BACKGROUND: Current assessment methods of penile cavernous fibrosis in animal models have limitations due to the inability to provide complex and volume analysis of fibrotic alterations. OBJECTIVE: The aim was to evaluate micro-computed tomography for assessment of cavernous fibrosis and compare it with histological, histochemical, immunohistochemical, and RT-PCR analysis. MATERIALS AND METHODS: A controlled trial was performed involving 25 New Zealand male rabbits with induced testosterone deficiency by orchidectomy. Penile samples were obtained before and after 7, 14, 21, and 84 days from orchidectomy. We consistently performed (a) gray value analysis of corpora cavernosa 3D models reconstructed after micro-computed tomography, (b) morphometry of smooth muscles/connective tissue ratio, collagen type I/III ratio, and area of TGF-beta-1 expression in corpora cavernosa, and (c) RT-PCR of TGF-beta-1 expression. RESULTS: Micro-computed tomography allowed visualization of penile structures at a resolution comparable to light microscopy. Gray values of corpora cavernosa decreased from 1673 (1512-1773) on the initial day to 1184 (1089-1232) on the 21st day (p < 0.005). However, on the 84th day, it increased to 1610 (1551-1768). On 21st and 84th days, there was observed a significant decrease in smooth muscle/connective tissue ratio and a significant increase in collagen type I/III ratio (p < 0.05). TGF-beta1 expression increased on the 84th day according to immunohistochemistry (p < 0.005). RT-PCR was impossible to conduct due to the absence of RNA in obtained samples after micro-CT. DISCUSSION AND CONCLUSIONS: Micro-computed tomography provided 3D visualization of entire corpora cavernosa and assessment of radiodensity alterations by gray value analysis in fibrosis progression. We speculate that gray value changes at early and late fibrosis stages could be related to tissue reorganization. RT-PCR is impossible to conduct on tissue samples studied by micro-CT due to RNA destruction. We also suggest that micro-computed tomography could negatively affect the immunohistochemical outcome, as a significant increase of TGF-beta-1 expression occurs later than histological fibrotic signs.

The combustion of solid fuel at power plants pollutes adjacent areas with potentially toxic elements (PTEs), which increases risks to public health in the vicinity of these facilities. The proposed paper presents the results of a geochemical study of PTEs (Cr, Mn, Ni, Cu, Zn, Cd, and Pb) contamination in the vicinity of Novocherkassk Power Plant (NPP) as it relates to environmental and human health risks. The impact zone of NPP is pronounced for a distance of approximately 7 km northwest of the enterprise-the second largest coal power plant in Southern Russia. Data from monitoring sites lead us to conclude that spatial patterns of soil pollution are strongly influenced by the peculiarities of local atmospheric circulation, while the characteristics of soils within the study area play a secondary role. The highest levels of PTEs and their exchangeable forms exceed both regional background and sanitary and hygienic standards within a radius of 3 km to the west of the plant, which corresponds to a zone of soils contaminated with Cr, Ni, Cu, Zn, Cd, and Pb. The carcinogenic risk to human health slightly exceeds the permissible standard of 1 × 10-6 for soils in close vicinity of the enterprise due to the potential human intake of Ni, Cd, and Pb. The results of the health risk assessment indicate no noncarcinogenic risks for adults, while for children, they are low.

RESUMEN

Micro-CT visualization allows reconstruction of eye structures with the resolution of light microscopy and estimation of tissue densities. Moreover, this method excludes damaging procedures and allows further histological staining due to the similar steps in the beginning. We have shown the feasibility of the lab-based micro-CT machine usage for visualization of clinically important compartments of human eye such as trabecular outflow pathway, retina, iris and ciliary body after pre-treatment with iodine in ethanol. We also identified the challenges of applying this contrasting technique to lens, cornea, and retina and proposed alternative staining methods for these tissues. Thereby this work provides a starting point for other studies for imaging of human eyes in normal and pathological conditions using lab-based micro-CT systems.

The effects of bulk- and nano-CuO were monitored on barley (Hordeum sativum L.) in hydroponic conditions. The anatomical and cyto-/morphometric parameters of plants, exposed to both types of CuO in different doses (300 and 2000 mg/L) were recorded. The germination rate, root and shoot lengths decreased in a dose-dependent manner. Exposure to nano-CuO significantly increased Cu content in the H. sativum roots; however, the translocation rates of dissolved Cu were low and showed less accumulation in above-ground tissues. The differences between nano- and bulk-CuO treated plants were sufficiently evident, but at lower concentrations, these differences were non-significant. The relative seed germination inhibition was noted up to 11% and 22% under the high dose of bulk- and nano-CuO, respectively; however, at low dose, it was non-significant. The relative root length was reduced 3.6 fold by bulk- and 1.5 fold by nano-CuO, and shoot lengths decreased 1.6 fold by bulk- and 1.4 fold by nano-CuO under the high dose after growth of 30 days. It indicated more morphological effects on H. sativum caused by bulk- than the nano-CuO. The cytomorphometric analysis indicated the average cortex cell, total cortex, and total central cylinder areas of root cells and the average areas of chlorenchyma leaf cells were increased as compared to control in both bulk- and nano-CuO treated plants. It showed destructive effects of nano- and bulk-CuO on cellular organizations of H. sativum anatomy. Thus, at the low dose, the minimal effects of nano-CuO were observed than the bulk. Therefore, the finding could be interest for the safe application of nano-CuO.

The progressive increase in nanoparticles (NPs) applications and their potential release into the environment because the majority of them end up in the soil without proper care have drawn considerable attention to the public health, which has become an increasingly important area of research. It is required to understand ecological threats of NPs before applications. Once NPs are released into the environment, they are subjected to translocation and go through several modifications, such as bio/geo-transformation which plays a significant role in determination of ultimate fate in the environment. The interaction between plants and NPs is an important aspect of the risk assessment. The plants growing in a contaminated medium may significantly pose a threat to human health via the food chain. Metal oxide NPs ZnO and CuO, the most important NPs, are highly toxic to a wide range of organisms. Exposure and effects of CuO and ZnO NPs on soil biota and human health are critically discussed in this study. The potential benefits and unintentional dangers of NPs to the environment and human health are essential to evaluate and expected to produce less toxic and more degradable NPs to minimize the environmental risk in the future.

RESUMEN

Vapor bubble nucleation on a microrough surface wetted by a volatile, incompressible liquid has been analyzed. The work of formation of a critical nucleus in a surface cavity has been evaluated. Concave sites of a surface with negative curvature can decrease the height of the activation barrier for the formation of a critical nucleus. The average density of the nucleation sites has been evaluated for a surface whose (small) deviation from a plane is specified by a Gaussian random function.

RESUMEN

This paper reports on noninertial cavitation that occurs beyond the zone close to the horn tip to which the inertial cavitation is confined. The noninertial cavitation is characterized by collating the data from a range of measurements of bubbles trapped on a solid surface in this noninertial zone. Specifically, the electrochemical measurement of mass transfer to an electrode is compared with high-speed video of the bubble oscillation. This gas bubble is shown to be a "noninertial" event by electrochemical surface erosion measurements and "ring-down" experiments showing the activity and motion of the bubble as the sound excitation was terminated. These measurements enable characterization of the complex environment produced below an operating ultrasonic horn outside of the region where inertial collapse can be detected. The extent to which solid boundaries in the liquid cause the frequencies and shapes of oscillatory modes on the bubble wall to differ from their free field values is discussed.
