RESUMEN

The follow-up of glioma patients after therapeutic intervention remains a challenging topic, as therapy-related changes can emulate true progression in contrast-enhanced magnetic resonance imaging. 18F-fluoroethyl-tyrosine (18F-FET) is a radiopharmaceutical that accumulates in glioma cells due to an increased expression of L-amino acid transporters and, contrary to gadolinium, does not depend on blood-brain barrier disruption to reach tumoral cells. It has demonstrated a high diagnostic value in the differentiation of tumoral viability and pseudoprogression or any other therapy-related changes, especially when combining traditional visual analysis with modern radiomics. In this review, we aim to cover the potential role of 18F-FET positron emission tomography in everyday clinical practice when applied to the follow-up of patients after the first therapeutical intervention, early response evaluation, and the differential diagnosis between therapy-related changes and progression.

Differences in PCDD/Fs concentrations were registered among adult bank voles (Myodes glareolus) inhabiting forest in the vicinity of the Landfill Salariyevo (Moscow, Russia) and caught in early spring (over-winter survivors) and late summer. The levels of highly toxic congeners and WHO-TEQ05 in samples of wintering voles were much lower than those in samples got in summer. This difference was investigated analyzing PCDD/Fs in sexually matured animals and offspring obtained in vivarium. Practically, no significant differences of PCDD/Fs concentrations and general toxicity were revealed among sexes. The main impact to these seasonal differences in PCDD/Fs accumulation was assigned to the types of functional development and activity, diet variation as natural growth of all voles occurs on the same territory but in seasonally diverse conditions. So, the species functional ecology (lifestyle and seasonal features) should be considered in practical application of animal models from natural populations for local monitoring of PCDD/Fs body burden, and individual functional type of ontogenesis among cyclomorphic mammalian species, especially. Thus, the results obtained during monitoring should be carefully interpreted for a proper environmental management.

The physically based melt of the low elevation Eastern Siberian glaciers is poorly understood due to the lack of direct micrometeorological studies. We used an automatic meteorological station to record the meteorological and energy characteristics of the Sygyktinsky Glacier, south Eastern Siberia (56.8° N, 117.4° E, 2,560 m a.s.l.), during two ablation seasons and computed the surface energy balance (SEB) for 30-min intervals. The glacier ablation was both modeled and measured by stakes and a thermistor cable. The net radiation (Rnet) was the main contributor (71-75 W m-2, 89-95%) to the SEB (79 W m-2, 100%), followed by sensible (2-4 W m-2, 3-5%) and latent (2-3 W m-2, 2-4%) heat fluxes. The net shortwave radiation was the main positive component of Rnet, while the net longwave radiation was weak and either negative (- 15 W m-2 in 2019) or positive (4 W m-2 in 2020). The small proportion of turbulent fluxes in the SEB is explained by the low wind speed (1.2 m s-1). The glacier ablation was found to be more sensitive to changes in shortwave radiation and wind speed, suggesting the need to consider the atmospheric conditions of the ablation period (summer snowfalls, cloudiness, wind speed) when analyzing long-term trends in glacial changes.

Background In the past 3 decades, the arterial switch procedure has replaced the atrial switch procedure as treatment of choice for transposition of the great arteries. Although survival is superior after the arterial switch procedure, data on pregnancy outcomes are scarce and transposition of the great arteries after arterial switch is not yet included in the modified World Health Organization classification of maternal cardiovascular risk. Methods and Results The ROPAC (Registry of Pregnancy and Cardiac disease) is an international prospective registry of pregnant women with cardiac disease, part of the European Society of Cardiology EURObservational Research Programme. Pregnancy outcomes in all women after an arterial switch procedure for transposition of the great arteries are described. The primary end point was a major adverse cardiovascular event, defined as combined end point of maternal death, supraventricular or ventricular arrhythmias requiring treatment, heart failure, aortic dissection, endocarditis, ischemic coronary events, and thromboembolic events. Altogether, 41 pregnant women (mean age, 26.7±3.9 years) were included, and there was no maternal mortality. A major adverse cardiovascular event occurred in 2 women (4.9%): heart failure in one (2.4%) and ventricular tachycardia in another (2.4%). One woman experienced fetal loss, whereas no neonatal mortality was observed. Conclusions Women after an arterial switch procedure for transposition of the great arteries tolerate pregnancy well, with a favorable maternal and fetal outcome. During counseling, most women should be reassured that the risk of pregnancy is low. Classification as modified World Health Organization risk class II seems appropriate.

Polyethyleneimine, poly-L-lysine, chitosan and some others cationic polymers have been thoroughly studied as nucleic acid delivery systems in gene therapy. However, the drug release from these systems proceeds at a very low rate due to extremely high binding between a carrier and gene material. To reduce these interactions and to enhance drug release, we developed a set of amphiphilic polypeptides containing positively and negatively charged amino acids as well as a hydrophobic one. The copolymers obtained were characterized by size-exclusion chromatography, static light scattering, HPLC amino acid analysis and 1HNMR spectroscopy. All copolymers formed particles due to a self-assembly in aqueous media. Depending on polypeptide composition, the formation of particles with hydrodynamic diameters from 180 to 900 nm was observed. Stability of polymer particles, loading and release efficiency were carefully studied. Cellular uptake of the particles was efficient and their cytotoxicity was negligible. The application of polymer carriers, containing siRNA, to vascular endothelial growth factor (VEGF-A165) silencing of ARPE-19 cells was successful. The gene silencing was confirmed by suppression of both messenger RNA and protein expression.
