RESUMEN

AIMS: To determine the prevalence of Aeromonas species in freshwater fish farms, factors affecting their prevalence and virulence factors associated with each species. METHODS AND RESULTS: In a cross-sectional study from 128 farms in four districts of Uttar Pradesh, India, 11 species of Aeromonas were identified by gyrB sequencing including the first report of Aeromonas crassostreae from fish. Four species of Aeromonas were more prevalent (MP) in fish farms, A. veronii bv. sobria (50.0%) was the highest, followed by A. caviae (18.8%), A. veronii bv. veronii (11.7%) and A. dhakensis (7.0%). The less prevalent (LP) species were A. hydrophila, A. media, A. jandaei, A. allosaccharophila, A. salmonicida, A. crassostreae and A. taiwanensis. Spatial variation in the prevalence of Aeromonas species was observed. Dominance of biovar sobria ranged from 33.3 to 68.6%, notably lesser in farms with on-farm biosecurity measures. The prevalence of biovar veronii was significantly associated with pangas fish, rainy season and farms with on-farm biosecurity measures. The prevalence of LP species was significantly higher in mrigal fish and winter season. Multiple virulence factors (>6) were detected in 70.2% of the Aeromonas species. Significant association of ß-hemolysin, DNase, slime production, act, ahh1, aexT and lip was observed with different species of Aeromonas. Moreover, 75.8% of Aeromonas species possessed one or more enterotoxins genes (act/alt/ast). CONCLUSION: Significant association of spatio-temporal variables, host fish species and on-farm biosecurity measures were observed on the prevalence of some of the Aeromonas species in freshwater fish farms. Most of the Aeromonas species harboured virulence factors indicating their potential for pathogenicity. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first study that determined the prevalence and identified the factors that affect the prevalence of Aeromonas species in freshwater fish farms. This information will be useful in managing Aeromonas infection in fish and their risks to public health.

The inhibitory effects of ammonium chloride (NH4Cl) and chlorpromazine hydrochloride on betanodavirus were evaluated on Sahul Indian sea bass kidney (SISK) cell line. The cytotoxicity of different concentrations of NH4Cl (0.1 mM, 1 mM, 10 mM, 100 mM and 500 mM) and chlorpromazine hydrochloride (1 µM, 10 µM, 100 µM, 200 µM and 500 µM) were assessed in SISK cells using different cytotoxic assays. Among the selected concentrations, 0.1 mM, 1 mM and 10 mM of NH4Cl and chlorpromazine hydrochloride at the dose of 1 µM, 10 µM and 100 µM were found to be non-toxic to the SISK cell line and same were chosen for the trials against nodavirus. The presence of nodavirus in the infected cells was confirmed by cytopathic effect (CPE) and RT-PCR (Reverse transcriptase PCR). NH4Cl of 1 mM and 10 mM, and chlorpromazine hydrochloride of 10 µM and 100 µM could successfully inhibit betanodavirus infection in SISK cells, which was confirmed by indirect ELISA and real-time PCR analysis. The result further suggested that the chlorpromazine hydrochloride drug could be more effective in inhibiting the betanodavirus with much lower dose than NH4Cl which was more effective at a higher dose. The present study thus suggested that NH4Cl and chlorpromazine hydrochloride drugs could be successfully used for controlling the nodavirus infection in aquaculture.

Monogenea (gill parasite) is a major problem in aquaculture that reduces the growth of cultured fish and adversely affects the economy. The present study was performed to evaluate the impact of various degrees of Thaparocleidus sp. (dactylogyrids, monogenean) infestation on haematological and serum biochemical parameters of Pangasianodon hypophthalmus. A standard cohabitation study, following complete randomized design in triplicate, was conducted to obtain low, moderate and high degrees of infestation in P. hypophthalmus along with the control (uninfested) group. Blood and serum were studied for haematological (total erythrocyte count (TEC), haemoglobin (Hb), packed cell volume (PCV), total leucocyte count (TLC) and indices viz. mean cell volume (MCV), mean cell haemoglobin (MCH), mean cell haemoglobin concentration (MCHC) and erythrocyte osmolarity brittleness (EOB)) and serum biochemical parameters (serum glutamate oxaloacetate transaminase (SGOT), serum glutamate pyruvate transaminase (SGPT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), lactate, total bilirubin and creatinine. Significant (p < 0.05) increase in TEC, Hb, TLC, EOB, SGOT, SGPT, LDH, lactate, bilirubin, and creatinine were noticed in moderate to high monogenean-infested group in comparison to the control group. However, significant (p < 0.05) reduction in MCH, and MCV and no difference (P > 0.05) in PCV were noticed in high degree parasitized group in comparison to the control group. The results of altered haematological and serum biochemical parameters in various degrees of monogenean-infested groups signify the density dependent physiological responses and changes in cells of the blood. The data of serum enzymes in the present study would be valuable for assessing the health status of the host and facilitate as a potential biomarker in relation to various degrees of monogenean infestation.

Modulation of innate immune activity and oxidative stress response of Pangasianodon hypophthalmus through experimental infection with (Thaparocleidus sp.) dactylogyrid monogenean was studied. A standard cohabitation method was used to infect healthy experimental fish. After 14 days, dactylogyrid (gill monogenean) infested fish were sampled and categorised into three different infected groups namely (T1) low (<10 mean dactylogyrid per gill arch per fish), (T2) moderate (10-49 mean dactylogyrid per gill arch per fish) and (T3) high (>50 mean dactylogyrid per gill arch per fish) along with a control group T0 (un-infested fish). Serum and tissues (gills and liver) were collected from experimental fish and analyzed for markers of innate immune and oxidative stress, respectively. The results showed that respiratory burst activity, myeloperoxidase level, serum lysozyme, α-2 macroglobulin and total serum immunoglobulin level were significantly (p < 0.05) elevated in fish with different degrees of parasite infestation compared to the control (un-infested group). Similarly, cellular oxidative biomarkers superoxide dismutase, catalase, glutathione-S-transferase and Na+-K+-ATPase activities of gills and liver were significantly (p < 0.05) elevated in dactylogyrid infested fish in comparison to the control. However, significantly decreased level of albumin, albumin to globulin ratio, total serum antiprotease and ceruloplasmin were observed in fish infested with low degree of dactylogyrids, while no significant differences in these parameters were observed between moderately infested and the control groups. The results suggested that varying degree of gill monogenean dactylogyrid infestation not only modulated the innate immune response of P. hypophthalmus by lowering albumin, total serum antiprotease and ceruloplasmin and inducing respiratory burst activity, phagocytic activity, myeloperoxidase, lysozyme, α-2 macroglobulin and total immunoglobulins, but also the oxidative stress biomarkers. The baseline data obtained in the present study will be valuable in understanding the host-parasite relationship and the dynamics of innate, oxidative stress responses and susceptibility of P. hypophthalmus to different degrees of parasitosis.

Identification of elasmobranchs by conventional taxonomy is difficult due to similarities in morphological characters. Species-specific molecular markers are good choice for identifying species irrespective of it's life stage. Recently, mitochondrial cytochrome c oxidase subunit I (COI) gene got global recognition as a barcode gene to discriminate all animals up-to species level. In this study, mitochondrial COI partial gene was used to develop DNA barcodes for 18 species of elasmobranchs (10 species of sharks and 8 species of rays). The COI barcodes clearly distinguished all the species with high interspecific distance values than intraspecific values. The average interspecific and intraspecific distance values are 8.6% and 0.3% for sharks, respectively and 12.4% and 0.63% for rays, respectively using K2P method. The Neighbor-Joining tree showed distinct clusters shared by the species of same genera. The COI barcodes were also used to estimate allopatric divergences for selected species across broad geographical locations and found that Sphyrna lewini, Aetobatus narinari and Neotrygon kuhlii have cryptic diversity.

White spot syndrome virus (WSSV) is one of the major viral pathogens affecting shrimp aquaculture. Four proteins, WSSV199, WSSV 222, WSSV 249 and WSSV 403, from WSSV are predicted to encode a RING-H2 domain, which in presence of ubiquitin conjugating enzyme (E2) in shrimp can function as viral E3 ligase and modulate the host ubiquitin proteasome pathway. Modulation of host ubiquitin proteasome pathway by viral proteins is implicated in viral pathogenesis. In the present study, a time course expression profile analysis of WSSV Open Reading Frame (ORF) 199 and Penaeus monodon ubiquitin conjugating enzyme (PmUbc) was carried out at 0, 3, 6, 12, 24, 48 and 72 h post WSSV challenge by semi-quantitative RT-PCR as well as Real Time PCR. EF1α was used as reference control to normalize the expression levels. A significant increase in PmUbc expression at 24 h post infection (h.p.i) was observed followed by a decline till 72 h.p.i. Expression of WSSV199 was observed at 24 h.p.i in WSSV infected P. monodon. Since the up-regulation of PmUbc was observed at 24 h.p.i where WSSV199 expression was detected, it can be speculated that these proteins might interact with host ubiquitination pathway for viral pathogenesis. However, further studies need to be carried out to unfold the molecular mechanism of interaction between host and virus to devise efficient control strategies for this chaos in the shrimp culture industry.
