the DCAD of a dairy cow's diet has an important impact on the effect of AS on ABB and calcium metabolism with respect to the duration and amount of change. The target regions of DCAD should be clearly below 100 mEq/kg dry matter to ensure the desired effect on ABB and calcium metabolism. Extremely negative DCAD should be avoided to minimize the risk of clinical acidosis induced by AS.
