RESUMEN

The maintenance of fluid homeostasis is necessary for function of the neural retina; however, little is known about the significance of potential fluid management mechanisms. Here, we investigated angiopoietin-4 (Angpt4, also known as Ang3), a poorly characterized ligand for endothelial receptor tyrosine kinase Tie2, in mouse retina model. By using genetic reporter, fate mapping, and in situ hybridization, we found Angpt4 expression in a specific sub-population of astrocytes at the site where venous morphogenesis occurs and that lower oxygen tension, which distinguishes peripheral and venous locations, enhances Angpt4 expression. Correlating with its spatiotemporal expression, deletion of Angpt4 resulted in defective venous development causing impaired venous drainage and defects in neuronal cells. In vitro characterization of angiopoietin-4 proteins revealed both ligand-specific and redundant functions among the angiopoietins. Our study identifies Angpt4 as the first growth factor for venous-specific development and its importance in venous remodeling, retinal fluid clearance and neuronal function.

Laboratory rats are generally fed ad libitum, although this method is associated with obesity and an increased frequency of spontaneous tumours. It has been challenging looking for ways to limit feed consumption in group-housed rats without any setbacks to animal welfare and scientific results. The diet board, as a method of dietary restriction, was used in the present study. Diet board feeding allows group housing and should result in enhanced welfare compared with traditional methods of dietary restriction. With respect to animal model robustness and translatability of results it is important that the feeding regime does not affect diurnal rhythmicity of biological parameters. In the present study the effects of diet board feeding on diurnal rhythms of blood glucose, serum ghrelin, faecal immunoglobulin A (IgA) and faecal corticosterone were assessed. The diet board did not alter diurnal rhythms, and adds weight to the use of this method for dietary restriction which should benefit animal health and the validity of scientific results generated from the animals.

Environmental enrichment provides laboratory animals with novelty and extra space, allowing different forms of multisensory stimulation ranging from social grouping to enhanced motor activity. At the extreme end of the spectrum, one can have a super-enriched environment. Environmental enrichment is believed to result in improved cognitive and sensorimotor functions both in naïve rodents and in animals with brain lesions such as those occurring after a stroke. Robust behavioral effects in animals which have suffered a stroke are probably related not only to neuronal plasticity in the perilesional cortex but also in remote brain areas. There is emerging evidence to suggest that testing restorative therapies in an enriched environment can maximize treatment effects, e.g., the perilesional milieu seems to be more receptive to concomitant pharmacotherapy and/or cell therapy. This review provides an updated overview on the effect of an enriched environment in stroke animals from the practical points to be considered when planning experiments to the mechanisms explaining why combined therapies can contribute to behavioral improvement in a synergistic manner.

Outbred NIH/S male mice were housed from weaning in groups of 4 without enrichment (control) or with nesting material (nest), nesting material and a box (nest-and-box), or nesting material and a tube (nest-and-tube) as environmental modification. The aim of the study was to investigate effects of widely recommended nesting material and additional shelters on male mice. The aggressiveness of the mice in their home cages clearly increased in the nest group, as assessed by the number of wounds. In the nest group, fighting was a stressful situation for the mice, leading to changes in weight gain and in the weights of the thymus, adrenals, spleen, and epididymal adipose tissue. Moreover, the agonistic behavior of these mice toward an intruder was increased both in individual tests (an intruder with the individual mouse) and group tests (an intruder with a group of mice). The provision of a box or tube as a shelter, in addition to nesting material, prevented intracage fighting and did not lead to alterations in the weight gain or organ weights of the mice. However, the agonistic behavior of mice with shelters was slightly increased in behavioral tests. Anxiety in the elevated plus-maze was not affected by any of the housing systems. In conclusion, the agonistic behavior of NIH/S mice, an aggressive strain, seemed to be easily enhanced by these environmental modifications. The suitability of any enrichment should be carefully evaluated, especially when highly aggressive mice are used.
