RESUMEN

Attractivity and selectivity of two types of traps with synthetic, long-lasting, bisexual generic attractants were compared to conventional light traps to promote their wider use, as an easy-to-use standardised method for entomology. The targeted herbivorous Macroheterocera species playing important role in ecosystems as food source for higher trophic levels (e.g. predatory arthropods, birds and mammals), while other hand they can cause significant economic loss in agriculture. Data on their population dynamic and composition of their assemblages are necessary for both nature conservation and efficient pest management. Light- and semiochemical-baited traps with semisynthetic- (SBL = the acronym stands for semisynthetic bisexual lure) and synthetic lures (FLO = the acronym stands for floral lure of synthetic floral compounds) were used in species rich area of West Ukraine, and in all 10,926 lepidopterans trapped were identified. The attractivity of the light trap was highest with 252 species caught, while traps with semiochemicals captured 132 species including 28 exclusively caught only by them. The qualitative selectivity of light vs. semiochemical-baited traps differed considering both taxa and habitat preferences in such a way that they completed each-other. Differences in quantitative selectivity were also proved even in case of pest species. The parameters of methods varied depending on the phenological phases of the studied assemblages. Considering the revealed attractivity and selectivity, the parallel use of the two methods can offer improved reliable data for conservation biology and pest management.

During field tests implemented in Transcarpathia (West Ukraine) in 2015, 6501 specimens belonging to 26 Brachyceran fly families were collected with traps baited with generic lures (originally developed for noctuid moths) based on fermenting liquid and floral compounds. Isoamyl alcohol-based baits generally attracted more flies than phenylacetaldehyde-based baits and unbaited controls; however, the phenylacetaldehyde-based traps were the most attractive to the Empididae and Milichiidae families. The isoamyl alcohol-based semisynthetic lure showed significant attractivity to the families of Muscidae, Ulidiidae, Sarcophagidae, Calliphoridae, Sciomyzidae, Heleomyzidae, Drosophilidae, Phoridae and Platystomatidae. Additionally, isoamyl alcohol-based semisynthetic lure is the first reported attractant of the Sciomyzidae family. Since our phenylacetaldehyde-based floral lure was also attractive to Heleomyzidae flies, both types of lures can be seen as the first known attractants of this family.

The diurnal bees, lepidopterans, and other pollinators are among the most studied flower-visiting insect taxa. They mostly play distinct functions in temperate grasslands and ecotones of grassland-forest mosaics (such as in forest steppes). Although orthopterans are widely distributed in these habitats, however, their flower visitation is nearly unknown, especially in the temperate zone. During the development of traps with chemical lures to catch Lepidoptera pests, large numbers of Orthoptera were caught that provide a chance for studying the flower visitation and odor and indirectly the host plant preference of seven temperate zone Tettigoniidae species. Data on the attractivity of isoamyl alcohol-based semisynthetic lures for Meconema thalassinum and efficiency of phenylacetaldehyde-based lures on Leptophyes albovittata and Phaneroptera falcata were reported for the first time. Additionally, analysis of nature photos collected from internet sources, as part of a passive citizen science also supports the revealed preference of these species. Based on photos, the studied orthopterans mainly visit Asteraceae species including the most preferred Tanacetum vulgare, Pulicaria dysenterica, Achillea millefolium, Solidago canadensis, and Centaurea scabiosa. Based on catches of volatile traps, the first data were recorded on the attractivity of phenylacetaldehyde- and isoamyl alcohol-based lures on three temperate zone Orthoptera species. Results of a passive citizen science study strengthen these results that may increase the knowledge on the host plant and habitat preference of Orthoptera species.

Grape production worldwide is increasingly threatened by grapevine trunk diseases (GTDs). No grapevine cultivar is known to be entirely resistant to GTDs, but susceptibility varies greatly. To quantify these differences, four Hungarian grape germplasm collections containing 305 different cultivars were surveyed to determine the ratios of GTDs based on symptom expression and the proportion of plant loss within all GTD symptoms. The cultivars of monophyletic Vitis vinifera L. origin were amongst the most sensitive ones, and their sensitivity was significantly (p < 0.01) higher than that of the interspecific (hybrid) cultivars assessed, which are defined by the presence of Vitis species other than V. vinifera (e.g., V. labrusca L., V. rupestris Scheele, and V. amurensis Rupr.) in their pedigree. We conclude that the ancestral diversity of grapes confers a higher degree of resilience against GTDs.

OBJECTIVES: Recording and reproducing mandibular movements have been of key importance in the practice of dentistry for over a century. Recently, it has become possible to use digital technologies for these tasks. This study presents a preliminary method to try to identify the mandibular instantaneous centres of rotation based solely on intraoral scanners. METHODS: The dentitions of four participants were scanned, multiple inter-occlusal registrations and buccal scans were performed in closed and opened positions. Blender software was used to align the meshes during the post-scan digital workflow. Bite alignment accuracy was assessed and then improved with a strict exclusion protocol. An automated algorithm was used to find rotations between closed stage and open stage meshes. RESULTS: Our exclusion protocol reduced the bite alignment error significantly (p = 0.001) and the root-mean-square error value of the meshes decreased from 0.09 mm (SD = 0.15) to 0.03 mm (SD = 0.017). However, the remaining translational error caused an unexpectedly large shift in the axis of rotation (mean = 1.35 mm, SD = 0.77) with a 41.83: 1 ratio. As found in other studies, our results showed even a small amount of error during registration can shift the axis of rotation a large amount. This phenomenon will compromise the results of common pantographic methods which assume a rotation axis of the condyle. It also adds valuable information to the concept of instantaneous centers of rotation by revealing their true characteristics.

The northeastern part of the Carpathian Lowland is one of the most species-rich parts of the region, despite the long-term and sometimes extreme changes in land use. The fragmented remains of formerly contiguous forests maintain a significant part of the wildlife living in different oak, mixed oak, and gallery forest, and even faunas of connected grassland habitats. To reveal how the parallel Carpathian and Pannonian effects form the local insect fauna, the Noctuoidea assemblages of eight forest patches were assessed and compared, based on the distribution data of 479 species. Three types of assemblages were designated, with similar diversity but different compositions. The most unique and valuable ones can be found in the colline area of Transcarpathia, where the high habitat diversity supported by the topography promotes the coexistence of the species of humid forest fauna and many steppic and southern elements. The protection and planned management of these remained forest patches and their network in international cooperation can maintain a significant part of the fauna of the affected regions.

Levocetirizine dihydrochloride active ingredient was microencapsulated using nano spray-drying technology for preparing microparticles containing topical gel against edema. Hydroxyl propyl methyl cellulose (HPMC) was used as a carrier polymer during spray drying. The active ingredient content of the nano spray-dried products was 52.81% (w/w) and 51.33% (w/w) for ex vivo and in vivo experiments, respectively, and the average particle size was 2.6 µm. X-ray diffraction analysis indicated an amorphous state of the active ingredient embedded in the amorphous matrix of the polymer. Dermal oil gels composed of Miglyol 812 gelated by Dermofeel viscolid included 5% (w/w) (for ex vivo) and 10% (w/w) (for in vivo) active ingredient without or with 0.05% (w/w) menthol penetration enhancer. Qualitative ex vivo penetration studies using a confocal Raman microscopic correlation mapping were executed on human abdominal skin. The results showed that the active ingredient was enriched in the epidermis and upper dermis layer of the skin using oleogel loaded with the nano spray-dried drug-HPMC composite. Menthol addition to the oleogel resulted in the concentration of levocetirizine in the dermis. In vivo tests were performed on a mouse model of croton oil-induced ear edema. Negative control and Fenistil-treated groups were compared using the prepared oil gels with and without menthol. Without penetration enhancer, 20 µL of our oil gel loaded with nano spray-dried levocetirizine dihydrochloride composite showed similar effectiveness to the same volume of Fenistil gel, while 5 µL menthol containing sample was sufficient to eliminate the skin irritation similarly to 20 µL Fenistil.

The addition of synthetic eugenol and benzyl acetate to the known floral chemical and moth attractant phenylacetaldehyde synergized the attraction of Amata phegea (Lepidoptera: Amatidae). Traps baited with the ternary blend caught ca. four times more A. phegea moths than traps baited with phenylacetaldehyde alone. Both female and male moths were attracted; in a preliminary test, the female numbers caught were almost double compared to the males. Most A. phegea were caught when the blend was formulated in a dispenser with medium release rates. Traps baited with the ternary lure in polyethylene bag dispensers detected a single well-pronounced peak in seasonal trapping, suggesting that this multicomponent bisexual lure could be efficient enough to be applied to the detection and monitoring of female and male A. phegea.

In the first part of this two-piece publication, the isolation, identification and in vitro characterization of ten endophytic Trichoderma isolates were reported. Here we report the ability of two different mixes of some of these isolates (Trichoderma simmonsii, Trichoderma orientale and Trichoderma gamsii as well as of Trichoderma afroharzianum and T. simmonsii) to colonize and stimulate the growth of grapevines. Two commercial vineyards about 400 km away from the site of isolation were used as experimental fields, from which the strains of three Trichoderma species were re-isolated up to four years after rootstock soaking treatment with conidiospores, performed before planting. The treatments decreased the overall percentage of lost plants of about 30%, although a low number of lost plants (about 5%) were observed also in the control plot. For all cultivars and clones, the Trichoderma treatments significantly increased both the bud burst ratio and bud burst vigor index. In addition, the grape must parameters such as the Brix degrees, as well as the extract, the D-glucose and the D-fructose concentrations all appeared to be improved, suggesting a potentially higher ethanol content of the produced wine. We conclude that grapevine-endophytic Trichoderma isolates promote plant growth, which could be a useful feature for sustainable agriculture in general and integrated plant production in particular.

The caddisfly fauna of the Transcarpathian part of the Pannonian Lowland was poorly studied formerly. Here, we present the results of a six-year survey (2015-2020) carried out in four sampling sites of the Ukrainian part of the Bereg Plain and provide the actualised checklist of this area. Actually, 7346 specimens of 53 caddisfly species were collected. The number of known caddisfly species increased from 13 to 61. Two species Hydropsycheguttata and Parasetodesrespersellus, which formerly were considered extinct in the Pannonian Ecoregion, were detected and another especially rare species (e.g. Cyrnusflavidus) was also recorded. The fauna of the region cover a significant part of both Hungarian and Ukrainian caddisfly fauna. Assemblages of four characteristic habitat types of the region showed significant differences considering their quantitative and qualitative composition, substrate, current, hydrological- and feeding types. The high diversity and natural value of the small lowland watercourses were proven using a new Caddisfly Conservation Index (CCI) calculated, based on vulnerability and rarity of species. The fauna and assemblages showed a unique character mainly independent from large rivers of the region.

BACKGROUND: The fusion of computer tomography and deep learning is an effective way of achieving improved image quality and artifact reduction in reconstructed images. OBJECTIVE: In this paper, we present two novel neural network architectures for tomographic reconstruction with reduced effects of beam hardening and electrical noise. METHODS: In the case of the proposed novel architectures, the image reconstruction step is located inside the neural networks, which allows the network to be trained by taking the mathematical model of the projections into account. This strong connection enables us to enhance the projection data and the reconstructed image together. We tested the two proposed models against three other methods on two datasets. The datasets contain physically correct simulated data, and they show strong signs of beam hardening and electrical noise. We also performed a numerical evaluation of the neural networks on the reconstructed images according to three error measurements and provided a scoring system of the methods derived from the three measures. RESULTS: The results showed the superiority of the novel architecture called TomoNet2. TomoNet2 improved the quality of the images according to the average Structural Similarity Index from 0.9372 to 0.9977 and 0.9519 to 0.9886 on the two data sets, when compared to the FBP method. This network also yielded the best results for 79.2 and 53.0 percent for the two datasets according to Peak-Signal-to-Noise-Ratio compared to the other improvement techniques. CONCLUSIONS: Our experimental results showed that the reconstruction step used in skip connections in deep neural networks improves the quality of the reconstructions. We are confident that our proposed method can be effectively applied to other datasets for tomographic purposes.

This paper reports on the identification and in vitro characterization of several Trichoderma strains isolated from the Tokaj Wine Region in North-East Hungary. Ten isolates were analyzed and found to consist of six individual species-T. gamsii, T. orientale, T. simmonsii, T. afroharzianum, T. atrobrunneum and T. harzianum sensu stricto. The growth potential of the strains was assessed at a range of temperatures. We also report here on the in vitro biocontrol properties and fungicide tolerance of the most promising strains.

Lepidopteran assemblages were studied at 16 sampling sites in the Hajdúság Region between 2013 and 2020. Although studies targeted development of synthetic phenylacetaldehyde-based and semi-synthetic isoamyl alcohol-based baits for pest monitoring, traps caught 179 species belonging to the Sphingidae, Thyatiridae, Geometridae, Erebidae and Noctuidaefamilies. Most species were pests or widely distributed generalists, but there were also many rare habitat specialists, for example, silvicol species, whose appearance was unexpected in the recently less forested region. The specificity of the two bait types tested differed notably both on family and subfamily levels. Semi-synthetic baits performed better and attracted a wide range of noctuids belonging mainly to the Xyleninae and Noctuinae subfamilies, while synthetic phenylacetaldehyde-based lures showed specificity to Plusiinae subfamilies with lower number of sampled species. Our data fill a gap of knowledge since the fauna studied formerly was nearly unknown and brings attention to the alternative use of volatile traps of agricultural pests in faunistical studies.

The dried bean beetle, Acanthoscelides obtectus, is an economically important, worldwide pest of legume crops including dry beans, Phaseolus vulgaris. Assessment of A. obtectus infestation levels in pre-harvest field crops and post-harvest granaries is difficult to achieve because there is no effective monitoring tool for early detection so that interventions can be deployed as needed. Because A. obtectus is a generic pollen and nectar feeder, we adopted an electrophysiological (EAG) screening approach, using the antennae of female A. obtectus to identify physiologically active, volatile phytochemicals, which could then be investigated for their attractiveness to A. obtectus in laboratory behavioral assays and preliminary field tests. Of the 27 compounds tested in EAG screening, 5 compounds, i.e., methyl anthranilate, methyl eugenol, benzyl alcohol, (RS)-lavandulol, and 2-phenylethanol, elicited stronger EAG responses than the standard (1-phenylethanol). In 4-arm olfactometer bioassays, female A. obtectus preferred the olfactometer arm containing the odor of either methyl anthranilate or benzyl alcohol compared to the solvent control. In preliminary field tests using these 2 compounds as a binary mixture, at least 5 times as many beetles were caught on baited traps compared to non-baited traps. The field data also suggested that benzyl alcohol was primarily responsible for the field activity of the blend. We hypothesize that the attraction of A. obtectus to the combined benzyl alcohol/methyl anthranilate and the single benzyl alcohol baits is connected to the species` nectar- and pollen-feeding behaviour and not to its intraspecific communication. To our knowledge, this is the first evidence that A. obtectus behavior in the field can be modified by the deployment of plant-derived semiochemicals.

Immune homeostasis in peripheral tissues is, to a large degree, maintained by the differentiation and action of regulatory T cells (Treg) specific for tissue Ags. Using a novel mouse model, we have studied the differentiation of naive CD4+ T cells into Foxp3+ Treg in response to a cutaneous Ag (OVA). We found that expression of OVA resulted in fatal autoimmunity and in prevention of peripheral Treg generation. Inhibiting mTOR activity with rapamycin rescued the generation of Foxp3+ T cells. When we varied the level of Ag expression to modulate TCR signaling, we found that low Ag concentrations promoted the generation of Foxp3+ T cells, whereas high levels expanded effector T cells and caused severe autoimmunity. Our findings indicate that the expression level of tissue Ag is a key determinant of the balance between tissue-reactive effector and peripheral Foxp3+ T cells, which determines the choice between tolerance and autoimmunity.

Genetic activation of hedgehog/glioma-associated oncogene homolog (HH/GLI) signaling causes basal cell carcinoma (BCC), a very frequent nonmelanoma skin cancer. Small molecule targeting of the essential HH effector Smoothened (SMO) has proven an effective therapy of BCC, though the frequent development of drug resistance poses major challenges to anti-HH treatments. In light of recent breakthroughs in cancer immunotherapy, we analyzed the possible immunosuppressive mechanisms in HH/GLI-induced BCC in detail. Using a genetic mouse model of BCC, we identified profound differences in the infiltration of BCC lesions with cells of the adaptive and innate immune system. Epidermal activation of Hh/Gli signaling led to an accumulation of immunosuppressive regulatory T cells, and to an increased expression of immune checkpoint molecules including programmed death (PD)-1/PD-ligand 1. Anti-PD-1 monotherapy, however, did not reduce tumor growth, presumably due to the lack of immunogenic mutations in common BCC mouse models, as shown by whole-exome sequencing. BCC lesions also displayed a marked infiltration with neutrophils, the depletion of which unexpectedly promoted BCC growth. The study provides a comprehensive survey of and novel insights into the immune status of murine BCC and serves as a basis for the design of efficacious rational combination treatments. This study also underlines the need for predictive immunogenic mouse models of BCC to evaluate the efficacy of immunotherapeutic strategies in vivo.

The box tree moth, Cydalima perspectalis, is an invasive pest in Europe causing damage on Buxus species. In this study, we aimed to develop a "bisexual" lure to attract both female and male moths. Based on a previous screening bioassay we tested methyl salicylate, phenylacetaldehyde and eugenol as potential attractants in different combinations. The trapping results showed that both binary and ternary blends attracted male and female moths. Catches with these blends were comparable to catches with the synthetic pheromone. Subsequently we carried out single sensillum recordings, which proved the peripheral detection of the above-mentioned compounds on male and female antennae. To identify synergistic flower volatiles, which can be also attractive and can increase the trap capture, we collected flower headspace volatiles from 12 different flowering plant species. Several components of the floral scents evoked good responses from antennae of both females and males in gas chromatography-electroantennographic detection. The most active components were tentatively identified by gas chromatography coupled mass spectrometry as benzaldehyde, cis-ß-ocimene, (±)-linalool and phenethyl alcohol. These selected compounds in combination did not increase significantly the trap capture compared to the methyl salicylate- phenyacetaldehyde blend. Based on these results we discovered the first attractive blend, which was able to attract both adult male and female C. perspectalis in field conditions. These results will yield a good basis for the optimization and development of a practically usable bisexual lure against this invasive pest.

Agriotes ustulatus is an economically important click beetle in Europe. A female-produced pheromone, (E,E)-farnesyl acetate, has been identified and is used for monitoring and detecting males. More recently, a floral lure targeting females with modest, but significant, activity has been described. Based on preliminary data, we hypothesized, that similar to the effects on the congeneric A. brevis, addition of the pheromone to the floral lure should improve female A. ustulatus catches. Also, as click beetles have been reported to respond to white light, we studied possible interactions between visual and chemical cues. In field trials, the addition of the synthetic pheromone to the floral lure resulted in a dramatic increase in the number of females trapped, whereas male catches remained unaffected and equal to those in traps baited with pheromone only. A white visual cue did not influence trap catches. Maximum catches of both sexes of A. ustulatus can be achieved using the pheromone and the floral lure inside the same trap. Furthermore, the compounds can be formulated in a single polyethylene bag dispenser, making handling of the trap easier. Due to a much larger proportion of females in the catch, this improved trap may be a promising tool for semiochemical-based, environmentally sound agricultural practice against this important pest.

RESUMEN

BACKGROUND: Phenylacetaldehyde is a flower volatile and attractant for among others the European corn borer Ostrinia nubilalis. The addition of 4-methoxyphenethyl alcohol has recently been reported to increase O. nubilalis catches four to five times, yielding a bisexual lure for the species. RESULTS: The bisexual lure significantly outperformed synthetic pheromone lures of O. nubilalis at 11 out of 13 experiments conducted in Bulgaria, Hungary, Italy, Slovenia and Turkey. CONCLUSION: The bisexual lure will provide growers with a new and efficient method for detection and monitoring of O. nubilalis. © 2017 Society of Chemical Industry.
