OBJECTIVE: Canine babesiosis, an infectious disease transmitted by Dermacentor reticulatus, is exhibiting growing importance in Germany. The aim of this study was to display the increased incidence of canine babesiosis in the Rhine-Main area in Hesse, with special focus on the accumulation in the district of Groß-Gerau. MATERIAL AND METHODS: The retrospective study included dogs presented to the veterinary hospital between October 2018 and December 2020 and diagnosed with canine babesiosis on the basis of a positive Babesia spp.-PCR. RESULTS: A total of 697 dogs were tested by Babesia spp.-PCR during this time period. Of these, 81 (12 %) were positive.Sequencing was performed in 14 of the 81 dogs (17 %) (B. canis n = 13, B, vulpes n = 1). A simultaneous anaplasmosis infection was detected in 2 dogs. Strikingly, babesiosis cases occurred throughout the year with accumulations in March/April as well as in October.Evaluation of a complete blood cell count revealed pancytopenia in 44 of the 81 animals (54 %). Anemia was present in 66 (82 %), thrombocytopenia in 76 of the 81 patients (94 %). Only 2 of the 81 positive cases showed no hematological changes. Hyperbilirubinemia was found in 66 of 73 measured bilirubin levels (90 %).All animals were treated with two injections of imidocarb-diproprionate (Carbesia®) in 14-day intervals. Follow-up PCR was performed in 37 of the 81 patients (46 %). In the majority of cases (92 %), successful therapy was confirmed by a negative Babesia-PCR. A total of 6 of the 81 patients (7 %) were euthanized during the treatment period. The reasons for euthanasia were progressive renal disease, high-grade intravascular hemolysis necessitating multiple blood transfusions, and development of splenic and renal abscesses. CONCLUSION: In dogs with clinical signs such as apathy, pyrexia and hemoglobinuria, as well as hematologic abnormalities comprising anemia, thrombocytopenia as well as pancytopenia, babesiosis needs to be included in the list of differential diagnoses. Testing should be initiated accordingly regardless of the season, however especially in spring and autumn.

Although atrial dilation is a common finding in feline cardiac disease, detailed investigations of atrial pathology are rare in cats. The aim of the study was to investigate the correlation between pathohistological findings, morphometric data and the degree of dilation of the left atrial appendage (LAA) in 53 cats. Based on the LAA volume, the samples were grouped into normal/control (group 0, ≤1 ml [n = 9]), mildly dilated (group 1, >1 to ≤2 ml [n = 16]), moderately dilated (group 2, >2 to ≤3 ml [n = 14]) and markedly dilated (group 3, >3 ml [n = 14]) groups, independent of the underlying disease. Samples from the LAA and the left atrium (LA) were histologically evaluated using haematoxylin and eosin- and Picrosirius red-stained sections, and morphometrically analysed using an image analysis system. The degree of endo-, myo- and epicardial fibrosis was directly correlated with increased LAA dilation. Due to cardiomyocyte hyperplasia and hypertrophy, the mean thickness of the atrial wall was significantly greater in groups 1 and 2 than in group 0. Conversely, group 3 had a lower mean atrial wall thickness than groups 1 and 2, which was attributed to increased transmural fibrosis and cardiomyocyte atrophy. These findings reflect intensive dynamic remodelling processes during LA and LAA dilation, indicating that reversibility appears to be limited in cases of severe left atrial dilation.

Norwegian Forest cats (NFCs) are often listed as a breed predisposed to cardiomyopathy, but the characteristics of cardiomyopathy in this breed have not been described. The aim of this preliminary study was to report the features of NFC cardiomyopathy based on prospective echocardiographic screening of affected family groups; necropsy findings; and open-source breed screening databases. Prospective examination of 53 NFCs revealed no murmur or left ventricular (LV) outflow tract obstruction in any screened cat, though mild LV hypertrophy (defined as diastolic LV wall thickness ≥5.5mm) was present in 13/53 cats (25%). Gross pathology results and histopathological sections were analysed in eight NFCs, six of which had died of a cardiac cause. Myocyte hypertrophy, myofibre disarray and interstitial fibrosis typical of hypertrophic cardiomyopathy were present in 7/8 cats, but endomyocardial fibrosis suggestive of restrictive cardiomyopathy was also present in the same cats. Pedigree data analysis from 871 NFCs was supportive of a familial cardiomyopathy in this breed.

A 7 yr old mixed-breed dog was presented with a choroidal melanoma of the left eye that had penetrated the sclera, producing an orbital mass. Bradycardia was detected on auscultation. The bradycardia resolved after exenteration of the orbit and was therefore presumed to be associated with the oculocardiac reflex.

The histomorphological findings and immunohistochemical expression of matrix metalloproteinases (MMPs-1, 2, 9 and 14) and their tissue inhibitors (TIMPs-2, 3 and 4) are reported in the parietal (pTV) and septal leaflets (sTV) of the tricuspid valves in normal dogs and dogs with chronic valvular disease (CVD). The layers of the normal sTV were not as well defined as in the pTV and the spongiosa of the sTV contained abundant mucopolysaccharides (MPS) and adipocytes. In CVD, there was expansion of the spongiosa of the pTV due to deposition of MPS, leading to formation of nodules along the free edge. In CVD, there was fibrosis of the atrialis of the sTV and formation of nodular deposits of MPS in the spongiosa and ventricularis, mainly affecting the proximal and middle parts of the leaflet. In dogs with normal pTV and sTV, MMPs-1 and 14 and TIMPs-2, 3 and 4 were expressed, while MMPs-2 and 9 were absent. In mild CVD, expression of MMPs-2, 9 and 14 were increased in the pTV, whereas small foci within the spongiosa contained MMP-9 and TIMP-3 positive cells. In advanced CVD, MMP-14 also was increased in the pTV. In mild CVD, there was increased expression of MMPs-1 and 2 and TIMP-2, but decreased expression of TIMP-4, in the sTV. Small foci with expression of MMP-14 and TIMPs-2, 3 and 4 were also present in the sTV in mild CVD. In advanced CVD, there was increased expression of MMPs-2 and 9 and TIMP-2 in the sTV. In CVD there is upregulation of various MMPs in the pTV, whereas there is a complex alteration in expression of MMPs and TIMPs in the sTV.

This study aimed to characterize the composition and distribution of the extracellular matrix (ECM) components in normal canine mitral valves (MV) and in chronic heart valve disease (CVD). MV of 50 dogs (normal (n=9), mild (n=13), moderate (n=17), severe (n=11) CVD) were investigated macroscopically, histologically (H.-E., picrosirius red) and immunohistochemically (collagen I, III, IV, V, VI, elastin, laminin, fibronectin, heparan sulphate). In normal MV, ECM components were expressed in a typical layered pattern. In mild CVD, basement membrane components (laminin, collagen IV, fibronectin) were increased. Advanced CVD was characterized by myxomatous nodular lesions displaying a marginal and a central region comprised mainly of collagen I, VI and fibronectin in the former and collagen I and III in the latter. Collagen IV and laminin appeared multifocally in marked CVD. In conclusion, not only an accumulation of proteoglycans, but also a distinctly altered expression of basement membrane components, and collagens characterizes CVD.

While the pathogenesis of chronic valvular disease (CVD) in dogs remains unclear, alterations in the activity of specific metalloproteinase enzymes and their inhibitors within the valve stroma are suspected of having a role. This study describes the immunohistochemical distribution pattern of matrix metalloproteinase (MMP) types 2, 9 and 14 and their tissue inhibitors, termed tissue inhibitors of metalloproteinase (TIMP), types 2 and 3, in normal canine mitral valves (MVs) (n=10) and in dogs with mild (n=7), moderate (n=14) and severe (n=9) CVD. In normal MVs, MMP-2 and -14, and TIMP-2 were expressed in isolated stromal cells. Tissue inhibitor of metalloproteinase-3 exhibited moderate intracellular and mild extracellular expression. With increasing severity of CVD, the expression of MMP-2 decreased. The number of stromal cells expressing MMP-14 increased, predominantly in the margins of the nodular lesions. Tissue inhibitor of metalloproteinase-2 and -3 expression increased both intra- and extracellularly. Matrix metalloproteinase-9 was not detected in normal or diseased valves. In conclusion, CVD was characterised by alterations in the distribution and intensity of valvular MMP and TIMP expression, suggesting that depressed catabolism and the accumulation of extracellular matrix components within affected valves contributes to their structural alteration and consequent loss of function.
