RESUMEN

Bulimulus bonariensis is considered a species of relevance to agribusiness, having been declared a pest with indirect damage because of its negative effects on several crops such as soybeans, chickpeas, and corn in central and northern Argentina. The objective of this work was to analyze the growth pattern of a population born under laboratory conditions, to explore population aspects such as survival and mortality, to estimate the age and size at gonadal maturity and first reproduction, and to contribute to the knowledge of the reproductive biology of this gastropod. From the clutches obtained, the basic biologic parameters were calculated and the individuals hatched under laboratory conditions counted and measured every two weeks. The clutches contained an average of 44 eggs, which took about 13.7 days to hatch at a birth rate of 41.82%. The growth pattern in the five clutches was analyzed individually, and the logistic model used was the one with the highest degree of fit to that observed growth pattern, followed by the Gompertz model, and finally the von Bertalanffy model. In addition, the models were applied to the 102 specimens analyzed together as a cohort, where the best fitting model was also proved to be the logistic growth model. A concave type III survival curve was obtained from the horizontal life table. The cohort was reduced by 48% during the first 50 days after birth. Beyond one month of hatching, life expectancy gradually increased and remained high between 65-302 days of life. After day 330, life expectancy decreased and only 13.72% exceeded one year of birth, with an average length of 16.68 mm. The last specimen died after 23 months at a total length of 20.24 mm, and the life expectancy was estimated at almost three years. In addition, it was inferred that gonadal maturity, when these gastropods reach 12 mm of total shell length, is reached after 200 days of life. Therefore, the individuals that are born are able to reproduce for the first time a year after birth, when they have the approximate size of 16.68 mm.

Bulimulus bonariensis, a snail distributed over a large part of Argentina, has generated negative effects on the agribusiness. We accordingly conducted a seasonal sampling during 2018-2019 in order to characterize the gametic cycle and establish the size of this gastropod at gonadal maturity. Three reproductive stages were identified: the mature in autumn; the spawn, which begins in winter but occurs mainly in the spring, followed by the post-spawn in summer, where an absence of gonadal rest was evidenced. In the fall, in maturity resorption was observed in addition to abundant primary spermatocytes, secondary spermatocytes, and growing vitellogenic oocytes. At the same time, could correlate the degree of spermioviduct development with the stage of gonadal development. Moreover, we used a logistic regression to calculate the size at gonadal maturity, which was established at the total shell length of 12 mm. In addition, we found that at the beginning of reproductive development those gastropods are protogynous hermaphrodites, but after reaching gonadal maturity became simultaneous hermaphrodites. Finally, B. bonariensis also proved to be an iteroparous species. The information provided here will be essential for delineating and establishing population control strategies.

Reduced sleep duration and sleep deprivation have been associated with cognitive impairment as well as decreased white matter integrity as reported by experimental studies. However, it is largely unknown whether differences in sleep duration and sleep quality might affect microstructural white matter and cognition. Therefore, the present study aims to examine the cross-sectional relationship between sleep duration, sleep quality, and cognitive performance in a naturalistic study design, by focusing on the association with white matter integrity in a large sample of healthy, young adults. To address this, 1,065 participants, taken from the publicly available sample of the Human Connectome Project, underwent diffusion tensor imaging. Moreover, broad cognitive performance measures (NIH Cognition Toolbox) and sleep duration and quality (Pittsburgh Sleep Quality Index) were assessed. The results revealed a significant positive association between sleep duration and overall cognitive performance. Shorter sleep duration significantly correlated with fractional anisotropy (FA) reductions in the left superior longitudinal fasciculus (SLF). In turn, FA in this tract was related to measures of cognitive performance and was shown to significantly mediate the association of sleep duration and cognition. For cognition only, associations shift to a negative association of sleep duration and cognition for participants sleeping more than 8 hr a day. Investigations into subjective sleep quality showed no such associations. The present study showed that real-world differences in sleep duration, but not subjective sleep quality are related to cognitive performance measures and white matter integrity in the SLF in healthy, young adults.

We aimed to extend our knowledge on the relationship between physical fitness (PF) and both white matter microstructure and cognition through in-depth investigation of various cognitive domains while accounting for potentially relevant nuisance covariates in a well-powered sample. To this end, associations between walking endurance, diffusion-tensor-imaging (DTI) based measures of fractional anisotropy (FA) within brain white matter and cognitive measures included in the NIH Toolbox Cognition Battery were investigated in a sample of n = 1206 healthy, young adults (mean age = 28.8; 45.5% male) as part of the human connectome project. Higher levels of endurance were associated with widespread higher FA (pFWE < 0.05) as well as with enhanced global cognitive function (p < 0.001). Significant positive relationships between endurance and cognitive performance were similarly found for almost all cognitive domains. Higher FA was significantly associated with enhanced global cognitive function (p < 0.001) and FA was shown to significantly mediate the association between walking endurance and cognitive performance. Inclusion of potentially relevant nuisance covariates including gender, age, education, BMI, HBA1c, and arterial blood pressure did not change the overall pattern of results. These findings support the notion of a beneficial and potentially protective effect of PF on brain structure and cognition.

The exotic land gastropod Ovachlamys fulgens (Gude, 1900), aka the jumping snail, was registered in Eldorado city, Misiones province, constituting the first documented record of that species in Argentina-and for all of South America as well. Identities of the individuals were confirmed through morphological examination, and by a DNA sequencing of the cytochrome oxidase subunit I gene. DNA sequences for the mitochondrial 16S-rRNA and the nuclear 18S-rRNA genes were also obtained from the morphologically-confirmed specimens to gain insights into the genetic background of this species in Argentina, and as additional markers for enhancing the rapidity of identification by governmental authorities who are responsible for managing the presence of exotic species. Different size classes suggested that the species had successfully reproduced and become established in the locality. Ovachlamys fulgens is emphasized in the literature as being a serious pest to orchids. Misiones province harbors a high species richness of orchids, representing about one half of the known species in Argentina. Owing to the use of the orchid as an ornamental plant, the commercial trade could have facilitated the introduction of the species into Argentina. Control measures, preventive education, and surveillance are needed to prevent the further spread of this snail.

Heleobia piscium (d'Orbigny, 1835), a member of the Cochliopidae family found only in South America, is distributed from Entre Ríos, Delta del Paraná, and the littoral of the Río de la Plata down as far as to Punta Indio (Buenos Aires), the southernmost limit of the snail's geographical distribution. To date, little information is available regarding the reproductive cycle of species within this family either in Argentina or throughout South America. The present work analyzed the histology of the reproductive system of the gonochoric species H. piscium and determined the stages oogenesis and spermatogenesis under natural conditions. Specimens of H. piscium were collected in the Multiple-Use Natural Reserve Isla Martín García, located in the Upper Río de la Plata estuary to the south of the mouth of the Uruguay River. The gametogenic cycle in both sexes was found to consist of the following stages: early maturation, maturation, and evacuation. The maturation period was found to extend from January to October and evacuation of the gametes to start in November and end in February (summer in the Southern Hemisphere). The results indicated the H. piscium exhibit a reproductive cycle without a resting period.

Fast excitatory synaptic transmission that is contingent upon N-methyl d-aspartate receptor (NMDAR) function contributes to core information flow in the central nervous system and to the plasticity of neural circuits that underlie cognition. Hypoactivity of excitatory NMDAR-mediated neurotransmission is hypothesized to underlie the pathophysiology of schizophrenia, including the associated cognitive deficits. The neurosteroid pregnenolone (PREG) and its metabolites pregnenolone sulfate (PregS) and allopregnanolone in serum are inversely associated with cognitive improvements after oral PREG therapy, raising the possibility that brain neurosteroid levels may be modulated therapeutically. PregS is derived from PREG, the precursor of all neurosteroids, via a single sulfation step and is present at low nanomolar concentrations in the central nervous system. PregS, but not PREG, augments long-term potentiation and cognitive performance in animal models of learning and memory. In this report, we communicate the first observation that PregS, but not PREG, is a potent (EC50 ∼2 pM) enhancer of intracellular Ca(2+) that is contingent upon neuronal activity, NMDAR-mediated synaptic activity, and L-type Ca(2+) channel activity. Low picomolar PregS similarly activates cAMP response element-binding protein (CREB) phosphorylation (within 10 minutes), an essential memory molecule, via an extracellular-signal-regulated kinase/mitogen-activated protein kinase signal transduction pathway. Taken together, the results are consistent with a novel biologic role for the neurosteroid PregS that acts at picomolar concentrations to intensify the intracellular response to glutamatergic signaling at synaptic but not extrasynaptic, NMDARs by differentially augmenting CREB activation. This provides a genomic signal transduction mechanism by which PregS could participate in memory consolidation of relevance to cognitive function.

The species of the genus Drepanotrema, most of them endemic to the Neotropical region, belong to the Planorbidae. Of the nine species of this genus, six are found in Argentina. The present investigation analysed the individual growth of Drepanotrema cimex in Arenalcito pond (34°11 S, 58°15 W), Martín García Island Natural Reserve of Multiple Uses, Buenos Aires Province, Argentina. A total of 14 samples were collected (n = 1931) from February 2006 to June 2007. Five environmental variables were measured: water temperature, dissolved oxygen (mg L1), electrical conductivity (µS cm1), total dissolved solids, and pH. The individual growth of the species was analysed mathematically by means of the von Bertalanffy equation, where: Lt = 5,6(1-e2,0592 (t0,293)). The parameter L was estimated by the Ford-Walford Method. The population of D. cimex was characterised by a complex and dynamic size structure throughout the annual cycle. The analysis of the curves revealed unimodal (2006) and polymodal (April to June 2007) distributions, which pattern served to identify the existence of cohorts within the population studied.

Este artigo analisa o crescimento individual de Drepanotrema cimex na lagoa de Arenalcito, Reserva Natural de Usos Múltiples Ilha Martín García, Buenos Aires, Argentina (34°11 S - 58°15 W). A amostragem foi realizada entre fevereiro de 2006 e junho de 2007. Durante o curso das campanhas foram obtidas 14 amostras (n = 1931). Cinco variáveis ambientais foram medidas: temperatura da água, oxigênio dissolvido (mg L1), condutividade elétrica (uS cm1), sólidos dissolvidos e pH. O crescimento individual das espécies foi analisada matematicamente por meio da equação de von Bertalanffy em que: Lt = 5,6 (1-e2, 0592 (t0, 293)). O paràmetro L foi estimada pe lo método de Ford-Walford. A população de D.cimex foi caracterizada por uma estrutura complexa e dinâmica de tamanho ao longo do ciclo anual. A análise das curvas mostrou distribuições unimodais (2006) e polimodais (abril-junho 2007), identificando a existência de coortes na população estudada.

BACKGROUND: The γ-aminobutyric acid (GABA) type A receptor (GABA(A)R) contains the recognition sites for a variety of agents used in the treatment of brain disorders, including anxiety and epilepsy. A better understanding of how receptor expression is regulated in individual neurons may provide novel opportunities for therapeutic intervention. Towards this goal we have studied transcription of a GABA(A)R subunit gene (GABRB1) whose activity is autologously regulated by GABA via a 10 base pair initiator-like element (ß(1)-INR). METHODS: By screening a human cDNA brain library with a yeast one-hybrid assay, the Polycomblike (PCL) gene product PHD finger protein transcript b (PHF1b) was identified as a ß(1)-INR associated protein. Promoter/reporter assays in primary rat cortical cells demonstrate that PHF1b is an activator at GABRB1, and chromatin immunoprecipitation assays reveal that presence of PHF1 at endogenous Gabrb1 is regulated by GABA(A)R activation. RESULTS: PCL is a member of the Polycomb group required for correct spatial expression of homeotic genes in Drosophila. We now show that PHF1b recognition of ß(1)-INR is dependent on a plant homeodomain, an adjacent helix-loop-helix, and short glycine rich motif. In neurons, it co-immunoprecipitates with SUZ12, a key component of the Polycomb Repressive Complex 2 (PRC2) that regulates a number of important cellular processes, including gene silencing via histone H3 lysine 27 trimethylation (H3K27me3). CONCLUSIONS: The observation that chronic exposure to GABA reduces PHF1 binding and H3K27 monomethylation, which is associated with transcriptional activation, strongly suggests that PHF1b may be a molecular transducer of GABA(A)R function and thus GABA-mediated neurotransmission in the central nervous system.

N-methyl D-aspartate (NMDA) receptors (NMDARs) mediate fast excitatory synaptic transmission and play a critical role in synaptic plasticity associated with learning and memory. NMDAR hypoactivity has been implicated in the pathophysiology of schizophrenia, and clinical studies have revealed reduced negative symptoms of schizophrenia with a dose of pregnenolone that elevates serum levels of the neuroactive steroid pregnenolone sulfate (PregS). This report describes a novel process of delayed-onset potentiation whereby PregS approximately doubles the cell's response to NMDA via a mechanism that is pharmacologically and kinetically distinct from rapid positive allosteric modulation by PregS. The number of functional cell-surface NMDARs in cortical neurons increases 60-100% within 10 minutes of exposure to PregS, as shown by surface biotinylation and affinity purification. Delayed-onset potentiation is reversible and selective for expressed receptors containing the NMDAR subunit subtype 2A (NR2A) or NR2B, but not the NR2C or NR2D, subunits. Moreover, substitution of NR2B J/K helices and M4 domain with the corresponding region of NR2D ablates rapid allosteric potentiation of the NMDA response by PregS but not delayed-onset potentiation. This demonstrates that the initial phase of rapid positive allosteric modulation is not a first step in NMDAR upregulation. Delayed-onset potentiation by PregS occurs via a noncanonical, pertussis toxin-sensitive, G protein-coupled, and Ca(2+)-dependent mechanism that is independent of NMDAR ion channel activation. Further investigation into the sequelae for PregS-stimulated trafficking of NMDARs to the neuronal cell surface may uncover a new target for the pharmacological treatment of disorders in which NMDAR hypofunction has been implicated.

The aim of this study was to determine whether the dry needling of myofascial trigger points (MTrPs) is superior to placebo in the prevention of pain after total knee arthroplasty. Forty subjects were randomised to a true dry needling group (T) or to a sham group (S). All were examined for MTrPs by an experienced physical therapist 4-5 hours before surgery. Immediately following anesthesiology and before surgery started, subjects in the T group were dry needled in all previously diagnosed MTrPs, while the S group received no treatment in their MTrPs. Subjects were blinded to group allocation as well as the examiner in presurgical and follow-up examinations performed 1, 3, and 6 months after arthroplasty. Subjects in the T group had less pain after intervention, with statistically significant differences in the variation rate of the visual analogue scale (VAS) measurements 1 month after intervention and in the need for immediate postsurgery analgesics. Differences were not significant at 3- and 6-month follow-up examinations. In conclusion, a single dry needling treatment of MTrP under anaesthesia reduced pain in the first month after knee arthroplasty, when pain was the most severe. Results show a superiority of dry needling versus placebo. An interesting novel placebo methodology for dry needling, with a real blinding procedure, is presented.

INTRODUCTION: The Argentine Society of Transplants has set in motion an activity called Grand Rounds to Provide Feedback on Procurement Activities with the purpose of informing, training, and creating awareness by providing information about the evolution of transplanted patients. OBJECTIVE: To measure, describe and analyze the impact that the transplant results presented at the rounds had on participants. RESULTS: One hundred and fifty-eight surveys were conducted. The respondents' average age was 40.06 years (95% confidence interval [CI 95%] 38.39-41.73) including 52.08% women (CI 95%: 43.92%-60.24%); 77.14% physicians (CI 95%: 84.09%-70.18%); 7.85% nurses (CI 95%: 3.4%-12.31%;) and 4.28% surgical nurses and other professional (CI 95%: 0.93%-7.64). When asked how comforted they felt when they learnt about a transplantation, the score prior to the round was 4 (CI 95%: 3.79-4.20) and after it, 4.8 (CI 95%: 4.69-4.90; P < .05). When asked about the transparency of the donation process, the average score before the round was 4.16 (CI 95%: 3.96-4.36) and after it, 4.76 (CI 95%: 4.67-4.86; P < .05). When asked how proactive they considered themselves before, before the average score was 3.54 (CI 95% 3.30-3.78) and post-round, 4.53 (CI 95%: 4.38-4.67; P < .05). When asked if they thought that this activity might be useful for them to manage future donors, 97.41% (CI 95%: 89%-96.98%) of respondents answered affirmatively. The answers to the question whether they thought this activity might increase donation were affirmative in 92.99% of cases; "Don't know", 6.36%; and negative in less than 1%. When consulted about their feelings, the preferred one was "happiness" for 46.82% of respondents; followed by "satisfaction" for 29.36% and "emotion" for 23.80%. The analysis of the open-ended questions revealed that the rounds were perceived as closure of the procurement-transplantation process. CONCLUSIONS: This widely accepted tool was viewed as a vehicle to interconnect links in the work process from organ procurement to transplantation.

RESUMEN

This report aims to facilitate the implementation of the Three Rs (reduction, refinement and replacement) in the testing of vaccines for regulatory and other purposes. The focus is predominantly on identification of reduction and refinement opportunities in batch potency testing but the principles described are widely applicable to other situations that involve experimental infections of animals. The report should also help to interpret the requirements of the European Pharmacopoeia with regard to the use of alternative tests, humane endpoints and other refinements. Two specific worked examples, for batch potency testing of Clostridium chauvoei and canine leptospira, with recommendations for harmonisation of international test requirements for these and other vaccines, are provided as appendices online.

gamma-Aminobutyric acid type B receptors (GABA(B)Rs) mediate both slow inhibitory synaptic activity in the adult nervous system and motility signals for migrating embryonic cortical cells. Previous papers have described the expression of GABA(B)Rs in the adult brain, but the expression and functional significance of these gene products in the embryo are largely unknown. Here we examine GABA(B)R expression from rat embryonic day 10 (E10) to E18 compared with adult and ask whether embryonic cortical neurons contain functional GABA(B)R. GABA(B)R1 transcript levels greatly exceed GABA(B)R2 levels in the developing neural tube at E11, and olfactory bulb and striatum at E17 but equalize in most regions of adult nervous tissue, except for the glomerular and granule cell layers of the main olfactory bulb and the striatum. Consistent with expression differences, the binding affinity of GABA for GABA(B)Rs is significantly lower in adult striatum compared with cerebellum. Multiple lines of evidence from in situ hybridization, RNase protection, and real-time PCR demonstrate that GABA(B)R1a, GABA(B)R1b, GABA(B)R1h (a subunit subtype, lacking a sushi domain, that we have identified in embryonic rat brain), GABA(B)R2, and GABA(B)L transcript levels are not coordinately regulated. Despite the functional requirement for a heterodimer of GABA(B)R subunits, the expression of each subunit mRNA is under independent control during embryonic development, and, by E18, GABA(B)Rs are negatively coupled to adenylyl cyclase in neocortical neurons. The presence of embryonic GABA(B)R transcripts and protein and functional receptor coupling indicates potentially important roles for GABA(B)Rs in modulation of synaptic transmission in the developing embryonic nervous system.
