RESUMEN

The mechanisms of action of pyrimidine nucleoside derivatives on model lipid membranes of various compositions were studied. A systematic analysis of the tested agents' effects on the membrane physicochemical properties was performed. Differential scanning microcalorimetry data indicated that the ability of nucleoside derivatives to disorder membrane lipids depended on the types of nucleoside bases and membrane-forming lipids. The 5'-norcarbocyclic uracil derivatives were found to be ineffective, while N4-alkylcytidines demonstrated the most pronounced effects, significantly decreasing the dipalmitoylphosphocholine melting temperature and cooperativity of phase transition. The elongation of hydrophobic acyl radicals potentiated the disordering action of N4-alkylcytidines, while an increase in hydrophilicity due to replacing deoxyribose with ribose inhibited this effect. The ability of compounds to form transmembrane pores was also tested. It was found that 5-alkyluridines produced single, ion-permeable pores in phosphatidylglycerol membranes, and that methoxy-mycolic acid and trehalose monooleate potentiated the pore-forming activity of alkyloxymethyldeoxyuridines. The results obtained open up perspectives for the development of innovative highly selective anti-tuberculosis agents, which may be characterized by a low risk of developing drug resistance due to the direct action on the membranes of the pathogen.

Affinity chromatography is a widely used technique for antibody isolation. This article presents the successful synthesis of a novel affinity resin with a mutant form of protein A (BsrtA) immobilized on it as a ligand. The key aspect of the described process is the biocatalytic immobilization of the ligand onto the matrix using the sortase A enzyme. Moreover, we used a matrix with primary amino groups without modification, which greatly simplifies the synthesis process. The resulting resin shows a high dynamic binding capacity (up to 50 mg IgG per 1 mL of sorbent). It also demonstrates high tolerance to 0.1 M NaOH treatment and maintains its effectiveness even after 100 binding, elution, and sanitization cycles.

Monitoring of antimicrobials residues in food of animal origin is performed by control laboratories to ensure public health, and knowledge of the stability of antimicrobials during storage is essential for the reliability of results. For stability studies, analysis of incurred samples is preferential to fortified samples due to the possible conversion of antimicrobial metabolites back to parent compounds during sample preparation, storage, and analysis of the incurred samples, resulting in an increased concentration of the analyte. We have analyzed the concentrations of 13 antimicrobials from 8 groups (tetracyclines, fluoroquinolones, phenicols, sulfonamides, aminoglycosides, penicillins, macrolides, and nitroimidazoles) at different time points of freeze-storage (1 week; 1, 2, and 3 months) using HPLC-MS/MS. Incurred samples were prepared from muscle tissue, liver, kidneys, eggs, and milk taken from different animals (cows, pigs, poultry, goats, and fish). Incurred and fortified samples of honey were investigated as well. The results have shown that all analytes in all samples were stable during the investigated periods regardless of animal species, matrix, and concentration levels.

The rapid increase in the antibiotic resistance of microorganisms, capable of causing diseases in humans as destroying cultural heritage sites, is a great challenge for modern science. In this regard, it is necessary to develop fundamentally novel and highly active compounds. In this study, a series of N4-alkylcytidines, including 5- and 6-methylcytidine derivatives, with extended alkyl substituents, were obtained in order to develop a new generation of antibacterial and antifungal biocides based on nucleoside derivatives. It has been shown that N4-alkyl 5- or 6-methylcytidines effectively inhibit the growth of molds, isolated from the paintings in the halls of the Ancient Russian Paintings of the State Tretyakov Gallery, Russia, Moscow. The novel compounds showed activity similar to antiseptics commonly used to protect works of art, such as benzalkonium chloride, to which a number of microorganisms have acquired resistance. It was also shown that the activity of N4-alkylcytidines is comparable to that of some antibiotics used in medicine to fight Gram-positive bacteria, including resistant strains of Staphylococcus aureus and Mycobacterium smegmatis. N4-dodecyl-5- and 6-methylcytidines turned out to be the best. This compound seems promising for expanding the palette of antiseptics used in painting, since quite often the destruction of painting materials is caused by joint fungi and bacteria infection.

The emergence of drug-resistant strains of pathogenic microorganisms necessitates the creation of new drugs. A series of uridine derivatives containing an extended substituent at the C-5 position as well as C-5 alkyloxymethyl, alkylthiomethyl, alkyltriazolylmethyl, alkylsulfinylmethyl and alkylsulfonylmethyl uridines were obtained in order to explore their antimicrobial properties and solubility. It has been shown that new ribonucleoside derivatives have an order of magnitude better solubility in water compared to their 2'-deoxy analogues and effectively inhibit the growth of a number of Gram-positive bacteria, including resistant strains of Mycobacterium smegmatis (MIC=15-200 µg/mL) and Staphylococcus aureus (MIC=25-100 µg/mL). Their activity is comparable to that of some antibiotics used in medicine.

OBJECTIVE: Evaluate the change in participant emergency care knowledge and skill confidence after implementation of the WHO-International Committee of the Red Cross (ICRC) Basic Emergency Care (BEC) course. DESIGN: Pretest/post-test quasi-experimental study. SETTING: Mechnikov Hospital in Dnipro, Ukraine. PARTICIPANTS: Seventy-nine participants engaged in the course, of whom 50 (63.3%) completed all assessment tools. The course was open to healthcare providers of any level who assess and treat emergency conditions as part of their practice. The most common participant profession was resident physician (24%), followed by health educator (18%) and prehospital provider (14%). INTERVENTIONS: The 5-day WHO-ICRC BEC course. PRIMARY AND SECONDARY OUTCOME MEASURES: Change in pre-course and post-course knowledge and skill confidence assessments. Open-ended written feedback was collected upon course completion and analysed for common themes. RESULTS: Participant knowledge assessment scores improved from 19 (IQR 15-20) to 22 (IQR 19-23) on a 25-point scale (p<0.001). Participant skill confidence self-assessment scores improved from 2.5 (IQR 2.1-2.8) to 2.9 (IQR 2.5-3.3) on a 4-point scale (p<0.001). The most common positive feedback themes were high-quality teaching and useful skill sessions. The most common constructive feedback themes were translation challenges and request for additional skill session time. CONCLUSIONS: This first implementation of the WHO-ICRC BEC course for front-line healthcare providers in Ukraine was successful and well received by participants. This is also the first report of a BEC implementation outside of Africa and suggests that the course is also effective in the European context, particularly in humanitarian crisis and conflict settings. Future research should evaluate long-term knowledge retention and the impact on patient outcomes. Further iterations should emphasise local language translation and consider expanding clinical skills sessions.

Background and Aim: Although Enterococcus faecalis and Enterococcus faecium are common members of human and animal gut microbiota, their resistance to different antimicrobials makes them important pathogens. Multidrug-resistant enterococci often contaminate foods of animal origin at slaughterhouses. The World Health Organization and the World Organization for Animal Health recommend including animal-derived enterococci in antimicrobial resistance (AMR) monitoring programs. This study aimed to fill a literature gap by determining the current AMR prevalence of E. faecalis and E. faecium from different food-producing animals in Russia. Materials and Methods: Samples of biomaterial were taken from chickens (n=187), cattle (n=155), pigs (n=49), turkeys (n=34), sheep (n=31), and ducks (n=31) raised at 28 farms in 15 regions of Russia. Isolates of E. faecalis (n=277) and of E. faecium (n=210) (487 isolates in total; 1 isolate per sample) were tested for resistance to 12 antimicrobials from 11 classes using the broth microdilution method. Three criteria were used for the interpretation of minimum inhibitory concentration: Epidemiological cutoff values (ECOFFs) from the European Committee on Antimicrobial Susceptibility Testing (EUCAST) and Clinical and Laboratory Standards Institute (CLSI) clinical breakpoints. The AMR cloud online platform was used for data processing and statistical analysis. Results: A difference of >10% was found between E. faecalis and E. faecium resistance to several antimicrobials (erythromycin, gentamycin, tetracycline, chloramphenicol, ciprofloxacin, and streptomycin). In total, resistance to most antimicrobials for enterococci isolates of both species taken from turkeys, chicken, and pigs was higher than cattle, sheep, and ducks. The highest levels were found for turkeys and the lowest for ducks. Among antimicrobials, resistance to bacitracin and virginiamycin was 88-100% in nearly all cases. High levels of clinical resistance were found for both bacteria species: Rifampicin (44-84%) from all animals, tetracycline (45-100%) from poultry and pigs, and erythromycin (60-100%), ciprofloxacin (23-100%), and trimethoprim-sulfamethoxazole (33-53%) from chickens, turkeys, and pigs. No vancomycin-resistant isolates were found. Most isolates were simultaneously resistant to one-three classes of antimicrobials, and they were rarely resistant to more than three antimicrobials or sensitive to all classes. Conclusion: Differences in resistance between enterococci from different farm animals indicate that antimicrobial application is among the crucial factors determining the level of resistance. Conversely, resistance to rifampicin, erythromycin, tetracycline, and ciprofloxacin found in enterococci from farm animals in our study was notably also found in enterococci from wild animals and birds. Our results may be partly explained by the intrinsic resistance of E. faecium and E. faecalis to some antimicrobials, such as trimethoprim/sulfamethoxazole and bacitracin.

Reindeer herding is a vitally important agricultural sector in the Russian Far North. It is believed that Northern ecosystems readily accumulate persistent pollutants because of trophic chains and climate features peculiar to the region. Reindeers graze on vast areas, and their seasonal migrations to distances of up to hundreds of kilometers in the North-South direction increase the likelihood of crossing a locally polluted area. Here, we present the results of a large-scale nationwide study of reindeer liver, kidneys, and muscle pollution by dioxins, cadmium, and mercury. Samples were taken in 2015-2020 from 41 locations in 8 reindeer-herding regions of Russia. Dioxins were determined in 383 samples of liver and 13 of muscle, and cadmium and mercury-in 505 samples of liver, 315 of kidneys, and 22 of muscle. Dioxin pollution has shown a clear geographical trend, i.e., liver concentrations of dioxins steadily decrease from the Western to the Eastern parts on the Russian Far North, with the highest concentration of 76.5 pg/g of fat WHO-TEQ. The discovered trend may be explained by the pattern of chemical plants' localization and by the density of reindeer population (as known from the literature sources). The highest concentrations of metals were found in kidneys (7.3 mg/kg of cadmium and 1.1 mg/kg of mercury). The contribution of local sources to cadmium and mercury pollution was found to be less than expected. We also suggest that reindeer liver may serve as a good additional indicator of environmental pollution by dioxins and heavy metals.

BACKGROUND AND AIM: Commensal Escherichia coli is an important indicator of antimicrobial resistance (AMR) in animals and food of animal origin. Therefore, it was recommended by the World Health Organization and OIE for inclusion in resistance surveillance programs. At the same time, the data on E. coli isolates from animals in Russia are scarce. The aim of this work was to determine the current prevalence of resistance and genetic markers of non-pathogenic commensal E. coli collected from major food-producing animals (poultry, pigs, and cows) in different regions of Russia and to compare these data with data from other countries to prioritize antimicrobials for limiting their use according to the National Action Plan. MATERIALS AND METHODS: Samples (n=306) were collected from biomaterial of chicken, turkey, cows, and pigs raised on 11 farms in the European part of Russia, Siberia, and North Caucasus. Isolates (n=306) of E. coli were tested for resistance to 11 antimicrobials from ten classes using the broth microdilution method. MICs were interpreted against EUCAST microbiological and clinical breakpoints. For data analysis and statistical processing, the AMRcloud online platform was used. The data are presented in comparison with other countries. RESULTS: In Russia, higher levels of microbiological and clinical resistance of E. coli to critically important antimicrobials, including colistin, cefotaxime, and ciprofloxacin, were found compared to other countries, especially in poultry: About 30% of isolates from chicken were resistant to colistin, 8% to cefotaxime, and 88% to ciprofloxacin according to EUCAST ECOFFs. Only 10% of isolates from cows were resistant to cefotaxime. About 47% of isolates of E. coli from chicken had a moderate relative resistance for ampicillin and 56% for tetracycline. For most antimicrobials, isolates from cows demonstrated a lower resistance than isolates from poultry and pigs. All tested isolates from chicken, turkey, and pigs showed a simultaneous microbiological resistance to at least three classes of antimicrobials. No pan-resistant isolates were found. CONCLUSION: High levels of AMR of commensal E. coli from poultry, especially for critically important drugs, are a matter of concern and should be taken into account when choosing antimicrobials to be restricted for use in animal husbandry according to the National Action Plan.

APHC3 is an analgesic polypeptide that was found in the sea anemone (Heteractis crispa), and contains 56 amino acid residues. This polypeptide is of interest for the development of medications for diseases, associated with inflammatory or neuropathological processes, as well as its use as an analgesic. This work presents an innovative biotechnological method for APHC3 production. We have constructed a recombinant plasmid intended for biosynthesizing the fusion protein consisting of a chitin-binding domain, DnaB mini-intein from Synechocystis sp. capable of undergoing pH-dependent self-cleavage, and the target peptide. In the process of biosynthesis the fusion protein aggregates and forms the inclusion bodies that are welcomed since APHC3 is a cytotoxic peptide. The target peptide recovery process developed by us involves 3 chromatographic steps. The method developed by us enables to produce 940 mg of the recombinant APHC3 from 100 g of the inclusion bodies. The method is straightforward to implement and scale up. The recombinant APHC3 activity and effectiveness as an analgesic was proved by animal testing.

Phosphoribosylpyrophosphate synthetase (PRPPS) from the thermophilic bacterial strain Thermus thermophilus HB27 catalyzes the synthesis of phosphoribosylpyrophosphate from ribose 5-phosphate and ATP, and belongs to the class I PRPPSs. The three-dimensional structure of the recombinant enzyme was solved at 2.2 Šresolution using crystals grown in microgravity from protein solution containing ATP, magnesium and sulfate ions. An ADP molecule was located in the active site of each subunit of the hexameric enzyme molecule and sulfate ions were located in both the active and allosteric sites. It was found that the catalytic loop that restricts the active-site area and is usually missing from the electron-density map of class I PRPPSs adopts different conformations in three independent subunits in T. thermophilus PRPPS. A closed conformation of the active site was found in one of subunits where the highly ordered catalytic ß-hairpin delivers the Lys and Arg residues that are essential for activity directly to the ADP molecule, which occupies the ATP-binding site. A comparison of the conformations of the catalytic loop in the three independent subunits reveals a possible mode of transition from the open to the closed state of the active site during the course of the catalyzed reaction.

Thymosin ß4 is a 43 amino acid long peptide with an acetylated N-terminal serin that has a high potential as a remedy for healing ulcers, wounds and burns. Although protein biosynthesis offers attractive opportunities in terms of a large-scale production, currently thymosin ß4 is mainly produced by chemical synthesis. The problems that hinder the successful commercialization of the biotechnological approach are associated with the small peptides expression and N-terminal acetylation. This work presents an innovative biotechnological method for thymosin ß4 production that employs the peptide acetylation in vivo. A genetically engineered construct was created, where the Tß4 coding sequence fused with the intein Mxe GyrA sequence and chitin-binding domain was combined with the acetyltransferase coding sequence to form a polycistronic construct under a stringent control of T7 promoter. This plasmid construct provided for the expression of the Tß4-intein fusion protein. In the process of the post-translational modification in vivo formyl methionine was completely removed from the target peptide N-terminus and followed by the Tß4 precursor N-terminal acetylation. The use of the intein-mediated expression system made it possible to extract thymosin ß4 in only 2 chromatographic runs. The method is straightforward to implement and scale up.
