RESUMEN

This paper starts with a brief history of the birth of the field of radioecology during the Cold War with a focus on US activity. We review the establishment of the international system for radiation protection and the science underlying the guidelines. We then discuss the famous ICRP 60 statement that if "Man" is protected, so is everything else and show how this led to a focus in radioecology on pathways to "Man" rather than concern about impacts on environments or ecosystems. We then review the contributions of Radiation Research Society members and papers published in Radiation Research which contributed to the knowledge base about effects on non-human species. These fed into international databases and computer-based tools such as ERICA and ResRad Biota to guide regulators. We then examine the origins of the concern that ICRP 60 is not sufficient to protect ecosystems and discuss the establishment of ICRP Committee 5 and its recommendations to establish reference animals and plants. The review finishes with current concerns that reference animals and plants (RAPs) are not sufficient to protect ecosystems, given the complexity of interacting factors such as the climate emergency and discusses the efforts of ICRP, the International Union of Radioecologists and other bodies to capture the concepts of ecosystem services and ecosystem complexity modelling in radioecology.

RESUMEN

Norway's Centre of Excellence for Environmental Radioactivity (CERAD) research programme included studies on transfer of radionuclides in various ecosystems within the context of environmental risk assessment. This article provides highlights from 10 years of research within this topic and summarises lessons learnt from the process. The scope has been extensive, involving laboratory-based experiments, field studies and the implementation of transfer models quantifying radionuclide uptake directly from the surrounding environment and via food chains. Field studies have had a global span and have, inter alia, covered sites contaminated with radionuclides associated with particles, ranging from nanoparticles to fragments, due to nuclear accidents (e.g., Chornobyl and Fukushima accidents) along with sites having enhanced levels of naturally occurring radioactive materials (e.g., Fen Complex in Norway and Taboshar in Tajikistan). Focus has been put on speciation and kinetics in determining radionuclide behavior and fate as well as on the influence of environmental factors that are potentially critical for the transfer of radionuclides. In particular, seasonal factors have been shown to greatly affect the dynamics of 137Cs and 90Sr bioaccumulation and loss in freshwater fish. The work has led to the collation of organism-specific (i) parameters important for kinetic models, i.e., uptake and depuration rates, and (ii) steady-state concentration ratios, CRs, where the use of stable analogue CRs as proxies for radionuclides has been brought into question. Dynamic models have been developed and applied for radiocaesium transfer to reindeer, radionuclide transfer in Arctic marine systems, transfer to fish via water and feed and commonly used agricultural food-chain transfer models applied in the context of nuclear emergency preparedness. The CERAD programme should contribute substantially to the scientific community's understanding of radionuclide transfer in environmental systems.

The aim of this study was to assess the exposures received by firefighters engaged in extinguishing the large-scale wildfires in the most contaminated areas of the Ukrainian part of the Chornobyl Exclusion Zone in 2016 and 2020. The assessments are based on measurements of radionuclide airborne concentrations in the breathing zones of workers and at the aerosol sampling stations of the automated radiation monitoring system operated by SSE Ecocenter. During the wildfires, the radionuclide airborne concentrations increased by orders of magnitude compared to the background levels, reaching maximum values in the firefighting area of 1.20 ± 0.01 Bq m-3 for 90Sr, 0.18 ± 0.01 Bq m-3 for 137Cs, (1.8 ± 0.3) ∙10-4 Bq m-3 for 238Pu, (4.5 ± 0.7) ∙10-4 Bq m-3 for 239-240Pu, and (8.0 ± 1.3) ∙10-3 Bq m-3 for 241Pu. The internal effective doses to firefighters due to inhaled radionuclides did not exceed 2 µSv h-1 and were 3-5 times lower compared to the external dose of gamma radiation. Thus, the time of firefighting in the ChEZ will be limited by the external dose.

An anonymous web-based survey was developed to check different aspects (SHAMISEN SINGS project): stakeholder awareness and perceptions of available mobile applications (apps) for measuring ionising radiation doses and health/well-being indicators; whether they would be ready to use them in the post-accidental recovery; and what are their preferred methodologies to acquire information etc. The results show that participation of the citizens would be most beneficial during post-accident recovery, providing individual measurements of external ionizing dose and health/well-being parameters, with possible follow-up. Also, participants indicated different preferences for sources to gain knowledge on ionising radiation and for the functions that an ideal app should have. The level of awareness and readiness to use apps to measure ionising radiation dose depended on two main aspects: individual differences (age & gender) and whether people were from countries affected by the previous major accidents. We concluded that stakeholders could have benefits from the data management plan: (1) it potentiates resilience at individual and community level; (2) citizens' measurements contribute to environmental monitoring and public health screening; (3) linkages between different types of data (environmental exposure, individual behavioural diaries, and measurements of health indicators) allow to perform more rigorous epidemiological studies.

A combination of synchrotron-based elemental analysis and acute toxicity tests was used to investigate the biodistribution and adverse effects in Daphnia magna exposed to uranium nanoparticle (UNP, 3-5 nm) suspensions or to uranium reference (Uref) solutions. Speciation analysis revealed similar size distributions between exposures, and toxicity tests showed comparable acute effects (UNP LC50: 402 µg L-1 [336-484], Uref LC50: 268 µg L-1 [229-315]). However, the uranium body burden was 3- to 5-fold greater in UNP-exposed daphnids, and analysis of survival as a function of body burden revealed a ∼5-fold higher specific toxicity from the Uref exposure. High-resolution X-ray fluorescence elemental maps of intact, whole daphnids from sublethal, acute exposures of both treatments revealed high uranium accumulation onto the gills (epipodites) as well as within the hepatic ceca and the intestinal lumen. Uranium uptake into the hemolymph circulatory system was inferred from signals observed in organs such as the heart and the maxillary gland. The substantial uptake in the maxillary gland and the associated nephridium suggests that these organs play a role in uranium removal from the hemolymph and subsequent excretion. Uranium was also observed associated with the embryos and the remnants of the chorion, suggesting uptake in the offspring. The identification of target organs and tissues is of major importance to the understanding of uranium and UNP toxicity and exposure characterization that should ultimately contribute to reducing uncertainties in related environmental impact and risk assessments.

BACKGROUND: The SHAMISEN (Nuclear Emergency Situations - Improvement of Medical And Health Surveillance) European project was conducted in 2015-2017 to review the lessons learned from the experience of past nuclear accidents and develop recommendations for preparedness and health surveillance of populations affected by a nuclear accident. Using a toolkit approach, Tsuda et al. recently published a critical review of the article by Cléro et al. derived from the SHAMISEN project on thyroid cancer screening after nuclear accident. MAIN BODY: We address the main points of criticism of our publication on the SHAMISEN European project. CONCLUSION: We disagree with some of the arguments and criticisms mentioned by Tsuda et al. We continue to support the conclusions and recommendations of the SHAMISEN consortium, including the recommendation not to launch a mass thyroid cancer screening after a nuclear accident, but rather to make it available (with appropriate information counselling) to those who request it.

The role of insects in the transfer of radionuclides has received little attention, despite their key role in nutrient cycling within ecosystems. In this study, we investigated the trophic food chain transfer of radiocaesium (137Cs) from reindeer meat to the blowfly Calliphora vicina (Robineau-Desvoidy) (Diptera: Calliphoridae) and further from blowfly pupae to the parasitoid wasp Nasonia vitripennis (Walker) (Hymenoptera: Pteromalidae). Radiocaesium was transferred to blowfly larvae during their feeding stage, with the highest whole-organism to food source concentration ratios (CRwo-fs) being found in actively feeding third instar blowfly larvae, with CRwo-fs in the range of 0.68-0.90. CRwo-fs in blowfly larvae at later developmental stages (i.e., post-feeding stage, prepupal stage, pupal stage, and adult stage) were significantly lower, with CRwo-fs in adult flies in the range of 0.07-0.10. Modelling of the data indicated that >70 % of the radiocaesium present in third instar feeding stage larvae was removed exponentially with excreta prior to pupariation. Furthermore, on average 49 % of the radiocaesium activity concentration assimilated in the metamorphing blowfly pupa was retained in the pupal case after the adult blowfly had emerged. When blowfly pupae were parasitised by parasitoid wasps, a greater proportion of radiocaesium was retained in the parasitised pupal case, with only 2-3 % of the activity concentration in the meat being found in the wasps, providing further evidence to support that radiocaesium is biodiluted in insect food chains. Our results indicate that carcass decomposition driven by insects directly affects the fate of radiocaesium retained in vertebrate carcasses and we discuss these findings in connection to radiocaesium cycling and dispersion in terrestrial ecosystems.

BACKGROUND: Reproductive effects of ionizing radiation in organisms have been observed under laboratory and field conditions. Such assessments often rely on associations between exposure and effects, and thus lacking a detailed mechanistic understanding of causality between effects occurring at different levels of biological organization. The Adverse Outcome Pathway (AOP), a conceptual knowledge framework to capture, organize, evaluate and visualize the scientific knowledge of relevant toxicological effects, has the potential to evaluate the causal relationships between molecular, cellular, individual, and population effects. This paper presents the first development of a set of consensus AOPs for reproductive effects of ionizing radiation in wildlife. This work was performed by a group of experts formed during a workshop organized jointly by the Multidisciplinary European Low Dose Initiative (MELODI) and the European Radioecology Alliance (ALLIANCE) associations to present the AOP approach and tools. The work presents a series of taxon-specific case studies that were used to identify relevant empirical evidence, identify common AOP components and propose a set of consensus AOPs that could be organized into an AOP network with broader taxonomic applicability. CONCLUSION: Expert consultation led to the identification of key biological events and description of causal linkages between ionizing radiation, reproductive impairment and reduction in population fitness. The study characterized the knowledge domain of taxon-specific AOPs, identified knowledge gaps pertinent to reproductive-relevant AOP development and reflected on how AOPs could assist applications in radiation (radioecological) research, environmental health assessment, and radiological protection. Future advancement and consolidation of the AOPs is planned to include structured weight of evidence considerations, formalized review and critical assessment of the empirical evidence prior to formal submission and review by the OECD sponsored AOP development program.

A series of 131I tracer experiments have been conducted at two research stations in Norway, one coastal and one inland to study radioiodine transfer and dynamics in boreal, agricultural ecosystems. The hypothesis tested was that site specific and climatological factors, along with growth stage, would influence foliar uptake of 131I by grass and its subsequent loss. Results showed that the interception fraction varied widely, ranging from 0.007 to 0.83 over all experiments, and showing a strong positive correlation with biomass and stage of growth. The experimental results were compared to various models currently used to predict interception fractions and weathering loss. Results provided by interception models varied in the range of 0.5-2 times of the observed values. Regarding weathering loss, it was demonstrated that double exponential models provided a better fit with the experimental results than single exponential models. Normalising the data activity per unit area to remove bio-dilution effects, and assuming a constant single loss rate gave weathering half-times of 22.8 ± 38.3 and 10.2 ± 8.2 days for the inland and coastal site, respectively. Whilst stable iodine concentrations in grass and soil were significantly higher (by approximately a factor of 5 and 7 times for grass and soil respectively) at the coastal compared to the inland site, it was not possible to deconvolute the influence of this factor on the temporal behaviour of 131I. Nonetheless, stable iodine data allowed us to establish an upper bound on the soil to plant transfer of radioiodine via root uptake and to establish that the pathway was of minor importance in defining 131I activity concentrations in grass compared to direct contamination via interception. Climatological factors (precipitation, wind-speed and temperature) appeared to affect the dynamics of 131I in the system, however the decomposition of these collective influences into specific contributions from each factor remains unresolved and requires further study. The newly acquired data on the interception and weathering of radioiodine in boreal, agricultural ecosystems and the reparametrized models developed from this, substantially improve the toolbox available for Norwegian emergency preparedness in the event of a nuclear accident.

A combination of synchrotron radiation-based elemental imaging, in vivo redox status analysis, histology, and toxic responses was used to investigate the uptake, biodistribution, and adverse effects of Ce nanoparticles (CeO2 NP; 10 nm; 0.5-34.96 mg Ce L-1) or Ce(NO3)3 (2.3-26 mg Ce L-1) in Caenorhabditis elegans. Elemental mapping of the exposed nematodes revealed Ce uptake in the alimentary canal prior to depuration. Retention of CeO2 NPs was low compared to that of Ce(NO3)3 in depurated individuals. X-ray fluorescence (XRF) mapping showed that Ce translocation was confined to the pharyngeal valve and foregut. Ce(NO3)3 exposure significantly decreased growth, fertility, and reproduction, caused slightly reduced fecundity. XRF mapping and histological analysis revealed severe tissue deformities colocalized with retained Ce surrounding the pharyngeal valve. Both forms of Ce activated the sod-1 antioxidant defense, particularly in the pharynx, whereas no significant effects on the cellular redox balance were identified. The CeO2 NP-induced deformities did not appear to impair the pharyngeal function or feeding ability as growth effects were restricted to Ce(NO3)3 exposure. The results demonstrate the utility of integrated submicron-resolution SR-based XRF elemental mapping of tissue-specific distribution and adverse effect analysis to obtain robust toxicological evaluations of metal-containing contaminants.

RESUMEN

The objective of this paper is to present the results of discussions at a workshop held as part of the International Congress of Radiation Research (Environmental Health stream) in Manchester UK, 2019. The main objective of the workshop was to provide a platform for radioecologists to engage with radiobiologists to address major questions around developing an Ecosystem approach in radioecology and radiation protection of the environment. The aim was to establish a critical framework to guide research that would permit integration of a pan-ecosystem approach into radiation protection guidelines and regulation for the environment. The conclusions were that the interaction between radioecologists and radiobiologists is useful in particular in addressing field versus laboratory issues where there are issues and challenges in designing good field experiments and a need to cross validate field data against laboratory data and vice versa. Other main conclusions were that there is a need to appreciate wider issues in ecology to design good approaches for an ecosystems approach in radioecology and that with the capture of 'Big Data', novel tools such as machine learning can now be applied to help with the complex issues involved in developing an ecosystem approach.

Adverse health outcomes of ionizing radiation given chronically at low dose rates are highly debated, a controversy also relevant for other stressors. Increased knowledge is needed for a more comprehensive understanding of the damaging potential of ionizing radiation from all dose rates and doses. There is a lack of relevant low dose rate data that is partly ascribed to the rarity of exposure facilities allowing chronic low dose rate exposures. Using the FIGARO facility, we assessed early (one day post-radiation) and late (recovery time of 100-200 days) hepatic genome-wide transcriptional profiles in male mice of two strains (CBA/CaOlaHsd and C57BL/6NHsd) exposed chronically to a low dose rate (2.5 mGy/h; 1200h, LDR), a mid-dose rate (10 mGy/h; 300h, MDR) and acutely to a high dose rate (100 mGy/h; 30h, HDR) of gamma irradiation, given to an equivalent total dose of 3 Gy. Dose-rate and strain-specific transcriptional responses were identified. Differently modulated transcriptional responses across all dose rate exposure groups were evident by the representation of functional biological pathways. Evidence of changed epigenetic regulation (global DNA methylation) was not detected. A period of recovery markedly reduced the number of differentially expressed genes. Using enrichment analysis to identify the functional significance of the modulated genes, perturbed signaling pathways associated with both cancer and non-cancer effects were observed, such as lipid metabolism and inflammation. These pathways were seen after chronic low dose rate and were not restricted to the acute high dose rate exposure. The transcriptional response induced by chronic low dose rate ionizing radiation suggests contribution to conditions such as cardiovascular diseases. We contribute with novel genome wide transcriptional data highlighting dose-rate-specific radiation responses and emphasize the importance of considering both dose rate, duration of exposure, and variability in susceptibility when assessing risks from ionizing radiation.

Adaptation of the nematode Caenorhabditis elegans towards NM300K silver nanoparticles (Ag NPs) has previously been demonstrated. In the current study, the sensitivity to a range of secondary stressors (CeO2 NP, Ce3+, Cu2+, Cd2+, and Paraquat) following the multigenerational exposure to silver nanoparticles (Ag NPs NM300K) or AgNO3 was investigated. This revealed improved tolerance to the ROS inducer Paraquat with higher fecundity after pre-exposure to Ag NP, indicating an involvement of reactive oxygen species (ROS) metabolism in the adaptive response to NM300K. The potential contribution of the antioxidant defenses related to adaptive responses was investigated across six generations of exposure using the sod-1::GFP reporter (GA508), and the Grx1-roGFP2 (GRX) biosensor strains. Results showed an increase in sod-1 expression by the F3 generation, accompanied by a reduction of GSSG/GSH ratios, from both AgNO3 and Ag NP exposures. Continuous exposure to AgNO3 and Ag NP until the F6 generation resulted in a decreased sod-1 expression, with a concomitant increase in GSSG/GSH ratios. The results thus show that despite an initial enhancement, the continuous exposure to Ag caused a severe impairment of the antioxidant defense capacity in C. elegans.

Many radiation protection actions carry a multitude of direct and indirect consequences that can impact on the welfare of affected populations. Health surveillance raises ethical challenges linked to privacy and data protection, as well as questions about the net benefit of screening. The SHAMISEN project recognized these issues and developed specific recommendations to highlight ethical challenges. Following a brief overview of ethical issues related to accident management, this paper presents the SHAMISEN recommendations: R1 The fundamental ethical principle of doing more good than harm should be central to accident management; and R4 Ensure that health surveillance respects the autonomy and dignity of affected populations, and is sensitive to any inequity in the distribution of risks and impacts. While a holistic approach to accident management means that decisions will be complicated by different values, perceptions and uncertainties about outcomes, addressing ethical issues could help ensure that the assumptions and potential conflicts behind eventual decisions are as transparent as possible.

Gamma radiation produces DNA instability and impaired phenotype. Previously, we observed negative effects on phenotype, DNA methylation, and gene expression profiles, in offspring of zebrafish exposed to gamma radiation during gametogenesis. We hypothesize that previously observed effects are accompanied with changes in the expression profile of non-coding RNAs, inherited by next generations. Non-coding RNA expression profile was analysed in F1 offspring (5.5 h post-fertilization) by high-throughput sequencing 1 year after parental irradiation (8.7 mGy/h, 5.2 Gy total dose). Using our previous F1-γ genome-wide gene expression data (GSE98539), hundreds of mRNAs were predicted as targets of differentially expressed (DE) miRNAs, involved in pathways such as insulin receptor, NFkB and PTEN signalling, linking to apoptosis and cancer. snRNAs belonging to the five major spliceosomal snRNAs were down-regulated in the F1-γ group, Indicating transcriptional and post-transcriptional alterations. In addition, DEpiRNA clusters were associated to 9 transposable elements (TEs) (LTR, LINE, and TIR) (p = 0.0024), probable as a response to the activation of these TEs. Moreover, the expression of the lincRNAs malat-1, and several others was altered in the offspring F1, in concordance with previously observed phenotypical alterations. In conclusion, our results demonstrate diverse gamma radiation-induced alterations in the ncRNA profiles of F1 offspring observable 1 year after parental irradiation.

This paper explores how health concerns of populations living in contaminated areas following radiological accidents can be considered in developing health surveillance. The research was performed in the framework of the SHAMISEN project, and aimed at identifying the impacts on, and challenges associated with, living and social conditions of affected populations. These objectives were achieved through the analysis of specific Case Studies in different situations observed after the Chernobyl accident (Belarus and Norway) and the activities carried out after the Fukushima accident. It incorporates an analysis of testimonies of medical experts and local stakeholders from contaminated territories in Japan within two Case Studies as well as through a dedicated workshop jointly organised with Fukushima Medical University in Japan in March 2016. The analysis addresses the following topics:Thus, this paper outlines key lessons learned from each of these topics, by providing tangibles examples from the analysis of the various Case Studies.

This paper, the last in the Special Issue (SI) on the SHAMISEN project, presents an overview of the SHAMISEN Recommendations for Preparedness and Health Surveillance of Populations Affected by a Radiation Accident. The recommendations are based on the lessons learnt from previous nuclear accidents, and the engagement activities with different stakeholder groups, described in the other papers of this SI. The SHAMISEN project developed a total of 28 recommendations. They include general recommendations, applicable across all phases of an accident, and specific recommendations for each of the three main phases: preparedness, early and intermediate, and long-term recovery. The recommendations are subdivided by topic: health surveillance, epidemiological studies, dose reconstruction, evacuation, and training of and communication with health personnel and other actors involved in liaising with affected populations. Each recommendation is divided into 3 sections - why, how and who - thus providing background and concrete advice as to how each SHAMISEN recommendation should be implemented and by whom. It is notable that many recommendations are also applicable to other disaster types, including the current SARS-CoV-2 pandemic.

Exposure of the thyroid gland to ionizing radiation at a young age is the main recognized risk factor for differentiated thyroid cancer. After the Chernobyl and Fukushima nuclear accidents, thyroid cancer screening was implemented mainly for children, leading to case over-diagnosis as seen in South Korea after the implementation of opportunistic screening (where subjects are recruited at healthcare sites). The aim of cancer screening is to reduce morbidity and mortality, but screening can also cause negative effects on health (with unnecessary treatment if over-diagnosis) and on quality of life. This paper from the SHAMISEN special issue (Nuclear Emergency Situations - Improvement of Medical And Health Surveillance) presents the principles of cancer screening, the lessons learned from thyroid cancer screening, as well as the knowledge on thyroid cancer incidence after exposure to iodine-131. The SHAMISEN Consortium recommends to envisage systematic health screening after a nuclear accident, only when appropriately justified, i.e. ensuring that screening will do more good than harm. Based on the experience of the Fukushima screening, the consortium does not recommend mass or population-based thyroid cancer screening, as the negative psychological and physical effects are likely to outweigh any possible benefit in affected populations; thyroid health monitoring should however be made available to persons who request it (regardless of whether they are at increased risk or not), accompanied with appropriate information and support.
