Free (epi) catechin, quercetin, (epi) gallocatechin, flavonol glycosides and condensed tannins were identified according to their molecular mass, characteristic product ions and retention times in extracts obtained from leaves and branches of Maytenus ilicifolia (Congorosa) by mass spectrometry. The in vitro anthelmintic activity against cattle gastrointestinal nematodes of Congorosa extract was determined using the Egg Hatch Inhibition Assay. Additionally, commercial quercetin, gallocatechin and epicatechin were evaluated. Although total phenolics, total tannins and condensed tannins contents were lower in branches extract than in leaves extract, the EC50 were 0.065 mg/mL and 0.890 mg/mL for branches and leaves extract, respectively. Moreover, the use of polyvinylpyrrolidone as a blocking agent of tannins, did not change significantly the EC50 for branches extract, but significantly changed for leaves extract. Quercetin and gallocatechin EC50 values were in the range 0.03-0.05 mg/mL and epicatechin showed 100% inhibition of the egg hatching process at 0.004 mg/mL.

The sensitivity (Se) and specificity (Sp) of three diagnostic tests for the detection of Campylobacter fetus venerealis (Cfv) using field samples were estimated using a Bayesian latent class model (BLCM), accounting for the absence of a gold standard. The tests included in this study were direct immunofluorescence antibody test (IFAT), polymerase chain reaction (PCR), and real-time PCR (RT-PCR). Twelve farms from two different populations were selected and bull prepuce samples were collected. The IFAT was performed according to the OIE Manual. The conventional PCR was performed as multiplex, targeting the gene nahE for C. fetus species identification and insertion element ISCfe1 for Cfv identification. The RT-PCR was performed as uniplex: one targeting the gene nahE for C. fetus and the other targeting the insertion ISCfe1 (ISC2) for Cfv. Results from the BLCM showed a median Se of 11.7% (Bayesian credibility interval (BCI): 1.93-29.79%), 53.7% (BCI: 23.1-95.0%), and 36.1% (BCI: 14.5-71.7%) for IFAT, PCR, and RT-PCR respectively. The Sp were 94.5% (BCI: 90.1-97.9%), 97.0% (BCI: 92.9-99.3%), and 98.4% (BCI: 95.3-99.7%) for IFAT, PCR, and RT-PCR respectively. The correlation between PCR and RT-PCR was positive and low in samples from both sampled population (0.63% vs 8.47%). These results suggest that diagnostic sensitivity of the studied tests is lower using field samples than using pure Cfv strains.

A systematic review and meta-analysis (MA) were performed to summarize all scientific evidence for the effects of castration in male beef cattle on welfare indicators based on cortisol concentration, average daily gain (ADG), and vocalization. We searched five electronic databases, conference proceedings, and experts were contacted electronically. The main inclusion criteria involved completed studies using beef cattle up to one year of age undergoing surgical and non-surgical castration that presented cortisol concentration, ADG, or vocalization as an outcome. A random effect MA was conducted for each indicator separately with the mean of the control and treated groups. A total of 20 publications reporting 26 studies and 162 trials were included in the MA involving 1814 cattle. Between study heterogeneity was observed when analysing cortisol (I2=56.7%) and ADG (I2=79.6%). Surgical and non-surgical castration without drug administration compared to uncastrated animals showed no change (P≥0.05) in cortisol level. Multimodal therapy for pain did not decrease (P≥0.05) cortisol concentration after 30min when non-surgical castration was performed. Comparison between surgical castration, with and without anaesthesia, showed a tendency (P=0.077) to decrease cortisol levels after 120min of intervention. Non-surgical and surgical castration, performed with no pain mitigation, increased and tended to increase the ADG by 0.814g/d (P=0.001) and by 0.140g/d (P=0.091), respectively, when compared to a non-castrated group. Our MA study demonstrates an inconclusive result to draw recommendations on preferred castration practices to minimize pain in beef cattle.

BACKGROUND: On two farms it was noted that after routine treatment with monepantel, fecal egg counts failed to drop. This was accompanied by lambs mortality due to Haemonchus contortus infection. The aim of this work was to evaluate the efficacy of monepantel to control gastrointestinal nematodes (GIN) in two sheep farms, in Uruguay. FINDINGS: A Fecal Egg Count Reduction Test (FECRT) was subsequently performed at the Experimental Stations Glencoe of INIA Tacuarembó (Farm 1) and Sheep Unit of INIA La Estanzuela (Farm 2) using the World Association for the Advancement of Veterinary Parasitology guidelines. On Farm 1 the FECRT was performed using 6-8 month old Corriedale or Merino Dohne x Corriedale male lambs naturally infected with GIN. On day 0 pre-treatment, three groups of 15 lambs each were selected, blocked by fecal egg count level (FEC) and randomly assigned to one of the following: Group 0 = untreated control, Group 1 = treated with monepantel (Zolvix®, Novartis Animal Health Inc.) from stock previously purchased; Group 2 = treated with monepantel from stock provided by the supplier, at the recommended dose of 2.5 mg/kg of body weight. Fecal samples were collected directly from the rectum from each lamb on day 0 and on day 9 post-treatment. On Farm 2, the FECRT was conducted on a group of 8 month old male lambs Milchschaff x Finn. At this farm, 10 lambs were randomly allocated to be treated with monepantel (Group 1) and 10 lambs were randomly allocated to remain as untreated control (Group 0) using the same protocols as Farm 1. On farm 1 the FECR was 0.0% (95% CI = 0.0-49.0) and 42.0% (95% CI = 0.0-75.0) for Group 1 and Group 2 respectively. For Farm 2, the FECR was 82.1% (95% CI = 36.0-99.0). Haemonchus spp was the resistant genus. CONCLUSIONS: Poor effcicacy of monepantel in treating GIN parasites was demonstrated on both farms.
