Occurrence of an unpleasant interpersonal event in daily life may cause an individual to experience unpleasant emotions and recall memories regarding it. These emotions, manifesting in daily social interactions, are often complex and mixed. In the laboratory, autobiographical recall is frequently used to induce emotions; however, it often involves recalling memories associated with a specific discrete emotion (e.g., sadness). To examine the neural activity of emotions similar to real-life experiences, we examined neural activity while recalling memories of stressful interpersonal events in daily life, without specifying a discrete emotion. Of the 23 university students recruited, 21 were analyzed and asked to freely recall memories and answer a series of questions on a monitor concerning their recalled memories while their neural activity was measured with functional magnetic resonance imaging. Amygdala activity increased while receiving the instructions, followed by a decrease in activity. This indicates that the participants' arousal and vigilance initially increased in response to a novel stimulus, and then decreased by habituation. Disgust and anger, which frequently occur as negative interpersonal feelings, were most prominently produced with strong associations with each other. More importantly, activation of the right amygdala while responding to questions regarding the recalled memories was positively correlated with disgust or anger only when not controlling for anger or disgust, respectively. These results indicate that responding to questions facilitated the generation of a mixed emotional response compared to during free recall alone. Furthermore, disgust and anger as a mixed emotion can synergistically activate amygdala.

RESUMEN

This perspective article discusses the importance of evidence-based psychotherapy and highlights the usefulness of near-infrared spectroscopy (NIRS) in assessing the effects of psychotherapeutic interventions as a future direction of clinical psychology. NIRS is a safe and non-invasive neuroimaging technique that can be implemented in a clinical setting to measure brain activity via a simple procedure. This article discusses the possible benefits and challenges of applying NIRS for this purpose, and the available methodology based on previous studies that used NIRS to evaluate psychotherapeutic effects. Furthermore, this perspective article suggests alternative methodologies that may be useful, namely, the single- and multi-session evaluations using immediate pre- and post-intervention measurements. These methods can be used to evaluate state changes in brain activity, which can be derived from a single session of psychotherapeutic interventions. This article provides a conceptual schema important in actualizing NIRS application for evidence-base psychotherapy.

The emotions that people experience in day-to-day social situations are often mixed emotions. Although autobiographical recall is useful as an emotion induction procedure, it often involves recalling memories associated with a specific discrete emotion (e.g., sadness). However, real-life emotions occur freely and spontaneously, without such constraints. To understand real-life emotions, the present study examined characteristics of emotions that were elicited by recalling "stressful interpersonal events in daily life" without the targeted evocation of a specific discrete emotion. Assuming generation of mixed and complex emotions, emotional groups with relatively strong correlation of multiple emotions according to surprise, fear, anger, disgust, sadness, and happiness were expected. Seventy-two university students (35 males, mean age: 19.69 ± 1.91 years; 37 females, 20.03 ± 2.42) participated in the study. In the emotion induction procedure, participants freely recalled memories as per the instructions on a monitor, and then responded silently to a series of questions concerning any one recalled incident. Assessments of emotional states using emotion scales and another item indicated that validated emotional changes had occurred during the task. Inter-correlations between six emotions demonstrated an emotional group consisting of disgust and anger, which frequently occur as negative interpersonal feelings, and that of fear and sadness. This indicated generation of mixed and complex emotions as experienced in social life. Future studies concerning relationships between these emotions and other factors, including neurophysiological responses, may facilitate further understanding about relationships between mental and physiological processes occurring in daily life.

BACKGROUND/AIMS: Monensin, an Na ionophore, increases intracellular Na ([Na]i). Alteration of [Na]i influences ion transport through the sarcolemmal membrane. So far, the effects of monensin on ventricular myocytes have not been examined in detail. The main objective of this study was to elucidate the mechanism via which monensin-evoked increases in [Na]i affect the membrane potential and currents in ventricular myocytes of guinea pigs. METHODS: Membrane potentials and currents were measured using the whole-cell patch-clamp technique in single myocytes. The concentration of intracellular Ca ([Ca]i) was evaluated by measuring fluorescence intensity of Fluo-4. RESULTS: Monensin (10-5M) shortened the action potential duration (APD) and reduced the amplitude of the plateau phase. In addition, monensin decreased the sodium current (INa) and shifted the inactivation curve to the hyperpolarized direction. Moreover, it decreased the L-type calcium current (ICa). However, this effect was attenuated by increasing the buffering capacity of [Ca]i. The Na-Ca exchange current (INa-Ca) was activated particularly in the reverse mode. Na-K pump current (INa-K) was also activated. Notably, the inward rectifying K current (IK1) was not affected, and the change in the delayed outward K current (IK) was not evident. CONCLUSION: These results suggest that the monensin-induced shortened APD and reduced amplitude of the plateau phase are primarily due to the decrease in the ICa, the activation of the reverse mode of INa-Ca, and the increased INa-K, and second due to the decreased INa. The IK and the IK1 may not be associated with the abovementioned changes induced by monensin. The elevation of [Na]i can exert multiple influences on electrophysiological phenomena in cardiac myocytes.

BACKGROUND: Childhood obesity has become a global epidemic and the etiology of maladaptive ingestive behavior in children warrants further research. Mounting evidence suggests that the caudate is associated with body weight gain and obesity in adults. In adolescents, however, how caudate-related neural networks are associated with body weight gain is unclear because their central nervous systems are still developing. OBJECTIVES: The current longitudinal resting-state functional magnetic resonance imaging (rs-fMRI) study was conducted to investigate the hypothesis that caudate-related neural networks have a role in weight gain in adolescents. METHODS: The study included 20 healthy adolescents with a mean age of 17.5 ± 2.0 years and a mean body mass index of 20.6 ± 2.1 who underwent baseline rs-fMRI then follow-up rs-fMRI approximately 1 year later. Body mass index (BMI) was measured at both timepoints. Seed-based functional connectivity analysis was utilized to analyze caudate-related functional connectivity (FC) using the caudate as a seed. Associations between caudate-related FC and BMI at baseline were assessed, as were associations between change in BMI and caudate-related FC between baseline and follow-up. RESULTS: At baseline, greater caudate-lateral prefrontal cortex FC was correlated with lower BMI (family wise error-corrected p < 0.05). Compared to the baseline, increased FC in the caudate-lateral prefrontal cortex at follow up were negatively associated with increased BMI (p < 0.05). CONCLUSION: Given that the lateral prefrontal cortex and caudate are associated with inhibitory control, the caudate-lateral prefrontal cortex FC may have a preventive effect on weight gain in adolescents. The results of the current study suggest that developing inhibitory control would lead to the prevention of childhood obesity.

Background: Schizophrenia is a disabling illness. Social cognition and interaction training (SCIT) seeks to improve patients' social functioning by alleviating deficits in social cognition. SCIT has shown promise in improving social cognition in patients with schizophrenia, but has not yet been studied in Japan. Design: An assessor-masked, randomized, parallel-group clinical trial was conducted to compare the feasibility and efficacy of SCIT with treatment as usual (TAU). Setting: Participants were recruited from outpatient clinics at the National Center of Neurology and Psychiatry and four other hospitals in Japan. Participants: Seventy-two patients diagnosed with schizophrenia or schizoaffective disorder consented to participate in the trial. Procedure: Participants were randomly allocated to either a SCIT subgroup or a TAU subgroup. SCIT is a manual-based group intervention that is delivered in 20-24-h-long weekly sessions. Groups include two to three clinicians and four to eight patients. Hypotheses: We hypothesized that SCIT would be found to be feasible and that patients who were randomized to receive SCIT would exhibit improvements in social cognition. Results: Data from 32 participants in each subgroup were entered into analyses. The persistence rate in the SCIT subgroup was 88.9%, and the average attendance rate was 87.0%. Intrinsic motivation was significantly higher in the SCIT subgroup than the TAU group during the first half of the program. Mixed effects modeling of various outcome measures revealed no significant interaction between measurement timepoint and group in any measures, including social cognition, neurocognition, symptom severity, and social functioning. In the case of the social cognition measure, significant change was observed only in the SCIT subgroup; however, the interaction between timepoint and group failed to reach significance. In an exploratory subgroup analysis, a shorter duration of illness was found to be associated with significantly better improvement on the social cognition measure in the SCIT subgroup compared with the TAU subgroup. Conclusions: In terms of the primary objective, the relatively low dropout rate observed in the present study suggests that SCIT is feasible and well tolerated by patients with schizophrenia in Japan. This view is also supported by participants' relatively high attendance and intrinsic motivation.

The current study examined whether cerebral blood flow (CBF) changes in the anterior ventral medial prefrontal cortex (VMPFC) represent responses to emotional stimuli instead of cognitive control of emotion during distraction tasks, as suggested by our previous near-infrared spectroscopy (NIRS) study. The functional distinction of MPFC regions is occasionally difficult due to their multiple functions. Here, we addressed some remaining concerns from our previous distraction studies, and employed the distraction paradigm, in which negative or neutral images are presented as emotional stimuli; a rest period is included in one condition and working memory (distraction) tasks in the other. To extract the components of CBF changes associated with responses to negative emotional stimuli (defined as emotion-related CBF changes), we subtracted the CBF changes obtained in the neutral condition from those in the negative condition. We found that emotion-related CBF changes in the anterior left VMPFC were significantly greater in the rest condition than in the distraction task condition. These findings imply that the distraction task decreased unnecessary brain activities, resulting in decrease in the emotion-related CBF changes. They support our previous findings, indicating that CBF changes in anterior VMPFC regions, measured using NIRS, are specifically associated with responses to emotional stimuli.

When near-infrared spectroscopy (NIRS) is used to measure emotion-related cerebral blood flow (CBF) changes in the prefrontal cortex regions, the functional distinction of CBF changes is often difficult because NIRS is unable to measure neural activity in deeper brain regions that play major roles in emotional processing. The CBF changes could represent cognitive control of emotion and emotional responses to emotional materials. Supposing that emotion-related CBF changes in the prefrontal cortex regions during distraction are emotional responses, we examined whether oxygenated hemoglobin (oxyHb) decreases. Attention-demanding tasks cause blood flow decreases, and we thus compared the effects of visually paced tapping with different tempos, on distraction. The results showed that the oxyHb level induced by emotional stimulation decreased with fast-tempo tapping significantly more than slow-tempo tapping in ventral medial prefrontal cortex regions. Moreover, a Global-Local task following tapping showed significantly greater local-minus-global response time (RT) difference scores in the fast- and mid-tempo condition compared with those in the slow-tempo, suggesting an increased attentional focus, and decreased negative emotion. The overall findings indicate that oxyHb changes in a relatively long distraction task, as measured by NIRS, are associated with emotional responses, and oxyHb can be decreased by successfully performing attention-demanding distraction tasks.

This study investigated the neural processing underlying the cognitive control of emotions induced by the presentation of task-irrelevant emotional pictures before a working memory task. Previous studies have suggested that the cognitive control of emotion involves the prefrontal regions. Therefore, we measured the hemodynamic responses that occurred in the prefrontal region with a 16-channel near-infrared spectroscopy (NIRS) system. In our experiment, participants observed two negative or two neutral pictures in succession immediately before a 1-back or 3-back task. Pictures were selected from the International Affective Picture System (IAPS). We measured the changes in the concentration of oxygenated hemoglobin (oxyHb) during picture presentation and during the n-back task. The emotional valence of the picture affected the oxyHb changes in anterior parts of the medial prefrontal cortex (MPFC) (located in the left and right superior frontal gyrus) and left inferior frontal gyrus during the n-back task; the oxyHb changes during the task were significantly greater following negative rather than neutral stimulation. As indicated in a number of previous studies, and the time courses of the oxyHb changes in our study, activation in these locations is possibly led by cognitive control of emotion, though we cannot deny it may simply be emotional responses. There were no effects of emotion on oxyHb changes during picture presentation or on n-back task performance. Although further studies are necessary to confirm this interpretation, our findings suggest that NIRS can be used to investigate neural processing during emotional control.
