RESUMEN

The current study aimed to determine the polyphenol compounds in Nigella sativa (NS) and Eruca sativa (ES) seeds, and evaluate the impact of their addition either as a sole additive or in combination on the growth performance, digestibility, some rumen and blood parameters and antioxidative status of Barki lambs. Forty-eight male lambs (27.18 ± 0.22 kg, 5-6 months), were divided into 4 balanced groups. The experimental diets were randomly distributed to the control group (CON); fed alfalfa hay plus concentrate feed mixture at a ratio of 30:70% without additives, while, NSD, ESD, and NESD groups: fed CON diet plus 2% NS, 2% ES or 1% NS + 1% ES, respectively as a ratio from total mixed ration (TMR). Results indicated that rutin and catechin were the most phenolic compounds observed either in NS or ES seeds. The NS and ES-supplemented groups recorded the highest (P < 0.05) values for dry matter digestibility, nutritive values, average daily gain, and the best feed conversion ratio. However, growth performance, nutritive value, and all nutrient digestibility except for dry matter were not significantly altered with the NESD group. Concentrations of ruminal NH3-N and TVFA were significantly (P < 0.05) reduced with the NESD group, with no significant differences in pH values among different groups. Values of blood parameters showed significant increases in WBCs, PCV, and T-AOC, and decreases in cholesterol, triglycerides, and MDA with the addition of NS and ES seeds or both. Therefore, the addition of NS and ES seeds is recommended to improve lambs' health and antioxidant status.

RESUMEN

This study investigated the impact of auditory stimuli on muscular activation patterns using wearable surface electromyography (EMG) sensors. Employing four key muscles (Sternocleidomastoid Muscle (SCM), Cervical Erector Muscle (CEM), Quadricep Muscles (QMs), and Tibialis Muscle (TM)) and time domain features, we differentiated the effects of four interventions: silence, music, positive reinforcement, and negative reinforcement. The results demonstrated distinct muscle responses to the interventions, with the SCM and CEM being the most sensitive to changes and the TM being the most active and stimulus dependent. Post hoc analyses revealed significant intervention-specific activations in the CEM and TM for specific time points and intervention pairs, suggesting dynamic modulation and time-dependent integration. Multi-feature analysis identified both statistical and Hjorth features as potent discriminators, reflecting diverse adaptations in muscle recruitment, activation intensity, control, and signal dynamics. These features hold promise as potential biomarkers for monitoring muscle function in various clinical and research applications. Finally, muscle-specific Random Forest classification achieved the highest accuracy and Area Under the ROC Curve for the TM, indicating its potential for differentiating interventions with high precision. This study paves the way for personalized neuroadaptive interventions in rehabilitation, sports science, ergonomics, and healthcare by exploiting the diverse and dynamic landscape of muscle responses to auditory stimuli.

The Ataxia telangiectasia and Rad3-related (ATR) inhibitor ceralasertib in combination with the PD-L1 antibody durvalumab demonstrated encouraging clinical benefit in melanoma and lung cancer patients who progressed on immunotherapy. Here we show that modelling of intermittent ceralasertib treatment in mouse tumor models reveals CD8+ T-cell dependent antitumor activity, which is separate from the effects on tumor cells. Ceralasertib suppresses proliferating CD8+ T-cells on treatment which is rapidly reversed off-treatment. Ceralasertib causes up-regulation of type I interferon (IFNI) pathway in cancer patients and in tumor-bearing mice. IFNI is experimentally found to be a major mediator of antitumor activity of ceralasertib in combination with PD-L1 antibody. Improvement of T-cell function after ceralasertib treatment is linked to changes in myeloid cells in the tumor microenvironment. IFNI also promotes anti-proliferative effects of ceralasertib on tumor cells. Here, we report that broad immunomodulatory changes following intermittent ATR inhibition underpins the clinical therapeutic benefit and indicates its wider impact on antitumor immunity.

AIM: To develop normative data for meibomian gland dysfunction (MGD) parameters, using non-contact meibography technique of Sirius Costruzione Strumenti Oftalmici (CSO) machine, in an Egyptian population sample. METHODS: Observational, cross-sectional, analytic study, in which 104 Egyptian volunteers were included. Both upper lids were examined, using "Sirius CSO" machine. Each eyelid was given a degree of meibomian gland loss (MGL), which was calculated by the software of the machine. RESULTS: Mean percentage MGL in right upper lid was of 30.9%±12.6%, and that of left upper lid was 32.6%±11.8%. Thirty-four volunteers (32.7%) had first-degree MGL in their right upper lid, and 67.3% had second-degree loss. One volunteer (1%) had zero-degree MGL in left upper lid, 28 (26.9%) had first-degree loss, and 75 (72.1%) had second-degree loss. Degree of MGL in right upper eyelid was not related to age, but degree of MGL in left upper eyelid increased with age. There was statistically significant difference between both genders for degree of MGL in right eye (P=0.036) and in left eye (P=0.027). CONCLUSION: Noncontact meibography is a useful non-invasive tool for diagnosing MGL. MGL is diagnosed in 100% of apparently normal individuals; 26.9%-32.7% of which have first-degree MGL, and 67.3%-72.1% have second-degree MGL.

PURPOSE: Children and young adults with neuromuscular disorders have a high incidence of both spine and hip deformities. The aim of this study was to evaluate the outcome of either primary scoliosis or hip surgery in children and young adults with neuromuscular disorders. METHODS: A retrospective study was conducted on all children and young adults with neuromuscular-related synchronous hip subluxation/dislocation and scoliosis undergoing hip or scoliosis surgery in our institution between 2012 and 2021 with a minimum follow-up of 24 months. Demographic and operative data were collected; radiological parameters were measured preoperatively and postoperatively at final follow-up. RESULTS: Forty neuromuscular patients with synchronous hip displacement and scoliosis were included. Twenty patients with an average age of10.2 years had hip correction surgery performed primarily, with a mean follow-up of 54.9 (24-96) months. The other 20 patients with an average age of 12.4 years had scoliosis correction first, with a mean follow-up of 40 (24-60) months. In the "Hip first" group, pelvic obliquity, hip MP and Cobb angle were 16.8°, 71%, and 49°, respectively. At final follow-up, the mean pelvic obliquity and Cobb angles significantly progressed to 27.2° (p = 0.003) and 82.2° (p = 0.001), respectively. Eighteen patients (90%) required scoliosis correction after the hip surgery. In the "Scoliosis first" group, the mean pelvic obliquity, hip MP and Cobb angle were 21.2°, 49% and 65.5°, respectively. At final follow-up, both pelvic obliquity and Cobb angle significantly improved to 8.44° (p = 0.002) and 23.4° (p = 0.001), respectively. In 11/20 (55%) patients, the hip MP had significantly increased following the spinal surgery to 62% (p = 0.001), but only 5/20 (25%) patients underwent hip surgery after scoliosis correction. CONCLUSION: In neuromuscular patients presenting with synchronous hip displacement and scoliosis deformity, corrective scoliosis surgery is associated with a significant correction of pelvic obliquity and lower rates of secondary hip surgery. On the other hand, primary hip surgery does not reduce the risk of pelvic obliquity and scoliosis deformity progression.

RESUMEN

Despite being very common, lateral condyle fractures in children are rarely associated with acute nerve injuries. We present the case of a 10-year-old, left-handed male child who presented with a left lateral humeral condyle fracture associated with radial nerve injury. The patient was managed by open reduction and internal fixation plus radial nerve exploration, which was found entrapped in the fracture site. The patient achieved full recovery after 16 weeks. We report this case to present the approach and the operative findings and to emphasize the importance of a preoperative clinical examination in addition to preoperative planning to achieve a favorable outcome.

Two natural betaine sources; dehydrated condensed molasses fermentation solubles (Bet1) and Betafin®, a commercial anhydrous betaine extracted from sugar beet molasses and vinasses (Bet2); were used to investigate their impact on rumen fermentation parameters and lactation performance of lactating goats. Thirty-three lactating Damascus goats, with an average weight of 37 ± 0.7 kg and their age ranged from 22 to 30 months (2nd and 3rd lactation season), were divided into three groups, each group contained 11 animals. The control group (CON) was fed ration without betaine. While the other experimental groups were fed a control ration supplemented either with Bet1 or Bet2 to provide a 4 g betaine/kg diet. Results confirmed that betaine supplementation improved nutrient digestibility and nutritive value, and increased milk production and milk fat contents with both Bet1 and Bet2. Significant increases in concentration of ruminal acetate were observed in betaine-supplemented groups. Goats fed dietary betaine non-significantly recorded higher concentrations of short and medium-chain fatty acids (C4:0 to C12:0), and significant lower concentrations of C14:0 and C16:0 in milk. Also, both Bet1 and Bet2 non-significantly decreased the blood concentrations of cholesterol and triglycerides. Therefore, it could be concluded that betaine can improve the lactation performance of lactating goats and produce healthy milk with beneficial characteristics.

Globally, microplastics (MPs) contamination in aquatic organisms is emerging as an alarming phenomenon. In the present study, we investigated MPs in three commercially important fishes (Bombay duck Harpadon nehereus, ribbon fish Trichiurus lepturus and hairfin anchovy Setipinna phasa) in fresh and dried conditions collected from two sites (Chattogram and Kuakata) of the Bay of Bengal. It was evident that fresh T. lepturus ingested highest amount of MPs through the gills (6.41 mps/g) from Chattogram followed by in the gastrointestinal tract, GIT (6.20 mps/g) and in the muscle (1.20 mps/g) from Kuakata. Among the fresh fishes, H. nehereus from Kuakata accumulated highest amount of MPs (0.21 mps/g), while S. phasa from Kuakata contained the least amount of MPs (0.06 mps/g). On the other hand, among the dried fishes, T. lepturus from Kuakata contained highest amount of MPs (46.00 mps/g), while S. phasa from Kuakata retained lowest amount of MPs (2.17 mps/g). Strangely, all the dried fishes showed significantly higher amount of MPs compared to fresh fishes from both the locations. Fiber was the most dominant type of shape of MPs which accounted 66 %, followed by fragment (27.38 %), microbeads (3.59 %), film (1.48 %), foam (1.31 %) and pellet (0.25 %). Size-wise, the major portion (39.66 %) of MPs was present to be in size range less than 0.5 mm followed by 37.67 % in the size range of 0.5-1.0 mm group and rest 22.67 % within 1.0-5.0 mm. Red (41.55 %) colored MPs was the most prominent, followed by brown (22.11 %), blue (16.32 %), pink (11.69 %), purple (5.10 %), and green (2.25 %). Among polymer types, low-density polyethylene (LDPE) was the most common (38 %), followed by polystyrene (PS-22 %), polyvinyl chloride (PVC-16 %), polyamide (PA-13 %) and ethylene-vinyl acetate (EVA-9 %). The present study confirms high occurrence of MPs in the dried fishes over the fresh fishes from the Bay of Bengal, with high potential of trophic transfer to the human body.

Pathologically activated neutrophils (PMN) with immunosuppressive activity, which are termed myeloid-derived suppressor cells (PMN-MDSC), play a critical role in regulating tumor progression. These cells have been implicated in promoting tumor metastases by contributing to premetastatic niche formation. This effect was facilitated by enhanced spontaneous migration of PMN from bone marrow to the premetastatic niches during the early-stage of cancer development. The molecular mechanisms underpinning this phenomenon remained unclear. In this study, we found that syntaphilin (SNPH), a cytoskeletal protein previously known for anchoring mitochondria to the microtubule in neurons and tumor cells, could regulate migration of PMN. Expression of SNPH was decreased in PMN from tumor-bearing mice and patients with cancer as compared with PMN from tumor-free mice and healthy donors, respectively. In Snph-knockout (SNPH-KO) mice, spontaneous migration of PMN was increased and the mice showed increased metastasis. Mechanistically, in SNPH-KO mice, the speed and distance travelled by mitochondria in PMN was increased, rates of oxidative phosphorylation and glycolysis were elevated, and generation of adenosine was increased. Thus, our study reveals a molecular mechanism regulating increased migratory activity of PMN during cancer progression and suggests a novel therapeutic targeting opportunity.

Cancer immunotherapy often depends on recognition of peptide epitopes by cytotoxic T lymphocytes (CTLs). The tumor microenvironment (TME) is enriched for peroxynitrite (PNT), a potent oxidant produced by infiltrating myeloid cells and some tumor cells. We demonstrate that PNT alters the profile of MHC class I bound peptides presented on tumor cells. Only CTLs specific for PNT-resistant peptides have a strong antitumor effect in vivo, whereas CTLs specific for PNT-sensitive peptides are not effective. Therapeutic targeting of PNT in mice reduces resistance of tumor cells to CTLs. Melanoma patients with low PNT activity in their tumors demonstrate a better clinical response to immunotherapy than patients with high PNT activity. Our data suggest that intratumoral PNT activity should be considered for the design of neoantigen-based therapy and also may be an important immunotherapeutic target.

The objective of this study was to determine whether vision-occluded progressive resistance training would increase upper-extremity movement performance using the one-repetition maximum (1-RM) bench press. Participants (n = 57) were recruited from a historically black college and university (HBCU), cross-matched by sex, age (±1 year), 1-RM (±2.27 kg), 1-RM/weight (±0.1), and 1-RM/lean mass ratio (±0.1), and randomly assigned to either the experimental group (vision occluded) or the control group. Participants performed resistance training for 6 weeks prior to beginning the study, and 1-RM was assessed the week prior to the beginning of the study. Weight and body composition were measured using a BOD POD. Of the 57 participants who started the study, 34 completed the study (Experimental = 16, Control = 18) and were reassessed the week after completing the 6-week-long training protocol. Using a combination of Mann−Whitney U and Wilcoxon signed-rank tests, we found that when accounting for changes in lean muscle mass, individuals who trained with their vision occluded reported significantly greater improvements in 1-RM strength compared to those who did not (p < 0.05). The findings from our study suggest that vision-occluded progressive resistance training increases upper-extremity performance when assessed using the bench press. These findings may have significant practical implications in both sports and rehabilitation, as these techniques may be used to enhance performance in athletes and/or improve rehabilitation effectiveness.

Literature suggests that anxiety affects gait and balance among young adults. However, previous studies using machine learning (ML) have only used gait to identify individuals who report feeling anxious. Therefore, the purpose of this study was to identify individuals who report feeling anxious at that time using a combination of gait and quiet balance ML. Using a cross-sectional design, participants (n = 88) completed the Profile of Mood Survey-Short Form (POMS-SF) to measure current feelings of anxiety and were then asked to complete a modified Clinical Test for Sensory Interaction in Balance (mCTSIB) and a two-minute walk around a 6 m track while wearing nine APDM mobility sensors. Results from our study finds that Random Forest classifiers had the highest median accuracy rate (75%) and the five top features for identifying anxious individuals were all gait parameters (turn angles, variance in neck, lumbar rotation, lumbar movement in the sagittal plane, and arm movement). Post-hoc analyses suggest that individuals who reported feeling anxious also walked using gait patterns most similar to older individuals who are fearful of falling. Additionally, we find that individuals who are anxious also had less postural stability when they had visual input; however, these individuals had less movement during postural sway when visual input was removed.

We examined microplastics (MP) in two commercially important dried fish, Bombay duck (Harpadon nehereus) and ribbon fish (Trichiurus lepturus), collected from two sites on the Bay of Bengal (Cox's Bazar and Kuakata). The number of MP found in dried Bombay duck and ribbon fish from Kuakata was significantly higher (41.33 g-1 and 46.00 g-1, respectively) than the MP present in samples collected from Cox's Bazar (28.54 g-1 and 34.17 g-1, respectively). Fibers were the most common type of MP identified in all samples (41-64%), followed by fragments (22-34%), microbeads (9-16%), films (3-4%), foams (1-4%), and pellets (0-2%). ATR-FTIR analysis revealed three different types of MP polymer - polyethylene (35-45%), polystyrene (20-30%) and polyamide (30-45%) in the dried fish samples. The study confirms the presence of high MP loads in dried fish from the Bay of Bengal, with high potential of trophic transfer of MP to the human body.

BACKGROUND: Obesity is a growingly impacting human health concern. Laparoscopic sleeve gastrectomy (LSG) is an effective treatment for morbid obesity. However, the general anesthesia (GA) used in this major surgery has its documented drawbacks in obese patients with high risk. On the other hand, combined thoracic spinal-epidural anesthesia (CTSEA), a modern regional anesthesia procedure, has the advantages of both spinal and epidural anesthesia but without their shortcomings. This prospective study is a case experience that assesses the feasibility of CTSEA as an anesthesia option for laparoscopic sleeve gastrectomy (LSG). METHODS: A total of 100 patients were recruited for LSG as a management procedure for morbid obesity, which was performed under CTSEA. Perioperative events, functional parameters, and patients' satisfaction scores were recorded. RESULTS: Our prospective study showed successful use of CTSEA in 99% of the patients, except for one patient (1%) in whom CTSEA was converted into GA due to severe pain and anxiety. Few adverse events occurred and were managed accordingly. The satisfaction score revealed that 94% of the patients were satisfied. CONCLUSIONS: CTSEA was a successful anesthetic alternative procedure for LSG surgery.

BACKGROUND: Solar lentigines are skin lesions manifested by benign dark pigmentation causing a cosmetic problem in many patients. Several treatment modalities used for the management of solar lentigines. Side effects and rates of recurrence may be associated with them. OBJECTIVE: Treating solar lentigines with two different techniques of pulsed dye laser (PDL) and evaluation of the results both clinically and via the examination of ultrastructural changes by electron microscopy. PATIENTS AND METHODS: This study was conducted on 22 subjects with solar lentigines and having Fitzpatrick III-IV skin types, was managed by the use of PDL after enrolling them into two groups. Group I (one stacked PDL was used) and Group II (treated by stacked PDL in two sessions, 1 month apart). At baseline and 6 months after treatment, two punch biopsies with a diameter of 2 mm were taken from all patients. All taken biopsies were prepared for light and electron microscopic examinations. RESULTS: Both PDL techniques induced significant better clinical and histological outcomes. No one demonstrated any postoperative complications such as post-inflammatory hyperpigmentation and scarring. CONCLUSIONS: The two techniques of PDL are efficient for solar lentigines treatment.

Quality fish feed is the prime need for successful aquaculture. Feed qualities determine the fish flesh quality including appearance, color, odor, flavor, texture, nutritive value, and shelf-life. Nowadays, consumers are very much concerned about various issues regarding way of fish farming, types of feed ingredients used etc. The current study was conducted to assess the heavy metal contents and nutritional composition of some selected commercial fish feeds used in Bangladesh. The major heavy metal concentrations and proximate composition (moisture, crude protein, crude lipid, ash, crude fiber, and carbohydrate) of the collected feed samples were analyzed. The results showed that the feeds contained a number of heavy metals in varying proportions. The highest concentrations (mg/kg) of heavy metals such as lead (Pb), cadmium (Cd), chromium (Cr), copper (Cu), and zinc (Zn) analyzed in fish feed samples were 0.189, 0.027, 1.023, 0.303, and 1.468, respectively. There were significant differences between the nutritive values provided by feed companies and the values observed in this finding. The present study recommends that adequate measures are required to be taken by commercial fish feed manufacturers to ensure the nutritional quality of feed as well as to avoid the contamination of feed from heavy metals. Otherwise, fish and human, the ultimate consumer, may be predisposed to the assimilation and accumulation of the assessed heavy metals.

This study presents the analysis of airborne gamma-ray spectrometry and aeromagnetic data to delineate the distribution of the radioactive elements and determine their anomalous zones, subsurface structures controlled mineralized zones, and 3D magnetic inversion to give a full view of basement relief. According to statistical treatment of airborne gamma-ray spectrometry measurements, the highest values of potassium and equivalent thorium associated with Nakhiel Formation are 3.2% and 12.8 ppm respectively. Meanwhile, the equivalent uranium associated with phosphate beds attains 19.1 ppm. The spatial distribution of the relatively high concentrations of eU, eU/K and eU/eTh are recorded in adjacent areas in Upper Cretaceous deposits as > 12, 25 ppm and 5 ppm for eU, eU/K and eU/eTh, respectively. According to field measurements, the highest values of eU associated with phosphate beds and black Shale are 117 ppm and 20 ppm respectively. Meanwhile, the highest value of potassium associated with Quaternary sediments is 2.9%. Moreover, the highest value of eTh associated with Taref Formation is 16.2 ppm. The geochemical studies of collected samples from the anomalous zones showed the presence of secondary uranium in addition to rare earth elements. Meanwhile, the black shale trace elements are measured and showed high concentrations of tungsten. The magnetic data analysis is performed using tilt derivative and 3D magnetic modeling methods. Most of the deduced structures are trending in NW-SE, and NE-SW directions. The depths to the basement rocks are ranged from 0.475 km to 1.07 km below the ground surface.

PURPOSE: To study the impact of smell loss on quality of life in patients with Coronavirus Disease 2019 (COVID-19), and assess the importance of olfaction before and after the loss of smell. In addition, to assess the impact of smell loss on psychological well-being and distress, and to assess coping strategies used by COVID-19 patients with smell loss. METHODS: This study was a cross-sectional study. A total of 487 COVID-19 positive patients with anosmia were recruited. All participants filled in the validated Multi-Clinic Smell and Taste Questionnaire, and the validated General Well-being Schedule. RESULTS: Negative impacts of smell loss, associated risks, interference with daily activities, and deterioration in well-being were common. The importance of the sense of smell was evaluated (In relation to other senses) as higher in the period after the loss than before the loss (p < 0.001). All aspects of health-related QOL are statistically significant (p < 0.001), with the exception of financial security and friendship, which are not statistically significant (p = 0.129, p = 0.334), respectively. Psychological well-being was negatively affected, and the use of both problem- and emotion-focused strategies was common. CONCLUSION: COVID-19 Patients with loss of smell have significant reductions in health-related QOL. Their loss of smell directly affects daily activities related to the olfactory function. Therefore, priority should be given to diagnose and treat the loss of smell. Patients who have recently developed smell loss may be offered a combination of the problem- and emotion-focused strategies to cope with their condition.

BACKGROUND: Chronological age estimation is a challenging marker in the field of forensic medicine. The current study aimed to investigate the accuracy of signal joint T-cell receptor rearrangement excision circles (sjTRECs) quantification and telomere length measurement as methods for estimating chronological age. METHODS: Telomere length was estimated in the DNA derived from the buccal cells through estimating the telomeric restriction fragment (TRF) length using TeloTTAGGG Telomere Length Assay while the sjTRECs quantification was carried out on DNA isolated from the blood samples using qPCR. RESULTS: The TRF length was shortened with increased age (r = -0.722, p < 0.001). The sjTRECs were also decreased with increased age (r = -0.831, p < 0.001). Stronger coefficient and lower standard error of the estimate was obtained when multiple regression analysis for age prediction based on the values of both methods was applied (r = -0.876, p < 0.001).

BACKGROUND AND OBJECTIVE: The use of aromatic and herbal plants as a feed additive became a new trend in feeding dairy cows to enhance animal productivity. This study was conducted to evaluate the effect of adding some herbal and aromatic plants (Melissa, Fennel and Caraway) at 0.7% of total DM of rations. MATERIALS AND METHODS: Three herbal plants were evaluated in this study (melissa, fennel and caraway) by using thirteen Holstein Friesian cows. Cows were in 3rd and 5th lactation seasons were used as one experimental group by swing over design in lactation experiment (control then T1 then T2 then T3 then control). Nutrients digestibility, milk production, milk composition and blood constituents were determined. RESULTS: Data indicated that nutrients digestibility and nutritive value were improved with herbal plants addition, especially with caraway treatment which recorded significantly (p>0.05) the highest values. In the same trend, Actual milk yield, 4% FCM and fat content were significantly (p>0.05) increased when cows fed rations supplemented with fennel and caraway. The addition of melissa decreased the concentration of Total Saturated Fatty Acids (TSFA) in the milk. Normal ranges for blood parameters with no negative impact on animal health were observed with experimental rations. CONCLUSION: It could be concluded that caraway addition at 0.7% of the total dry matter of dairy rations, has a great potential to improve animal productivity and enhance the quality of milk fatty acids profile.
